概要:2型自然リンパ球細胞(ILC2)は、適応免疫反応を増強し、かつ組織微環境における炎症性「アラルミン」インターロイキン33(IL−33)に対する第1のレス

GFP+腫瘍細胞を探求していた。フローサイトメトリー分析は、[A9+ベクター対照]腫瘍を持つ動物の肝臓(約32%)及び副腎(約16%)において最高の割合のGFP陽性細胞を検出した。IL−33によるA9細胞のトランスフェクションは、肝臓では約0.15%、及び副腎では約2.63%までGFP+細胞の平均数を減少させた(図10)。単一の緑色細胞は、肺及び血液中で検出されたが、脳試料はすべて、GFPを含まなかった。循環腫瘍細胞は、遠位部位に播種するようにプログラム化されていない、局所的増殖の可能性を有する腫瘍(TC1)を持つ動物の試験された組織及び臓器のすべてにおいて検出されなかった。この見解は、原発性腫瘍形態と転移性腫瘍形態との間で明らかな形質学的分化を示した、切除された腫瘍の病理組織学的試験によって支持された。転移性細胞の核は、さらに多形性であった。悪性細胞は、より小さい細胞質含量、及びより大きな核/細胞質の比率、密度の高い、均一に分散された固体構造の形成を有した(図11a)。原発性腫瘍は、隣接組織(図11bにある預託組織)の中に分布しており、スポンジに似た「柔らかい」構造を形成し、それらを悪性腫瘍細胞増殖と明らかに区別した。さらに、原発性腫瘍は、線維芽細胞及びコラーゲン沈着からなる、明確な線維性被膜によって封入された(図11c)。この被膜の存在は、原発性腫瘍が出現部位で局在化した状態を可能にし、原発性腫瘍と悪性腫瘍との間で形質学的及び生物学的分化の基本的な基準を定義する。対照的に、悪性腫瘍は、通常、被膜を有せず、そのため、遠位臓器に播種することが可能である。それ故に、免疫選択によるMHC−I欠失表現型を自然発生的に獲得したマウス肺腫瘍モデル(A9)は、移植時に、インビボでその転移性特性を保持することができ、MHC−I欠失表現型を、疾患の浸潤的拡散を有する悪性の可能性と相関させることを可能にした。さらに、IL−33によって誘導された変化は、腫瘍の発達の臨床経過を変化させることができた。まとめると、これらの観察は、IL−33遺伝子相補が、悪性遺伝子発現のプログラミングを修正することができることを示唆している。結果として、腫瘍成長速度、疾患の転移拡散、及びその重症度を低下させることによって癌細胞集団の転移の可能性に影響を及ぼす。
好中球、好酸球等の多数の多形核白血球の浸潤を刺激するサイトカイン及びケモカイン等の炎症性メディエータを生成する。脂肪組織の急性炎症は、原発性[TC1+ベクター]及び遺伝的に補完された転移性腫瘍における好中球浸潤を特徴とした。したがって、炎症細胞及びメディエータは、原発性[TC1+ベクター]及び[A9+IL−33]腫瘍の微環境において上昇するが、転移性腫瘍[A9+ベクター]においては上昇せず、これは、プロスタグランジン及びロイコトリエンファミリー等の炎症関連遺伝子、ならびにインターロイキン(IL)関連遺伝子の著しい下方制御を示す、我々のマイクロアレイデータと一致している。転移性組織における炎症反応の重要な誘引物質の欠如は、好酸球のためにギムザ染色を用いて実証された。好酸球の蓄積の2つの焦点領域は、悪性腫瘍の周辺に隣接した正常な組織内に認められた。IL−33の発現によってもたらされた因子の放出は、微環境を大幅に修正し、好酸球が組織を流れるのを可能にした。IL−33が修正された腫瘍の部分を通してランダムに分布した好酸球の著しい浸潤があった(図19)。これらの観察は、系におけるIL−33の存在が、腫瘍微環境内で炎症反応を変化させ得、自然及び適応レベルのマウス肺癌における保護的な抗腫瘍免疫を媒介し得ることを示唆している。

IL−33の発現は、悪性癌において減少する:転移性遺伝子特徴のネットワークに対処するために、比較マイクロアレイ分析は、Two Colour Agilent Microarray Technologyを用いて、マウス肺及び前立腺癌の先行の非転移性及び転移性の細胞株において行われた。転移性/非転移性細胞株における遺伝子の異常発現は、腫瘍における免疫認識から免疫回避への移行の機構を特徴付け、理解するために手段を提供する。選択された遺伝子の翻訳の可能性を検証するために、主要なヒト組織データが、考慮された。遺伝子候補が、ヒト前立腺癌の転移性形態で減少することが確認された。具体的には、European Biostatistics Institute(アクセス日:2013年1月30日)によって作成されたGene Expression Atlasから得られたデータを用いて、対象となる遺伝子が、良性及び原発性前立腺腫瘍と比較して、転移性前立腺癌において下方制御されることが見出された。データの遺伝子発現レベル及び/またはタンパク質レベルの間で一致しており、それは、転移に対して良好な予測子であり得ることを示す。

現行の免疫療法アプローチは、主に、免疫抑制を抑制及び/または免疫活性化を促進する戦略に基づいている。異なるアプローチの組み合わせは、永続的な臨床反応をもたらす、異なるステップの癌免疫サイクルに影響を及ぼし得る。腫瘍特異的な免疫反応(抗原依存性因子)の活性化と共に、抑制腫瘍微環境(抗原非依存性因子)の改変を組み合わせることを目的として、インビトロで活性化したILC2細胞の養子移入を実施した。

細胞株
