Abstract
Description
フラビウイルス属のウイルスは、最も重要な節足動物媒介性のヒト病原体であり、予防的治療や治癒的治療が利用できない南米における現在のジカ爆発などのますます深刻な蔓延を引き起こしている。ジカVの現在のメディアへの影響に加えて、社会に最も高い報酬を課すフラビウイルス疾患は、アミノ酸配列が30〜35%異なる血清型DENV1〜4と呼ばれる4つの異なるウイルスによって引き起こされるデング熱である。世界の年間発生率は、3億9000万の症例であり、このうち、9600万人は、臨床的に明らかであり1、約25000人が死亡していると推定されている。グローバリゼーション、Aedes蚊ベクターの広がり、都市計画の不十分な計画、最近まで認可されたワクチンまたは抗デング治療薬がなかったことなど、いくつかの要因がパンデミックを引き起こしている2。ジカVはまた、Aedes蚊によっても広がり、フラビウイルスの中でそのエンベロープタンパク質は他のフラビウイルスよりもDENV(42〜46%発散、図1A)のアミノ酸配列に最も近い。
フラビウイルスは、カプシド(C)前駆体膜タンパク質(prM)およびエンベロープ(E)、ならびに7つの非構造タンパク質(NS1、NS2A、NS2B、NS3、NS4A、NS4B、およびNS5)の3つの構造タンパク質を有する直径50nmである、比較的単純なポジティブセンスの一本鎖RNAウイルスである(図1B)。EおよびprMは、ウイルスの糖タンパク質の殻を形成し、Eは宿主細胞の結合および細胞への侵入7に関与する。ウイルス粒子の組み立ておよび成熟は、DENVについて最も徹底的に研究されている。小胞体中の粒子の形態形成中、Eの180個のコピーは、1:1の様式で、prMの180個のコピーと会合して、「未成熟ビリオン」をそれらの特徴となるスパイク状の外観7〜9を与える、60三量体(ヘテロ六量体)スパイクを形成する(図2A)。トランスゴルジネットワーク内のprMは、それが分泌されるまで、ビリオンと会合したままの状態で膜アンカー型Mの切り株およびPRを生成し、宿主がコードされたフリンプロテアーゼによって切断される8、10、11。宿主細胞から分泌されると、prは、離れて、「成熟ビリオン」を残し、Eの180個のコピーを含む平滑な構造は、2、3、および5倍軸の周りに20面体対称を有する90ヘッドトゥーテール二量体に配置される(図2B)。
1種の血清型のデングによる感染は、その血清型との再感染への終生免疫の世代をもたらすが、他の血清型ではもたらさない18〜20。4種すべてのデング血清型がしばしば共循環するか、または周期的に互いに置き換わるので、複数の感染が流行国では典型的である。十分に管理された疫学研究は、大部分の重度のデング熱感染が、二次的または連続的なデング熱感染を経験している個体において生じることを示している21〜23。
過去数十年にわたるデング熱感染の指数関数的増加は、デングワクチンの探索を必要であるが、この目標を達成することは非常に困難であることを示している。成功したワクチンは、デング熱にさらされていないか、またはこれまでにデング熱感染を経験している個体において、4種すべてのデング血清型、好ましくは1または2用量で保護的で耐久性のある免疫応答を誘導する必要があるであろう。同時に、ワクチンは上記の増強または病原性免疫応答を誘発することを避ける必要があり得る。
「EDE特異的中和抗体の構造研究により、融合ループの露出した主鎖および2つの保存されたグリカン鎖(N67およびN153結合グリカン)を含む、認識決定基がE二量体界面で血清型不変部位に見出されることが明らかになったことを言及する(Rouvinski et al.,2015)。これらの2つのグリコシル化部位は、他のフラビウイルスにおいて高度に保存されていない。さらに、ZIKVは、N67結合グルシレーション部位を持たず、N154結合グリコシル化部位(DENVのN153結合グリコシル化部位と同等)は、単離されたZIKV株の一部には存在しない(表S2)。さらに、DENV EDE mAb結合にとって重要なbストランド、150ループ、ijループ、およびA鎖のいくつかの残基は、ZIKVおよび他のフラビウイルスにおいて保存されていない(図S2)。重要なことに、ZIKV sE構造は、EDE抗体の結合領域に独自に正電荷のパッチを示すので(図S1)、EDE特異的抗生物質はZIKV感染に対して効果的でない可能性がある。しかしながら、高度に保存された融合ループを標的とする他
の既知のフラビウイルスFLE特異的抗体は、2A1−G6の中和プロファイルによって確認されるように、ZIKVを中和することができる。」

−一方のsE単量体が、グリコシル化部位を導入する少なくとも1つの変異を有し、変異したアミノ酸残基が、X官能基を担持する改変糖でグリコシル化され、

両方の変異した残基が、具体的にはクリック化学によって、第1のsE単量体の糖のX官能基を他方のsE単量体の糖のY官能基と反応させることによって、改変糖類を介して一緒に結合している。
T200、K202、E203、L308、K310、Q323、W391、F392(DENV−2)、またはジカPF13のT315、K373、S70、S72、Q77、R99、G104、M68、R252、D83、Q253、またはフラビウイルス、例えばジカウイルスもしくはデングウイルスの等価残基のうちの1つ以上は、EDEへの化合物の結合に必要であると考えられる。それ故に、一実施形態では、EDEを含むエンベロープタンパク質または構造は、これらの残基のうちの1つ以上、またはこれらのすべてを含む。

DENV血漿はZIKVと交差反応する
血清型1〜4による二次DENV感染後6ヶ月に採取した個体からの血漿を、捕捉ELISAによってZIKVおよびDENVへの結合について試験した。すべての場合において、DENV免疫血漿は、DENVおよびZIKVの両方に結合した(図1a)。アフリカ(HD78788)またはフランスポリネシア(PF13)に由来するウイルス株への結合には、顕著な差はなかった(図1b)。
DENV感染の特徴の1つは、二次感染時の病気の重篤度の増加である。これの説明の1つは、前のDENV感染を対象にする既存の抗体がオプソニン作用するが、二次感染を中和しない抗体依存性増強である。オプソニン化ウイルスは、より高いウイルス複製を引き起こす単球およびマクロファージなどのFc受容体発現骨髄細胞による取り込みの標的となる。
DENV感染患者から単離された形質膜芽細胞から生成された、DENVエンベロープタンパク質に反応する145のヒトモノクローナル抗体のプールを以前に作成した34。これらの抗体の詳細なエピトープマッピングは3つの広い反応性を示した。十分に記載された融合ループエピトープ(FLE)に反応する約1/3の抗体は、1/3は明確にマッピングされなかったが、融合ループ抗体のように、それらはウエスタンブロットによりエンベロープタンパク質に反応した(これらはエンベロープ残基W101の変異に対して感受性ではなかったため、非FLEと称される)。最後に、約40の抗体群はウエスタンブロットによってエンベロープタンパク質に反応せず、無傷ウイルス粒子にのみ結合した。これらの抗体は、凍結電子顕微鏡法およびX線結晶学によって、基本のヘッドトゥーテール二量体を構成する2つのエンベロープタンパク質単量体の界面に形成された立体配座の四次エピトープに結合することが示されており、その90はDENV糖タンパク質殻に正20面体対称で配置されている34、38。この新しいエピトープをE二量体エピトープ(EDE)と称し、これは、E中のN結合型グリカンN153の除去に対して感受性に基づいてEDE1およびEDE2の2つの群に細分された(EDE2結合はN153の除去によって減少したが、EDE1は減少しなかった)。いくつかのEDE抗体は、4種すべてのDENV血清型に対して完全に交差反応性であり、ピコモル範囲の感染を中和することができた。
融合ループおよびEDE mAbは、EDEのフットプリントが融合ループ領域も網羅するため、重複するエピトープを有する。EDE抗体がポリクローナル抗DENV血漿により誘発されたADEを克服することができるかどうかを試験するために、抗DENV血漿の濃度を高めてインキュベートしたZIKVに抗DENV EDE1 mAbの滴定を加えた(図6)。融合ループ抗体は効果を示さなかったが、ZIKVの結合力が低い752−2B2を除くEDE1 mAbは、752−2C8および753(3)C10をそれぞれ0.091±0.007および0.034±0.006μg/mlの力価で50%阻害を生じるPF13感染のADEを強力に阻害することができた。EDE1 mAbよりもZIKVの結合力が低いEDE2 mAbは、このアッセイにおいてADEを阻害することができなかった。これらの研究は、EDE1抗体がZIKV ADEを強力に阻害することができ、存在する場合、十分なレベルでインビボで保護し得ることを実証する。
南アメリカにおけるZIKVウイルス感染の、関連するギランバレー症候群および小頭症を伴う最近の爆発は大きな懸念である15、16、17。ギランバレー症候群は、比較的まれな合併症であり、ZIKVに感染した個体の0.024%に影響を及ぼすと推定されるが、ZIKV流行の規模のために、これは依然として多数の症例につながる15。多くの研究は、依然として小頭症の正確な原因を理解するために実行する必要があるが、これが発達中の脳の子宮内感染によって引き起こされることがますます明らかになっている17、39、40、41、42。ジカは、動物モデルにおいて、胎盤に感染し、成長を妨げることを示し、胎盤を通過して脳に感染し得ることも示されている43、44、45。さらに、インビトロZIKVは、神経細胞培養に感染し、神経球の発達を破壊し得る46、47。母体の感染後の神経学的損傷の正確なリスクは依然として決定されていないが、初期の研究では、フランス領ポリネシアからの他の報告は約1%のリスクがあるが、これは最初の3カ月間で最大22%であり得ることを示唆している48、49。
試料
書面によるインフォームドコンセントと、タイのKhon KaenおよびSiriraj病院の倫理委員会と英国のKhon Kaen and Siriraj倫理委員会の承認を得て血液サンプルを収集した。DENV感染の血清型は、ウイルスゲノムのRT−PCR検出によって決定した。デング熱病の回復から6ヵ月後にサンプルを採取した。
ベロ細胞(AFRIMSからの贈呈品)、293T、およびU937細胞を、それぞれ、MEM、DMEM、およびRPMI−1640中、37℃で培養した。C6/36細胞(AFRIMSからの贈呈品)を、Leibovitz L−15中、28℃で増殖させた。すべての培地は、10%熱不活性化ウシ胎仔血清、100単位/mlのペニシリン、および100μg/mlのストレプトマイシンを含有した。すべての細胞株は、マイコプラズマ汚染がなかった。
すべてのウイルスは、C6/36細胞で増殖させた。ZIKV株HD78788(アフリカ系統)は、Anavaj Sakuntabhaiによって提供された。ZIKV株PF13/251013−18(PF13)は、2013年にフランスポリネシアでZIKV発生中に患者から単離された。DENV−1(ハワイ)、DENV−2(16681)、DENV−3(H87)、およびDENV−4(1−0093)は、AFRIMSからの贈呈品であった。上清を含むウイルスを、−80℃で保存する前に、4℃、2000rpmでの遠心分離によって清澄化した。ウイルス力価は、ベロ細胞のフォーカス形成アッセイによって決定した34。すべてのウイルスストックは、マイコプラズマ汚染がなかった。
免疫グロブリンG1の重鎖および軽鎖を含む一対のプラスミドをポリエチレンイミン法により293T細胞に同時トランスフェクションし、無タンパク質培地で培養した。抗体を含む培養上清を5日後に採取した。
MAXISORPイムノプレート(442404;NUNC)を、マウス抗Eタンパク質、4G2(ZIKVに交差反応する融合ループマウスAb)でコーティングした。プレートを、3%BSAで1時間ブロックし、続いてウイルス上清でインキュベートした。1時間後、10μg/mlの抗DENV humAbまたは連続希釈血漿を添加した。この反応は、ALPコンジュゲート抗ヒトIgG抗体(A9544;Sigma)およびPNPP基質によって可視化した。NaOHで反応を停止させ、吸光度を405nmで測定した。エンドポイント力価(EPT)は、擬似抗原で得られた平均OD値の2倍に相当する相反的な血漿希釈として定義された。
フォーカス減少中和アッセイ(FRNT)を用いて、抗体の中和能を判定した。ウイルスを37℃で1時間、抗体または血漿サンプルの段階希釈物とともにインキュベートした。次いで、混合物をベロ細胞に添加し、2日間(ZIKVの場合)または3日間(DENVの場合)インキュベートした。次いで、記載されるように、フォーカス形成アッセイを実施した34。簡潔に述べると、ベロ細胞を、抗E mAb 4G2で染色し、続いてペルオキシダーゼコンジュゲートヤギ抗マウスIg(P0047; Sigma)で染色した。病巣(感染細胞)は、ペルオキシダーゼ基質、DABを添加することによって可視化した。フォーカス減少率を計算し、50%のFRNTは、SPSSパッケージからのプロビットプログラムを用いて計算した。
連続希釈された抗体または血漿サンプルを、U937細胞に添加する前に、37℃で1時間、ウイルスとともにインキュベートした。インキュベーション2(ZIKVの場合)または3日後(DENVの場合)、上清を採取し、ウイルス力価をフォーカス形成アッセイによって測定した。倍数増加は、抗体の存在下/非存在下でのウイルス力価との比較によって計算した。

ジカウイルスは、ブラジルの新生児における小頭症およびフランスのポリネシアの成人におけるギランバレー症候群に関連していた最近の大発生まで、ヒトの重症疾患に関連していなかったフラビウイルス属の一員である。ジカウイルスはデングウイルスに関連しており、デング患者から単離され、立体配座エピトープを標的とする抗体のカテゴリーがジカウイルスを強力に中和することを本明細書に報告している。ジカウイルスのエンベロープタンパク質と複合体を形成するこれらの抗体の2つの結晶構造は、保存されたエピトープの詳細を明らかにし、これはウイルス成熟中のエンベロープタンパク質二量体と前駆体prMタンパク質との相互作用部位でもある。ジカおよびデングウイルスの免疫複合体の比較は、ジカおよびデングウイルスを同時に防御する強力な中和抗体を誘発できるユニバーサルワクチンの合理的でエピトープを焦点する設計の基礎をなす。
ウイルスRNAポリメラーゼNS5のアミノ酸配列を用いた主要なヒト病原性フラビウイルスの系統発生解析は、蚊媒介性脳炎ウイルス群とのZIKVのクラスタリングを示している(図1a)。興味深いことに、このクラスター化は、Eタンパク質のアミノ酸配列が考慮され、ZIKVがDENV群で分岐する場合には異なる。このクラスタリングが抗体との相互作用に反映されるかどうかを調べるために、昆虫細胞において産生される組換え可溶性ZIKV細胞外ドメイン(ZIKV sE)に対して不十分な中和、交差反応性FLE、および強力に中和するEDE MAbの親和性を測定するために、Octet装置を用いたバイオ層干渉法(BLI)を使用した(オンライン方法を参照のこと)。DENV sEとは対照的に、サイズ排除クロマトグラフィー(SEC)によってモニタリングされるように溶液中で本質的に単量体であり、EDE抗体断片30による結合の際に二量体に変換されたZIKV sEは、SECで二量体として挙動した(ED.図1b)。
結晶化条件、得られた結晶、および構造決定は、オンライン法の節に記載され、ED表1に要約されている。ZIKV sEとEDE1 C8およびEDE2 A11との複合体の結晶は、それぞれ、scFvおよびFab断片で得られた。構造の平均分解能は、非配位ZIKV sE二量体の構造に対して2.7Åおよび2.9Å(それぞれ)および3.1Åである。回折パターンは、3つの結晶において異方性であり、3つの直交方向の分解能限界は、ED表1に引用されている。非配位ZIKVの構造では、溶液中の単量体として挙動する、細菌で産生され、インビトロで再折りたたまれたZIKV sEの最近決定された構造に反して、150ループが順序付けられ31、グリカンがループの構造に役立ち、SECの二量体を観察したので、sE二量化も促進することを示す。
ZIKV SEとの複合体におけるEDE1の総埋没表面積(BSA)は、DENV−2 SE複合体における約1300Å2と比較して約900Å2である(ED表3)。エピトープ領域の保存を図3dに示し、図3eおよび3fは、ZIKVおよびDENV sE上のC8フットプリントを比較する。DENV−2 sE構造で順序付けされたN67位のグリカン(図3c)は、フットプリント面積の全体的な差の約3分の2を占める。N67グリカンは、重鎖のフレームワーク領域2(FRH2)と相互作用し、ZIKV sE中に存在しないことは、これらの接触が結合に必須ではないことを示す。維持される相互作用の主な集団は、ドメインIIのβストランドbを中心とし、CDR H2、H3、およびL3からの側鎖はすべて、水素結合ドナー(NH原子)およびbdcβシート端部の受容体(主鎖カルボニル)を認識する(図3bおよび3c)。さらに、融合ループの主鎖(いくつかのグリシン残基を含む)およびCys74とCys105との間のジスルフィド結合は、CDR L1およびL3の芳香族側鎖によって囲まれている(ED図1も参照のこと)。これらの2つのCDRからの残基はまた、融合ループの厳密に保存された側鎖(Arg 99)または近く(Gln 77)も認識する。
A11抗体は、DENV−2 sEで見られたものとは非常に異なる角度で結合し、sE二量体の湾曲の差異を説明する。bストランドに沿った接触は保存されるが、抗体はこのストランドに対して異なる角度を作る(図4b)。C8と比較して、bストランドはその末端(残基71および73)でのみ認識されるが、C8は残基68(またはDENVでは67)からすべて認識する。150ループ上のグリカンとの接触もまた結合にとって重要であるため、A11の観察された傾斜結合は、DENVにおける153グリカンと比較して154グリカンのシフトした位置に関連している可能性が高い。水素結合相互作用の詳細は、3.8Åのより限定された分解能のため、DENV−2 sEとの複合体ではあまり明確に定義されていない。しかし、グリカンとは一連の異なる接触があることは明らかである(図4cおよびED図6b)。DENV2 sE/A11複合体において、グリカンは長いCDR3ループ中のα−ヘリックスによって認識される。ZIKV sEの場合には、グリカン部位の前に挿入が存在し、CDRH3αヘリックスと同じ相互作用をすることができないように、約6〜7Åのシフトを生じる。重要なことに、ビリオン上のZIKVの構造または非結合グリコシル化ZIKV sEとの比較は、150ループが良好に規則化していること(ED図6a)、およびDENV2ウイルスの場合と同様に、EDE1抗体によって不規則に誘導されることを示す。ED図6cは、グリカン鎖が所定の位置に残留し得るC8との衝突を示す。
これらの結果は、ジカウイルスとデングウイルスとの間に強力な交差中和のこれまでに認識されていない連結を提供し、したがって、伝統的な血清複合体を越えて抗原性フラビウイルス集団を同定する四次エピトープの構造的詳細を同定する。この関係は、ポリクローナル血清21を用いて認識されなかった保存されたエピトープによる強力な交差中和に基づいて、スーパー血清群を定義する。したがって、この研究は、この群のすべてのウイルスを防御できるユニバーサルワクチンの合理的な設計の基礎を築いている。

ZIKV sEタンパク質の組換え産生。組換えジカウイルスsEタンパク質(菌株H/PF/2013、GenBank受入番号KJ776791)は、前述42、43のように、Drosophila発現系(Invitrogen)中でタンデムStrepタグを用いて産生された。ジカsE配列(アミノ酸1〜408)を含む化学的に合成されたDNA断片(GeneArt)を、輸出シグナル配列BIP、エンテロキナーゼ切断部位、およびstrepタグをコードする発現ベクターpT38944にクローニングした。Drosophila Schneider2細胞を、選択のためにブラストサイジンを用いて安定にトランスフェクトした。タンパク質発現は、CuSO4の添加によって誘導され、上清は誘導から7〜10日後に収穫した。抗原は、製造業者の指示に従って、ストレプトアクチンカラム(IBA)を用いた親和性クロマトグラフィーによって精製した。最終精製ゲル濾過工程は、50mMのTris pH8、500mMのNaClで平衡化されたSuperdex increase 200 10/300 GLカラムを使用した。

4種の血清型のデングウイルス(DENV)に対するヒト抗体を強力に交差中和することが、最近単離され、構造的に特徴付けられている。例えば、WO2016/012800、Rouvinski et al(2015) Nature 520, 109−113、Dejnirattisai et al(2015) Nature Immunol 16, 170−177を参照のこと。これらの抗体は、これまでに発見されたE−二量体エピトープ(EDE)と呼ばれる高度に保存されたエピトープに結合し、これはジカウイルス(ZikaV)においても保存されており、ZikaVを強力に中和することも示されている。成熟したDENV粒子は、強力な「呼吸」挙動を有する準安定なE二量体の集合体であり、中和性ではないが疾病を増強する多くの抗体の生成を促進することを意味する。不完全に中和させる抗体応答の生成を最小限に抑えながら、安定化E−タンパク質二量体の生成に基づいてフラビウイルス、例えばフラビウイルスのジカ−デング群に対する防御免疫応答を惹起することができる抗原を開発するための逆ワクチン学アプローチを記載する。
デングおよび/または他のフラビウイルスに対するサブユニットワクチンを設計するために、逆ワクチン学アプローチをとることができる。これは、例えば、WO 2016/012800、Rouvinski et al(2015) Nature 520, 109−113、Dejnirattisai et al(2015) Nature Immunol 16, 170−177に記載されているように、EDEのような構造エピトープ、ならびにFLEおよびprMなどの線状エピトープを標的とする組換え抗体のパネルに基づくbnAb EDEエピトープの生成および構造的特徴付けを利用することができる。この計画の一般的な目的は、E二量体の安定したバージョンを生成し、反復構造/モデリングに基づいた設計プロセスを使用して、抗EDE応答の生成を特異的に標的とする免疫原を開発し、二量体の非EDE関連領域がより少ない防御性であるが感染を増強する抗体の生成を減少させることである。免疫原性は、ヒト免疫グロブリントランスジェニックマウス(例えば、Lee et al(2014)Nature Biotechnology Vol 32(4),356−363に記載されているマウスまたはEP1360287もしくはEP2264163に記載されているようなマウス)において試験することができ、インビボでの中和は、例えば、DENV感染のマウスモデルにおいて試験することができる。
A.ジスルフィド安定化変異体:sE二量体の安定性を改善するために、ジスルフィド結合形成のために可能な対の変異をトリアージするために構造ベースのアプローチ50を使用した。DENVからの二量体の結晶構造の分析は、二量体界面を横断する4.5Å下でCβ−Cβ距離で互いに対向する残基のペアの数を明らかにした。したがって、本発明者らは、一対のシステイン(そのうちの2つは、sE二量体の分子の2倍軸を横断する互いに対向する残基であり、システインに対する単一の置換のみを必要とする)による置換の6つの位置を同定した。変異体のうちの3つは、既に成功した共有結合DENV sE二量体発現をもたらし、EDEを繰り返し、EDE−mAbのパネルに結合しており(図4)、予備実験では免疫マウスの単量体Eと比較してより高い中和抗体力価を誘導する(図5)。ジカV sE二量体は4つのDENVのE二量体よりも安定であるが、FLE(融合ループエピトープ)抗体は依然としてジカVに結合し、これまでのデング熱感染による抗体がジカ病を増強する可能性があることを示唆している。したがって、FLEが曝露されないように、ジカV sE二量体をさらに安定化させることが重要である。これより、ジカVタンパク質に同じシステイン変異を導入し、例えば、ジカV−sEおよびDENV−sEジスルフィド安定化変異体を用いて免疫試験を並行して行うことができる。

抗EDE1 mAbクローン753(3)C10(C10)を、AG129マウスモデルにおいて、Zika感染からの保護を付与する能力について試験した。AG129マウスをB&K(Hull,UK)から入手し、Imperial CollegeのCBS施設で飼育した。すべての動物実験は、英国本社の認可の下、Imperial Collegeの倫理委員会のガイドラインに従って、収容レベル3施設で実施された。ウイルスストックを上記のように産生し、マウスモデルでの使用の前にVero細胞で滴定した。インターフェロン(IFN)−α/βおよびIFN−γ受容体(AG129マウス、雌、8〜10週齢)の両方が欠損した雌129/Svマウスに、ジカウイルス(ブラジリアン株PE243)による感染の24時間前に、精製ヒト抗EDE−1クローンC10またはアイソタイプ対照2−8Cを、200または50μg/マウスのいずれかで、腹腔内(腹腔内;200μL)投与した。マウスを、1.2×102FFU/マウスのジカPE243で腹腔内感染させた。PBSのみを投与したマウスを、実験対照として使用した。毎日の体重測定およびウイルス誘発性疾患の発症によって、マウスをモニタリングした。感染後2日目および4日目に、血液試料を採取した。血漿試料を、Vero細胞単層上のフォーカス形成アッセイを使用して、ウイルス負荷について滴定した。マウスは、人道的エンドポイントとしてプロジェクト認可の下で指定された、20%に近い体重減少および/または重度の病気の前に安楽死させた。元の体重の百分率は、感染直前の0日目の体重に基づいて計算した。体重測定値は、各実験群において3匹のマウスの平均+/− SEMとして表される。この実験を1回実施し、モノクローナル抗体C10によって保護されたことを示した。
Claims (64)
- 1つ以上のフラビウイルスに対して、個体にワクチン接種する際に使用するためのフラビウイルスエンベロープ二量体エピトープ(EDE)であって、前記EDEが、安定化組換えフラビウイルス、任意にデングウイルスおよび/またはジカ、エンベロープ糖タンパク質E細胞外ドメイン(sE)二量体であり、前記二量体が、前記2つのsE単量体間で少なくとも1つのジスルフィド鎖間結合で共有結合的に安定化される、ならびに/または各単量体の二量体界面またはドメインI(DI)/ドメインIII(DIII)リンカーにおいて、少なくとも1つのsE単量体のアミノ酸配列中の少なくとも1つのアミノ酸残基を、少なくとも1つのかさ高い側鎖アミノ酸で置換することによって非共有結合的に安定化され、前記2つのsE単量体間で少なくとも1つのスルフヒドリル反応性架橋剤で共有結合的に安定化される、ならびに/または、任意に、前記sE配列を分離する、リンカー領域、任意に、グリシンセリンリッチリンカー領域を有する単一ポリペプチド鎖として形成されることによって共有結合的に安定化される、ならびに/または、改変糖類を介して前記2つのsE単量体を連結することによって共有結合的に安定化される、ならびに/または、前記二量体が、DENV−1、DENV−2、DENV−3、DENV−4、ジカ、もしくは他のフラビウイルスのうちのいずれか1つもしくは2つからの天然および/もしくは変異エンベロープポリペプチドのホモ二量体もしくはヘテロ二量体であり、前記1つ以上のフラビウイルスが、ジカウイルス、ジカウイルスおよびデングウイルス、ジカウイルスおよび他のフラビウイルス、デング以外のフラビウイルスから選択される、EDE。

- 前記抗体またはその断片が、すべての血清型のデングウイルスおよびジカウイルスを中和し、任意に、すべての血清型のデングウイルスおよびジカウイルスを80%または90%または98%または100%中和し、任意に、すべての血清型のデングウイルスおよびジカウイルスを100%に中和し、任意に、すべての血清型のデングウイルスを同じ濃度の抗体または断片で100%中和する、請求項45〜51のいずれか一項に記載の使用のための抗体またはその断片。
- 前記抗体またはその断片が、1つ以上の血清型のデングウイルスおよび/またはジカウイルスを0.5〜0.01μg/mlの濃度で80、90、98、または100%中和する、請求項45〜52のいずれか一項に記載の使用のための抗体またはその断片。
- 前記抗体またはその断片が、すべての血清型のデングウイルスおよびジカウイルスを、0.5〜0.01μg/mlの濃度で80、90、98、または100%中和する、請求項45〜53のいずれか一項に記載の抗体またはその断片。
- 前記断片が、Fv断片、Fab様断片(例えば、Fab断片、Fab’断片、もしくはF(ab)2断片);またはドメイン抗体であるか、または前記抗体が、モノクローナル抗体または組換え抗体である、請求項45〜54のいずれか一項に記載の使用のための抗体またはその断片。
- 前記抗体が、ポリクローナル抗体またはその抗原結合部分である、請求項45〜54のいずれか一項に記載の使用のための抗体またはその断片。
- 前記抗体またはその断片が、混合物または抗体、任意に、
a)モノクローナル抗体もしくはその抗原結合部分の混合物、または
b)ポリクローナル抗体もしくはその抗原結合部分の混合物、または
c)混合物、またはモノクローナルおよびポリクローナル抗体もしくはその抗原結合部分、を含む組成物の一部として使用するためのものである、請求項45〜56のいずれか一項に記載の使用のための抗体またはその断片。 - 1つ以上のフラビウイルスによる感染の予防および/または治療のための方法における使用のための、請求項45〜57のいずれかに記載の抗体またはその断片をコードする核酸であって、前記1つ以上のフラビウイルスが、ジカウイルス、ジカウイルスおよびデングウイルス、ジカウイルスおよび他のフラビウイルス、デング以外のフラビウイルスから選択される、核酸。
- 前記抗体もしくはその断片または核酸が、上述の1つ以上のフラビウイルスによる感染の予防および/または治療のための方法における使用のためのものであり、前記治療が、抗体依存性増強(ADE)を減少させるためである、請求項45〜57のいずれか一項に記載の使用のための抗体もしくはその断片、または請求項58の使用のための核酸。
- 対象における1つ以上のフラビウイルスによる感染の診断またはモニタリングする際に使用するための、請求項1〜59のいずれか一項に記載のEDEまたはその抗体もしくはその断片であって、前記1つ以上のフラビウイルスが、ジカウイルス、ジカウイルスおよびデングウイルス、ジカウイルスおよび他のフラビウイルス、デング以外のフラビウイルスから選択される、EDEまたはその抗体もしくはその断片。
- 1つ以上のフラビウイルスによる感染に対してワクチン接種された対象における1つ以上のフラビウイルスによる感染に対するワクチン接種プロトコルの成功をモニタリングするためのインビトロ方法であって、
a)前記対象からの適切な生物学的試料を、請求項1〜29のいずれか一項に記載のEDEとインビトロで接触させるステップと、
b)前記生物学的試料中の前記二量体を対象とする中和抗体の量を判定するステップと、
c)ステップ(b)において判定された量を、前記対象に対して以前に得られた前記二量体を対象とする抗体の量と比較するステップと、を含み、
前記二量体を対象とする中和抗体の量の有意な増加が、前記ワクチン接種プロトコルの成功のマーカーを構成し、
前記1つ以上のフラビウイルスが、ジカウイルス、ジカウイルスおよびデングウイルス、ジカウイルスおよび他のフラビウイルス、デング以外のフラビウイルスから選択される、インビトロ方法。 - 1つ以上のフラビウイルスによる感染を患っている患者を、請求項45〜57のいずれか一項に記載の抗体もしくはその断片、または請求項58に記載の核酸での治療、あるいはその用量の増加を必要とする可能性が高いとして特定するための方法であって、前記対象における抗EDE抗体および抗融合ループ抗体のレベルの決定を含み、前記EDEが、請求項1〜29のいずれかに記載のとおりであり、前記1つ以上のフラビウイルスが、ジカウイルス、ジカウイルスおよびデングウイルス、ジカウイルスおよび他のフラビウイルス、デング以外のフラビウイルスから選択される、方法。
- 前記対象が主に抗融合ループ抗体を有する場合、前記対象が前記抗体、断片、または核酸で治療、任意に高用量でそれを必要とする、請求項62に記載の方法。
- 前記対象が抗EDE抗体を有しないか、または特に低レベルの抗EDE抗体を有する場合、前記対象が、高用量の前記抗体、断片、または核酸を必要としていると見なされる、請求項62または63に記載の方法。
