Abstract
Description

b)加熱デバイスに含まれる実質的に非磁性の加熱アダプタの上側面上の対応する受け部分の中にマルチウェルプレートのウェルを挿入するステップであって、受け部分がマルチウェルプレートのウェルを受けるように成形される、ステップと;
c)ステップa)またはb)のうちの任意のステップの前、その最中、またはその後で、液体試料、および結合表面を有する磁性粒子の懸濁液を、複数のウェルの少なくとも一部分に個別にまたはまとめて加えるステップと;
d)加熱アダプタの上側面上で受け部分の間にある加熱要素を用いてウェル内の液体試料を所定の温度まで加熱して、生物学的標的物質が磁性粒子の表面に結合されるときの条件を利用して、液体試料および磁性粒子をインキュベートするステップと;
e)加熱アダプタの下側面内の対応する凹部の中に磁気分離プレートの複数の磁石を導入するステップであって、各磁石が、加熱アダプタを通る、対応するウェルの内側空間に対して磁場勾配を印加することを目的として上記ウェルの外側壁に十分に近接させられ、それにより磁性粒子のペレットを形成する、ステップと;
f)磁力により磁性粒子のペレットを保持しながら、ピペッタの複数のピペット先端部を用いてそれぞれのウェルから液体を抜き取るステップと
を含む。
g)ウェルに溶出緩衝液を加えて、その中で磁性粒子を再懸濁するステップと;
h)溶出緩衝液を用いて磁性粒子から生物学的標的物質を溶出するステップと
をさらに含む。

Claims (14)
- 液体試料(220)中の生物学的標的物質を処理するためのシステムであって:
− 開いている頂部および閉じている底部を備える複数のウェル(210)を有するマルチウェルプレート(200)であって、前記複数のウェル(210)の少なくとも一部が結合表面を有する磁性粒子(230)の懸濁液を有する、マルチウェルプレート(200)と;
− 所定の幾何学的構成の複数の磁石(320)を有する磁気分離プレート(310)を備える磁気分離デバイス(300)であって、前記マルチウェルプレート(200)を基準として前記磁気分離プレート(310)を移動させるためのアクチュエータ(330)をさらに備える、磁気分離デバイス(300)と;
− 上側面(105)、下側面(115)、および前記上側面(105)の方に向かって開いている複数の受け部分(110)、を有し、前記受け部分(110)が前記マルチウェルプレート(200)の前記ウェル(210)を受けるように成形される、実質的に非磁性の加熱アダプタ(120)と、前記加熱アダプタ(120)の前記上側面(105)上で前記受け部分(110)の間にある加熱要素(130)とを備える加熱デバイス(100)と、
を備えており、
前記加熱アダプタ(120)が、前記磁気分離プレート(310)の前記複数の磁石(320)の前記所定の幾何学的構成に対応する位置に、前記下側面(115)の方に向かって開いている凹部(125)をさらに備え、前記受け部分(110)および前記凹部(125)が、挿入された前記磁石(320)により前記対応する挿入されたウェル(210)に対して磁場勾配を印加するのを可能にするために十分に近接する、
システム。 - ピペッタ(500)、制御ユニット、およびデータ管理ユニットの群から選択される1つまたは複数の要素をさらに備える、請求項1に記載のシステム。
- 前記加熱デバイス(100)が温度センサ(140)をさらに備える、請求項1または2に記載のシステム。
- 前記加熱要素(130)が、導電性の抵抗性材料と、熱移動媒体を含む流体システムまたはマイクロ流体システムとからなる群から選択される1つまたは複数の要素を備える、請求項1から3のいずれか1項に記載のシステム。
- 前記加熱要素(130)が前記加熱アダプタ(120)の前記上側面上に印刷される、請求項1から4のいずれか1項に記載のシステム。
- 前記加熱要素(130)が前記加熱アダプタ(120)の前記上側面(105)上にスパッタされるかまたは真空蒸着される、請求項1から5のいずれか1項に記載のシステム。
- 前記マルチウェルプレート(200)が標準的なSBSのフォーマットに従う寸法を有する、請求項1から6のいずれか1項に記載のシステム。
- 前記磁気分離デバイス(300)が、前記加熱デバイス(100)および前記マルチウェルプレート(200)を受けるためのフレーム(360)をその上側面にさらに備える、請求項1から7のいずれか1項に記載のシステム。
- 前記加熱アダプタ(120)が、アルミニウムおよびその合金、銅、鉄鋼、銀、アルミナセラミック、ならびに炭化ケイ素からなる群から選択される1つまたは複数の材料で作られる、請求項1から8のいずれか1項に記載のシステム。
- 液体試料(220)中の生物学的標的物質を処理するための方法であって:
a)開いている頂部および閉じている底部を備える複数のウェル(210)を有するマルチウェルプレート(200)を提供するステップと;
b)加熱デバイス(100)に含まれる実質的に非磁性の加熱アダプタ(120)の上側面(105)上の対応する受け部分(110)の中に前記マルチウェルプレート(200)の前記ウェル(210)を挿入するステップであって、前記受け部分(110)が前記マルチウェルプレート(200)の前記ウェル(210)を受けるように成形される、ステップと;
c)前記ステップa)またはb)のうちのいずれかのステップの前、その最中、またはその後で、前記液体試料(220)、および結合表面を有する磁性粒子(230)の懸濁液を、前記複数のウェル(210)の少なくとも一部分に個別にまたはまとめて加えるステップと;
d)前記加熱アダプタ(120)の前記上側面(105)上で前記受け部分(110)の間にある加熱要素(130)を用いて前記ウェル(210)内の前記液体試料(220)を所定の温度まで加熱して、前記生物学的標的物質が前記磁性粒子(230)の前記表面に結合されるときの条件を利用して、前記液体試料(220)および前記磁性粒子(230)をインキュベートするステップと;
e)前記加熱アダプタ(120)の下側面(115)内の対応する凹部(125)の中に磁気分離プレート(300)の複数の磁石(320)を導入するステップであって、各磁石(410)が、前記加熱アダプタ(120)を通る、対応するウェル(210)の内側空間に対して磁場勾配を印加することを目的として前記ウェル(210)の外側壁に十分に近接させられ、それにより磁性粒子(230)のペレット(240)を形成する、ステップと;
f)磁力により磁性粒子(230)の前記ペレット(240)を保持しながら、ピペッタ(500)の複数のピペット先端部(510)を用いて前記それぞれのウェル(210)から前記液体を抜き取るステップと
を含む
方法。 - g)前記ウェル(210)に溶出緩衝液を加えて、前記ウェル(210)内で前記磁性粒子(230)を再懸濁するステップと;
h)前記溶出懸濁液を用いて前記磁性粒子(230)から前記生物学的標的物質を溶出するステップと
をさらに含む、請求項10に記載の方法。 - 前記磁性粒子(230)の前記結合表面からの前記生物物質の溶出が、前記加熱アダプタ(120)の前記上側面(105)上で前記受け部分(110)の間にある前記加熱要素(130)を用いて前記ウェル(210)内の前記溶出緩衝液を所定の温度まで加熱すること、および前記磁性粒子(230)の前記表面から前記生物学的標的物質が溶出されるときの条件を利用して前記溶出緩衝液および前記磁性粒子(230)をインキュベートすることを伴う、請求項11に記載の方法。
- 前記磁性粒子(230)の表面上で前記生物学的標的物質を保持しながら、前記磁性粒子(230)の表面から望ましくない成分を除去するために前記ウェルに洗浄緩衝液を加えて、磁力により磁性粒子(230)の前記ペレット(240)を保持しながら前記それぞれのウェル(210)から前記液体を抜き取るステップを、ステップf)の後にさらに含む、請求項10から12のいずれか1項に記載の方法。
- 前記磁性粒子(230)の表面に前記生物物質を結合させた状態で前記磁性粒子(230)を洗浄するステップが、前記加熱アダプタ(120)の前記上側面(105)上で前記受け部分(110)の間にある前記加熱要素(130)を用いて前記ウェル(210)内の前記懸濁液を所定の温度まで加熱して、前記磁性粒子(230)の前記表面に対して前記生物学的標的物質が結合された状態を維持するときの条件を利用して前記懸濁液をインキュベートするステップを伴う、請求項13に記載の方法。
