Abstract
Description
本発明の目的は、高効率でロバストな、経済的かつスケーラブルな組換えヒトG−CSFの精製のための下流プロセスを提供することである。
i.大腸菌の高細胞密度発酵及び封入体の単離によるrHu−GCSFの生成ステップと、
ii.G−CSFを放出するための、ステップ(i)から得られた封入体の可溶化ステップと、
iii.ステップ(ii)から得られたG−CSFタンパク質のリフォールディングステップと、
iv.ステップ(iii)から得られたG−CSFタンパク質の清澄及び精製ステップと、
v.任意選択的に、ステップ(iv)から得られた精製されたG−CSFをPEGに結合させるステップと、
vi.任意選択的に、ステップ(v)から得られた結合(ペグ化)G−CSFを、陽イオン交換クロマトグラフィーを用いて精製するステップと、を含み、
可溶化された封入体から精製されたGCSFを得るためのプロセスの収率は50〜70%の範囲であり、SEC−HPLCによって分析されるG−CSFの純度は98%以上であり、
凝集体等の不純物の合計は2%以下である。RPHPLCによって分析した場合のG−CSFの純度は96.5%以上であり、
酸化GCSF、還元GCSF、f−met GCSFの合計は3.5%以下であり、
GCSFからPEG−GCSFを生成するプロセスの収率は60〜75%の範囲であり、SEC−HPLCによって分析した場合のPEG−GCSFの純度は95%以上である。
本発明のGCSFの精製は、発酵ブロスの可溶性画分と微粒子画分とを分離してバイオマスを収集することによるrHu−GCSFの生成によって実行されてもよい。分離は、好ましくは遠心分離、濾過によって、より好ましくは遠心分離によって行われてもよい。
細菌封入体の可溶化は、適切に最適化されなければ、さらなるプロセスステップの収率及び精製されるタンパク質の品質にまで影響を及ぼし得る重大なステップである。
タンパク質をリフォールディングするために使用されるリフォールディング緩衝液は、通常、リフォールディング溶液の伝導率を増加させるために塩酸グアニジニウム及びアルギニン等の化学物質からなる。このリフォールディング溶液の処理は、しばしば、イオン交換クロマトグラフィーステップの前に限外濾過/透析濾過ステップを有する必要性を要し、下流プロセス操作に余計な時間、複雑性、及び支出が加わる。これらの制約に留意して、本発明のリフォールディングプロセスは、いずれの余分な調整ステップも行うことなく、リフォールディング溶液をイオン交換クロマトグラフィーに直接充填することができるように開発される。
酸性化したリフォールディングG−CSFタンパク質溶液は、任意選択的に、精製のためにクロマトグラフィーに供する前に、形成され得るあらゆる沈殿物及び微粒子状物質を除去するために濾過及び/または遠心分離に供されてもよい。
精製されたrHu−GCSF(原薬)は、酸性pHの好適な緩衝液中でポリエチレングリコールに結合させてもよい。
結合させたペグ化G−CSF混合物は、平衡化緩衝液で希釈されてもよく、pHは3.5〜5.0の範囲に調整され、より好ましくは、pHは4.0〜4.5に調整されてもよい。

5%ソルビトール及び0.004%ポリソルベート80を含有する、pH4.0の10mMの酢酸ナトリウム緩衝液中に配合された本発明のプロセスから得られるG−CSFまたはフィルグラスチムを含む。
ステップ1:大腸菌の高細胞密度発酵によるrHu−GCSFの生成
好適な抗生物質を含有する前接種培地に、冷凍ストックから再生したペトリ皿の大腸菌細胞を接種する。培養物を30℃で12時間、250rpmでインキュベートする。好適な抗生物質を含有する基本培地に大腸菌を接種することによって前接種材料及びそれに続いて接種材料を調製し、30℃で12時間、250rpmでインキュベートする。600nmでの出発光学密度が約0.1になるように、調整済みの基本塩培地を含有する発酵槽に接種培養物を接種する。所望の光学密度600nmに達したら、1mMのIPTGを加えることにより培養を誘導する。誘導した流加培養中、誘導後に、誘導した大腸菌細胞を含有するブロスを、4℃及び6000rpmでの30分間の遠心分離によって回収する。回収した細胞ペレットをリン酸緩衝食塩水(PBS)で洗浄する。洗浄したペレット細胞をさらなる処理まで−80℃で保存する。
発酵から得られ、溶解緩衝液中で15倍に希釈された冷凍細胞ペレット19.0gは、100mMのトリス、20mMのEDTA、250mMのNaCl、及びpH8.0を含む。溶液を室温(RT)で30分間室撹拌し続ける。
1gの封入体を計量し、50mMのトリス、5mMのEDTA、及び8Mの尿素をpH10で含む20mlの可溶化緩衝液に可溶化した。1時間の可溶化後、40mMのシステインを加えてジスルフィド結合を還元し、pHを10.0に再調整し、連続的に撹拌しながら室温で60分間可溶化を維持した。可溶化後、11000gの遠心分離を30分間使用して、可溶化された封入体溶液を清澄した。
清澄及び可溶化された封入体溶液を、50mMのトリス、0.5Mの尿素、5%ソルビトール、及び2mMのシスチンからなる10体積のリフォールディング緩衝液中に徐々に加えることによりリフォールディングした。希釈はポンプを一定流速で使用して行われる。可溶化試料をリフォールディング緩衝液に加えた後、リフォールディングのために試料を一晩(約16時間)室温(25℃)に維持した。

リフォールディングした清澄タンパク質溶液を、20mMの酢酸ナトリウムと等価の1Mの酢酸ナトリウム(pH4.0)の原液の添加により酸性化し、最終的に2Mの塩酸によってpHを最大pH4.0までに調整して30分間維持した。

酸性化後、0.2μフィルタを用いた精密濾過により溶液を濾過して沈殿物を除去し、予め平衡化した2mlの陽イオン交換カラムに充填した。40mMの酢酸ナトリウムからなる平衡化緩衝液(pH4.0)の5CVでカラムを洗浄し、UV280からベースラインまでの吸光度を引き出した。40mMの酢酸ナトリウムを含有する洗浄緩衝液(pH5.5)でカラムをさらに洗浄し、生成物に関連する不純物を除去した。洗浄後、精製rHu−GCSFを回収し、40mMの酢酸ナトリウム、200mMの塩化ナトリウムからなる溶出緩衝液(pH5.4)を通過させることによってカラムから溶出させた。以下の分析方法を用いて、充填、フロースルー、洗浄、及び溶出を含む全てのクロマトグラフィー試料を分析する。

e)LAL試験を用いたエンドトキシンの定量
精製された標的タンパク質を含有するHIC画分を、10mMのナトリウム(pH4.0)、5%ソルビトール、0.004%ポリソルベート80からなる所定の製剤緩衝液中で緩衝液交換し、10kDaの限外濾過膜を使用して最大1mg/mlまでさらに濃縮した。最終rHu−GCSF濃縮溶液を0.2ミクロンで滅菌濾過し、次に使用するまで2〜8℃で保存した。

5%ソルビトール及び0.004%Tween 80を含有する10mMの酢酸ナトリウム緩衝液(pH4.0)中に1〜1.2mg/ml濃度で存在する精製rHu−GCSFを、pH5.0の1Mの酢酸ナトリウムの添加により約5.0のpHに調整する。最終緩衝液強度は、約50mMである。3倍量(重量ベース)のmPEG−20kDa−CHOを、ペグ化前の反応混合物中に存在するrHu−GCSFの量まで反応混合物mPEG−20kDa−PEG−アルデヒドに加える。直ちに、20mMのシアノ水素化ホウ素ナトリウム[Na(CN)BH3]を反応混合物に加える。反応混合物を室温[約25℃]で一晩撹拌する。プロセス中、モノペグ化フィルグラスチムの割合を確認するために品質分析が行われ、14時間後に概して75〜80%のモノペグ化フィルグラスチムが得られ、pH8.0の1Mのトリスで反応を停止させて、さらなる処理まで2〜8℃で保存する。

Claims (13)
- G−CSFの単離及び精製のためのプロセスであって、
i.大腸菌(E.coli)の高細胞密度発酵及び封入体の単離によるrHu−GCSFの生成ステップと、
ii.G−CSFを放出するための、ステップ(i)から得られた封入体の可溶化ステップと、
iii.ステップ(ii)から得られた前記G−CSFタンパク質のリフォールディングステップと、
iv.ステップ(iii)から得られたG−CSFタンパク質の清澄及び精製ステップと、
v.任意選択的に、ステップ(iv)から得られた精製されたG−CSFをPEGに結合させるステップと、
vi.任意選択的に、ステップ(v)から得られた前記結合(ペグ化)G−CSFを、陽イオン交換クロマトグラフィーを用いることによって精製するステップと、を含み、
前記プロセスの収率は50〜70%の範囲であり、SEC−HPLCによって分析されるG−CSFの純度は98%以上であり、
凝集体等の不純物の合計は2%以下であり、RPHPLCによって分析されるG−CSFの純度は96.5%以上であり、
酸化GCSF、還元GCSF、f−met GCSFの合計は3.5%以下であり、
GCSFからPEG−GCSFを生成するプロセスの収率は60〜75%の範囲であり、SEC−HPLCによって分析した場合のPEG−GCSFの純度は95%以上である、プロセス。 - 存在する酸化GCSFの量は1.5%未満であり、還元GCSFは1%未満であり、f−met GCSFは1.5%未満である、請求項1のステップ(vi)に記載のプロセス。
- 存在する凝集体の量は2%未満である、請求項1のステップ(vi)に記載のプロセス。
- GCSFのプロセス収率は50%以上であり、PEG−GCSFのプロセス収率は60%以上である、請求項1のステップ(vi)に記載のプロセス。
- 大腸菌の高細胞密度発酵によって生成される細胞は、溶解緩衝液によって溶解され、
前記溶解緩衝液のpHは、7.5〜8.5の範囲であり、
前記単離は、実験室規模の超音波破砕機、高圧細胞ホモジナイザー、遠心分離、及び濾過、または組み合わせ、好ましくは前記実験室規模の超音波破砕機と高圧細胞ホモジナイザーとの組み合わせを含む群から選択される計器によって行われ、
前記単離された封入体は、トリス−EDTA−トリトン緩衝液、トリス−EDTA−DOC緩衝液、トリス−NaCl−尿素緩衝液、トリス緩衝液、及びトリトンX−100、または混合物、好ましくはトリス−EDTA、尿素、NaCl、及びトリトンX−100の混合物を含む群から選択される緩衝液で洗浄され、
前記トリスは30〜50mMの範囲で存在し、EDTAは3〜7mMの範囲であり、尿素は0.1〜0.5Mの範囲であり、NaClは1〜2Mの範囲であり、トリトンX100は0.8〜1.5%の範囲であり、
前記洗浄緩衝液のpHは7.5〜8.5の範囲であり、
温度は20〜25℃の範囲であり、インキュベーション時間は60〜90分の範囲である、請求項1のステップ(i)に記載のプロセス。 - 前記単離された封入体は、尿素、緩衝液、及び還元剤の可溶化混合物中で可溶化され、
前記還元剤は、20〜100mMの範囲で存在するシステインであり、
前記緩衝液は、トリス、EDTA、及び尿素の混合物であり、
尿素の濃度は6〜8Mの範囲であり、トリスは20〜50mMの範囲であり、EDTAは2〜7mMの範囲であり
前記可溶化混合物のpHは9.5〜10.5の範囲であり
前記可溶化された封入体は、8000〜15000gの範囲の遠心分離によってG−CSFタンパク質を放出するように清澄され、
前記可溶化された封入体によって放出された前記G−CSFタンパク質は、RP−HPLCによって純度について推定され、
放出されたG−CSFの前記純度は25〜50%の範囲であることが分かる、請求項1のステップ(ii)に記載のプロセス。 - 前記リフォールディング緩衝液は、アルギニンを含有し、
前記G−CSF可溶化タンパク質は、尿素、ソルビトール、リフォールディング緩衝液、及び酸化剤を含むリフォールディング溶液中での希釈によってリフォールディングされ、
前記酸化剤は、シスチン、システイン、酸化グルタチオン(GSSH)、シスタミンを含む群から選択され、好ましくは、前記酸化剤はシスチンであり、
シスチンは0〜3mMの範囲で存在し、
尿素は0.4M〜1Mの範囲で存在し、
前記G−CSFタンパク質は、RP−HPLCによって純度について推定され、
G−CSFの前記純度は、40〜70%の範囲であり、
リフォールディングされたG−CSFの収率は、85%〜95%の範囲であることが分かり、
前記G−CSFタンパク質溶液は、酸性化剤の存在下で酸性化され、
前記酸性化剤は、酢酸ナトリウム、酢酸、塩酸、及びオルトリン酸を含む群から選択され、好ましくは、前記酸性化剤は塩酸であり、
酸性化したリフォールディングタンパク質溶液のpHは、3.5〜4.5の範囲であり、
前記リフォールディングステップの収率は、50〜90%の範囲であり、好ましくは、前記収率は60〜85%である、請求項1のステップ(iii)に記載のプロセス。 - 前記G−CSFタンパク質は、G−CSFタンパク質の溶出液を得るために清澄及び精製され、
前記G−CSFタンパク質溶液は、精製のためにクロマトグラフィーに供する前に、沈殿物及び微粒子状物質を除去する清澄のために濾過及び遠心分離に供され、
前記清澄は、限外濾過、透析濾過、精密濾過、深層濾過、接線流濾過、全流濾過、及び遠心分離を含む群から選択され、好ましくは、前記清澄は精密濾過または遠心分離によって行われ、
前記遠心分離プロセスは、バッチモード及び連続モードから選択され、好ましくは、前記遠心分離プロセスは連続モードであり、
前記精製の方法は、2つのクロマトグラフィー法を含み、
前記クロマトグラフィー法は、イオン交換クロマトグラフィー及び疎水性相互作用であり、
前記イオン交換クロマトグラフィーは、陽イオン交換クロマトグラフィー及び陰イオン交換クロマトグラフィーから選択され、好ましくは陽イオン交換クロマトグラフィーであり、より好ましくは弱陽イオン交換クロマトグラフィーであり、
前記陽イオン交換クロマトグラフィーは、固定相及び移動相を含み、
前記固定相は、SPセファロースセファロースFF、CMセファロースFF、SPセファロースHP、フラクトゲルSO3、及びフラクトゲルSE HiCap、より好ましくはCMセファロースを含む群から選択され、好ましくは、前記固定相はCMセファロースFFであり、
前記移動相は、酢酸ナトリウム、酢酸、リン酸ナトリウム、オルトリン酸、または混合物を含む群から選択され、好ましくは、混合物は酢酸ナトリウム及び酢酸を含み、
移動相の濃度は20〜50mMであり、pHは3.5〜4.5の範囲であり、
前記GCSFタンパク質は、4.0〜6.0の範囲、好ましくは4.5〜5.6の範囲のpHの、酢酸ナトリウム、酢酸、及びNaClを含む緩衝液を使用して陽イオン交換カラムから溶出され、
溶出モードは、段階勾配、直線勾配、及び両方の組み合わせを含む群から選択され、好ましくは、溶出に使用される勾配は段階勾配であり、
前記溶出は、100〜250cm/時間、より好ましくは150〜200cm/時間の流速で行われ、
溶出液の動的結合能は、10〜20mg/ml、好ましくは15〜18mg/mlの範囲であり、
陽イオン交換クロマトグラフィーステップからのG−CSFのRP−HPLC純度は、90%〜97%の範囲であり、
陽イオン交換クロマトグラフィーステップからの前記G−CSFタンパク質の収率は、90%〜98%の範囲であり、
rHu−G−CSFを含有するイオン交換クロマトグラフィー溶出液は、pH2.0〜7.0、好ましくはpH4.0〜pH6.0の範囲の酸性条件下で、疎水性相互作用クロマトグラフィーの固定相に適用され、
希釈溶液中の硫酸アンモニウムの有効濃度が0.5〜1.5Mの範囲、好ましくは0.7〜1.0Mの範囲に留まることを確実にするために、前記イオン交換クロマトグラフィー溶出液が硫酸アンモニウムで希釈され、
イオン交換クロマトグラフィーは、固定相及び移動相を含み、
前記硫酸アンモニウムで処理されたG−CSFを含有するイオン交換クロマトグラフィー溶出液は、好ましくはpH2.0〜pH7.0、より好ましくはpH4.0〜pH6.0の範囲の酸性条件で疎水性相互作用クロマトグラフィーの固定相に適用され、
前記固定相は、フェニルセファロースFF、ブチルセファロース、Nuviac−Prime、HEAハイパーセルハイパーセル、PPAハイパーセルハイパーセル、フェニルセファロースHP、MEPハイパーセル、及びカプト・アドヘア、好ましくはフェニルセファロースFFを含む群から選択され、
疎水性相互作用クロマトグラフィーの移動相は、4.5〜6、より好ましくは5.2〜5.8の範囲のpHの、酢酸ナトリウム、酢酸、及び硫酸アンモニウムの混合物を含み、
前記溶出は、95〜300cm/時間、好ましくは100〜250cm/時間の流速で行われ、
前記精製されたGCSFタンパク質は、4〜6、より好ましくは4.0〜5.0の範囲のpHの、酢酸ナトリウム及び酢酸を含む移動相を使用して、疎水性相互作用クロマトグラフィーの前記固定相から溶出され、
溶出モードは、段階勾配、直線勾配、及び両方の組み合わせを含む群から選択され、好ましくは、溶出に使用される勾配は段階勾配であり、
前記溶出液は、95〜300cm/時間、好ましくは100〜250cm/時間の流速で得られ、
疎水性相互作用クロマトグラフィーから得られるG−CSFタンパク質の純度は、96.5%〜99.5%の範囲であり、
疎水性相互作用クロマトグラフィーから得られる前記G−CSFタンパク質の収率は、80%〜90%の範囲である、請求項1のステップ(iv)に記載のプロセス。 - 前記精製されたG−CSFタンパク質を、任意選択的に緩衝液中でポリエチレングリコール(PEG)に結合させ、
前記緩衝液は、酢酸ナトリウム、酢酸リン酸塩、リン酸水素カリウム、リン酸二水素カリウム、リン酸水素二ナトリウム、フタル酸水素カリウム、四ホウ酸ナトリウムを含む群から選択され、好ましくは、前記緩衝液は酢酸ナトリウムであり、
前記結合反応のpHは、pH5.0〜5.8の範囲であり、
前記結合反応は、3〜5倍高い量(w/w)の20kD PEGアルデヒドを加えることによって行われ、
前記結合反応は、シアノ水素化ホウ素ナトリウム、トリアセトキシ水素化ホウ素ナトリウム、水素化ホウ素ナトリウム、亜ジチオン酸ナトリウム、ハイドロサルファイトナトリウムを含む群から選択される還元剤の存在下で行われ、好ましくは、前記還元剤はシアノ水素化ホウ素ナトリウムであり、
前記還元剤は、10〜30mMの範囲で存在し、好ましくは、前記還元剤は20mMであり、
反応混合物は、室温で12〜18時間、好ましくは12〜16時間撹拌され、
結合反応からのモノペグ化rHu−GCSFの収率は、65〜85%の範囲であり、好ましくは、前記収率は70〜75%であり、
前記モノペグ化rHu−GCSF反応混合物を保存するための温度は、1〜10℃の範囲であり、好ましくは、前記温度は2〜8℃である、請求項1のステップ(v)に記載のプロセス。 - 前記結合させたペグ化G−CSFタンパク質は、任意選択的に、陽イオン交換クロマトグラフィーによってモノペグ化G−CSFを得るために精製され、
前記結合させたペグ化G−CSFは、pHを3.5〜5.0の範囲に調整するように平衡化緩衝液で希釈され、好ましくは、前記pHは4.0〜4.5の範囲に調整され、
前記陽イオン交換クロマトグラフィーは、固定相及び移動相を含み、
前記固定相は、MacroCap−SP及びSPセファロースFF、フラクトゲルSO3を含む群から選択され、好ましくは、前記固定相はMacroCap−SPであり、
前記移動相は、酢酸ナトリウム、酢酸、NaCl、及びそれらの混合物から選択され、好ましくは、前記移動相は混合物であり、
前記酢酸ナトリウムは10〜50mMの範囲で存在し、NaClは0.25〜0.75Mの範囲で存在し、前記移動相のpHは3.5〜5の範囲で存在し、好ましくは、前記pHは4.0〜4.5であり、
溶出モードは、段階勾配、直線勾配、及び両方の組み合わせを含む群から選択され、好ましくは、溶出に使用される勾配は段階勾配であり、
前記精製されたモノペグ化r−Hu−GCSFは、100〜150cm/時間、好ましくは100cm/時間の流速で回収され、
陽イオン交換クロマトグラフィーからのモノペグ化GCSFの収率は、80〜95%の範囲であり、好ましくは、前記範囲は85〜90%であり、
前記精製されたモノペグ化rHu−GCSFは、10kDaの限外濾過膜によって9〜12mg/mlの範囲に濃縮され、好ましくは前記精製されたモノペグ化rHu−GCSFは、11.0〜11.5mg/mlの範囲に濃縮され、
得られた前記精製されたモノペグ化rHu−GCSFは、1〜10℃の範囲の温度で、好ましくは2〜8℃で保存される、請求項1のステップ(vi)に記載のプロセス。 - 顆粒球の増殖及び分化のための、液体の非経口静脈内製剤中における、請求項1に記載の単離及び調製されたGCSFの使用。
- 薬学的に許容される賦形剤とともに、請求項1に記載のように得られるGCSFを含む薬学的組成物。
