Re=LVavgρ/μ
[式中、Lは関連長さ、μは流体粘度、ρは流体密度、およびVavgは流れの平均速度である]として定義されるレイノルズ数(Re)を特徴とすることができるが、これらの流れ特性は、反応チャンバー中で変動し、反応チャンバー内の流れは、1種または複数種の磁界などの外部源により操作することができる。チャネルの小さい寸法のために、Reは通常、100より遥かに小さく、多くの場合1.0未満である。このレイノルズ数の枠組みでは、流れは完全な層流であり、乱流は発生しない。乱流への移行は通常、レイノルズ数2,000の範囲で発生する。
蛍光分子またはビオチン分子または磁気微小粒子に結合した抗体を使ったCHO分泌細胞の標識に基づいて、多量の組換え型治療薬を分泌する哺乳動物細胞の迅速で効率的なキャプチャーを可能とする方法の開発が、本発明を例示するために本明細書で詳細に記載される。
該方法の開発を容易にし、細胞選別の能力を評価するために、治療用タンパク質、すなわち、免疫グロブリン、ならびに、抗体を分泌する細胞をより容易に追跡するための蛍光レポータータンパク質の両方を発現する第1のレポーター細胞を設計した。治療用免疫グロブリンガンマ(IgG)および抗生物質選択マーカーのための発現ベクター、ならびに蛍光タンパク質、「高感度緑色蛍光タンパク質」または「高感度青色蛍光タンパク質2」(EGFPまたはeBFP2)コードプラスミドをCHO細胞に同時遺伝子導入した。種々のレベルの免疫グロブリンを安定に発現しているポリクローナル集団をBFPおよび表面IgGディスプレイに基づいてFACSにより選別し、その後、IgG産生をELISAにより評価した(図1)。同時に、GFPとIgGまたはBFPとIgGを発現しているモノクローナルCHO細胞集団(例えば、細胞クローン)を限界希釈法により選択した。IgG分泌をELISAアッセイにより評価した。種々のレベルの表面IgGを発現しているが、低/中レベルのIgG産生クローンを、参照細胞集団として選択した。
−CHO−M懸濁細胞(IgGなし、GFPなし)
−F206−IgG+、GFP+:高IgGディスプレイヤーおよび高産生細胞、望ましいクローン。
−BS2−IgG+、BFP+:中IgGディスプレイヤー、中IgG産生細胞、望ましくないクローン。
−BLC−IgG+、BFP+:中IgGディスプレイヤー、中IgG産生細胞、望ましくないクローン。
−BHB−IgG+、BFP+:極高IgGディスプレイヤー、中IgG産生細胞、望ましくないクローン。
IgGを発現しないか、または種々の既知のレベルのIgGを発現している細胞集団と、多量のトラスツズマブ治療用IgGおよび同時発現GFPを分泌するF206クローン由来の所定の数の細胞とを混合した。この細胞を、ヒトIgGの定常部分に結合するビオチン標識二次抗体とインキュベートし、その後、ストレプトアビジンに結合した磁気微小粒子(Dynabeads MyOne T1(登録商標)、Invitrogen(登録商標)、#65601)とインキュベートした(図3)。それぞれの洗浄後、細胞の試料を保持し(回収1〜3と表される)、細胞培地中に入れ、選択せずに細胞を10日間増殖させた。その後、発現細胞から非発現細胞を識別するために、表面IgGディスプレイに関し細胞を評価した。図4に示すように、続けて洗浄する毎に、陰性細胞の割合が減り、3回目の洗浄後、ほとんど100%の陽性細胞が回収された。
マニュアルキャプチャープロセスが確立されると、MagPhase(登録商標)装置でそれを実行し、治療用ヒトIgGを発現しているCHO−M(Selexis(登録商標))細胞をキャプチャーすることが試みられた。
これらの調査の過程で、MagPhase(登録商標)による適切な取り扱いの観点、ならびにCHO細胞との特異的および非特異的相互作用の観点から、様々な種類の市販マイクロビーズおよびビーズ比率を試験し、最良の結果を与えると思われる条件を特定した。これには次のものを含めた。
強磁性マイクロビーズ:
−Chemicell(登録商標))FluidMAG(直径5.0μm)
−Chemicell(登録商標)SiMAG(直径1.0μmまたは2.0μm)
超常磁性マイクロビーズ:
−Dynabeads(登録商標)M280 2.8μm
−Dynabeads(登録商標)MyOne T1 1.0μm
−Ademtech(登録商標) 300nm
Dynabeads(登録商標)M−280およびMyOne T1:両方とも、MagPhase(登録商標)の種々の操作モード中、強磁性ビーズの存在下で操作することができた。しかし、より良好な空間配分が得られるという理由で、MyOne T1ビーズを選択した。M−280と比較して、それらのより弱い磁化は、プロセスの終わりでの強磁性ビーズからの分離および回収を容易にした。それらの1.0μmのサイズにより、2.8μmのマイクロビーズよりも、CHO細胞と結合するための特異的相互作用を可能とすることも明らかになった。
Chemicell(登録商標)FluidMAG 5.0μm:これらは、Chemicell(登録商標)SiMAGより磁気的に弱いが、一定の範囲の周波数および磁力、例えば、100〜200Hzおよび200〜300mA中で、効率的な混合を可能とした。下記で記載するように、これらの強磁性ビーズに対し、最適混合条件は150Hzおよび200mAであると定めることができた。下記の節で示すように、このような条件下で、混合または細胞キャプチャーモードにおいて、それらはチャンバー壁のまわりを回転し、均一で迅速な超常磁性ビーズの空間配分を可能とした。しかし、Chemicell FluidMAGは、このビーズのシリカ表面へ非特異的結合をする非発現CHO細胞との結合を減らすために、デンプン層でコートする必要があった。
高発現細胞を単離するために、MagPhase(登録商標)チャンバー中で強磁性および超常磁性粒子の両方を混合するというこの革新的手法のためのプロセスは、5つのステップで表すことができる。
最適化MagPhase(登録商標)細胞キャプチャー手順を使って、我々は、非発現細胞(CHO−M細胞)ならびに中、高、または極高IgGディスプレイヤー(すなわち、それぞれ、BS2、BLCおよびBHB細胞、図2参照)からの高産生細胞(すなわち、F206細胞)濃縮能を分析した。
7.1 MagPhase無菌キャプチャーおよびキャプチャーされた細胞/ビーズの分離タイミング最適化
MagPhase(登録商標)は非発現または中発現細胞から抗体高発現細胞を濃縮できることが確立されたので、我々は最初に、キャプチャーが無菌環境中で行い得るかどうかを試験した。この目的のために、層流フード中で、16mLの8%Javal溶液(Reactol(登録商標)lab、#99412)、16mLの10%Contrad90溶液(Socochim(登録商標)、#Decon90)および32mlの無菌Milli−Q水で洗浄することにより、元のMagPhase(登録商標)装置の内部液体処理マイクロ流体チャネルを最初に滅菌した。後の段階で、最適化プロセスおよび使い捨てカートリッジ設計が開発された際に、キャプチャーを行う前に、カートリッジをガンマ線照射(24Kグレイ)により滅菌した。
MagPhase(登録商標)を使った無菌キャプチャー後に、投入細胞をCB5フィードで処理した場合、1日目に回収された細胞は、MagPhase(登録商標)無菌キャプチャーに供さなかった細胞に比べて、F206細胞の5.6倍濃縮であった(図20A)。この濃縮は、類似の投入細胞混合物のMagPhase(登録商標)非無菌キャプチャーで得られた結果(図17A)と一致した。しかし、投入F206およびCHO−M細胞混合物を、MagPhase(登録商標)選別の前に、5%のCB5補充剤の存在下で前培養した場合、1日目に分離された細胞は、F206細胞の2倍濃縮に過ぎなかった(図20B)。ビーズから解離した細胞の回収前に、残存する細胞およびビーズ混合物をさらに3日目まで培養した場合、類似の結果が得られた。これは、細胞選別ステップの前にフィードが加えられた場合、フィードが細胞キャプチャーを妨害したことを示唆している。
上記MagPhase(登録商標)装置および方法により参照モノクローナル細胞の混合物をIgG発現細胞の濃縮効率に関し試験した。次に、我々は、MagPhase(登録商標)が、広範囲にわたる種々の多くの発現レベルのポリクローナル集団から高IgG分泌細胞の濃縮をも可能とするかどうかについて、決定することを必要とした。この目的のために、無菌MagPhase(登録商標)キャプチャーを行って、治療用トラスツズマブ抗体を安定に発現しているポリクローナル集団から高IgG分泌細胞をキャプチャーした。
血清由来ポリクローナル二次抗体の使用は、医薬品環境には適切ではないので、我々は、MagPhase(登録商標)キャプチャープロセスにおける、ビオチン化抗IgGモノクローナル抗体(mAb)の実現性についてさらに調査した。図23Aに示すように、2つの異なるモノクローナル抗体をMagPhase(登録商標)細胞キャプチャープロセスで試験することができた。CB5なしでキャプチャーされた細胞を培養した場合、Mabtech(登録商標)モノクローナル抗体の使用により、中および高IgGディスプレイヤーの両方を、対照細胞に比べて、1日目に2倍に濃縮した(図23A)。Mabtech(登録商標)mAbを使ってキャプチャーした細胞をCB5含有培地中で培養した場合、細胞中の中および高ディスプレイヤーの、それぞれ、1.4倍および1.6倍の増加が1日目に得られた(図24B)。細胞キャプチャー中にAcris mAbを使って、中および高ディスプレイヤー細胞のより低い濃縮が得られた。それに対応して、キャプチャーされた細胞のIgG比生産性は、Mabtech(登録商標)mAbを使った場合により高く、CB5フィードの存在下で細胞を溶出した場合、対照細胞より2倍高い生産性が得られた(図24C)。まとめると、これらのアッセイは、モノクローナル抗体を、MagPhase(登録商標)ベース無菌キャプチャーおよびポリクローナル集団から高分泌細胞の濃縮のために使用することができることを示す。
既知のバージョンのMagPhase(登録商標)選別プロセスは、汚染除去溶液を圧送することによるMagPhase(登録商標)の滅菌を必要とした。これらの既知のプロセスでは、マイクロ流体チャネルはいずれも使い捨てではなく、したがって、ボア汚染リスクがあり、薬理学的用途のための細胞選別に適合しないものにしている。本明細書で提示されるのは、特に、生細胞の無菌選別専用の新世代のMagPhase(登録商標)装置およびカートリッジであり、全ての液体および細胞取り扱い手順を、単回使用無菌カートリッジの閉ざされた一定の環境内で処理可能とする。

Claims (34)

- 入口チャネルを介して前記反応チャンバーの内容物を前記回収容器中に流すように、前記回収容器の空気ベント孔が、前記回収容器のベント孔を通して空気を圧送することにより前記反応チャンバー内の磁気標識細胞を回収するためのポンプに連結されている、請求項25に記載のシステム。
- 前記反応チャンバーの容積が10μl〜500μlである、請求項1〜26のいずれか1項に記載のカートリッジまたはシステム。
- 前記カートリッジが自己完結型かつ使い捨て型である、請求項1〜27のいずれか1項に記載のカートリッジまたはシステム。
- 1つの容器に、請求項19〜28のいずれか1項に記載のカートリッジを含み、キャプチャービーズおよびキャリアビーズが前記反応チャンバー中に含まれるか、またはさらなる容器で提供され、かつ別の容器に、前記カートリッジ中の前記キャプチャービーズおよびキャリアビーズの使用説明書を含むキット。
- 前記キャプチャービーズが超常磁性ビーズであり、前記キャリアビーズが強磁性ビーズであり、前記超常磁性ビーズの強磁性ビーズに対する比が、2:1〜50:1である、請求項29に記載のキット。
- 請求項1〜30のいずれか1項により識別され、好ましくは選択される細胞。
- 好ましくは、導入遺伝子発現産物を分泌する組換え型細胞を、20、40、60、80pcdを超えるレベルで含む単離細胞集団であって、単離細胞集団が単離された元の細胞集団を40%を超えて含まない、単離細胞集団。
- 前記分泌導入遺伝子が治療用タンパク質である、組換え型細胞の単離集団。
- 前記官能化磁気ビーズおよび任意で前記キャリアビーズと前記細胞の混合と、表面上に前記タンパク質をディスプレイしている細胞の前記識別および好ましくは選択との間の時間間隔が、1時間未満、30分未満、20分未満、15分未満または10分未満である、請求項1〜33のいずれか1項に記載の方法。
