細胞膜の透過性を検出するのに必要な最適なチャネル形状および検出周波数を決定するために、細胞/電解質等価回路を構築して、微細加工フローサイトメータでの単一細胞の電気インピーダンス応答をモデル化した。この回路モデルは、エレクトロポレーション中の細胞膜コンダクタンスの劇的な増加を説明するために前のモデルから適合された。このモデルは、図7Cに示すように、一対の電極間の緩衝液中に懸濁した細胞のインピーダンスの決定を可能にする。ここで、CDLは二重層容量である。RmおよびCmは、それぞれ細胞外媒体の抵抗および容量である。Cmemは細胞膜の容量であり、Riは細胞の内部抵抗である。エレクトロポレーションによる細胞膜透過処理の結果としての全体のインピーダンス変化を反映するため、可変RmemはCmemと平行して実施された。二層効果を含む膜透過処理細胞を説明するための結果として得られた全体的なインピーダンスの式を式1に示す。
電気透過処理された細胞膜の抵抗であるRporated_memは、以前に公表された数値モデルに基づき、従来の方法で近似され、細胞膜の0.1%のみが穿孔されていると推定される。
ここで、dは5nmの膜厚であり、σmは100μS/cmでの緩衝液伝導率である。
方法および材料
デバイスは、ガラス基板上の一対の平面電極と、ソフトリソグラフィによって製造されたポリジメチルシロキサン(PDMS)マイクロチャネルとからなる。デバイスの特徴を有するシリコンマスタモールドは、標準的なフォトリソグラフィ手順を用いて製造された。デバイスの主なチャネルは、長さ1mm、幅150μm、深さ10μmであり、250μm(長さ)×25μm(幅)×10μm(深さ)の寸法を有する狭窄部を組み込んでいる。簡潔には、PDMSポリマーと硬化剤との10:1の混合物を鋳型上に注ぎ、ネガ型レプリカを作製し、65℃で一晩硬化させた。インレットチャネル(直径0.5mm)およびアウトレットリザーバ(直径1.5mm)へのアクセスを形成するために、PDMSに穿孔した。チタン/プラチナ(Ti/Pt)平面電極は、金属の「リフトオフ」プロセスによって製造された。電極トレースをガラス基板上にリソグラフィでパターニングし、凹部を10:1の緩衝フッ化水素酸で約1分間、約2000Åの深さまでエッチングした。金属は、物理蒸着(KJL PVD75、Kurt J.Lesker Co.)によって堆積され、その後、電極トレースを残してアセトン中でフォトレジストを溶解した。得られたTi/Pt電極の幅は50μmであり、中心間距離は300μmであった。この距離は、SNR品質を損なうことなく、電気および光学分析のための十分な細胞通過時間を可能にした。パターニングされた電極を有するPDMSおよびガラス基板の表面を、100Wの電力で、酸素プラズマ下、700mTorr、250sccmのO2で60秒間処理した(PX−250、March Instruments)。活性化した基質を、立体顕微鏡(SZ61双眼ステレオズーム、オリンパス)を用いて整列させ、不可逆的に結合させた。銅線を、導電性エポキシを介して平面電極パッドに接合した。
自動細胞検出およびエレクトロポレーション
単一細胞の自動検出および各細胞への即時パルス印加が1.3細胞/秒のスループットで得られた。狭窄部長さを横切る細胞の移動により、チャネル内の一定の容積変位に起因した、安定したベースライン電流が得られる。250ミリ秒の推定細胞通過時間により、エレクトロポレーションおよびパルス後インピーダンス測定のための十分な時間窓が提供された。この代表的なプロットにおいて、垂直の赤線は、所定のエレクトロポレーションパルスの印加を示し、この場合、5.0ミリ秒で1.05kV/cmの電界である。パルスを投与した後、電流の急激な上昇が直ちに観察される。この電流の急上昇は、細胞膜コンダクタンスの増加に起因し、エレクトロポレーション誘発細胞膜透過処理の結果としての孔の形成の特徴である。細胞がチャネル狭窄部から離れると、電気信号は緩衝液のベースラインに戻る。このプロセスは、エレクトロポレーション領域を通過する各細胞について繰り返される。単一細胞検出のため、37dBの信号対雑音比(SNR)が測定された。システムは、光学的観察と比較して、各細胞の検出およびパルス化において97%の精度を維持した。エラーは、主に、通過中の各細胞に複数のパルスを印加することにつながる複数の細胞の偶発的なテールゲーティングに起因した。
エレクトロポレーションパルスの強度および持続時間を変化させることによって、我々は細胞膜透過処理の程度に特徴的な細胞インピーダンスの変化を示す。図10に示すように、5つの代表的な細胞につき測定された細胞電流応答が、パルス印加時に重畳される。パルス持続時間を0〜5.0ミリ秒に変化させながら電界を1.05kV/cmで一定に保つことにより、より長いパルス持続時間は、電極間の電流のより大きな急上昇を生じさせ、より高い膜透過処理の程度を示すようである。電界強度と持続時間の両方の関数としての細胞膜応答の完全な特徴付けを図11に表す。サイズ差による細胞間の差異を説明するために、細胞ベースライン(ΔIP)からの透過処理電流の変化を、最初に全細胞電流変位(ΔIC)によって正規化し、検出された細胞−電流ベースラインからの増加の百分率として表した。パルス持続時間の関数としてプロットすると、所与の電界強度につき、正規化された透過処理電流(ΔIP/ΔIC)とパルス持続時間との間に強い依存性が見られ、より長いパルス持続時間はより大きな透過処理をもたらした。同じパルス持続時間で異なる強度が印加される場合にはまた、電界強度の大きさに強い依存性が観察され、より強い電界がより大きな透過処理を導いた。0.58〜1.05kV/cmの範囲の電界では、パルス持続時間が1.0ミリ秒に達し、これを超えたときに、透過信号の急速な遷移が観察された。システムは電流の連続フローおよび動的測定を可能にするので、少なくとも200個の細胞が各パルス条件について測定され、総計で個々の細胞5,000個の分析に達した。
電気的に観察される細胞膜透過処理はまた、PIの細胞間核酸への結合の際に生じる蛍光強度の変化を記録することによって光学的にも確認された(図12)。光学カメラをロックインアンプセンサと同期させて、各パルス印加後にパルス化された細胞の連続画像を捕捉した。次に、これらの画像を、個々の細胞に基づいて、蛍光強度について評価した。より大きいパルス強度および持続時間は、より大きな細胞膜透過処理をもたらし、これは、穿孔された細胞膜を通って侵入するより多くのPIが、細胞質空間中で核酸と結合し、蛍光強度を上昇させることを可能にする。
所定のエレクトロポレーション処理を受ける単一細胞の生存率はまた、7AAD染色によるエレクトロポレーション−パルスパラメータと相関していた。生存細胞が膜再封のための時間を有するよう、細胞は、エレクトロポレーションパルスに曝されてから20分後に回収した。我々は、最高電界強度(1.05kV/cm)とパルス持続時間(5ミリ秒)の組み合わせが、死細胞からの分布への最も大きなシフトを引き起こすことを見出した。このことは、強力なエレクトロポレーション処理が細胞膜を不可逆的に損傷し、再封を妨げる可能性がより高いため、予想される。パルス強度または持続時間を減少させると、シフトの大きさが減少する。0.7kV/cmでエレクトロポレーション処理を受けたすべての細胞は、対照生存細胞の蛍光と同等の蛍光を保持し、透過後20分以内に細胞膜の完全な再封を示した。生存閾値は、図13D〜図13Fの垂直線によって示されるように、死細胞集団の平均蛍光強度の95%信頼下限を計算することによって、各細胞集団について決定された。図14は、各エレクトロポレーション条件での細胞の生存率を表す。全体の細胞生存率は、電界強度およびパルス持続時間の両方に応じて単調に減少する。より強いパルス条件は、細胞死をもたらす不可逆的な細胞膜損傷を引き起こす可能性がより高い一方で、中程度の条件(0.7kV/cm)で処理された細胞は回復の可能性があり、より高い集団生存率を示した。
Claims (34)
- 生体細胞をエレクトロポレーションするためのシステムにおいて、
緩衝液中の複数の生体細胞のフローを受容するように適合され、検出領域を含むマイクロ流体チャネル;
前記検出領域を横切る電界を印加するように適合された一対の電極;
前記電極を介して細胞検出信号および透過信号を生成することができる信号生成部;
前記検出領域のインピーダンスを検出するように適合された検出部;および
前記検出部によって検出されたインピーダンスに従って前記信号生成部を制御するように適合された制御部
を含むことを特徴とする、システム。 - 請求項1に記載のシステムにおいて、前記信号生成部が、前記細胞検出信号および前記透過信号を同時に生成することができることを特徴とする、システム。
- 請求項1に記載のシステムにおいて、前記検出部が、ロックインアンプを含むことを特徴とする、システム。
- 請求項3に記載のシステムにおいて、前記検出部が、前記検出領域内の細胞の蛍光を測定することができる撮像デバイスをさらに含むことを特徴とする、システム。
- 請求項1に記載のシステムにおいて、前記マイクロ流体チャネルが、前記検出領域を介して複数の生物細胞のフローを流体力学的にセンタリングすることができることを特徴とする、システム。
- 請求項1に記載のシステムにおいて、前記システムが、緩衝液のフローを受容するように適合された第2のマイクロ流体チャネルをさらに含み、前記第2のマイクロ流体チャネルが第2の検出領域を含むことを特徴とする、システム。
- 請求項6に記載のシステムにおいて、前記検出部が、前記第2の検出領域のインピーダンスを検出するように適合されることを特徴とする、システム。
- 請求項1に記載のシステムにおいて、前記信号生成部が、送達信号を生成することができることを特徴とする、システム。
- 請求項1に記載のシステムにおいて、前記制御部が、前記検出部によって検出されたインピーダンスに従って前記透過信号を生成するように前記信号生成部を制御することを特徴とする、システム。
- 請求項8に記載のシステムにおいて、前記制御部が、前記透過信号の生成を停止し、前記検出部によって検出されたインピーダンスに従って前記送達信号を生成するように前記信号生成部を制御することを特徴とする、システム。
- 緩衝液中で生体細胞をエレクトロポレーションする方法において、
前記生体細胞を検出領域に流体力学的に集束させること;
第1の電気信号を生成すること;
ベースライン閾値を超える前記検出領域のインピーダンス値の増加が前記生体細胞の存在を示す、前記検出領域のインピーダンス値を、前記第1の電気信号を用いて監視すること;
前記検出領域のインピーダンスの増加に応答して、第2の電気信号を生成すること;
前記検出領域のインピーダンス値が透過閾値よりも大きいかどうかを判定すること;および
前記検出領域のインピーダンス値の判定に応答して、前記第2の電気信号の少なくとも1つのパラメータを調整すること、
を含むことを特徴とする、方法。 - 請求項11に記載の方法において、
測定されたインピーダンス値が透過閾値に等しいという判定に応答して、前記第2の電気信号を停止すること;および
前記検出領域内に集束された前記細胞内への分子送達を引き起こす、第3の電気信号を生成すること、
をさらに含むことを特徴とする、方法。 - 請求項12に記載の方法において、
前記検出領域のインピーダンス値が前記透過閾値に等しいかどうかを判定すること;
前記検出領域のインピーダンス値が前記透過閾値と等しいものでないという判定に応答して、前記第3の電気信号の少なくとも1つのパラメータを調整すること、

- 請求項14に記載の方法において、前記第3の電気信号が、前記検出領域のインピーダンス値が前記ベースライン閾値に等しいという判定に応答して停止されることを特徴とする、方法。
- 請求項11に記載の方法において、前記ベースライン閾値が、緩衝液のみが流れている第2の検出領域のインピーダンス値を連続的に監視することによって決定されることを特徴とする、方法。
- 請求項11に記載の方法において、前記第2の電気信号の少なくとも1つのパラメータが、電界振幅、パルス持続時間、パルス列周波数、デューティサイクル、およびサイクル数からなる群から選択されることを特徴とする、方法。

- 請求項11に記載の方法において、前記透過閾値が、細胞死を引き起こさない最適な細胞透過処理に対応することを特徴とする、方法。
- 複数の生体細胞をエレクトロポレーションする方法において、
複数の生体細胞の連続フローを、検出領域を介して前記複数の生体細胞それぞれを通過する単縦列フローへと流体力学的に集束させること;
第1の電気信号を生成すること;
ベースライン閾値を超える前記検出領域のインピーダンス値の増加が前記複数の生体細胞それぞれの存在を示す、前記検出領域のインピーダンス値を、前記第1の電気信号を用いて監視すること;
前記検出領域のインピーダンスの増加に応答して、第2の電気信号を生成すること;
前記検出領域のインピーダンス値が透過閾値よりも大きいかどうかを判定すること;
前記検出領域のインピーダンス値の判定に応答して、前記第2の電気信号の少なくとも1つのパラメータを調整すること、
を含むことを特徴とする、方法。 - 請求項23に記載の方法において、
測定されたインピーダンス値が前記透過閾値に等しいという判定に応答して、前記第2の電気信号を停止すること;および
前記検出領域内に集束された前記複数の生体細胞それぞれの中への分子送達を引き起こす、第3の電気信号を生成すること、
