2.クリーム継続使用前後のオキシトシン変化解析
(1)方法

20−30代の女性35名を対象に、朝・夜の1日2回、洗顔後の日常のスキンケア後に、全顔へのクリームP又はQの塗布(約0.5 g)を4週間継続して頂いた。

全唾液の遠心分離後の上清(1.5〜3.0mL)と等量の0.1%(v/v)トリフルオロ酢酸(TFA)水溶液を混和した。遠心分離(3,000rpm、30min)後の上清を、Sep−pak C18カラム(200mg、3cc、Waters)に供し、下記のように抽出を行った。C18カラムに1mLの100%アセトニトリル(ACN)、次いで10mLの0.1%TFA水溶液(v/v)を通し、その後で0.1%TFA水溶液(v/v)と混和した全唾液(3.0〜6.0mL)を通し、10mLの0.1%TFA水溶液(v/v)で洗浄した後、3mLの(95%ACN/5%(0.1%TFA水溶液))(v/v)で溶出させた。溶出した溶液のACNをN2ガスで揮発させ、残った水溶液を凍結乾燥に供し、凍結乾燥品をOxytocin ELISA kit(Enzo)中の250μlのAssay Bufferに溶解して、上記キットを用いて定量した。Bio−rad Protein assay(BIO−RAD)で、ウシ血清アルブミンで作成した検量線を基に、抽出・濃縮前の唾液中のタンパク濃度(mg/mL)を定量し、単位タンパク量当たりのオキシトシン量を算出し、クリーム使用前の量を100とした変化率を算出した。

クリームP又はQ継続使用前後の唾液中のオキシトシン量の変化を検討した結果、図1に示すようにクリームP継続使用により有意な上昇が認められた。一方で、クリームQでは有意な上昇は認められなかった。このことから、コクを呈するクリームPを継続使用することにより、生体内のオキシトシンが上昇することが明らかになった。
