Karhanekらによって2010年3月25日に公開された「Functionalized Nanopipette Biosensor」と題する米国特許出願公開第2010/0072080号は、先端開口部を通過するときに検出されるタンパク質生体分子などの検体を含有し得る電解質溶液を保持するのに適した、ナノスケール寸法の円錐先端開口部を有する中空不活性、非生物学的構造体として例示されるナノピペットを含む、生体分子検出のための方法及び装置を開示している。
Pourmandらによって、2012年9月6日に公開された「Nanopore Device for Reversible Ion and Molecule Sensing or Migration」と題する米国特許出願公開第2012/0222958号は、電気化学的検出回路に使用するのに適したナノピペットを利用して、イオン移動及び結合を検出するための方法及び装置を開示している。キトサンは、最初にナノピペットに付着したPAA(ポリアクリル酸)層上で使用され、銅などのイオンの結合を測定するために使用される。
Actis et al. in “Functionalized nanopipettes: toward label−free, single cell biosensors,” Bioanalytical Reviews 1:177−185 (2010)は、DNA及びタンパク質を同定することができる無標識バイオセンサーとしてのナノピペットを開示している。

本発明は、外因性物質を必要とせずに、単一細胞内のpH及び細胞内のpHの変化を測定することができる手段及び装置を提供する。測定はリアルタイムで行われ、ナノピペットが細胞内に挿入されている間にpHの変化を追跡することができ、感受性回路は、例えば、細胞質、核、ミトコンドリアなどの細胞内のナノピペットのナノポア開口部でイオン電流を測定する。検出回路は、0.09pH単位の検出を示す記載された実施例で、約0.1〜0.01pH単位の高感度を提供する。さらに、システムは、約pH2〜11の間の大きなダイナミックレンジを有する。
試薬及び材料。キトサン(低分子量)、5−ニトロ−2−(3−フェニルプロピルアミノ)−安息香酸(NPPB)、二塩基性リン酸ナトリウム及び一塩基性ナトリウムをSigma Aldrichから購入した。塩化ナトリウム(ACSグレード)、塩酸、水酸化ナトリウム及び過酸化水素は、Fisher Scientificから入手した。氷酢酸は、Riedel−de−Haenから供給された。2−プロパノールはSpectrum Chemicalsから入手した。2’、7’−ビス−(2−カルボキシエチル)−5−(及び−6)−カルボキシフルオレセインアセトキシメチルエステル(BCECF−AM)をInvitrogenから購入した。ジメチルスルホキシド(無水)はFlukaから供給された。イーグル最小必須培地(MEM)、ダルベッコ改変イーグル培地(DMEM)及びトリプシンは、CellGroから購入し、胎児ウシ血清(FBS)及びペニシリン−ストレプトマイシンはGibcoから購入した。すべての水溶液は、18.2Ωcmの抵抗率を有する脱イオン水(Millipore、Synthesis System)中で調製される。
実施例 1:pH応答性石英ナノピペットセンサーの特性
ナノピペットの測定原理は、先端のイオン電流に基づいている。このイオン電流は、ナノピペットの細孔径及び表面電荷に大きく依存する。石英ナノピペットの表面電荷は、ガラス−液体界面でのシラノール基の解離により負である。石英は極めて酸性のpH値でプロトン化を受ける。石英のこれらの表面特性は、pHセンシング能を低下させるため、未修飾のナノペットでは非常に小さなpH変化を測定するには適切ではない。未修飾の石英表面の低い感度に関連する制限は、pH応答性ポリマー実体をナノピペット表面に取り込むことによって克服することができる。ここでは、pH感受性表面コーティングとしてキトサンを使用した。酸性pHで強い正電荷を有するキトサンは、静電相互作用によって負に帯電した石英表面上のヒドロキシル部分に引き付けられる。表面電荷の変化に加えて、キトサン層の厚さは、ナノピペットの感度を高め得るpHによって変化することが示されている。ナノピペット表面上のキトサン層の存在及び影響を評価するために、本発明者らは、表面修飾の結果としての電流応答の変化をモニタした。図1Aは、−0.5〜0.5Vの電位範囲(対Ag/AgCl参照電極)において10mMのPBS(pH7.0)で充填された未修飾の及びキトサン修飾された石英ナノペプチドの電気化学的トレースを示す。記録された電流応答は、キトサン修飾後に有意に減少する。ナノピペットチップの典型的な幾何学的形状は円錐形であり(図2A)、石英ナノピペットの孔径はSEMによって決定され、約97nmであることが判明した(図1B)。さらにSEM顕微鏡写真を撮影し、キトサン層の存在をさらに確認した(図2B)。キトサン修飾はナノピペットの内部で行われたため、集束イオンビームを使用してナノピペットを垂直にエッチングし、内部表面を露出させた。断面画像は、未修飾のナノピペットのものと比較した場合の、ナノピペット表面の内部のキトサン残基を示す(図1C及びD)。
固体ナノポアpHプローブを開発する我々のモチベーションは、単一細胞レベルで細胞内pHを測定し、それらの特徴的な代謝特性を有する癌細胞を同定することである。細胞内pH測定を行うために、キトサン修飾ナノピペットを細胞培養培地、MEM及びDMEMでさらにキャリブレーションした。細胞培地には様々なアミノ酸、ビタミン、その他の成分が含まれているため、最適な作業パラメーターはPBSに対して決定されたパラメーターとは異なっていた。走査された電位範囲は−0.2〜0.6Vであり、走査速度は0.1V/秒であった。pH変化に対するキトサン修飾ナノピペットの感度は、0.6Vで最も高かった。図6A〜6Bは、1X MEM及びDMEM溶液中のナノpHプローブのキャリブレーションを示す。培地中のナノpHプローブのキャリブレーションは、0.1M HClの20μlの連続添加により実施した。酸溶液を細胞培養培地に添加してから15秒後に測定を行い、均一溶液を得た。代表的なリニアスイープボルタモグラムを、MEM及びDMEM培地の酸滴定について図7A〜7Bに示す。これらの培地の成分が異なるので、それらの緩衝能はわずかに異なり、DMEMはMEMに比べてpH変化に対してより耐性がある。
細胞内pHの直接測定は、細胞のサイズが小さく、生理学的マトリックスの複雑さのために困難である。生理学的pHレベルはわずかにアルカリ性であるが、大きな集団及び細胞内区画の個々の細胞の細胞内pHレベルは不明である。従来、蛍光色素(例えば、BCECF−AM、オレゴングリーン)は、細胞内のpHの間接的な検出に利用されている。これらのpH指示薬は、大きな細胞集団にわたってpHの近似値を示すが、蛍光色素を使用することにはいくつかの欠点がある。i)狭いpH範囲による低感度、ii)高速光退色、iii)細胞毒性。さらに、特定の細胞小器官におけるこれらの色素の蓄積及びそれらの漏出の速度は、誤った解釈を導く可能性がある。MDA−MB−231細胞の細胞内pHを測定するための、従来のpH指示薬である、BCECF−AMを使用した我々の研究は、細胞内事象の正確かつ高感度な評価のために蛍光を使用することによる欠点を証明した。蛍光測定の細胞内pH測定を実施したこれらの研究では、細胞をBCECF−AMに曝露し、15分間インキュベートした。次いで、細胞を洗浄し、10分間、細胞内pHキャリブレーション緩衝液(pH7.5、6.5、5.5及び4.5)を含有するニゲリシンに曝露した。BCECF−AMは二重励起波長を有する。したがって、画像は458及び488nmで撮影した。各pH値の2つの励起波長について、明視野及び蛍光顕微鏡写真を得た。16〜23個の個々の細胞の蛍光強度を使用して、レシオメトリックキャリブレーション曲線を得た(データは図示せず)。1つの細胞群は、細胞内自己蛍光の存在を評価する陰性対照(BCECF−AMなし)として役立った。pH色素が存在しない場合、MDA−MB−231について観察可能な蛍光は存在しなかった。BCECF−AMに曝露された細胞を使用して、個々の細胞の細胞内pH値を推定した。10個の個々の細胞から得られた平均細胞内pH値は、6.78(±0.83)と算出された。しかし、BCECF−AM曝露後に撮影した顕微鏡写真は、細胞体上の蛍光強度が変化することを明らかにした(データは図示せず)。蛍光強度は細胞が厚くなるほど高かった。さらに、個々の細胞において互いに近接している任意の2つの領域は、pH値の大きな変動を有することが見出された。これらの変動は、(i)蛍光色素の不均一な分布または蓄積に起因する可能性があるか、(ii)蛍光色素の他の分子との交差反応性に起因する可能性がある。蛍光測定のもう1つの欠点は、培地に頻繁な変化を必要とする試料調製工程であり、これは細胞にストレスを与え、基底細胞内レベルを変化させる可能性がある。さらに、蛍光色素の使用では、目的の化合物と共にこれらの色素が存在すると、細胞の生理機能を変化させることによって、または試験すべき化合物と交差反応させることによって、誤った実験の結論が生じる可能性があるため、薬物試験、または毒性測定などの治療過程にわたる単一細胞の連続的な調査をすることはできない。言い換えれば、従来の蛍光プローブでは、治療剤、チャネル活性化剤、または毒素の細胞への影響を評価するための時間の経過にわたる単一の細胞の連続的な調査を行うことはできない。
本発明のナノpHプローブは、薬物治療中の細胞内pH変化をモニタするために使用することができる。この目的のために、周知の塩化物チャネル遮断薬、5−ニトロ−2−(3−フェニルプロピルアミノ)−安息香酸(NPPB)の添加の間、単一の細胞での連続モニタリングのために、このナノpHプローブを配置した。NPPBは、腎上皮細胞及びマクロファージ細胞における塩化物チャネルを遮断することが以前に示されており、結果として細胞内環境の酸性度が上昇する。従来、pHの変化は、蛍光色素(BCECF−AM)の導入によって間接的に測定されてきた。したがって、この医薬操作試験は、ナノpHプローブのリアルタイム測定の能力を実証するだけでなく、pH検出に対する特異性を実証することにも役立つ。ベースラインを得るために、ナノpHプローブをMDA−MB−231細胞に挿入し、連続pH測定を7分間21秒ごとに行った。MDA−MB−231細胞におけるこのリアルタイムpHモニタリングは、測定中に値7付近で最小のドリフトを示した(図12、菱形)。NPPBの効果を研究するために、ナノpHプローブをMDA−MB−231細胞に挿入し、NPPB(無水DMSOで新たに調製)の100μMを細胞培地に添加する直前に細胞内pH記録を開始した。図12の正方形は、7分間のNPPB曝露の結果としてのpH変化を示す。細胞内pHレベルは、NPPBの導入後、最初の2分以内に有意に低下し、2.5まで低下した。測定されたpHレベルは、NPPB導入の4分後に安定化した。このpHの上昇は、アポトーシスにより、細胞体の収縮をもたらす可能性があり、ナノpHプローブのチップを細胞培地に曝露することになる。ナノpHプローブを使用した3つの個々のMDA−MB−231細胞の細胞内pH測定は、NPPB曝露後のリアルタイムpH変化を示しただけでなく、薬物応答に関して細胞間の変動も示した(図13)。
上記の装置は、細胞内の成分の酸化または還元に応答するナノピペット上のキトサン層に付着した層でさらに修飾することができる。
上記の装置は、ナノpHプローブの多重アレイでさらに構成することができる。さらに、アレイで使用される様々なナノピペットの内部に、多数の表面認識材料を追加することができる。ナノピペット構造数は様々であってもよく、それらのすべてがキトサンpHセンシングコーティングを含むわけではない。

Claims (28)
- (a)i)支持体上の細胞を貫通するためのマイクロマニピュレータ及び検出装置に動作可能に接続可能であり、(ii)その中に作用電極を含み、(iii)水素イオンを選択的に吸収するポリマーコーティングを含有する、ナノピペット構造と、
(b)溶液中の前記作用電極と参照電極との間に異なる電圧を印加するように構成され、異なる電圧下で前記作用電極と前記参照電極との間のイオン電流を測定するようにさらに構成された増幅回路にさらに接続される前記ナノピペット構造と、
(c)前記増幅回路によって測定された異なるイオン電流を前記ナノピペット構造の外側の細胞内のpH値と相関させるための論理手段とを含む、単一細胞内のpHを測定する装置。 - 前記マイクロマニピュレータ及び検出装置が、SICM(走査型イオン伝導顕微鏡)及び単一細胞の中へ移動するためのナノピペットを制御するxyzコントローラを含む、請求項1に記載の装置。
- 前記増幅回路が、利得制御及びイオン電流を検出するためのローパスフィルタを有する検出回路を含む、請求項1または2に記載の装置。
- 単一の論理手段に接続されたナノピペット構造のアレイを含む、請求項1または2に記載の装置。
- 前記キトサンが約30,000〜60,000単位のモノマー数を有する、請求項4に記載の装置。
- 前記キトサンが、モノマーに結合したヘムタンパク質を含む、請求項5に記載の装置。
- 前記ポリマーコーティングが、スルホン化テトラフルオロエチレンコポリマー(Nafion(登録商標))、ポリ‐L‐リジン、及びアルギン酸塩からなる群から選択される、請求項1に記載の装置。
- 前記増幅回路が、前記参照電極に接続され、前記作用電極からの入力を有する増幅器からの入力に応答するポテンショスタットを含む、請求項1に記載の装置。
- 前記ポテンショスタットが、前記ポテンショスタットの参照電極にも接続されている対極に接続されている、請求項8に記載の装置。
- 前記作用電極及び前記対極が、Ag/AgClである、請求項8に記載の装置。
- (a)種々の電位におけるイオン電流対電位を測定する回路に電気的に接続され、単一細胞内にナノピペットを挿入するための挿入装置に取り付けられた前記ナノピペットと、
(b)整流値を周知のpH値と相関させるための論理手段であって、細胞内で取得された整流値を周知の整流値と相関させることができ、それによって測定されたpH値を同定する出力を提供する、論理手段と、
(c)前記ナノピペットの表面に直接結合し、水素イオンに対して多孔性であるキトサン材料の層を有する前記ナノピペットと、
(d)補助電極としても機能し、ポテンショスタットに接続された参照電極を含む回路とを含む、単一細胞内のpHを測定するための装置。 - 前記論理手段が、補助電極における前記電流を測定することによって、前記参照電極に対して所与の電位範囲での前記作用電極の前記電位を走査するようにプログラムされる、請求項11に記載の装置。
- フィルタ選択器及び感度選択回路によってブリッジされたi/V増幅器であって、前記構成要素が、前記電解液を通過する前記電流に基づいて前記検出可能な電流範囲を調整するように調整される、請求項11に記載の装置。
- (a)(i)支持体上の細胞を貫通するためのマイクロマニピュレータ及び検出装置に動作可能に接続可能であり、(ii)その中に作用電極を含み、(iii)水素イオンを選択的に吸収するポリマーコーティングを含有する、ナノピペット構造を調製することと
(b)溶液中の前記作用電極と参照電極との間に異なる電圧を印加するように構成され、異なる電圧下で前記作用電極と前記参照電極との間のイオン電流を測定するようにさらに構成された増幅回路に前記ナノピペット構造を接続することと
(c)前記増幅回路によって測定された異なるイオン電流を、前記ナノピペット構造の外側の細胞内のpH値と相関させるための論理手段に前記ナノピペット構造を接続することとを含む、単一細胞内のpHを測定するための装置を作る方法。 - 前記ポリマーコーティングが、キトサン材料層を前記ナノピペットに結合させることによって適用され、前記ナノピペットを通るイオン電流のI−V曲線を導き、測定する増幅器に前記作用電極を接続することをさらに含む、請求項14に記載の方法。
- (a)pHイオンに応答する内部層を有し、前記ナノピペット構造を通るイオン電流と、前記ナノピペット構造及び参照電極を含む電気化学セル内の様々な電位における電位とを測定するように構成されたポテンショスタットを含む回路に作用電極によって電気的に接続される、ナノピペット構造を提供することと、
(b)前記ナノピペット構造を前記電気化学セル内の生細胞に挿入することと、
(c)前記イオン電流を測定するために前記回路を使用することであって、前記電流が周知のpHに相関することと、を含む、細胞内のpHを測定する方法。 - 前記ナノピペットを挿入することが、SICM及びx−y−zコントローラを使用することを含む、請求項16に記載の方法。
- 前記回路が、利得制御及びイオン電流を検出するためのローパスフィルタを有する検出回路を含む増幅回路をさらに含む、請求項16または17に記載の方法。
- 前記内部層が、50nm〜150nmの直径の平均細孔径を有するキトサン材料の層を含む方法を含む、請求項16または17に記載の方法。
- 前記キトサンが約30,000〜60,000単位のモノマー数を有する、請求項19に記載の方法。
- 前記キトサンが、モノマーに結合したヘムタンパク質を含む、請求項20に記載の方法。
- 前記内部層が、スルホン化テトラフルオロエチレンコポリマー(Nafion(登録商標))、ポリ‐L‐リジン、及びアルギン酸塩からなる前記群から選択されるポリマーコーティングを含む、請求項16に記載の方法。
- 前記回路が、前記参照電極に接続され、前記作用電極からの入力を有する増幅器からの入力に応答するポテンショスタットを含む、請求項16に記載の方法。
- 前記ポテンショスタットが、前記参照電極に接続された対極に接続されている、請求項23に記載の方法。
- 前記作用電極及び前記対極が、Ag/AgClである、請求項23に記載の方法。
- 前記電圧が、0.5V〜0.7Vである、請求項23に記載の方法。
- 様々な電圧が、前記ポテンショスタット上に設定される、請求項26に記載の方法。
- 前記pH値が、癌性細胞で取得され、非癌性細胞のpHと比較される、請求項23に記載の方法。
