Claims (25)
- ヒト絨毛性ゴナドトロピンカルボキシ末端ペプチド(CTP)修飾ヒト成長ホルモン(hGH)ポリペプチドを製造する方法であって、
(a)所定の数の細胞に、前記CTP修飾hGHをコードするコード部分を含む発現ベクターを安定してトランスフェクトするステップであって、
i.前記トランスフェクトされた細胞が、前記CTP修飾hGHを発現及び分泌する、ステップと、
(b)前記CTP修飾hGHを過剰発現する細胞クローンを取得するステップと、
(c)前記クローンを溶液中で所定の規模まで増殖させるステップと、
(d)前記クローンを含有する前記溶液を収集するステップと、
(e)前記クローンを含有する前記溶液を濾過して、浄化された収集溶液を取得するステップと、
(f)前記浄化された収集溶液を精製して、所望の濃度の前記CTP修飾hGHを有する精製されたタンパク質溶液を取得するステップと、を含み、
それによりCTP修飾hGHを製造し、前記製造されたCTP修飾hGHのアミノ酸配列が配列番号7に記載される、方法。 - 前記CTP修飾hGHタンパク質の純度は、少なくとも90%である、請求項1に記載の方法。
