From the above, it was clarified that the hearing loss due to noise exposure was significantly suppressed in the diet group containing silicon particles as compared with the normal diet group. Further, in the group containing the silicon fine particles, the hearing ability after 1 week of noise exposure was significantly higher than that in the normal diet group, and it was revealed that the silicon fine particles were effective for the restoration of the hearing ability. In addition, no significant difference was observed between the two in normal hearing before noise exposure.
C.硫黄関連化合物に基づいた多変量解析
C−1 サンプル調製
C57BL/6Jマウス(雄、7週齢)を日本SLCより入手した。シリコン微粒子投与群には、上記シリコン微粒子含有食を、コントロール群には、通常の飼料(通常食)(オリエンタル酵母工業株式会社製、型番AIN93M)を各群5匹ずつ1週間与えた。各々のマウスについて、深麻酔下で大腸を摘出し、盲腸、結腸及び直腸の3つに分けた。腸管内包物を取り出した各部位の一部(約2cm)をまとめて、重量を計測した。計測後、粉末状ドライアイスにて急速凍結し、マウス1匹の大腸サンプルとする。1群5匹の総計10匹の凍結大腸サンプルは、サルファーインデックス分析(株式会社ユーグレナ)に用いた。後日同様にサンプル調製を行い、1群5匹の総計10匹の凍結大腸サンプルを調製し、同様にサルファーインデックス分析(株式会社ユーグレナ)に用いた。
C. Multivariate analysis C-1 sample preparation based on sulfur related compounds
C57BL/6J mice (male, 7 weeks old) were obtained from Japan SLC. The silicon microparticle-administered group was fed with the above-mentioned silicon microparticle-containing diet, and the control group was fed with a normal feed (normal diet) (manufactured by Oriental Yeast Co., Ltd., model number AIN93M), 5 animals per group for 1 week. For each mouse, the large intestine was removed under deep anesthesia and divided into three parts, the cecum, colon and rectum. Part of each site (about 2 cm) from which the intestinal tract inclusion was taken out was put together and weighed. After the measurement, the sample is rapidly frozen with powdered dry ice to obtain a large intestine sample of one mouse. A total of 10 frozen large intestine samples of 5 animals per group were used for the sulfur index analysis (Euglena Co., Ltd.). Samples were prepared in the same manner as the following day, and a total of 10 frozen large intestine samples of 5 animals per group were prepared and similarly used for sulfur index analysis (Euglena Co., Ltd.).
C−2 分析前処理
1回目のサンプル調製で得られた同群の凍結マウス大腸サンプル(5つ)を合わせ、内部標準化合物を含むメタノール抽出液を添加し(1 ml /g(臓器))、ペッスルですり潰した。その後、遠心分離を行い、上清100μlをサンプルとした。遠心後のサンプル上清100μlに対して硫黄化合物標識試薬等を添加し(計130μl)、懸濁した。遠心分離した上清(87μl)を遠心型エバポレーターで乾固した。水60μlに再懸濁後遠心分離した上清5μlをサルファーインデックス分析用サンプルとした。2回目のサンプル調製で得られたサンプルも同様に処理しサルファーインデックス分析用サンプルを得た。サルファーインデックス分析に用いたサンプルは、シリコン微粒子投与群は2サンプル(5匹からの混合サンプルが2つ)、コントロール群も2サンプル(5匹からの混合サンプルが2つ)である。

The samples used for the sulfur index analysis were 2 samples (2 mixed samples from 5 animals) in the silicon fine particle administration group and 2 samples (2 mixed samples from 5 animals) in the control group.
C−3 サルファーインデックス分析
調製したサンプルに含まれる硫黄化合物は、サルファーインデックスメソッドを用いてLC MSMS 8040(島津製作所製)で分析を行った。具体的には、表1及び表2の測定対象化合物種うち、内部標準化合物(No. 53; Camphorsulfonate)およびチオール基修飾剤(No. 40; Monobromobimane)を除く、全61種の硫黄関連化合物種で相対定量を実施した。相対定量には、得られたマスクロマトグラムのピーク面積(内標化合物で標準化)を用いた。大腸サンプルにおいて計35種の化合物が検出された。検出された硫黄関連化合物データに基づく多変量解析に基づく、サンプル間の類似度のマッピング解析(Rソフトveganパッケージを活用)を行った。

上記C-3で検出された35種類の硫黄関連化合物に基づき各サンプルについて多変量解析を行った結果、シリコン微粒子投与群とコントロール群は下記の10の化合物によって区別することができた。シリコン微粒子投与群及びコントロール群の下記10化合物による多変量解析結果(各2つのサンプルの解析結果の平均値)を図6に示す。
