Abstract
Description
筋萎縮のうち、加齢による進行性の筋萎縮(筋力の低下を伴うこともある)は、サルコペニアと呼ばれている。サルコペニアは、高齢者に、寝たきり、転倒、不動や認知症などの有害事象を引き起こし、この有害事象により更にサルコペニアが進行するという悪循環をもたらすことが知られている。
そのため、筋萎縮を予防及び治療することで、高齢者においては活動性を維持し、日常生活動作レベルを確保し、更には、上述の有害事象を防止することが期待されている。
筋萎縮の予防及び治療に関しては、ミオスタチン(Myostatin)を標的とすることの有用性が示されている(非特許文献1及び2)。
1.筋萎縮の予防剤及び/又は治療剤のスクリーニング方法であって、
以下の工程(1)〜(3):
(1)試験物質と筋肉細胞とを接触させる工程、
(2)該筋肉細胞内のSmad2及び/又はSmad3の量及び/又は活性を測定する工程、及び、
(3)該筋肉細胞内のSmad2及び/又はSmad3の量の上昇を抑制する試験物質、及び/又は、該筋肉細胞内のSmad2及び/又はSmad3の活性を抑制する試験物質を選択する工程
を含む、スクリーニング方法。
2.工程(1)の筋肉細胞が骨格筋細胞である、前記1に記載のスクリーニング方法。
3.Smad2がリン酸化Smad2である、及び/又は、Smad3がリン酸化Smad3である、前記1又は2に記載のスクリーニング方法。
4.工程(2)において、Smad2及び/又はSmad3の量を測定し、工程(3)において、筋肉細胞内のSmad2及び/又はSmad3の量の上昇を抑制する試験物質を選択する、前記1〜3のいずれか1項に記載のスクリーニング方法。
5.工程(2)において、Smad2及び/又はSmad3の量を、ウェスタンブロッティングにて測定する、前記1〜4のいずれか1項に記載のスクリーニング方法。
6.工程(2)において、筋肉細胞内のSmad2及び/又はSmad3の活性を測定し、工程(3)において、筋肉細胞内のSmad2及び/又はSmad3の活性を抑制する試験物質を選択する、前記1〜3のいずれか1項に記載のスクリーニング方法。
7.筋萎縮がサルコペニアである、前記1〜6のいずれか1項に記載のスクリーニング方法。
8.筋萎縮がスポーツ外傷の治療より生じた筋萎縮である、前記1〜6のいずれか1項に記載のスクリーニング方法。
9.前記1〜8のいずれか1項に記載のスクリーニング方法を行なうためのキットであって、筋肉細胞中のSmad2及び/又はSmad3の量又は活性を測定するための試薬を含むことを特徴とするキット。
以下の工程(1)〜(3):
(1)試験物質と筋肉細胞とを接触させる工程、
(2)該筋肉細胞内のSmad2及び/又はSmad3の量及び/又は活性を測定する工程、及び、
(3)該筋肉細胞内のSmad2及び/又はSmad3の量の上昇を抑制する試験物質、及び/又は、該筋肉細胞内のSmad2及び/又はSmad3の活性を抑制する試験物質を選択する工程
を含んでいる。
「筋萎縮」とは、安静状態が長く続くことによって筋肉が体積減少を起こし、萎縮した状態をいう。
筋萎縮の原因としては、加齢、不動、無重力、長期の臥床、長期あるいは大量のステロイド治療、癌によるカヘキシー、栄養飢餓、四肢の骨折や筋、腱及び靭帯の断裂を治療するための固定等が挙げられる。前述の四肢の骨折や筋、腱及び靭帯の断裂には、スポーツ外傷により生じたものが含まれる。
筋萎縮のうち、加齢により発症する進行性の筋萎縮はサルコペニアと呼ばれている。本発明は、サルコペニアの予防剤及び/又は治療剤のスクリーニングに好適に用いることができる。
また、本発明は、スポーツ外傷の治療(四肢の骨折や筋、腱及び靭帯の断裂の固定)により生じた筋萎縮の予防剤及び/又は治療剤のスクリーニングにも好適に用いることができる。
筋萎縮は主に骨格筋で発症する。筋萎縮は、あらゆる部位の骨格筋で発症しうるが、腓腹筋や大腿筋で発症しやすく、発症による影響も大きい。
筋萎縮は、筋肉を有するあらゆる動物で起こりうるが、本発明は、哺乳動物、特にヒトにおける筋萎縮の予防剤及び/又は治療剤のスクリーニングに好適に用いることができる。
「筋萎縮の予防剤」には、筋萎縮の発症を阻害する剤の他、筋萎縮の発症を遅延させる剤等が含まれる。
「筋萎縮の治療剤」には、筋萎縮の症状を改善又は緩和する剤の他、発症した筋萎縮の進行を遅延させる剤等が含まれる。
「スクリーニング」とは、上述の筋萎縮の予防剤及び/又は治療剤を、試験物質(候補物質)の中から選択することをいう。
工程(1)では、試験物質と筋肉細胞とを接触させる。
筋肉細胞としては、あらゆる動物由来のものを使用できるが、哺乳動物(例えば、マウスやヒト)由来の筋肉細胞を用いることが好ましい。
筋肉細胞としては、筋萎縮を発症しうる筋肉に由来する細胞を特に制限なく用いることができる。筋肉細胞の具体例としては、骨格筋細胞、心筋細胞、血管や腸管の平滑筋細胞等が挙げられる。筋肉細胞としては、骨格筋細胞が好ましい。
骨格筋細胞が由来する部位は特に限定されないが、筋萎縮が発症しやすい腓腹筋や大腿筋に由来する骨格筋細胞が好ましい。
筋肉細胞は、筋萎縮を発症している筋肉組織から採取した細胞であってもよく、筋萎縮を発症していない筋肉組織から採取した細胞であってもよい。
また、筋肉細胞は、採取した細胞自体であってもよく、プライマリー細胞であってもよく、株化された細胞であってもよい。また、筋肉細胞は、間葉系幹細胞から誘導した筋肉細胞であってもよく、iPS細胞やES細胞から誘導した筋肉細胞であってもよい。
試験物質と接触させた後の筋肉細胞は、その生存を維持できる任意の条件下でインキュベートしてもよい。インキュベート条件は、筋肉細胞の培養条件として知られているものを特に制限なく用いることができる。
工程(2)では、試験物質と接触させた筋肉細胞内のSmad2及び/又はSmad3の量及び/又は活性を測定する。
「Smad2」及び「Smad3」は、いずれも、ミオスタチン、IGF−1やTGF−βといったサイトカインによる細胞内シグナル伝達に関与することが知られているタンパク質性転写因子である。なお、Smad2のアミノ酸配列は、例えば、アクセッション番号「NM_001003652」でGenBankに登録されている。また、Smad3のアミノ酸配列は、例えば、アクセッション番号「NM_001145102」でGenBankに登録されている。
(1)ミオスタチンが筋肉細胞表面のレセプターActRIIBに結合して、ActRIIBの細胞内ドメインをリン酸化する。
(2)リン酸化されたActRIIBが、別の細胞表面レセプターActRIBをリン酸化し、更にActRIBとの会合体を形成する。
(3)前記の会合体が、Smad2及びSmad3をリン酸化する。
(4)リン酸化されたSmad2及びSmad3が、Smad4と会合体(Smad2とSmad4との会合体、Smad3とSmad4との会合体や、Smad2とSmad3とSmad4との会合体)を形成する。
(5)前記の会合体が筋肉細胞の核内へ移行して、筋肉を構成するタンパク質の分解に関わる遺伝子(Atrogin-1及びMuRF-1)の発現を促進する。
ところで、後述の実施例で示されるように、筋萎縮を発症している筋肉細胞では、Smad2及びSmad3の量がミオスタチンからは独立した様式で上昇している。
したがって、本発明のスクリーニング方法は、ミオスタチンを標的とする筋萎縮治療剤とは異なる機序で作用する医薬品の開発に用いることができる。
また、Smad2とSmad3の両方の量及び/又は活性を測定してもよい。
また、工程(2)では、Smad2及び/又はSmad3(以下、「Smad2/3」ともいう)の量のみを測定してもよく、活性のみを測定してもよく、量と活性の双方を測定してもよい。
Smad2/3の量は、リン酸化されたSmad2/3の量として測定してもよい。
例えば、Smad2/3のタンパク質としての量を直接測定してもよく、当該タンパク質量に対応するSmad2/3のmRNA量を指標に測定してもよい。
Smad2/3のタンパク質としての量は、例えば、Smad2/3に特異的な抗体を用いたウェスタンブロッティングやELISA等を使用して測定することができる。
Smad2/3のmRNA量は、例えば、リアルタイムPCRや、リポーター遺伝子アッセイ等を使用して測定することができる。リポーター遺伝子アッセイは、例えば、Smad2/3をコードする遺伝子の下流にリポーター遺伝子(例えば、ルシフェラーゼ遺伝子)を組み込むことにより、行なうことができる。
Smad2/3の活性は、当該技術分野で一般的に用いられている転写活性の定量方法を特に制限なく用いて測定することができる。具体例としては、リポーターアッセイ法やリアルタイムPCR法等が挙げられる。
工程(3)では、工程(2)の測定結果に基づき、筋肉細胞内のSmad2/3の量の上昇を抑制する試験物質、及び/又は、該筋肉細胞内のSmad2及び/又はSmad3の活性を抑制する試験物質を、筋萎縮の予防剤及び/又は治療剤として選択する。
工程(2)の測定結果と比較される対象(対照)は、工程(1)で用いた試験物質と接触させていない筋肉細胞のSmad2/3の量及び/又は活性であってもよく、工程(1)で用いた試験物質とは異なる物質(例えば、Smad2/3の活性を低下させることが知られている公知物質(例えば、SIS3(Smad3 Inhibitor))と接触させた筋肉細胞のSmad2/3の活性であってもよい。
「Smad2/3の量の上昇を抑制する」とは、「Smad2/3の量を変化させない」ことと、「筋肉細胞内のSmad2/3の量を低下させる」こととをいう。
工程(1)で使用した試験物質が、筋肉細胞内のSmad2/3の量を変化させなかった場合、当該試験物質は、筋萎縮を予防するための医薬品(例えば、Smad2/3の量を筋萎縮発症量(発症レベル)未満に維持する医薬品)や、筋萎縮を治療するための医薬品(例えば、筋萎縮の進行を遅延させる医薬品)の候補となる。
工程(1)で使用した試験物質が、筋肉細胞内のSmad2/3の量を低下させた場合、当該試験物質は、筋萎縮を予防及び/又は治療するための医薬品(例えば、Smad2/3のレベルを筋萎縮発症量(発症レベル)未満へと低減する医薬品)の候補となる。
工程(1)で使用した試験物質が、筋肉細胞内のSmad2/3の活性を抑制した(低下させた)場合、当該試験物質は、筋萎縮を予防及び/又は治療するための医薬品(例えば、Smad2/3の活性を筋萎縮が発症する活性(発症レベル)未満へと低減する医薬品)の候補となる。
上記の試薬としては、筋肉細胞中のSmad2/3の量又は活性を測定するために当該技術分野で一般的に用いられているものを特に制限なく用いることができる。例えば、Smad2/3の量をリアルタイムPCRにより測定する場合には、Smad2/3のプライマーが試薬として挙げられる。Smad2/3の量をウェスタンブロッティングやELISAにより測定する場合には、Smad2/3に対する特異的抗体が試薬として挙げられる。
また、本発明のキットは、前述の試薬の他、測定手段に固有の試薬、例えば、ウェスタンブロッティングやELISAでは固相ブロッキング剤等を含むことができる。
更に、本発明のキットは、本発明のスクリーニング方法を実施する手順を記載した説明書(指示書)を含んでいてもよい。
切断処置1週間後に腓腹筋の重量を測定した。坐骨神経切断側の筋肉重量は0.817±0.055g/マウス体重であり、非切断側の筋肉重量は1±0.057g/マウス体重であった。非切断側の筋肉重量を1とした場合の、坐骨神経切断側の相対筋肉重量を図1に示す。
図1に示されるように、座骨神経切断側で有意な筋肉重量の減少が見られた。
非切断側の各遺伝子の発現量を1とした場合の、坐骨神経切断側の各遺伝子の相対発現量を図2に示す。図2(a)はAtrogin-1に関する結果を示し、図2(b)はMuRF-1に関する結果を示す。
図2に示されるように、座骨神経切断側では、Atrogin-1及びMuRF-1の発現が有意に上昇していた。Atrogin-1及びMuRF-1は、いずれも筋肉を構成するタンパク質の分解に関わるので、その過剰発現は筋萎縮につながるものである。
(1)及び(2)の結果は、マウスの坐骨神経切断側では、坐骨神経切断に起因する筋肉の安静状態により、筋萎縮が生じたことを示している。
同内容の実験を2組(#1, #2)で施行した。
結果を図3に示す。なお、beta-actinの発現を内部比較コントロールとして用いた。
図3に示されるように、坐骨神経切断側(すなわち、筋萎縮を起こしている筋肉)において、Smad2及びSmad3の発現の増加(タンパク質としての量の増加)が認められた。また、リン酸化Smad2及びSmad3についても、坐骨神経切断側で発現の増加が認められた。
したがって、筋肉細胞内のSmad2及び/又はSmad3の量の上昇の抑制及び/又はSmad2及び/又はSmad3の活性の抑制の有無を指標として、筋萎縮の予防剤及び/又は治療剤をスクリーニングすることができる。
(1)8週齢のミオスタチンコンディショナルノックアウトマウス (Mstn cKO, Mx Cre/Mstnflox/flox)(5匹)と、コントロールマウス(control, Mstnflox/flox)(5匹)に、polyIpolyCを投与した。なお、polyIpolyCは、Creの発現誘導からミオスタチンを欠損させるために投与した。
投与から3週間後に、各マウスに坐骨神経切断側(denerve)と非切断側(control)とを作成した。坐骨神経切断により、坐骨神経切断側の骨格筋は安静状態となった。
切断処置1週間後に、腓腹筋の重量を測定した。ミオスタチンコンディショナルノックアウトマウスについて、坐骨神経切断側の筋肉重量は0.774±0.068g/マウス体重であり、非切断側の筋肉重量は1±0.063g/マウス体重であった。コントロールマウスについて、坐骨神経切断側の筋肉重量は0.756±0.069g/マウス体重であり、非切断側の筋肉重量は1±0.1g/マウス体重であった。各マウスについて、非切断側の筋肉重量を1とした場合の、坐骨神経切断側の相対筋肉重量を図4に示す。
図4に示されるように、座骨神経切断側における筋肉重量の有意な減少がミオスタチンコンディショナルノックアウトマウス及びコントロールマウスの双方において見られた。
各マウスについて、非切断側の各遺伝子の発現量を1とした場合の、坐骨神経切断側の各遺伝子の相対発現量を図5に示す。
図5に示されるように、ミオスタチンコンディショナルノックアウトマウス及びコントロールマウスの双方において、座骨神経切断側では、Atrogin-1及びMuRF-1(いずれも、筋肉を構成するタンパク質の分解に関わる遺伝子)の発現が有意に上昇していた。
結果を図6に示す。なお、beta-actinの発現を内部比較コントロールとして用いた。
図6に示されるように、ミオスタチンコンディショナルノックアウトマウスの坐骨神経切断側(すなわち、筋萎縮を起こしている筋肉)において、Smad2及びSmad3の発現の増加(タンパク質としての量の増加)が認められた。
(1)〜(3)の結果は、ミオスタチンコンディショナルノックアウトマウス(すなわち、ミオスタチンを発現できないマウス)の坐骨神経切断側(すなわち、筋萎縮を起こしている筋肉)において、Smad2及びSmad3の発現の増加が起きたことを示している。これは、筋萎縮におけるSmad2及びSmad3のタンパク質の量レベルでの増加は、ミオスタチンに依存しないことを示している。
切断処置1週間後に、腓腹筋の重量を測定した。
野生型マウス(WT)について、非切断側の筋肉重量を1とした場合の坐骨神経切断側の相対筋肉重量は0.779であった。
Smad3ノックアウトマウス(Smad3 KO)について、非切断側の筋肉重量を1とした場合の坐骨神経切断側の相対筋肉重量は0.89であった。
Smad2及びSmad3ダブルノックアウトマウス(DKO)について、非切断側の筋肉重量を1とした場合の坐骨神経切断側の相対筋肉重量は1.033であった。
非切断側の筋肉重量を1とした場合の、坐骨神経切断側の相対筋肉重量を図7に示す。
図7に示されるように、Smad3の発現が抑制されていないマウス(WT)では、座骨神経切断側で有意な筋肉重量の減少が見られた。一方、Smad3の発現を抑制したマウス(Smad3 KO及びDKO)では、座骨神経切断側における統計学的に有意な筋肉重量の減少は見られなかった。
非切断側(control)の各遺伝子の発現量を1とした場合の、坐骨神経切断側(denerve)の各遺伝子の相対発現量を図8に示す。
図8に示されるように、Smad3の発現が抑制されていないマウス(WT)では、座骨神経切断側において、Atrogin-1及びMuRF-1(いずれも、筋肉を構成するタンパク質の分解に関わる遺伝子)の発現が有意に上昇していた。一方、Smad3の発現を抑制したマウス(Smad3 KO及びDKO)では、座骨神経切断側における統計学的に有意なAtrogin-1及びMuRF-1の上昇は見られなかった。
したがって、Smad3の量の上昇の抑制又はSmad3の活性抑制の有無を指標として、筋萎縮の予防剤及び/又は治療剤をスクリーニングすることができる。
Claims (9)
- 筋萎縮の予防剤及び/又は治療剤のスクリーニング方法であって、
以下の工程(1)〜(3):
(1)試験物質と筋肉細胞とを接触させる工程、
(2)該筋肉細胞内のSmad2及び/又はSmad3の量及び/又は活性を測定する工程、及び、
(3)該筋肉細胞内のSmad2及び/又はSmad3の量の上昇を抑制する試験物質、及び/又は、該筋肉細胞内のSmad2及び/又はSmad3の活性を抑制する試験物質を選択する工程
を含む、スクリーニング方法。 - 工程(1)の筋肉細胞が骨格筋細胞である、請求項1に記載のスクリーニング方法。
- Smad2がリン酸化Smad2である、及び/又は、Smad3がリン酸化Smad3である、請求項1又は2に記載のスクリーニング方法。
- 工程(2)において、Smad2及び/又はSmad3の量を測定し、
工程(3)において、筋肉細胞内のSmad2及び/又はSmad3の量の上昇を抑制する試験物質を選択する、請求項1〜3のいずれか1項に記載のスクリーニング方法。 - 工程(2)において、Smad2及び/又はSmad3の量を、ウェスタンブロッティングにて測定する、請求項1〜4のいずれか1項に記載のスクリーニング方法。
- 工程(2)において、筋肉細胞内のSmad2及び/又はSmad3の活性を測定し、
工程(3)において、筋肉細胞内のSmad2及び/又はSmad3の活性を抑制する試験物質を選択する、請求項1〜3のいずれか1項に記載のスクリーニング方法。 - 筋萎縮がサルコペニアである、請求項1〜6のいずれか1項に記載のスクリーニング方法。
- 筋萎縮がスポーツ外傷の治療より生じた筋萎縮である、請求項1〜6のいずれか1項に記載のスクリーニング方法。
- 請求項1〜8のいずれか1項に記載のスクリーニング方法を行なうためのキットであって、筋肉細胞中のSmad2及び/又はSmad3の量又は活性を測定するための試薬を含むことを特徴とするキット。
