Abstract
Description
1)癌にかかっている若しくは癌になるリスクにある患者から、サンプルを得ること;
2)そのサンプルを、NME7ファミリーのメンバーのレベル若しくはNME7ファミリーのメンバーをコードする核酸のレベルを検出若しくは測定することができる分析にかけること;
3)コントロール患者若しくはコントロール細胞でのレベルに対して、試験サンプルでのNME7ファミリーの測定されたメンバー、あるいはNME7ファミリーメンバーをコードする核酸のレベルを比較すること;
4)NME7ファミリーのメンバー、あるいはNME7ファミリーのメンバーをコードする核酸のレベルが、コントロールに比べ上昇していることを決定すること; そして
5)もし、該テストが比較されたコントロールが、以前、癌と診断されたドナー由来ならば、該試験サンプルのドナーは、癌にかかっているか若しくは癌の進行を持っていると結論を下すこと。この方法では、循環系、あるいは組織でのNME7ファミリーのメンバーの検出は、患者での癌の指標でありうる。NME7ファミリーのメンバーは、NME7、NME7b、NME7-X1、あるいはNME7-AB様タンパク質でありうる。
患者において、NME7ファミリーのメンバー若しくはMUC 1 *の存在の検出すること;そして
該NME7ファミリーのメンバー若しくはMUC1*発現を示す患者に、抗NME7若しくは抗MUCl *抗体、あるいは複数抗体を投与すること。該NME7ファミリーメンバーは、NME7、NME7b、NME7-X1あるいはNME7-AB様タンパク質でありうる。
1)癌を持っている疑いがある、又は発癌の危険がある、又は転移性癌になる危険がある、患者からサンプルを得ること;
2)NME7ファミリーのメンバー若しくはNME7ファミリーのメンバーをコードする核酸の量を測定することであって、そこでは測定されたレベルはコントロールサンプルで測定されたものに対し優位に高いこと;
3)該患者が、癌を保持する(発癌しているともいう)又はより攻撃的若しくは転移性癌を進展したと決定すること;
4)該患者に、NME7ファミリーのメンバーの発現を抑える、NME7の開裂を阻害する、あるいはその標的へのNME7結合を阻害する、有効な量の治療薬を投与すること。該NME7ファミリーのメンバーの標的は、MUC1*でありうる。該NME7ファミリーのメンバーの標的はMUC1*細胞外ドメインのPSMGFR部分でありうる。該NME7ファミリーのメンバーは、NME7、NME7b、NME7-X1、あるいはNME7-AB様タンパク質でありうる。

2i及びよりナイーブな状態へ幹細胞を変える他の試薬と同様にNME7-AB、NME7-X1は、さらにより転移性の状態に癌細胞を形質転換することを引き起こす。任意のもの、あるいはこれらの試薬のコンビネーションでの処置の後、癌細胞はより高い生着率を持っており、腫瘍を試験動物に生じさせるのに100,000倍までより少ない細胞を要求する。したがって、本願で開示された方法は、患者の試験動物の中への原発性腫瘍細胞の異種移植を可能にするために使用することができる。
癌増殖あるいはより転移性の状態への移行を阻害するこれらの抗体は、抗癌治療薬として使用のために選ばれ、癌の治療若しくは予防のために患者に投与されうる。選択された抗体は、例えば、ヒトキメラ抗体あるいは完全ヒト化抗体を設計若しくは作ることによりさらに最適化されうる。このアプローチの効果を実証するために、本発明者らは、その癌性機能に重大であると推定されたNME7の複数領域から複数NME7ペプチドを選んだ。その後、本発明者らは、これらのペプチドを使用して、抗体を生成し、次に、次のことへのそれらの能力に関し、免疫化ペプチド及び抗体をテストした: a) 癌増殖を阻害すること; そして、b) 癌細胞から転移性癌細胞へ誘導移行を阻害すること。NME7ペプチドは、抗体産生のための免疫化剤、及び阻害剤自体として選択された(図19及び例11)。ペプチドA1 (配列番号: 141)、A2(配列番号: 142)、Bl(配列番号: 143)、B2(配列番号: 144)及びB3(配列番号: 145)、そこでAは、ペプチドが由来するドメインを指す、つまり、NDPK Aドメインをさし及びBはそのペプチドがNDPK Bドメインに由来することをさす(図15)。各々のペプチドは、免疫原として使用され、多クローン性抗体の産生のために2羽のウサギ各々に注入された。免疫化ウサギ血液から回収された抗体は、免疫化ペプチドで調製されたカラムで精製された。その後、精製された抗体は、ヒトNME7への結合能に関してテストされた。得られた抗体はすべて、所望のように、ヒトNMElではなくヒトNME7に結合する(図26 A-B、実施例12)。これらの結果は、その配列がNME7で見つかるが、NMElにおいては正確に同一でないペプチドを選ぶことによって、NMElではなくNME7に特異的に結合する抗体が生成されることを示す。NMElは健康な機能を有するので、NMElに影響をしない抗体を生成することは、ほとんどの場合、好適である。該抗体は、また、MUC1*細胞外ドメインペプチドへのNME7の結合を阻害するそれらの能力に関してテストされた。図27に示されるELISA実験は、該抗体が、それらがNMElの結合を阻害したよりはるかに多くのMUC1*細胞外ドメインペプチドへのNME7-ABの結合を阻害したことを示す。

実験のこのシリーズでは、本発明者らは、幹細胞と癌細胞中のNME6及びNME7の発現をプローブした。さらに、本発明者らはMUC1*をNME7の標的であると確認した。NMEl、NME6及びNME7の存在・不存在を決定するために、本発明者らは、細胞溶解物で、最初にウェスタンブロット分析を行なった。図1Aで、抗MUCl*抗体でコートされた表面上においてNME1二量体中で培養された(レーン1)又はMEFs上においてbFGF中で(レーン2)若しくはT47Dヒト乳癌細胞溶解物で(レーン3)若しくは陽性コントロールとしてのNME1−wtで培養された、BGOlvヒト胚性幹細胞からの溶解産物が、SDS-PAGEによって分離され、その後、抗NMEl特異抗体でプローブされた。結果は、NMEl二量体あるいはbFGF中で培養されたかどうかで、ヒトES細胞及びT47D癌細胞で、NMElが強く発現されることを示す。同じ細胞溶解物は、SDS-PAGEによって分離され、次に、抗NME6特異抗体(Abnovaからの抗NME6)でプローブされた。NME6は検出されなかった(データは示されない)が、それは、その後、もっと濃縮されたサンプルで検出された(図2参照)。

- 該ペプチドは、以下の表(SEQ ID NOS:141〜145)にリストしたペプチドの1若しくはそれ以上に、少なくとも80%相同である、請求項1に記載の剤。
- 該ペプチドは、以下の表(SEQ ID NOS: 88〜145)にリストされたものから選ばれる、請求項1に記載の剤。
- 該ペプチドは、以下の表(SEQ ID NOS: 141〜145)にリストされたものから選ばれる、請求項6に記載の剤。
- 該ペプチドは、以下の表(SEQ ID NOS: 88〜145)にリストされたペプチドに、高度に相同である、あるいはN末端若しくはC末端で7つまでのアミノ酸残基が付加若しくは欠失されたものである、請求項1に記載の剤。
- 該ペプチドは、スペーサー若しくはリンカーによって別のペプチドに結合される、請求項1に記載の剤。
