蓋付V底試験管(旭硝子株式会社製)に、5mmol/L塩化マグネシウム含有50mmol/L Tris−HCl緩衝液(和光純薬工業株式会社製、pH7.5)0.2mLに、2.5mg/mLウシ血清アルブミン溶液(商品名:アルブミン ウシ血清由来、製品番号:A2153、シグマアルドリッチジャパン合同会社製)0.1mL、0.1mg/mLホスホジエステラーゼ溶液(商品名:Phosphodiesterase cyclic Nucleotide Activator−Deficient,From Bovine Heart,Crude、シグマアルドリッチジャパン合同会社製)0.1mL、及び酒粕抽出液、又は酒粕抽出液の酵母菌発酵物0.05mL添加し、混合溶液を37℃、5分間反応させた。反応後、前記混合溶液に0.5mg/mLサイクリックAMP(cAMP)溶液を0.05mL添加し、37℃、60分間反応させた後、3分間、沸騰水浴上で煮沸し、反応を停止させた。遠心分離機(装置名:KUBOTA7820、久保田商事株式会社製)にて、2,260×g、10分間、4℃で遠心分離を行い、上清中を下記の条件下においてHPLC分析を行った。さらに、上記と同様の方法で空試験を行った。
