37 CFR 1.53(b)の下に出願された本非仮特許出願は、2015年6月29日出願の米国仮特許出願第62/186236号の米国特許法119(e)に基づく利益を主張し、この仮出願の全体は引用により本明細書に包含される。
本発明は、概してPI3K阻害化合物であるtaselisib(GDC−0032)によるがんの治療に関する。本発明はまた、in vitro、in situ、及びin vivoでの哺乳動物細胞又は関連する病的状態の診断又は治療のためのtaselisibの使用方法に関する。
GDC−0032(taselisib)
(a)taselisibを用いた患者から得られた生物学的試料を処理すること;及び
(b)p110アルファタンパク質の枯渇を検出すること;
を含み、生物学的試料におけるp110アルファタンパク質の枯渇が、taselisibによる治療に応答するであろう患者を同定する。p110アルファタンパク質の枯渇は、化合物に対する患者の治療的応答性を示し、抗p110アルファ抗体への結合により測定されうる。試料中のp110アルファタンパク質に対する抗p110アルファ抗体の結合は、ウェスタンブロット分析、酵素結合免疫吸着法(ELISA)、ラジオイムノアッセイ(RIA)、免疫組織化学(IHC)、蛍光活性化セルソーティング(FACS)、又は逆相タンパク質アレイにより決定される。
(a)患者から得られた生物学的試料を、H1047R、C420R、H1047L、E542K、E545K及びQ546Rから選択される突然変異を含むPIK3CA突然変異状態について試験すること;
(b)PIK3CA突然変異を有する患者由来の生物学的試料をtaselisibと接触させ、p110アルファアイソフォームの枯渇を検出すること;及び
(c)taselisibを、PIK3CA突然変異を有する患者に投与すること
を含む、患者の治療法である。生物学的試料は循環性腫瘍細胞でもよい。taselisibと組み合わせて、患者には、5−FU、ドセタキセル、エリブリン、ゲムシタビン、GDC−0973、GDC−0623、パクリタキセル、タモキシフェン、フルベストラント、デキサメタゾン、ペルツズマブ、トラスツズマブエムタンシン、トラスツズマブ及びレトロゾールから選択される化学療法剤が投与されうる。患者は、HER2発現乳がん又はエストロゲン受容体陽性(ER+)乳がんに罹患している場合もある。がんは転移性でありうる。Taselisibは、アジュバント設定において患者に投与されてよい。

taselisibによるp110アルファ(p110a)の分解は、用量依存性、時間依存性、プロテアソーム依存性、及びユビキチン依存性に起こる。taselisibによる分解は、PIK3CAの突然変異細胞中のp110aに特異的であり;p85、p110デルタアイソフォーム、又はp110aは野生型細胞において観察されない。フィードバックに直面した経路抑制の予期されていなかった驚くべき恩恵は、1及び24時間経過時のpAKT及びpPRAS40のレベルにより測定される。これらの相関的観察は、taselisibによる治療選択の治療濃度域を広げうる(治療指数の上昇)。

アルペリシブ(BYL−719)
taselisibによって処理した生物学的試料におけるp110アルファの枯渇を検出及び測定するための上記方法に加えて、質量分析により直接的な検出を達成することができる。野生型及びH1047R突然変異型p110aタンパク質の混合物を発現するHCC1954乳がん細胞を、DMSO中500nMのtaselisib(GDC−0032)で24時間処理した。p110アルファタンパク質の免疫沈降法をを実施し、SDS−PAGEにより分離されてクーマシー染色されたタンパク質を補足した。p110アルファを含むGelブレンドを、ゲル内トリプシン消化に供し、分析用のトリプシンペプチドを生成した。DMSO(コントロール)又はtaselisibで処理した細胞由来のトリプシンペプチドは、対応する749.8282m/z(+/−10ppm)イオンの定量化に基づき、同等のレベルのQM(ox)NDAHHGGWTTKペプチド配列(配列番号7)を示した。taselisib処理した細胞由来のトリプシンペプチドは、DMSO処理細胞と比較して、393.6983m/z(+/− 10ppm)のイオンによって特徴づけられる、突然変異特異的ペプチドHGGWTTK(配列番号8)の欠損を示した。したがって、p110a H1047Rといった突然変異腫瘍に特異的なネオトリプシンペプチドを使用して、野生型形態と比較した場合の突然変異p110aの枯渇を実証することができる。

表1は複数のPI3K結合化合物の生物学的特性をまとめたものである。GDC−0032(Ndubaku et al(2013)Jour. Med. Chem. 56(11):4597-4610;Staben et al(2013)Bioorg. Med. Chem. Lett. 23 2606-2613;国際公開第2011/036280号;米国特許第8242104号;米国特許第8343955号)は、pan−PI3K阻害化合物 GDC−0941よりPI3Kアルファ突然変異がん細胞に対して強力である(Folkes et al(2008)Jour. of Med. Taselisib(GDC−0032)は、PI3K野生型アイソフォームとの相互作用とは異なるPI3Kアルファ突然変異アイソフォームとの重要な相互作用に起因して、PI3K野生型アイソフォームよりPI3Kアルファ突然変異アイソフォームについて高い選択性を有し、PI3Kアルファ突然変異アイソフォームの重要な突然変異特異的特徴と相互作用するための、原子及び官能基の精確な位置づけ及び配置を含みうる。このような相互作用は、PI3Kアルファ突然変異アイソフォームタンパク質との水素結合−ドナー、水素結合−アクセプター及び/又はファンデルワース力パートナーとして作用する官能基によって達成される(Staben et al (2013)Bioorg. Med. Chem. Lett. 23:2606-2613; Ndubaku et al (2013) Jour. Med. Chem. 56(11),4597-4610)。Taselisibは、低エネルギー立体構造での結合トポロジーに適合することができ、リガンド結合部位に効率的な極性及びファンデルワース相互作用をつくることができる。
特定の実施態様では、試料中のバイオマーカータンパク質の存在及び/又は発現レベル/量が、免疫組織化学(IHC)及び染色プロトコールを用いて試験される。組織切片のIHC染色は、試料中のタンパク質の存在を決定又は検出する信頼性の高い方法であることが示された。方法、アッセイ及び/又はキットのいずれかのいくつかの実施態様では、関連のバイオマーカーは突然変異p110アルファタンパク質である。いくつかの実施態様では、突然変異p110アルファは免疫組織化学により検出される。いくつかの実施態様では、個体由来の試料中における突然変異p110アルファバイオマーカーの発現の増大は、タンパク質発現の増大であり、さらなる実施態様では、IHCを用いて決定される。一実施態様では、バイオマーカーの発現レベルは、(a)抗体を用いて試料(例えば対象のがん試料)のIHC分析を実施すること;及びb)試料中のバイオマーカーの発現レベルを決定することを含む方法を用いて決定される。いくつかの実施態様では、IHC染色強度は基準との比較において決定される。いくつかの実施態様では、基準は参照値である。いくつかの実施態様では、基準は基準試料(例えば、コントロール細胞株染色試料又は非がん性患者由来の組織試料)である。
Taselisib(GDC−0032)は、がんを含む過剰増殖性疾患の治療において有用である。一実施態様では、突然変異PI3K腫瘍を有する患者がtaselisibで治療される。突然変異PI3K腫瘍は、乳房腫瘍、肺腫瘍、又は他の器官に見られる腫瘍である。
・バイオマーカーの同定に基づく診断の方法;
・患者がtaselisib、又はtaselisibと化学療法剤の組み合わせに応答すると思われるかどうかを決定する方法;
・taselisib、又はtaselisibと化学療法剤の組み合わせの除去を監視することにより治療有効性を最適化する方法;
・治療抵抗性突然変異の発生を監視することによりtaselisib、又はtaselisibと化学療法剤の組み合わせの治療レジメンを最適化する方法;及び
・taselisib又はtaselisibと化学療法剤療法の組み合わせによる治療から最大の恩恵を受けるであろう患者を同定し、taselisib又はtaselisibと化学療法剤療法の組み合わせによる治療に対する感受性及び応答性について患者を監視するための方法
を含む。

b)患者から得られた生物学的試料中のp110アルファのレベルと相関したp110アルファ又はバイオマーカーのレベルの変化を測定すること

本発明の化合物の薬学的組成物又は製剤は、taselisibと、薬学的に許容される担体、流動促進剤、希釈剤、及び賦形剤のうちの一又は複数とを含む。
Taselisibは、前悪性及び非腫瘍性又は非悪性の過剰増殖性疾患と共に、固形腫瘍又は造血性悪性腫瘍を含む過剰増殖性疾患の治療のための特定の化学療法剤と組み合わせて用いることができる。特定の実施態様では、taselisibは、組み合わせの同時投与のための単一の錠剤、ピル、カプセル、又は溶液として、化学療法剤と単剤に組み合わせられる。他の実施態様では、taselisibと化学療法剤は、投薬レジメン又は治療法の過程に従って、taselisibと、5−FU、ドセタキセル、エリブリン、ゲムシタビン、GDC−0973、GDC−0623、パクリタキセル、タモキシフェン、フルベストラント、デキサメタゾン、ペルツズマブ、トラスツズマブエムタンシン、トラスツズマブ及びレトロゾールより選択される化学療法剤との順次的投与のための、別個の錠剤、ピル、カプセル、又は溶液としての別個の製剤で投与される。化学療法剤は、抗過剰増殖特性を有するか、又は過剰増殖性疾患の治療に有用である。taselisibと化学療法剤の組み合わせは、相乗的特性を有しうる。薬学的組み合わせ製剤又は投与レジメンの化学療法剤は、好ましくは、互いに悪影響を与えないような、taselisibを補完する活性を有する。治療的組み合わせのこのような化合物は、意図した目的に有効な量で投与される。一実施態様では、この発明の薬学的製剤は、taselisibと本明細書に記載されるような化学療法剤とを含む。別の実施態様では、治療的組み合わせは、治療的有効量のtaselisibが一日二回から三週毎(q3wk)の範囲で投与され、治療的有効量の化学療法剤が、一日二回から三週毎の範囲で別個に、交互に投与される投与レジメンにより投与される。
本発明の治療的組み合わせは、治療される状態に適した任意の経路により投与することができる。適切な経路には、経口、非経口(皮下、筋肉内、静脈内、動脈内、吸入、皮内、髄腔内、硬膜外、及び点滴技術を含む)、経皮、直腸内、鼻腔内、局所(頬側及び舌下を含む)、膣内、腹腔内、肺内及び鼻腔内が含まれる。局所投与は、経皮パッチ又はイオントホレシス装置といった経皮投与の使用も含むことができる。薬物の製剤は、Remington's Pharmaceutical Sciences, 18th Ed., (1995) Mack Publishing Co., Easton, PAにおいて議論されている。薬物の他の例は、Liberman, H. 本発明の別の実施態様において、上記の疾患及び障害の治療に有用なtaselisibを含有する製品又は「キット」が提供される。一実施態様では、キットはtaselisibを含む容器を備える。キットは、容器上に又は容器に付属するラベル又は添付文書をさらに含みうる。「添付文書」という用語は、このような治療製品の使用に関する、指示、使用法、用量、投与、禁忌及び/又は注意事項についての情報を含む、治療製品の商品包装に通例含まれる説明書を指すために使用される。適切な容器は、例えばボトル、バイアル、シリンジ、ブリスターパッグなどを含む。容器はガラス又はプラスチックなどの様々な材料から形成されうる。容器は、病態を治療するのに有効なtaselisib又はその製剤を保持することができ、無菌アクセスポートを有しうる(例えば、容器は、皮下注射針によって穿孔可能なストッパーを有する静脈注射用溶液のバッグ又はバイアルとすることができる)。組成物中の少なくとも一の活性剤はtaselisibである。ラベル又は添付文書には、組成物が、がんなどの選択した病態の治療のために使用されることが示される。一実施態様において、ラベル又は添付文書に、式Iの化合物を含む組成物が、異常な細胞増殖から生じる障害を治療するために使用されうることが示される。ラベル又は添付文書には、この組成物が、他の障害を治療するために使用できることが示されてもよい。代替的に、又は追加的に、製造品は、薬学的に許容可能なバッファー、例えば注射用静菌水(BWFI)、リン酸緩衝生理食塩水、リンガー溶液及びデキストロース溶液を含む第二の容器をさらに含んでもよい。製造品は、他のバッファー、希釈剤、フィルター、針、及びシリンジを含め、商業的及び使用者の観点から望ましいその他の材料をさらに含んでもよい。
結合アッセイ:最初の偏光実験は、Analyst HT 96−384(Molecular Devices Corp,Sunnyvale、CA)において実施された。蛍光偏光親和性測定のための試料を、偏光バッファー(10mMのTris(pH7.5)、50mMのNaCl、4mMのMgCl2、0.05%のChaps、及び1mMのDTT)中最終濃度20ug/mLから開始して10mMのPIP2(Echelon−Inc.,Salt Lake City,UT)最終濃度まで1:3に段階希釈したp110アルファPI3K(Upstate Cell Signaling Solutions,Charlottesville,VA)の付加により調製した。室温で30分のインキュベーション時間の後、GRP−1及びPIP3−TAMRAプローブ(Echelon−Inc.,Salt Lake City,UT)の付加により反応を止めた。最終濃度はそれぞれ100nM及び5nMであった。384ウェルのブラックローボリュームProxiplates(登録商標)(PerkinElmer、Wellesley、MA.)においてローダミンフルオロフォア用の標準のカットフィルター(λex=530nm;λem=590nm)により読み取りを行った。蛍光偏光の値をタンパク質濃度の関数としてグラフ化した。EC50値は、KaleidaGraph(登録商標)ソフトウェア(Synergy software,Reading,PA)を用いて4パラメーター方程式にデータを当てはめることにより得られた。この実験は、続いて行われる阻害剤を用いた競合実験に用いられる適切なタンパク質濃度も確立する。
細胞の培養:細胞株を、10%のウシ胎子血清、100 U/mLのペニシリン、及び100μg/mLのストレプトマイシンを含むRPMI培地において、標準の組織培養条件下で増殖させた。HCC−1954及びHDQ−P1は、乳がん細胞株(American Type Culture Collection;Manassas,VA)である。HCC−1954及びHDQ−P1細胞を、6ウェルの組織培養プレートの各ウェルに、800,00細胞/ウェルで配置し、37℃で一晩インキュベートした。細胞を、各化合物に示された濃度で24時間インキュベートした。インキュベーションに続いて、細胞を、冷たいリン酸緩衝生理食塩水 (PBS)で一度洗浄し、プロテアーゼ阻害剤(F.Hoffman−LaRoche;Mannheim,Germany)、1mMのフェニルメチルスルホニルフルオリド、及びホスファターゼ阻害剤カクテル1及び2(Sigma−Aldrich;St.Louis,MO)を補充したBiosourceTM Cell Extraction Buffer(Invitrogen;Carlsbad,CA)中に溶解した。タンパク質濃度を、Pierce BCA Protein Assay Kit(Thermo Fisher Scientific;Rockford,IL)を用いて決定した。
1.培地中に約104個の細胞を含む細胞培養物100μlのアリコート(細胞株及び腫瘍種類については表3を参照)を、384ウェルの不透明な壁を有するプレートの各ウェルに配した。
2.培地のみで細胞を含まないコントロールウェルを準備した。
3.化合物を実験ウェルに加え、3〜5日間インキュベートした。
4.プレートを、約30分間室温に平衡化した。
5.各ウェル中に存在する細胞培養培地の体積に等しい体積のCellTiter−Glo(登録商標)試薬を加えた。
6.内容物をオービタルシェーカーで2分間混合し、細胞溶解を誘導した。
7.プレートを室温で10分間インキュベートし、発光シグナルを安定化させた。
8.発光を記録し、RLU=相対発光単位としてグラフに報告した。
9.ChouとTalalayの組み合わせ方法及びCalcuSyn(登録商標)ソフトウェアを用いるDose−Effect Analysis(Biosoft、Cambridge、UK)を使用して分析し、組み合わせ指標を得る。
細胞の培養:細胞株は、American Type Culture Collection(ATCC,VA)又はDeutsche Sammlung von Mikroorganismen und Zellkulturen GmbH(DMSZ,Germany)から得た。細胞株を、10%のウシ胎児血清、100単位/mlのペニシリン、及び100μg/mlのストレプトマイシンを補充したDMEM又はRPMI中で、5%のCO2下において37℃で培養した。MCF7−neo/HER2は、Genentech社で開発された、in vivoで選択される腫瘍細胞株であり、親MCF7ヒト乳がん細胞株に由来する。同質遺伝子的細胞株(SW48親、SW48E545K、及びSW48H1047R)を、Horizon Discovery Ltd.(Cambridge,UK)からライセンシングし、10%のウシ胎児血清、100単位/mlのペニシリン、及び100μg/mlのストレプトマイシンを補充したMcCoy's 5Aにおいて、5%のCO2下において37℃で培養した。
マウス:雌の重症複合免疫不全マウス(Fox Chase SCID(登録商標),C.B−17/IcrHsd,Harlan)又はヌードマウス(Taconic Farms,Harlan)は、週齢8から9であり、試験0日目の体重範囲は15.1から21.4グラムであった。動物に水(逆浸透、1ppm Cl)及び18.0%の粗タンパク質、5.0%の粗脂肪、及び5.0%の粗繊維からなるNIH 31 Modified and Irradiated Lab Diet(登録商標)を自由に摂らせた。マウスを、12時間の照明サイクル及び21〜22℃(華氏70〜72度)及び湿度40〜60%で静止マイクロアイソレーター内の照射ALPHA−Dri(登録商標)bed−o'cobs(登録商標)Laboratory Animal Beddingに収容した。PRCは、具体的には、耐性、飼育、外科手技、給餌及び流体の管理、並びに動物医薬ケアに関して、Guide for Care and Use of Laboratory Animalsの推奨に準拠する。PRCの動物のケア及び使用プログラムは、Association for Assessment and Accreditation of Laboratory Animal Care International(AAALAC)によって認証されており、これは実験動物のケア及び使用の許容基準への準拠を保証する。
腫瘍体積(mm3)=(w2×l)/2[w=腫瘍の幅、l=腫瘍の長さ(mm)]を用いて計算した。腫瘍重量は、1mgが腫瘍体積1mm3と等しいと仮定して予測されうる。
タンパク質アッセイ:タンパク質濃度を、Pierce BCA Protein Assay Kit(Rockford,IL)を用いて決定した。免疫ブロットの場合、等しいタンパク質の量を、NuPage Bis−Tris 4−12%勾配ゲル(Invitrogen;Carlsbad,CA)を用いた電気泳動により分離した;タンパク質を、InVitrogen社のIBlotシステム及びプロトコールを用いてニトロセルロース膜上に移した。ホスホ−Akt(Ser473)及びp110アルファに対する抗体はCell Signaling(Danvers,MA)から取得した。GAPDH及びベータ−アクチンに対する抗体はSigmaから取得した。
ウェスタンブロットの場合、4℃の5% w/vのBSA、1X TBS、0.1%のTween(登録商標)20中で稀釈した一次抗体と共に、穏やかに振とうしながら膜を一晩インキュベートする。

1. 20Xのリン酸緩衝生理食塩水(PBS):(#9808)1Lの1X PBSの調製:50mlの20X PBSを950mlのdH2Oに付加、混合。
2.10XのTris緩衝生理食塩水(TBS):(#12498)1Lの1X TBSの調製:100mlの10Xを900mlのdH2Oに付加、混合。
3.1X SDSの試料バッファー:Blue Loading Pack(#7722)又はRed Loading Pack(#7723)1/10容積の30X DTTを1容積の3X SDSローディングバッファーに加えることにより新鮮な3X 還元ローディングバッファーを調製。dH2Oで1Xに希釈。
4.10X Tris−Glycine SDSランニングバッファー:(#4050)1Lの1Xランニングバッファーの調製:100mlの10Xランニングバッファーを900mlのdH2Oに付加、混合。
5.10X Tris−Glycine転写バッファー:(#12539)1Lの1X転写バッファー:100mlの10X転写バッファーを200mlのメタノール+700mlのdH2Oに付加、混合。
6.Tween(登録商標)20(TBST)を含む10XのTris緩衝生理食塩水:(#9997)1Lの1X TBSTの調製:100mlの10X TBSTを900mlのdH2Oに付加、混合。
7.脱脂粉乳:(9999)。
8.ブロッキングバッファー:5% w/vの脱脂粉乳を含む1X TBST;150mlにつき、7.5gの脱脂粉乳を150mlの1X TBSTに付加、よく混合。
9.洗浄バッファー:(#9997)1X TBST。
10.ウシ血清アルブミン(BSA):(9998)。
11.一次抗体稀釈バッファー:5%のBSAを含む1X TBST;20mlにつき、1.0gのBSAを20mlの1X TBSTに付加、よく混合。
12.ビオチン化タンパク質ラダー検出パック:(#7727)。
13.事前染色タンパク質マーカー、広範囲(事前混合フォーマット):(#7720)。
14.ブロッティング用膜及び紙:(Cell Signaling Technology #12369)このプロトコールはニトロセルロース膜のために最適化された。孔サイズ0.2μmが一般に推奨される。
15.HRPにコンジュゲートした二次抗体:抗ウサギIgG、HRP結合抗体(#7074)。
16.検出試薬:SignalFireTM ECL試薬(#6883)。
試料調製のための一般的プロトコール
1.制御因子を含む新鮮な培地を所望の時間にわたって加えることにより細胞を処理する。
2.培養物から培地を吸引;細胞を1X PBSで洗浄;吸引する。
3.1X SDS試料バッファーを加えることにより(6ウェルプレートの1ウェルにつき100μl又は直径10cmのプレートにつき500μl)細胞を溶解させる。直ちにプレートから細胞をこすり取り、抽出物をエッペンチューブに移す。氷上に維持する。
4.10〜15秒間超音波処理して細胞溶解を完了させ、DNAをせん断する(試料粘度を低下させるため)。
5.20μlの試料95〜100℃に5分間加熱;氷上で冷却する。
6.5分間微量遠心分離する。
7.20μlをSDS−PAGEゲルにローディングする(10cm×10cm)。
注意:事前染色した分子量マーカーをローディング(#7720、10μl/レーン)し、電気泳動転写及びビオチン化タンパク質ラダー(#7727、10μl/レーン)を確認して分子量を決定することが推奨される。
8.ニトロセルロース膜への電気泳動転写(#12369).
注意:容積は10cm×10cm(100cm2)の膜のものである;異なる大きさの膜については、容積を適宜調節されたい。
I.膜のブロッキング
1.(任意)転写後、ニトロセルロース膜を、25mlのTBSで5分間室温で洗浄する。
2.膜を25mlのブロッキングバッファー中において1時間室温でインキュベートする。
3.15mlのTBSTを用いて三回、それぞれ5分間洗浄する。
II.一次抗体のインキュベーション
1.膜及び一次抗体を、10mlの一次抗体稀釈バッファー中において(製品データシートにおいて推奨される適切な希釈及び希釈剤で)、穏やかに振とうさせながら一晩4℃でインキュベートする。
2.15mlのTBSTを用いて三回、それぞれ5分間洗浄する。
3.10mlのブロッキングバッファー中において、穏やかに振とうさせながら、1時間室温で抗ウサギIgG、HRP結合抗体(1:2000の#7074)及び抗ビオチン、HRP結合抗体(1:1000−1:3000の#7075)と共に膜をインキュベートし、ビオチン化タンパク質マーカーを検出する。
4.15mlのTBSTを用いて三回、それぞれ5分間洗浄する。
5.検出(セクションD)を進める。
使用方法:
1.膜結合HRP(抗体コンジュゲート)を、TBSTにおいて三回、5分間洗浄する。
2.一部分2Xの試薬A及び一部分2Xの試薬B(例えば10mlにつき、5mlの試薬Aと5mlの試薬Bを加える)を希釈することにより1X SignalFireTM ECL試薬(#6883)を調製する。よく混合する。
3.膜と共に基質を1分間インキュベートし、余分な溶液を除去し(膜は濡れたまま)、プラスチックに包んでX−ray線フィルムに曝露する。
*肌に繰り返し曝露することは避ける。
既存の膜の再検出は、利用できる試料の量が限られているとき、複数のタンパク質を独立して免疫ブロットするために便利な手段である。最良の結果のためには、新鮮なブロットが常に推奨されることに注意されたい。再検出は、有効な方法でありうるが、ブロットの各再検出において、バックグランドシグナルが増加する可能性が存在する。加えて、新規抗体の結合に起因するシグナルが最初の免疫ブロッティング実験の残存シグナルではないように、再検出に先立って最初の抗体複合体の除去を確認することが推奨される。これは、ブロットをECL試薬に再曝露し、次の一次抗体を加える前にシグナルが存在しないことを確認することにより行うことができる。
注意:逆浸透脱イオン化(RODI)水又は同等に精製された水を用いて溶液を調製する。
1.洗浄バッファー:Tween(登録商標)20(TBST−10X)(#9997)を含むTris緩衝生理食塩水
2.ストリップバッファー:100mlを調製するために、0.76gのTrisベース、2gのSDS及び700μlのβ−メルカプトエタノールを混合する。脱イオン化H2Oを用いて100mlにする。HClを用いてpHを6.8に調整する。
1.フィルム曝露の後、膜を4回、それぞれ5分間TBST中で洗浄する。膜を乾燥させない場合に最良の結果が得られる。
2.ストリップバッファー中において(軽く振とうさせながら)膜を30分間50℃でインキュベートする。
3.膜を6回、それぞれ5分間TBST中で洗浄する。
4.(任意)元のシグナルが除去されたことを確認するために、膜を2回、それぞれ5分間10mlのTBSTを用いて洗浄する。膜をLumiGLO(登録商標)と共に、穏やかに振とうさせながら1分間室温でインキュベートする。膜から余分な現像液を排出させる。乾燥させない。プラスチックラップに包み、X線フィルムに曝露する。
5.再度膜を4回、それぞれ5分間TBST中において洗浄する。
6.ここで膜は再使用の準備が整う。ウエスタン免疫ブロット法プロトコールの「膜のブロッティング及び抗体のインキュベーション」工程において検出を開始する。
患者の腫瘍生検試料中における突然変異p110アルファのレベルを決定するために、様々な診断アッセイが利用可能である。一実施態様では、突然変異p110アルファのレベルは、免疫組織化学(IHC)により分析できる。腫瘍生検由来のパラフィン包埋組織切片をIHCアッセイに供し、以下の染色強度基準に合わせる:
スコア0−染色は観察されない、又は腫瘍細胞の10%未満に膜染色が観察される。
スコア1+−わずかな/ほとんど知覚できない膜染色が10%を上回る腫瘍細胞に観察される。細胞の膜の一部のみが染色される。
スコア2+−弱〜中程度の完全な膜染色が10%を上回る腫瘍細胞に観察される。
スコア3+−中〜強の完全な膜染色が10%を上回る腫瘍細胞に観察される。
液体クロマトグラフィー−タンデム質量分析(LC−MS/MS)分析を、それぞれDMSO(ビヒクル)又はGDC−0032(500nM)で24時間処理した三つの細胞株:HCC1954、HCC202及びHDQP1から免疫沈降させたp110アルファ(PIK3CA)タンパク質に対して実施した。各実験は、n=4の生物学的複写物について、一つの細胞株/処理当たり4〜6mgのタンパク質溶解物(合計6の試料/実験)から開始して実施された。PIK3CAの予想される移動に対応して、一つの試料当たり一つのゲル領域を、隣接レーンの精製済みp110アルファタンパク質の移動に基づいて切除した。ゲル片を〜1mm3の片に切断し、以下のようにしてゲル内消化させた。ゲル片を、50mMの炭酸水素アンモニウム/50%のアセトニトリルを用いて脱染し、100%のアセトニトリルを用いて脱水した後、50mMのジチオスレイトール(30分、50℃)及び50mMのヨードアセトミド(20分、室温)を用いてそれぞれ還元及びアルキル化した。ゲル片を再度脱水し、次いで氷上で2時間にわたり50mMの炭酸水素アンモニウム/5%のアセトニトリル消化バッファー中の20ng/ulのトリプシンで再度膨張させ、次いで37℃のオーブンに移して一晩インキュベーションした。消化されたペプチドをエッペンチューブに移し、ゲル片を、一度は50%のアセトニトリル/0.5%のトリフルオロ酢酸で、二回目は100%のアセトニトリルで、計二回抽出した。抽出物を消化されたペプチドと混合し、高速真空(speed−vac)乾燥させて完了した。試料を5%のギ酸/0.1%のヘプタフルオロ酪酸/0.01%の過酸化水素中において30分間再構成した後、LC−MS/MS分析した。

- taselisibによる治療のための患者を選択する方法であって:
(a)患者から得られた生物学的試料をtaselisibにより処理すること;及び
(b)p110アルファタンパク質の枯渇を検出すること;

a)taselisibを投与する工程;及び
b)患者から得られた生物学的試料中のp110アルファ又はp110アルファのレベルと相関したバイオマーカーのレベルの変化を測定する工程
