d)HCV複製のレベルを検出する工程を含み、
HCV複製の検出が、上記被験体がHCVに感染していることを指し示す、方法を提供する。
a).第1容器内の培地中のhADSCを候補化合物の非存在下でHCVと接触させる工程;
b).第1容器内の培地中のHCVのレベルを候補化合物の非存在下で決定する工程;
c).第2容器内の培地中のhADSCを候補化合物の存在下でHCVと接触させる工程;

別段の定義がない限り、本明細書中で用いられる科学技術用語および命名法は、本発明が関係する当業者によって一般に理解されるのと同じ意味を有する。概して、細胞培養、感染、分子生物学方法などのための手順は、当該技術分野において用いられる一般的な方法である。そのような標準的技法は、参考マニュアル、例えばSambrookら(1989、Molecular Cloning−A Laboratory Manual、Cold Spring Harbor Laboratories)およびAusubelら(1994、Current Protocols in Molecular Biology、Wiley、New York)などに見出すことができる。
本発明は、hADSCがHCVによる感染に対して許容性であるという発見に関する。
ヒト脂肪由来幹細胞は、中胚葉由来の多能性成体幹細胞であり、簡単に大量に得ることができる9。これらの細胞は、表皮成長因子様ファミリー21のメンバーであり脂肪生成に必須である特異的マーカーDLK−1(すなわち、Pref−1)を発現し、この発現は成熟脂肪細胞において完全に消滅する22〜24。ヒト脂肪生成DLK−1+細胞(hADSC)は複数の細胞系統に分化できるということが、相次ぐ証拠によって実証されており(総説については参考文献25、26参照)、hADSCは再生療法の考案に有望なツールとなっている。様々な解剖学的区画にある間葉系幹細胞がウイルスの感染に対して感受性であることも報告されている27〜32。しかし、ウイルス性疾患におけるhADSCの役割はまだ探究されていない。
一般に、hADSCは、当該技術分野において利用可能な周知の培地中で維持しかつ増殖させることができる。このような培地としては、限定はしないが、ケラチノサイトSFM(K培地)、ダルベッコ改変イーグル培地(登録商標)(DMEM)、DMEM F12培地(登録商標)、イーグル最少必須培地(登録商標)、F−12K培地(登録商標)、イスコフ改変ダルベッコ培地(登録商標)RPMI−1640培地(登録商標)、間葉系幹細胞基礎培地(ATCC(登録商標)PCS−500−030(商標))および間葉系幹細胞成長キット−低血清(ATCC(登録商標)PCS−500−040(商標))が挙げられる。
本発明によるHCVは、hADSCに感染することのできる任意のHCV、またはHCV感染個体から分離されることのできる任意のHCVとすることができる。一実施形態において、HCVは、遺伝子型1a、1b、2a、2b、2c、2d、3a、3b、3c、3d、3e、3f、4a、4b、4c、4d、4e、4f、4g、4h、4i、4j、5aおよび6aまたはそれらの任意の組み合わせからなる群から選択されるHCV遺伝子型の少なくとも1つである。別の実施形態において、HCVは、遺伝子型1a、1b、2a、2bおよび混合型2a+2bからなる群から選択されるHCV遺伝子型の少なくとも1つである。
関連する態様において、本発明はまた、HCVの増殖、またはHCV生活環分析の実施、またはHCV感染の診断、または抗ウイルス化合物の選抜、またはHCVに感染した被験体のHCVの特性評価のための、本明細書に記載のhADSCおよびhADSCの培養に適した培地を含むキットを提供する。
(臨床試料)
全ての臨床脂肪組織および肝臓試料は、Kaohsiung医科大学病院から施設研究委員会の承認を得て入手した(KMUH−IRB−960477、KMUH−IRB−960343およびKMUH−IRB−20120404)。手続きに先立ち、全てのドナーから書面によるインフォームドコンセントを得た。
HCV(+)脂肪組織は、肝細胞癌腫を切除するための手術創(開腹術)から得た。HCV(−)脂肪組織については、先に記載されているとおりに13、乳癌の乳房切除直後の乳房再建術を受けた女性の横軸型腹直筋皮弁から得た。脂肪吸引を受けている肥満者からもHCV(−)脂肪組織を得た(表1)。手術前に乳癌の管理のために補助化学療法または放射線療法を受けた患者は一人もいなかった。
HCV感染またはHCV未感染の個体から調製したRBC溶解した未分画SVF細胞をポリクローナルウサギ抗DLK1抗体(Abcam、米国)と共に4℃で30分間インキュベートした。記載されているとおりに10細胞を、0.8mmol/LのMgCl2、20mmol/LのHEPES、100U/mLのペニシリンおよび100μg/mLのストレプトマイシンを含有するHBSS中で2回洗浄し、磁気マイクロビーズに結合したヤギ抗ウサギIgG(Miltenyi Biotec Inc、Auburn、CA)と共に4℃で30分間インキュベートした。細胞懸濁液を洗浄し、MidiMACSセパレータ(Miltenyi Biotec)内のカラムに通し、その結果、DLK−1−細胞が通過し、DLK−1+細胞がカラムに保持された。両方の細胞画分をHBSS中で2回洗浄した。細胞生存率は、DLK−1+画分およびDLK−1−画分についてそれぞれ>96%および>97%であった。RT−PCRまたはqRT−PCRのためにRNAを抽出した。別個の実験において、細胞を免疫細胞化学のためにサイトスピンスライドに固定した(5〜7×103細胞/スライド)。試験管内感染の実験では、HCV(+)血清(HCVser)への曝露後、DLK−1+細胞を、表示の期間、培養および継代した(記載されているとおりに13継代培養した)。その後、RNAをウイルス5’−UTR転写物のRT−PCRまたはqRT−PCRのために抽出した。
この研究では、2つのプロトコルを採用した。
QIAamp(登録商標)ウイルスRNAミニキット(Qiagen、Basle、スイス)を使用してRNAを140μlのHCV(+)血清またはHCVser感染hADSC培養物の上清から抽出した。PureLink(登録商標)RNAミニキット(Ambion、Carlsbad、CA、米国)を製造業者の指示に従って使用して、細胞溶解物由来のRNAを単離した。その後、高容量cDNA逆転写キットを使用してRNAを一本鎖cDNAに変換し、続いてGoTaq Master Mix(Promega、WI、米国)を使用してPCRを行った。ウイルス複製物を定量するために、Applied Biosystems(登録商標)ViiA(商標)7リアルタイムPCRシステムを使用して、C型肝炎ウイルスアドバンスドキット(PrimerDesign Ltd.、英国)でPCRを行った。HCV特異的な逆方向プライマーおよび増幅プライマーは、ABIプライマー3.0 Express Soft Wordに従って設計した。記載されているとおりに35、プライマー5’−ACTCGCAAGCACCCTATCAG−3’を逆転写に使用し、PCRおよびリアルタイムPCRに使用するプライマーを、別のHCV遺伝子型の高度に保存された5’−非翻訳領域(UTR)に適合させた。
新鮮な脂肪組織を手術創から採集し、ホルマリンで固定し、パラフィンに包埋した。組織を5μmの切片に切断し、脱ろうし、その後、クエン酸緩衝液(10mMのクエン酸、pH6.0)に浸し、抗原回復のためにマイクロ波で加熱した。室温で30分間、5%のBSAでブロッキングした後、ポリクローナルウサギ抗DLK1抗体(1:150;カタログ番号ab21682、Abcam、米国)またはウサギIgG Ab(1:150、カタログ番号AB−105−C、R&D)を4℃で一晩適用し、続いてアルカリフォスファターゼ結合抗ウサギIgG二次抗体(1:500;Jason ImmunoResearch)を室温で1時間適用し、その後、fast red substrateシステム(シグマアルドリッチ)で発色させた。連続的なNS5染色のために、試料をPBSに10分間浸漬してカバーガラスを取り外し、スライドを0.3%のH2O2中で30分間、室温でインキュベートして、内因性ペルオキシダーゼに由来する非特異的バックグラウンドを減少させた。室温で30分間、5%BSAでブロッキングした後、マウス抗NS5抗体(1:200、クローンBGN/1246/5G7、カタログ番号0200−0423、AbD Serotec)またはマウスIgG1アイソタイプAb(1:100、カタログ番号14−4714、eBioscience)を4℃で一晩添加した。本発明者らの経験では、既に試料をパラフィンに包埋していたため、脂肪組織に対するNS5染色には細胞浸透手順が不要であった。その後、切片を西洋ワサビペルオキシダーゼポリマーQuanto試薬(抗マウス、既製、Thermo Scientific)と共に室温で7分間インキュベートし、UltraVision Quanto Detection System(発色用DAB基質含有;Thermo Scientific)で発色させた。その後、切片をヘマトキシリンで染色し、カバーガラスを元どおりに配置し、それによって細胞輪郭がわずかに変化した(図1Eに示すとおり)。TissueFAXS走査ソフトウェア(TissueGnostics)を使用して高品質顕微鏡(Zeiss)、高速コンピュータハードウェアおよび高解像度スクリーンによってDLK1およびNS5Aの染色の電子画像をキャプチャした。続いて、画像内の同じ領域を選択し、比較および共局在化の分析のために視覚化した。
未分画SVF細胞またはDLK−1+hADSCまたはDLK−1−細胞の免疫細胞化学のために、IHCの場合と同様の指針に従って、最適な発色時間をどの対照実験においても上記のとおりに予め決定した。表示の時点において細胞を回収し、サイトスピンによりポリリジン被覆ガラススライドに接着させ、続いて4%のホルマリンで20分間固定した。DLK−1染色のために、細胞を37℃で30分間、0.05%のトリプシン溶液によって抗原回復に供し、その後、DDWによって3回すすいだ。Ultra V Block緩衝液(UltraVision Quanto Detection Systemの中の試薬、Thermo、米国)で5分間ブロッキングした後、細胞を4℃で一晩ウサギ抗DLK1抗体またはウサギIgGと共にインキュベートし、続いてアルカリホスファターゼ結合抗ウサギIgG二次抗体と共に室温で1時間インキュベートし、fast red substrateシステム(Sigma−Aldrich)で5〜6分間(ほとんどの場合)発色させた。(図2Cに示すように同じスライド上で染色された)HCV特異的NS5の単染色または逐次染色のために、スライドをPBS中に10分間置いてカバーガラスを取り外し、封入剤を洗い流し、0.3%のTritox−100と1%のBSAとを含有するPBSで細胞を30分間浸透させブロッキングし、その後、UltraVision Quanto Detection System(Thermo Scientific、Fremont、CA、米国)を適用した。次いで、細胞をHydrogen Peroxide Block(Abcam、MA、米国)中で10分間インキュベートして、内因性ペルオキシダーゼに起因する非特異的なバックグラウンド染色を減少させた。洗浄後、細胞をUltra V Block(Thermo Scientific、MA、米国)と共に5分間インキュベートして非特異的なバックグラウンド染色をブロッキングし、マウスモノクローナル抗NS5抗体を含有する希釈緩衝液と共に4℃で一晩インキュベートした。マウスIgG1による染色を陰性対照として用いた。翌日、細胞をPrimary Antibody Amplifier Quanto溶液と共に10分間インキュベートし、さらに10分間、HRP Polymer Quantoと共にインキュベートし、各試薬適用の間にPBSで洗浄した。最後に、30μlのDAB Quanto Chromogenを1mlのDAB Quanto Substrateに添加し、旋回混合し、発色のために細胞に(ほとんどの場合)2〜3分間適用した。洗浄後、スライドに永久封入剤を載せ、カバーガラスで覆い、TissueFAXS顕微鏡(ZEISS)を使用して視覚化し、撮影した。
TEM研究のために、hADSCを回収し、記載されているとおりに36調製し、透過型電子顕微鏡(JEM2000 EXII;JEOL、東京、日本)で調べた。
PureLink(登録商標)RNAミニキット(Ambion、Carlsbad、CA、米国)を製造者の指示に従って使用してhADSCのRNAを単離した。高容量cDNA逆転写キット(Applied Biosystems)を使用してRNAを一本鎖cDNAに変換した。逆転写に使用した特異的プライマーは5’−ATGTACCCCATGAGGTCGGC−3’であった。PCRに使用したプライマーは、別のHCV遺伝子型のコアタンパク質領域に適合させた。第1回目のPCRには、以下の設定での熱プロファイルを用いて、(順方向)5’−CGCGCGACTAGGAAGACTTC−3’および(逆方向)5’−CGCGCGACGCGTAAAACTTC−3’を含有するプライマー混合物を使用した:94℃で2分間、続いて94℃で45秒間と55℃で45秒間と72℃で90秒間とを35サイクル、次いで最終伸長のために72℃で7分間。2回目のPCRにおける遺伝子型同定に使用した型特異的なアンチセンスプライマーは、5’−CCAAGAGGGACGGGAACCTC−3’(2a型)および5’−ACCCTCGTTTCCGTACAGAG−3’(2b型)であり、熱プロファイルは以下の設定とした:95℃で2分間、続いて95℃で30秒間と60℃で30秒間と72℃で30秒間とを30サイクル、次いで最終伸長のために72℃で7分間37、38。
懸濁液中の様々な継代の0.5〜1×105個のhADSCを4℃で1時間、マウス抗ヒトCD81モノクローナルAb(クローンJS−81、BD Biosciences)、抗LDL−R Ab(クローンC7、Millipore)、抗EGFR Ab(クローンLA1、Millipore)またはウサギポリクローナル抗SRB1 Ab(Novus Biologicals)で染色した。それぞれの対照は、マウスIgG1またはポリクローナルウサギIgGであった。洗浄後、細胞をさらにフルオレセインイソチオシアネート(FITC)結合二次Ab(Jackson ImmunoResearch Laboratories、PA、米国)と共にインキュベートし、Cell Quanta(商標)SC高解像度フローサイトメトリー(Beckman Coulter Fullerton、CA、米国)により分析した。細胞表面分子をブロッキングするために、(ウェル内に接着した)2x105個のhADSCを37℃で、表示用量(1〜100μg/ml)の抗体を含有する1mlの無血清K培地で前処理した。それぞれのアイソタイプ抗体による処理を対照として用いた。1時間後、未希釈のHCV(+)血清をエッペンドルフチューブ内に加えて、抗体存在下での3時間インキュベーションのためにMOIを0.2にした。その後、細胞を洗浄し、連続培養のために6cmのペトリ皿内に接種培養した。ApoE遮断のために様々な濃度の抗ApoE抗体(クローンE6D10)を、hADSCと共に3時間インキュベートする前に、記載のとおりに14HCV(+)血清に室温で1時間添加した。21日間の連続培養後に上清および細胞を回収し、RNAを抽出してウイルス5’−UTR転写物を定量した。別個の実験では、ウェル内のhADSCを表示用量のIFNα(Sigma−Aldrich、MO、米国)で16時間、K培地中で3回前処理し、その後、遺伝子型1a、1b、2aおよび2bのHCV(+)血清に曝露した。21日後、細胞溶解物中の5’−UTR転写物をqRT−PCRによって定量し、ビヒクル(PBS)対照で処理した細胞と比較した画分阻害として結果を計算した。
miR−122のRT−PCRのためのプライマーを調製し、記載されているとおりに39実施した。細胞からの全RNAをRNA抽出試薬REzol(商標)C&T(Protech、台北、台湾)で単離した。miR−122レベルを決定するために本発明者らは、TaqMan MicroRNA Reverse Transcriptionasキット(Applied Biosystems)を使用して抽出RNAを逆転写し、miR−122のTaqMan MicroRNA AssayによるリアルタイムPCR解析のテンプレートとしてcDNAを使用した。
オクルディンおよびクローディン−1に特異的なsiRNAを、記載されているとおりに15、40、41Dharmaconによって合成した。それらの各々の標的配列は、UAACAUUAGGACCUUAGAA(クローディン−1)およびGUGAAGAGUACAUGGCUGC(オクルディン)である。NPC1L1を記載のとおりに16調製し、DGAT−1のsiRNAを記載のとおりに19調製した。Xfectトランスフェクション試薬(Clontech)を使用して、特異的なsiRNAを6ウェル細胞プレートのウェル内のhADSCにトランスフェクトした。HCV感染は、siRNAトランスフェクト細胞をHCVserと共に37℃で3時間インキュベートし、その後HCVserをPBSで洗い流すことによって行った。記載されているとおりに15、19、40、41、トランスフェクション後48時間目の細胞をRT−PCRのために溶解して遺伝子サイレンシングの度合いを決定した。21日間の培養後にHCvser感染hADSCの細胞溶解物および上清を5’−UTRのqRT−PCRのために採集した。
Huh7.5細胞を、10%の熱不活性化ウシ胎児血清(Invitrogen)と0.1mMの非必須アミノ酸(Invitrogen)とを含有するDMEM(Invitrogen)中で培養した。試験管内ゲノムJFH−1 RNAを転写し、先に記載されているとおりに42電気穿孔によって細胞に送達した。その後、トランスフェクトした細胞を完全なDMEMに移し、3〜4日毎に継代した。通常の実践では、完全長JFH1 cDNAでトランスフェクトした細胞からの馴化培地を、10分間の遠心分離(3,000×g)によって浄化し、使用前に滅菌濾過(0.2μm酢酸セルロース、Millipore)した。より長期間の保存のために、HCVccを等分し、−80℃で保存した。4℃で一晩のインキュベーションのためにウイルスを、1/4体積の滅菌濾過済40%(w/v)ポリエチレングリコール−8000のPBS液の添加によって濃縮した。ウイルス沈殿物を遠心分離(8,000×g、15分)によって回収し、記載されているとおりに43PBS中に再懸濁した。
抗ウイルス薬であるリバビリン、シクロスポリンAおよびIFNαは全てSigma−Aldrichから入手した。テラプレビルは、米国マサチューセッツ州のSelleck Chemicalsから得た。段階用量のリバビリン、テラプレビルまたはシクロスポリンAを0日目にペトリ皿内のHCVser感染hADSCの培地に添加し、21日間培養した。IFNα処理に関しては、hADSCを16時間、表示用量のIFNαで前処理し、その後、HCVserと共にインキュベートした。その後、細胞溶解物のウイルス5’−UTR転写物を、ビヒクル対照で処理した細胞と比較した画分阻害として定量および計算した。ビヒクル対照は、リバビリンおよびIFNαの場合はPBS、シクロスポリンAおよびテラプレビルの場合は0.1%のDMSOとした。
HCVccおよびHCVadscの培地を、先に記載されているとおりにPEG−8000で濃縮した。いずれの試料も500μlの無血清培地に再懸濁させ、先に記載されているとおりに44、10mMのHepes(pH7.55)と150mMのNaClと0.02%のBSAとを含有する溶液を使用して調製した10%〜40%のイオジキサノール(それぞれ0.5ml)の連続イオジキサノール(OptiPrep、Axis−Shield、ノルウェー)密度勾配の上に積層した。勾配をSW−41ローター(Beckman Coulter)で6時間、4℃で40,000rpmで超遠心分離した。超遠心分離後、上部の勾配から17個の画分を回収した(各画分は500μl含んでいた)。最後に、QIAamp(登録商標)ウイルスRNAミニキット(QIAGEN、Basle、スイス)を使用して各画分から全RNAを単離した。RNAを、定量的RT−PCRによるHCV RNA検出のために使用した。
Quantikine(登録商標)ELISAヒトアポリポタンパク質B/ApoBイムノアッセイキットおよびQuantikine(登録商標)ELISAヒトアポリポタンパク質E/ApoEイムノアッセイキット(R&D Systems)を使用して、ApoBおよびApoEの発現を製造者の説明に従って検出した。HDLおよびLDL/VLDLのコレステロールを、HDLおよびLDL/VLDLコレステロールアッセイキット(Abcam)によって検出した。様々な浮遊密度の画分中のHDLおよびLDL/VLDLのApoB、ApoEおよびコレステロールの発現レベルを複製物に対して規格化した。
HCV関連またはHCV非関連の肝細胞癌腫のために外科的切除された肝臓検体から新鮮な非腫瘍性肝組織を採取し、PHHを単離し、記載されているとおりに45培養した。PHHを3日間接種培養して、コラーゲンI被覆6ウェルプレート上に細胞を付着させた。4日目に細胞を穏やかに洗浄し、HCVserまたはそれに対応するd21のhADSC増殖HCV(+)上清(1×104個のHCV5’−UTR複製物を含有する)と混合されたアルギニン不含WilliamsE培地(Invitrogen、CA、米国)中で終体積を0.5mlとして3時間培養した。感染後、細胞を洗浄し、1mlの培地中で感染後d5まで毎日新鮮な培地を交換しながらさらに培養した。その後、5’−UTRのRT−PCRのためのRNA抽出のために細胞を溶解した。
臨床上、HCV感染の興味深い特徴は、HCV(+)患者が慢性的に感染した肝臓内に過剰な脂肪蓄積、すなわち脂肪肝を有し得46、47、脂肪肝の重症度が肝線維症の割合と相関するようである48、という点である。また、最近の研究により、HCVのRNA複製は、飽和脂肪酸の利用可能性を高めることによって刺激され、多価不飽和脂肪酸または脂肪酸合成の阻害剤によって阻害される、ということが示されている49、50。これらの知見は、脂肪代謝がHCVの生活環において重要な役割を果たすことを示唆している。したがって本発明者らは、脂肪組織の細胞成分が生体内でのHCV感染に関与している可能性があるという仮説を立てた。
ナイーブHCV(−)hADSCが試験管内でのHCVser感染および複製に対して感受性であるか否かを調べるために、本発明者らはHCV(−)個体由来のhADSCを調製し、それらを培養下で継代した。(エッペンドルフチューブ内の)懸濁液中の第3継代(p−3)またはp−4の細胞を、最終体積を1mlとして0.2moi(すなわち、5×105個のhADSC細胞に対して1×105の5’−UTR複製数)のHCVser(表2)と共にインキュベートした。
hADSC産生ビリオン(「HCVadsc」と表示)の感染性を調べるために本発明者らは、「ドナー1」のp2のhADSCをHCVser−1bに感染させ、21日目に上清(「HCVadsc(1)」と表示)を回収した。0.22μmの細孔フィルタで濾過した後、HCVadsc(1)を使用して「ドナー2」のhADSCに感染させてHCVadsc(2)を作り、続いてそれを使用して「ドナー3」のhADSCに感染させた。結果は、HCVadscが、HCVserによる最初の感染に見られるように比較的一貫した複製効率で、様々なドナーのナイーブhADSCに対して感染性を有することを裏付けた(図3A)。最初にHCVser−1a、HCVser−2aおよびHCVser−2bに感染させたhADSCの上清に由来するHCVadscについても,同様の観察がなされた(データ示さず)。
HCVadscの物理的特性を評価するために本発明者らは、HCVser、HCVccおよびHCVadscの浮遊密度プロファイルを、記載されているとおりに43平衡遠心分離によって比較した。研究したいずれのウイルスも、遺伝子型2aに由来するものであった。以前の報告43、59、60と一致して、HCVserはより低い密度1.039(画分2)および1.080(画分7)の画分に大量のRNAを有していたが、HCVccの場合は1.132にピークがあった(画分13;図4A)。HCVadscでのRNAの最高量は密度1.080(画分7)で認められ、続いてより高い密度1.124および1.156(画分12および15)に2つのピークが認められた。興味深いことに、HCVadscの1.080のピークはHCVserのピークと同一であった(図4A)。したがって、HCVadscの物理的性質はHCVccよりもむしろ臨床分離物に類似している。

Claims (18)
- C型肝炎ウイルス(HCV)を増殖させるためのヒト脂肪由来幹細胞(hADSC)に基づくシステムであって、hADSCと、hADSCの培養に適した培地と、HCVとを含むシステム。
- 前記hADSCが、初代細胞または継代細胞、好ましくは第1〜15継代細胞、より好ましくは第1〜6継代細胞である、請求項1に記載のシステム。
- 前記HCVが、HCVに感染した個体の血液、血清、血漿もしくは体液に由来するか、または臨床HCV単離物である、請求項1または2に記載のシステム。
- 前記HCVが、遺伝子型1a、1b、2a、2b、2c、2d、3a、3b、3c、3d、3e、3f、4a、4b、4c、4d、4e、4f、4g、4h、4i、4j、5aおよび6aまたはそれらの任意の組み合わせ、好ましくは遺伝子型1a、1b、2a、2bまたは混合型2a+2bである、請求項1〜3のいずれか1項に記載のシステム。
- 前記システムがHCVの完全な複製を支援する、請求項1〜4のいずれか1項に記載のシステム。
- C型肝炎ウイルス(HCV)を増殖させる方法であって、HCVの複製に適した条件下でhADSCの培養に適した培地中でHCVを増殖させるためにhADSCを使用することを含む、方法。
- 前記hADSCが、初代細胞または継代細胞、好ましくは第1〜15継代細胞、より好ましくは第1〜6継代細胞である、請求項6に記載の方法。
- 前記HCVが、HCVに感染した個体の血液、血清、血漿もしくは体液に由来するか、または臨床HCV単離物である、請求項6または7に記載の方法。
- 前記HCVが、遺伝子型1a、1b、2a、2b、2c、2d、3a、3b、3c、3d、3e、3f、4a、4b、4c、4d、4e、4f、4g、4h、4i、4j、5aおよび6a、またはそれらの任意の組み合わせ、好ましくは遺伝子型1a、1b、2a、2bまたは混合型2a+2bである、請求項6〜8のいずれか1項に記載の方法。
- 前記方法がHCVの完全な複製を支援する、請求項6〜9のいずれか1項に記載の方法。
- HCVの増殖、またはHCV生活環分析の実施、またはHCV感染の診断、または抗ウイルス化合物の選抜、またはそのようなHCVに感染した被験体のHCVの特性評価のための、hADSCの使用。
- HCV感染の診断、またはHCVの増殖、またはHCV生活環分析の実施、または抗ウイルス化合物の選抜、またはそのようなHCVに感染した被験体のHCVの特性評価のための、キットの製造におけるhADSCの使用。
- 被験体のHCV感染を診断する方法であって、
a)hADSCを提供する工程、
b)hADSCの培養に適した培地中で、前記被験体から得た生体試料と共にhADSCをインキュベートする工程、
c)前記hADSCを、HCV複製を可能にするのに十分な時間培養する工程、および
d)HCV複製のレベルを検出する工程を含み、
HCV複製の検出が、前記被験体がHCVに感染していることを指し示す、方法。 - 前記生体試料が、血液、血清、血漿または体液に由来するものである、請求項13に記載の方法。
- 抗HCV化合物を選抜する方法であって、
a)第1容器内の培地中のhADSCを候補化合物の非存在下でHCVと接触させる工程;
b)前記第1容器内の前記培地中のHCVのレベルを前記候補化合物の非存在下で決定する工程;
c)第2容器内の培地中のhADSCを候補化合物の存在下でHCVと接触させる工程;
d)前記第2容器内の前記培地中のHCVのレベルを前記候補化合物の存在下で決定する工程;
e)前記候補化合物の存在下でのHCVのレベルを、前記候補化合物の非存在下でのHCVのレベルと比較する工程;および
f)前記候補化合物の存在下でのHCVレベルが前記候補化合物の非存在下でのHCVレベルよりも低い場合に前記候補化合物を抗HCV化合物として同定する工程を含む、方法。 - HCVのレベルが、HCV力価、HCV核酸のレベル、またはHCVポリペプチドのレベルを測定することによって決定される、請求項15に記載の方法。
- 前記候補化合物が、化学化合物、タンパク質、ペプチド、ペプチド模倣物、抗体、核酸、アンチセンス核酸、shRNA、リボザイムおよび小分子化学化合物からなる群から選択される少なくとも1つである、請求項15または16に記載の方法。
- 前記HCVが、遺伝子型1a、1b、2a、2b、2c、2d、3a、3b、3c、3d、3e、3f、4a、4b、4c、4d、4e、4f、4g、4h、4i、4j、5aおよび6aまたはそれらの任意の組み合わせからなる群、好ましくは遺伝子型1a、1b、2a、2bおよび混合型2a+2bからなる群から選択されるHCV遺伝子型の少なくとも1つである、請求項15〜17のいずれか1項に記載の方法。
