Abstract
Description
これは、天然であるか又は部分的に若しくは完全に合成により生産されるかにかかわらない免疫グロブリンを説明する。この用語は、抗体抗原結合部位を含む任意のポリペプチド又はタンパク質にも関する。本明細書では、抗体分子が、天然の供給源から単離する又はその精製によって得てもよいし、又は遺伝子組み換え若しくは化学合成によって得てもよいこと、及び抗体分子が非天然のアミノ酸を含み得ることを理解されたい。
これは、標的抗原に結合し、かつその全て又は一部に相補的である分子の一部を説明する。ある抗体分子では、抗原結合部位は、抗体抗原結合部位と呼ばれ、そして標的抗原に結合し、かつその全て又は一部に相補的である抗体の一部を含む。抗原が大きい場合は、抗体は、抗原の特定の部分のみに結合することができ、この部分はエピトープと呼ばれる。抗体抗原結合部位は、1つ以上の抗体可変ドメインによって提供することができる。抗体抗原結合部位は、好ましくは、抗体軽鎖可変領域(VL)及び抗体重鎖可変領域(VH)を含む。

本明細書に記載されるように、本発明によるコンジュゲートは、IL22、及び本血管形成に関連した抗原に結合する抗体分子を含む。抗体分子は、本明細書に記載されるように、好ましくはダイアボディ又はscFvであり、最も好ましくはダイアボディである。
本発明によるコンジュゲートをコードする単離核酸分子も提供される。核酸分子は、DNA及び/又はRNAを含み得、かつ部分的に又は完全に合成であり得る。本明細書に示されるヌクレオチド配列への言及は、特定の配列を有するDNA分子を包含し、かつ文脈上別段の解釈が必要でない限り、TがUで置換された特定の配列を有するRNA分子を包含する。
上記のように1つ以上の構築物を含む組換え宿主細胞も提供される。適切な宿主細胞としては、細菌、哺乳動物細胞、植物細胞、糸状菌、酵母、及びバキュロウイルス系、並びにトランスジェニック植物及び動物が挙げられる。

自己免疫疾患は、好ましくは、血管形成に関連し、かつ/又は血管形成によって特徴付けられる。自己免疫疾患は、血管形成によって特徴付けられる自己免疫疾患であり得、新生血管が、フィブロネクチンのED−Aアイソフォーム、フィブロネクチンのED−Bアイソフォーム、及び/又はテネイシンCを発現する。自己免疫疾患は、炎症性自己免疫疾患、即ち、炎症に関連し、かつ/又は炎症によって特徴付けられる自己免疫疾患であり得る。
「炎症性疾患及び/又は障害」は、炎症を伴い、かつ/又は炎症によって特徴付けられる疾患及び/又は障害を指す。炎症性疾患及び/又は障害は、好ましくは、血管形成に関連し、かつ/又は血管形成によって特徴付けられる。炎症性疾患及び/又は障害は、血管形成によって特徴付けられる炎症性疾患及び/又は障害であり得、新生血管が、フィブロネクチンのED−Aアイソフォーム、フィブロネクチンのED−Bアイソフォーム、及び/又はテネイシンCを発現する。
本発明のコンジュゲートは、抗炎症活性を有し、従って、炎症及び/又は自己免疫疾患の処置に応用されることが期待される。いかなる理論的な説明によっても限定されるものではないが、本発明のコンジュゲートは、実施例で実証されるように、新生血管の優れた標的化の結果として強力な抗炎症活性を示すことが期待される。従って、本発明のコンジュゲートは、患者、好ましくはヒト患者の処置の方法に使用されるように設計される。
IL22及び抗ED−A抗体F8を含む融合タンパク質のクローニング
muIL22(ハツカネズミ由来)及び抗ED−A抗体F8を含む抗体融合タンパク質をコードする遺伝子を、PCRアセンブリを用いて作製した。IL22をコードする配列(シグナルペプチド配列が欠損した)を、15個のアミノ酸グリシン−セリン−リンカー[(G4S)3]をコードする配列によって、ダイアボディの形態(GGSGG−リンカーによって連結された重鎖及び軽鎖)のF8抗体をコードする遺伝子のC末端(F8−IL22)又はN末端(IL22−F8)に連結した。IgG由来シグナルペプチドをコードする配列を、コードされた融合タンパク質の高収率生産を可能にするためにN末端に付加した。エンジニアリングされたNhel及びNotl制限部位を用いて、遺伝子を、pcDNA3.1哺乳動物細胞発現ベクターにクローニングした。遺伝子アセンブリの概略図が図1に示されている。muIL22−F8及びF8−muIL22融合タンパク質をコードするために使用される遺伝子の配列がそれぞれ、配列番号21及び22に示され、以下に報告される実験に利用される成熟muIL22−F8及びF8−muIL22融合タンパク質のアミノ酸配列はそれぞれ、配列番号23及び24に示されている。シグナルペプチドは、融合タンパク質の発現後に切断され、従って、成熟融合タンパク質の一部ではない。
muIL22を含む融合タンパク質を、PEI媒介トランスフェクションによりCHO−S細胞で一過性に発現させた。500×106個の細胞を、250mLの予備加温ProCHO−2培地(10% FBS、2% HT supplement、4% Ultraglutamine、1% 抗生物質−抗真菌溶液が添加された)で再懸濁した。融合タンパク質をコードする遺伝子を含む625μgのプラスミドを、150mM 滅菌NaCl溶液で希釈して12.5mLの総量にした。濾過水中、2.5mLの滅菌PEI溶液(ポリエチレンイミン、1g/L 線形、MW 25,000)を10mlの150mM 滅菌NaClと混合した。PEI混合物をプラスミド混合物に添加して、室温で10分間インキュベートした。必要なインキュベーション時間後に、混合物を調製した細胞に添加し、この細胞を、37℃の振盪機に載せて160rpmで4時間振盪した。4時間後に、250mLの予備加温PowerCHO−2培地(10% FBS、2% HT supplement、4% Ultraglutamine、1% 抗生物質−抗真菌溶液が添加された)を添加し、細胞を31℃の振盪機に載せて140rpmで6日間振盪した。次いで、融合タンパク質を精製した。同じ方法を利用して、huIL22を含む融合タンパク質も発現させることができる。
500mLのトランスフェクトCHO−S細胞懸濁液を、4℃で20分間、7000rpmにて遠心分離した。上清をフラスコにデカントして40℃で保存し、ペレットを廃棄した。上清を、PBSを用いて流量が最大2mL/分に調整されたポンプを用いて、プロテインA樹脂を含むカラム(プロテインAアガロースビーズ/樹脂、Sino Biological Inc.)の上のゲル濾過培地を含むカラム(SephadexTM G−25 Medium,GE Healthcare,#17−0033−02)に流した。続いて、ゲル濾過樹脂を廃棄した。プロテインAカラムを、分光光度計(NanoDrop 2000c,witec ag,OD280nm)を用いて決定される洗浄液の光学濃度が0.1未満になるまで400mLの「洗浄液A」(PBS中、100mM NaCl(分析用の塩化ナトリウム、Emsure(商標)、7547−14−5)、0.5mM EDTA pH8.0(Franziska Bootzによって親切に提供された)、0.1% Tween 20(ポリオキシエチレンソルビタンモノラウレート、Sigma−Aldrich(商標),#SZBA3190V))で洗浄し、次いで、分光光度計NanoDropで決定される洗浄液の280nmでの光学濃度が0.05未満になるまで400mLの「洗浄液B」(PBS中、100mM NaCl 0.5mM EDTA)で洗浄した。muIL22を含む融合タンパク質を、10mLの0.1M グリシン(pH3、Fluka(商標)、#BCBB2819)を用いて重力流で溶出した。画分を、1.5mLのエッペンドルフ管に1mLのアリコートとして収集し、即座に凍結させた。紫外分光測定によって確認される融合タンパク質を含む画分をプールし、透析膜(Spectra/Por(商標)Dialysis Membrane,MWCO 12’000−14’000,Spectrum laboratories)に移し、そして3〜4LのPBSで一晩透析した。翌日、融合タンパク質溶液をエッペンドルフ管に移し、更なる分析のために4℃で1〜2日間保存するか、又は液体窒素で急速凍結し、−80℃の冷凍器に移した。同じ方法を使用して、huIL22を含む融合タンパク質も精製することができる。
muIL22を含む融合タンパク質の脱グリコシル化を、Peptide−N−Glycosidase F(PNGase F,NEB P0704S)を用いて行って、N結合型糖タンパク質から複合オリゴ糖を除去した。変性条件下、15μgの融合タンパク質を、総量30μlの10×糖タンパク質変性緩衝液(NEB)と共に99℃で10分間インキュベートした。変性した融合タンパク質を6μlの10×Glycobuffer2(NEB)、6μlの10% NP−40、及び脱イオン水と総量60μlで混合した。3μlのPNGase Fの添加後、反応混合物を37℃で4時間インキュベートした。その後、全ての試料をSDS−PAGEによって分析した。脱グリコシル化の効果は、SDS−PAGEゲル中での移動度シフト及びバンドの鋭さとして確認できる。huIL22を含む融合タンパク質の脱グリコシル化も、同じ方法で行うことができる。
融合タンパク質のサイズ排除クロマトグラフィーを、AEKTA−FPLCシステム(GE healthcare)において電気泳動用緩衝液としてリン酸緩衝生理食塩水を含むsuperdex 200 5/150カラム(GE healthcare)を用いて行った。100μlのタンパク質溶液をループに注入して、自動的にカラムに注入した。280nmでの紫外吸光度を長時間評価した。
表面プラズモン共鳴(Biacore 3000 system,GE Healthcare)を用いて、muIL22を含む融合タンパク質のED−Aに対する結合親和性を分析した。マイクロセンサーチップ(CM5,GE Healthcare)を、1500共鳴単位被覆密度で、11A12、組換え的に発現されたED−Aで被覆した。表面プラズモン共鳴での分析では、シリンジフィルタユニット(Millex(登録商標)−GV,低タンパク質結合デュラポアメンブレン(Low protein binding durapore membrane),0.22μm,#N3HA70695)でタンパク質を濾過し、そしてその濃度を、分光光度計(NanoDrop 2000c,witec ag,OD280nm)で決定した。huIL22を含む融合タンパク質のBiacore分析も、同じ方法で行うことができる。
抗体部分の結合能力を、ELISAによって更に確認した。組換えEDAドメインを、マキシソープウェル(Nunc−Immuno)に室温で一晩固定した。結合の評価の日に、ウェルを、リン酸緩衝生理食塩水中、200μlの4%ミルク(ミルク−PBS)を用いて室温で2時間ブロッキングした。ブロッキング液の除去後、2%ミルク−PBS中、200μlの異なる抗体濃度をウェルに添加し、室温で1時間インキュベートした。その後、ウェルを、0.1% Tween−20を含む200μlのPBSで3回、そしてPBSで3回洗浄した。続いて、プロテインA HRP(GE healthcare)を含む200μlの2%ミルク−PBSで洗浄した。室温で40分間のインキュベーション後、POD基質(Roche)を添加した。H2SO4を用いて反応を停止させ、紫外分光光度計(SpectraMax Paradigm,Molecular Devices)を用いて450nm及び650nmでの吸光度の測定により測定値を得た。
muIL22を含む融合タンパク質中のmuIL22の活性を、HT29細胞中のSTAT3のリン酸化によって検証した。細胞を融合タンパク質と共にインキュベートし、そしてSTAT3のリン酸化を、ウェスタンブロット分析によって定量した。
muIL22を含む融合タンパク質のin vivo標的性能を、F9担癌マウスでの定量的生体分布試験によって評価した。タンパク質を、Iodogen法を用いて125Iで標識した。15μgの放射性ヨウ化融合タンパク質を、側尾静脈に静脈内(i.v.)注射した。注射の24時間後にマウスを屠殺し、臓器を摘出して秤量し、そして放射活性を、Packard Cobra γカウンターを用いて測定した。臓器の放射活性を、組織1グラム当たりの注射用量のパーセンテージとして表した(%ID/g±SEM)。同じアプローチを使用して、huIL22を含む融合タンパク質のin vivo標的性能を決定することができる。
muIL22−F8及びF8−muIL22融合タンパク質の特徴付け
精製融合タンパク質は、(1)SDS−PAGE及び(2)サイズ排除クロマトグラフィーを用いて確認されたように好ましい生化学特性を示した。クマーシー染色を用いるSDS−PAGE分析は、推定44kDaよりも僅かに高い広いタンパク質バンドを明らかにした(図2A及び図2B)。このシフトは、N結合型グリカンの存在によって引き起こされ、これは、PNGase Fを用いて除去することができ、バンドシフトが融合タンパク質の予想サイズになる(図2A及び図2B)。Superdex S200 5/150カラムを用いるサイズ排除クロマトグラフィー分析により、コンジュゲート調製物の均質性を更に確認した(図2C及び図2D)。
F9担癌マウスでmuIL22−F8及びF8−muIL22融合タンパク質の定量的生体分布試験により、融合タンパク質による優れた腫瘍標的が示された。融合タンパク質は、腫瘍の処置又は検出での使用を意図していないが、これは、融合タンパク質が、ED−Aを発現する組織(例えば、ED−Aを発現することが分かっている新生血管)を特異的に標的とすることを実証し、他の(健常な)組織では融合タンパク質の存在が非常に限定されている。これらの優れた標的特性は、治療への応用のためにIL22を含む融合タンパク質を利用する場合に有用であることが期待される。
