1.悪臭(イソ吉草酸)の定量と官能評価

(2) 96穴ウェルに残留したバイオフィルムに0.2%L-ロイシン(東京化成工業社製)添加培地を3時間接触させ、ジエチルエーテル(和光純薬工業社製)で培養液を抽出した。ジエチルエーテル部分を0.5mLバイアルに充填し、イソ吉草酸をGC/MSで定量した。また、イソ吉草酸の臭いを対象とした下記基準による官能評価により、悪臭強度を評価した。これらの結果を表1に示す。

2.バイオフィルム除去率
前記1(1)の後、96穴ウェルに残留したバイオフィルムをクリスタルバイオレット0.1%水溶液で染色した。イオン交換水を用いて各ウェルを2回ずつ洗浄後、99.5%エタノールを200μLウェルに添加して、染色したバイオフィルムを溶出し、570nmの吸光度を計測してバイオフィルムの残存量を算出した。
コントロール(比較例4)のバイオフィルム残存量を100%としたときの各実施例及び各比較例でのバイオフィルム残存量を差し引いた値をバイオフィルム除去率とした。また、このバイオフィルム除去率から、下記基準に従ってバイオフィルム除去能の判定を行った。
