材料及び方法
試薬及び抗体
[0095]AAV8−CBA−p75NTR−ECD−Fc(AAV−ECD−Fc)及びAAV8−CBA−GFP(AAV−GFP)のベクターは、Virovek(ヘイワード、CA、米国)によって産生及び精製した。pcDNA3ヒト完全長p75NTRは、Rainer Niedenthal博士、Institute for Physiological Chemistry/Biochemistry、Hannover Medical School、ハノーファー、ドイツ由来であった。合成Aβ42(カタログ番号62080)及びスクランブルAβ42(カタログ番号62046)は、American Peptide(サニーベール、CA、米国)から購入した。

[00117]操作者等及び研究者等は、群及び介入情報に対して完全盲検であった。動物は全て、12ヵ月齢で、モリス水迷路テスト、Y−迷路及びオープンフィールドを含む行動に関する課題に付した。モリス水迷路テストは、これまでに報告されるプロトコールに倣って、わずかな変更を伴って行った(Wang Y.J.他、2010年;Yau J.L.他、2007年;Markowska,A.L他、1993年)。簡潔に述べると、モリス水迷路テストは、連続して4日間の1日につき4つのプラットフォーム試行、続くプローブ試行から構成された。動作は、ビデオで記録して、画像分析ソフトウェア(ANY−迷路、Stoelting)によって分析した。プラットフォーム試行では、経路の距離及び待ち時間を測定し、プローブ試行では、四分円時間及びアニュラス横断を測定した(Markowska,A.L他、1993年)。自発的交替テスト及び新規アーム探索は、Y−迷路で実行した。自発交替テストに関して、マウスを、5分の期間中に迷路を通って自由に移動させた。交替は、重複する三重組での3つのアームへの連続的な進入と定義した(Sarter,M.他、1988年)。交替のパーセントは、考えうる交替(アーム進入−2の数と定義される)に対する実際の(総抗体)の比率に100を乗算したものとして算出した。新規アーム探索に関しては、選択した開始アーム(ホームアーム)の末端にマウスを配置させて、遮断したアーム(新規アーム)のうちの1つを用いて、5分間迷路を探索させた(Jung、W.R他、2008)。2時間の間隔後、マウスに、3つのアーム全てを自由に探査させた。新規アーム進入及び新規アームで費やした時間は、3分の検索試行中の3つのアーム全てにおける総時間のパーセントとして算出した。オープンフィールドテストに関して、各マウスをオープンフィールド装置の中央に配置した(Dellu,F.他、1992年;Conrad,C.D他、1999年)。経路は、コンピューター追跡システム(ライムライト(Limelight)、ActiMetrics)を使用して3分間追跡した。さらに、立ち上がり、身繕い、排便及び排尿を記録した。

[00162]AAV−GFP処置マウスと比較して、AAV−ECD−Fcで処置したマウスは、モリス水迷路テストにおいてより良好に行動した。これは、進行性プラットフォーム学習試行を用いたプラットフォーム上へ回避する回避待ち時間及び移動する距離、より多数のプラットフォーム区域横断(図9 a、b、c)の著しい低減に反映された。データは、AAV−ECD−Fc処置が、ADマウスにおいて空間学習及び記憶を著しく改善することを示す。さらに、AAV−ECD−Fc処置マウスはまた、AAV−GFP処置対照よりも、Y−迷路及びオープンフィールドテストにおいて、より良好に行動した。このことは、Y−迷路テストにおける新規アームへのより多くの進入及びより高い自発的交替に反映された(図9 d及びe)。AAV−ECD−Fc処置マウスの作業記憶が、対照マウスの作業記憶よりも良好であったことが示される。オープンフィールドテストでは、より長い移動距離が、AAV−GFP処置対照と比較して、AAV−ECD−Fc処置マウスにおいて観察され(図9 f)、ECD−Fc処置マウスは、対照ADマウスよりも良好な探索活動を有することを示している。しかしながら、水泳速度は、群間で有意には異ならず、マウスは、同等の自発運動活性を有することを示す。したがって、これらのデータにより、筋肉内p75ECD送達は、ADマウスにおいて認知低下を改善することが示される。

[00164]ミクログリオーシス及びアストロサイトーシスを含む脳炎症は、Aβに続くAD病理学の第2の特徴である。新皮質及び海馬におけるCD45(ミクログリオーシス)及びGFAP(アストロサイトーシス)染色の分画区域は、GFAP及びCD45陽性染色区域の分画の低減に反映されるように、AAV−GFP処置ADマウスと比較して、AAV−ECD−Fc処置ADマウスにおいて著しく低減された(図11)。したがって、学習及び記憶作業におけるより行動の改善並びに神経炎症の低減は、アミロイド荷重の低減に加えて、ECD−Fc処置の可能性の高い結末である。
[00165]タウ病理学を評価するために、pS396、pT231、pS199及びpS262のタウタンパク質のリン酸化を、上述するように、AAV−GFP処置対照と比較して、AAV−ECD−Fc処置ADマウスにおいて検査した。これらの部位でのリン酸化は、上述する抗pS396、pT231、pS262及びpS199抗体を使用して研究した。これらのリン酸化部位は、それらがADマウスにおけるタウ過剰リン酸化の共通部位であるため、本研究で選択した。皮質及び海馬におけるタウ−ホスホ−Ser396陽性ニューロンの分画区域は、GFP群の分画区域よりも、p75ECD処置群において著しく低かった(図12 a、b、c及びd)。セリン396、262、199及びスレオニン231を含む多重部位でのタウリン酸化のレベルは、処置群の脳において、一貫して且つ著しく減少した(図12 e及びf)。これらの見解により、p75ECDは、タウ過剰リン酸化に対して保護することが示される。
[00166]過剰リン酸化されたタウの凝集体は、アルツハイマー病又は前頭側頭型認知症(FTD)を患う脳において顕著である。凝集体は、FTDの家族性症例におけるタウ突然変異の同定後に、動物モデルにおいて再現させた。これには、本発明者等のこれまで精製したトランスジェニックモデルpR5が含まれ、それは、ニューロンにおいてFTD(P301L)突然変異微小管結合タンパク質タウ(MAPT)を発現する(Deters他、2008年)。マウスは、もつれ(NFT)形成、記憶障害及びミトコンドリア機能不全を含むタウ凝集を特徴とする。この実験では、3ヵ月齢のpR5マウスを、2つの異なる群に無作為に分けた。処置の最初の週の間に、10mg/kgのp75ECD−Fc又は5mg/kgのFc組換えタンパク質を、ip注射を介して2度/週、マウスに投与して、マウス血漿中に十分量のタンパク質を供給した。次に、マウスに、ip注射を介して、総処置期間3ヵ月の残りの期間、150〜200μ用量のPBS中において、2度/週、5mg/kgの組換えp75ECD−Fc又は2.5mg/kgの組換えFcタンパク質を付与した。
[00167]PR5マウスにおける学習及び記憶機能に対するp75ECD−Fcの効果を評価するために、12匹の3ヵ月齢のマウスを、2つの群に分けて、p75ECD−Fc又はFc組換えタンパク質を3ヵ月間付与した。並行して、野生型マウスを試験した。同年齢且つ同性の野生型マウスの群を対照として使用した。PR5マウスに関するモリス水迷路(MWM)テストにより、p75ECD−Fcが、Fc群と比較すると、学習試行の2日目、4日目及び5日目の間に回避待ち時間を(図13 A)並びに学習試行の2日目、3日目及び5日目の間に移動距離を(図13 B及びC)、著しく低減することが示された。野生型及びp75ECD−Fc群の学習プロセスは改善された。1日目に対する3日目、4日目及び5日目以内の回避待ち及び移動距離の著しい低減は、野生型群における学習プロセス中に見られた。p75ECD−Fc群において、回避待ち及び移動距離は、学習プロセス中に、1日目に対して4日目及び5日目以内に減少した一方で、学習の著しい改善は、学習プロセス中にFc群では見られなかった。しかしながら、p75ECD−Fcは、標的四分円における平均時間(図13 D)、追跡試行セッションにおけるプラットフォーム四分円における進入の数(図13 E)を変化させなかった。追跡試行におけるプラットフォームをまたぐ横断の数は、群間で不変であった(図13 F)。結果は、p75ECD−Fcが、PR5マウスにおいて学習のプロセスを改善されることができることを示す。
[00168]PR5マウスのADの特徴に対するp75ECD−Fcの効果を評価するために、p75ECD−Fc群及びFc処置群の両方由来の脳溶解物をウェスタンブロットに付して、ADバイオマーカータンパク質であるBACE1、sAPPβ及び完全長APP(APP.FL)を測定するための種々の一次抗体とともにインキュベートした。ウェスタンブロットデータにおけるPR5マウス脳におけるBACE1及びsAPPβの発現レベルは、p75ECD−Fcが、BACE1表現を著しく低減することを実証した(p<0.05)(図14 A及びB)。さらに、sAPPβのレベルが、PR5マウスのFc群と比較して、p75ECD−Fc処置群において減少することが観察された(図14 A及びC)。
[00169]p75ECD−Fcが、PR5マウスにおいてタウリン酸化を阻害するかどうかを試験するために、p75ECD−Fc又はFcのいずれかで処置したPR5マウス由来の脳溶解物を、ウェスタンブロット分析に付して、マウスリン−PHF−タウpSer202+Thr205モノクローナル(AT8)抗体を使用してタウリン酸化タンパク質を検出した。本発明者等のデータにより、p75ECD−Fcは、PR5マウスにおいてタウ(Ser202及びThr205)リン酸化を著しく低減することが示された(p<0.01)(図15 A及びB)。

- 治療有効量のp75細胞外ドメイン(p75ECD)又はその機能的断片、変異体、類似体若しくは誘導体をそれを必要とする対象に投与するステップを含む、脳アミロイド血管症(CAA)又はタウオパチーを処置及び/又は予防する方法。
- 前記p75ECD又はその機能的断片、変異体、類似体若しくは誘導体が、p75ECDである、請求項1に記載の方法。
- 前記p75ECDがp75ECD−Fc融合物である、請求項1又は2に記載の方法。
- 前記p75ECD−Fc融合物がp75ECD−Fc融合物アデノ随伴ウイルス(AAV)ベクターとして投与される、請求項3に記載の方法。
- 前記p75ECD−Fc融合物AAVベクターが、対象体重1kg当たり1×106〜1×1013個のウイルスゲノムの用量で投与される、請求項4に記載の方法。
- 前記p75ECD−Fc融合物AAVベクターが、対象体重1kg当たり1×108〜1×1013個のウイルスゲノムの用量で投与される、請求項4に記載の方法。
- 前記p75ECD−Fc融合物が組換えペプチドである、請求項3に記載の方法。

(b)前記被験対象の生物学的試料中のp75細胞外ドメイン(p75ECD)及び/又は完全長p75(p75FL)の濃度を測定するステップと、
(c)前記被験対象の生物学的試料中のp75ECD及び/若しくはp75FLの濃度、並びに/又はp75ECD:p75FLの比率を、認知障害のない対照対象又は認知障害のない対照対象群から得られた同等の生物学的試料中のp75ECD及び/若しくはp75FLの濃度、並びに/又はp75ECD:p75FLの比率と比較するステップと

前記被験対象におけるp75ECD及び/若しくはp75FLの濃度、並びに/又はp75ECD:p75FLの比率が、前記認知障害のない対照対象又は認知障害のない対照対象群におけるp75ECD及び/若しくはp75FLの濃度、並びに/又はp75ECD:p75FLの比率に比べて変更されている場合、アルツハイマー病(AD)と診断及び/又は予後予測される、被験対象においてアルツハイマー病を診断及び/又は予後予測する方法。 - (a)前記被験対象から生物学的試料を得るステップと、

前記被験対象におけるp75FLの濃度が、前記認知障害のない対照対象又は認知障害のない対照対象群に比べて増加している場合、アルツハイマー病と診断及び/又は予後予測される、請求項12に記載の方法。 - 前記被験対象におけるp75ECDの濃度が、前記認知障害のない対照対象又は認知障害のない対照対象群におけるp75ECDの濃度に比べて少なくとも50%減少している場合、ADと診断及び/又は予後予測される、請求項13に記載の方法。
- 前記被験対象におけるp75ECDの濃度が500pg/ml未満である場合、ADと診断及び/又は予後予測される、請求項13又は16に記載の方法。
- 前記被験対象におけるp75ECD:p75FLの比率が、前記認知障害のない対照対象又は認知障害のない対照対象群におけるp75ECD:p75FLの比率に比べて少なくとも50%減少している場合、ADと診断及び/又は予後予測される、請求項14に記載の方法。
- 前記被験対象におけるp75ECD:p75FLの比率が、前記認知障害のない対照対象又は認知障害のない対照対象群におけるp75ECD:p75FLの比率に比べて少なくとも75%減少している場合、ADと診断及び/又は予後予測される、請求項14又は18に記載の方法。
- 前記被験対象におけるp75FLの濃度が、前記認知障害のない対照対象又は認知障害のない対照対象群に比べて少なくとも50%増加している場合、アルツハイマー病と診断及び/又は予後予測される、請求項15に記載の方法。
