Abstract
Description

一部の実施形態では、患者試料に由来する核酸を配列決定してMfsd2a遺伝子における変異を究明する。当業者に周知である核酸の配列を決定するための任意の技法を使用することができる。DNAの配列決定の技法には、標識されたターミネータまたはプライマーを用いた従来のジデオキシ配列決定反応(Sanger法)及びスラブ電気泳動またはキャピラリー電気泳動におけるゲル分離が含まれる。一実施形態では、次世代(NextGen)の配列決定プラットフォームが本発明の実践にて有利に使用される。NextGen配列決定は、高処理能力の配列決定が可能である従来のSanger型以降の多数の配列決定法のいずれかを指す。NextGenの配列決定プラットフォームには、可逆的に終端となる標識されたヌクレオチドを用いた合成による配列決定、ピロ配列決定、454配列決定、標識されたオリゴヌクレオチドのプローブのライブラリに対する対立遺伝子特異的なハイブリッド形成、その後にライゲーションが続く標識されたクローンのライブラリに対する対立遺伝子特異的なハイブリッド形成を用いた合成による配列決定、重合工程の間での標識されたヌクレオチドの取り込みのリアルタイムのモニタリング、ポロニー配列決定、単一分子リアルタイム配列決定、及びSOLiD配列決定を挙げることができる。特定の配列決定プラットフォームの例には、454配列決定(Roche)(Margulies,Mら.2005,Nature,437,376−380);ヌクレオチド付加の際に解放されるピロリン酸(PPi)を使用するピロ配列決定が挙げられる。PPiはアデノシン5'−ホスホ硫酸の存在下でATPスルフリラーゼによってATPに変換される。ルシフェラーゼはATPを用いてルシフェリンをオキシルシフェリンに変換し、この反応は検出され、分析される光を生成する。使用することができるDNA配列決定法の別の例はSOLiD法(Applied Biosystems)である。SOLEXA(Illumina)配列決定は、フォールドバックPCRと係留したプライマーを用いた固相表面におけるDNAの増幅に基づく。使用することができる配列決定法の別の例には、Pacific Biosciencesの単一分子リアルタイム(SMRT(商標))法が挙げられる。使用することができる配列決定法のさらなる例は、ナノポア配列決定(Soni,G.V.及びMeller,A.(2007),Clin.Chem.53:1996−2001)である。使用することができる配列決定法の別の例には、DNAの配列決定を行うために化学感受性電界効果トランジスタ(chemFET)アレイ(たとえば、US20090026082に記載されたような)を使用することが関与する。

本発明の栄養組成物は従来の製剤化法及び食物生産法によって製造することができる。
[化学物質]
放射性標識していないリゾホスファチジルコリン、リゾホスファチジルエタノールアミン、リゾホスファチジルセリン及び他のリゾリン脂質はAvanti Polar Lipidsから購入した。放射性標識した1−パルミトイル2−リゾホスホコリン(LPC−[3H]パルミチン酸)、1−オレオイル2−リゾホスホコリン(LPC−[14C]オレイン酸)、1−ドコサヘキサエノイル2−リゾホスホコリン(LPC−[14C]DHA)及び[1−14C]ドコサヘキサエン酸([14C]DHA)はAmericans Radiochemicalsから購入した。クロロホルム(非標識)またはエタノール/トルエン(放射性標識)におけるリゾリン脂質を窒素ガスのもとで完全に乾燥させ、12%脂肪酸−遊離のBSA(Sigma)にて可溶化し、それを150mMのNaClに溶解した。LPC−[14C]−オレイン酸/LPC−オレイン酸混合物を調製するために、25μCiのLPC−[14C]−オレイン酸(比活性:55mCi/ミリモル)を乾燥させ、3mlの20mMの非標識のLPC−18:0/BSAに溶解した。LPC−[3H]−パルミチン酸)/LPC−パルミチン酸は、25μCiのLPC−[3H]−パルミチン酸(比活性:60Ci/ミリモル)を4mlの10mMのLPC−パルミチン酸に溶解することによって調製した。1−ドコサヘキサエノイルLPCは、5mMのCaCl2を含有するホウ砂緩衝液(pH8.5)におけるミツバチ毒PLA2(Sigma)による1,2−ジドコサヘキサエノイルPCの加水分解によって調製し、TLC法によって精製した。LPC−[14C]−DHA/LPC−DHA混合物を調製するために、10μCiのLPC−[14C]−DHAを非標識のLPC−DHA/BSAと10mMの最終濃度に混合した。[14C]−DHA/DHA混合物を調製するために、50μCiの[14C]−DHAを非標識のDHA/BSAと12.2mMの最終濃度に混合した。
Mfsd2aノックアウトマウスは以前記載された(Berger,J.H.,Charron,M.J.及びSilver,D.L.Major facilitator superfamily domain−containing protein 2a (MFSD2A) has roles in body growth,motor function,and lipid metabolism.PLoS One,7,e50629,doi:10.1371/journal.pone.0050629(2012))ように生成した。合計11%の脂肪を含有する高エネルギー餌5LJ5(PicoLab)でマウスを維持した。仔は3週齢で離乳し、高エネルギー餌で維持した。実験プロトコールはSingHealth Institutional Animal Care and Use Committeeによって認可された。
組織の脂質の分析のために、HET系の母に生まれた7〜8週齢のメスWT及びKOマウスの脳、肝臓及び心臓、並びに胎齢e18.5日の胎仔から脳を採取し、直ちに抽出まで液体窒素で凍結した。脂質抽出はクロロホルム/メタノール法に従った。細胞に基づいた遊離の脂肪酸の輸送については、BSA複合体における100μMのドコサヘキサエン酸、アラキドン酸、エイコサペンタエン酸、α−リノレン酸、リノール酸、オレイン酸、パルミチン酸で一晩処理した後のマウスMfsd2a及びモック対照を発現しているHEK293細胞からHIP緩衝液(容量当たり、ヘキサン:イソプロパノール3:2)を用いて脂質を抽出し、N2ガスのもとで乾燥させた。三連四重極/イオン捕捉質量分光分析(4000Qtrap,Applied Biosystem)と一体化した高速液体クロマトグラフィ(HPLC)1100システム(Agilent)によってリン脂質の種を測定した。以前記載されたように、個々のリン脂質種のレベルを分析し、PC−14:0/14:0、PE−14:0/14:0、PS−14:0/14:0(Avanti Polar Lipids,Alabaster,AL,USA)及びジオクタノイルPI(Echelon Biosciences,Inc.,Salt Lake City,UT,USA)を含むスパイクのある内部標準を用いて定量した。
放射性標識したLPCパルミチン酸(LPC−[3H]パルミチン酸)、オレイン酸(LPC−[14C]オレイン酸)及びドコサヘキサエン酸(LPC−[14C]DHA)またはTopFluor−LPC、TopFluor−LPE及びNBD−LPCを12%BSAに溶解し、それを150mMのNaClにて希釈した。Mfsd2a及び変異体の構築物を過剰発現しているHEK293細胞を用いて放射性標識したLPCまたは蛍光LPCの取り込みアッセイを調べた。手短には、リポフェクトアミン2000(Invitrogen)用いて、90〜95%の集密度のHEK293細胞にpcDNA3.1Mfsd2a(WT)、pcDNA3.1Mfsd2aD92A(D92A)、pcDNA3.1Mfsd2aD96A(D96A)またはpcDNA3.1(モック)のプラスミドで形質移入した。取り込みアッセイは形質移入の24時間後に実施した。リガンドとのインキュベートに先立って、形質移入されたHEK293細胞をアッセイ前に無血清DMEMで洗浄した。濃度及び時間に依存性のアッセイについては、放射性標識したLPCを事前に温めたDMEM培地で希釈した。ナトリウム依存性のアッセイについては、150mMのNaClまたは150mMの塩化コリンを伴った輸送緩衝液(5mMのKCl、10mMのHepes、pH7.4)にて放射性標識したLPCを希釈した。ナトリウム濃度依存性アッセイについては、150mMの一定のカチオンのモル濃度を維持するために、NaClの濃度の低下を塩化コリンで置き換えた。アッセイはすべて3つ組で37℃にて12穴プレートにおいて行った。
エタノールに溶解されたLPC−パルミチン酸を調製するために、0.75μCiのLPC−[3H]パルミチン酸をクロロホルム中のLPC−パルミチン酸で希釈した。混合物を乾燥させ、50μlのエタノールに溶解した後、上述のような150mMのナトリウムを伴った輸送緩衝液6mlを加えて50μMのLPC−パルミチン酸を有した。LPC−パルミチン酸ミセルを調製するために、0.75μCiのLPC−[3H]パルミチン酸をクロロホルム中のLPC−パルミチン酸で希釈した。混合物を乾燥させ、150mMのナトリウムを伴った輸送緩衝液6mlに溶解して100μMのLPC−パルミチン酸を有し、5分間氷上で超音波処理した。活性炭を加え、遠心分離してLPC−パルミチン酸の単量体を取り除いた。pcDNA3.1Mfsd2aプラスミドまたは対照としてのpcDNA3.1プラスミドによって過剰発現しているHEK293細胞にて輸送アッセイを37℃にて30分間同様に行った。
pcDNA3.1Mfsd2a(WT)、pcDNA3.1Mfsd2aD92A(D92A)、pcDNA3.1Mfsd2aD96AまたはpcDNA3.1(モック)のプラスミドによるHEK293細胞の形質移入の24時間後、無血清DMEM培地で細胞を1回洗浄し、その後、25μMの放射性標識したLPC−パルミチン酸と250μMの非放射性の競合相手の混合物を添加し、それを12%のBSAに溶解した。非放射性の競合相手を伴ったまたは伴わない試料にてBSAの総濃度を一定に保持した。アッセイは37℃で30分間行った。フォスコリン界面活性剤及びPAFのような他のLPC類似体との競合アッセイを、15分間を除いて同じ条件下で行った。細胞の生存に対する界面活性剤と生物活性のある脂質の否定的な効果を限定するために短い反応時間が必要だった。アッセイはすべて3つ組で37℃にて12穴プレートにおいて行った。
6〜8週齢のオス及びメスのマウスに、総容量150μlのリン酸緩衝化生理食塩水における75μlの20mMの放射性標識したLPC−[14H]オレイン酸、100μlの10mMのLPC−[14C]DHA/BSA複合体または82μlの12.2mMの[14C]DHA/BSA複合体を静脈内注射した。注射の2時間後、マウスを麻酔し、脳血管系に結合した血液及び脂質のトレーサーを取り除くために0.5%BSAを含有するPBSで5分間潅流した。脂質抽出のために組織を採取した。脂質抽出のために、組織を秤量し、類似の量の組織をクロロホルム/メタノール中にてホモジネートした。有機相の脂質をシンチラントと混合し、シンチレーションでカウントした。マウス当たり300μgのNBD−LPC/BSAまたは300μgのTopFluor−LPC/BSA複合体の注射によって同様の実験も行った。この実験では、PBSでマウスを5分間潅流し、その後、組織固定のために4%パラホルムアルデヒド(PFA)による15分間の潅流が続いた。厚さ40μmのWT及びKOの脳切片を調製し、TyphoonFLA9000スキャナー(Agilent)における緑色蛍光方式を用いて走査した。NBD−LPCの蓄積はその領域当たりの各切片の蛍光として表された。母親から胎仔への餌DHAの輸送も[14C]DHA/BSA複合体を用いて測定した。胎齢e18.5日の妊娠メスに[14C]DHA/BSA複合体(200μlの10mM[14C]DHA/BSA/マウス)のボーラスを強制投与した。強制投与の20時間後、胎仔の脳を採取し、秤量した。脂質抽出及び放射線活性の測定は上述のように行った。
pcDNA3.1Mfsd2a(WT)、pcDNA3.1Mfsd2aD92A(D92A)、pcDNA3.1Mfsd2aD96AまたはpcDNA3.1(モック)のプラスミドまたはヒトMfsd2a(Sport6プラスミドにおける)によって過剰発現しているHEK293細胞を無血清DMEM培地で1回洗浄し、その後、放射性標識した50μMのLPC[14C]オレイン酸、LPC[14C]DHA又は50μMのTopFluor−LPC及びNBD−LPCと共にインキュベートした。HIP緩衝液で30分間脂質を2回抽出した。窒素流で脂質を乾燥させ、クロロホルムで再構成させ、TLCプレート(Milipore)上でスポットを作らせた。リン脂質分離のための溶媒はクロロホルム/メタノール/アンモニア溶液(25%)(容積当たり50:25:6)だった。放射性標識したリン脂質のTLCプレートを30分間乾燥させ、Phosphorscreensに一晩暴露し、TyphoonFLA9000スキャナー(Agilent)で走査した。蛍光リン脂質のTLCプレートはTyphoonFLA9000スキャナーで走査し、Imagequantソフトウエアを用いて定量した。
HET系の両親で生まれた7.5〜8週齢の成熟オスWT及びKOマウスを深く麻酔し、経心腔で50mlの生理食塩水によって潅流し、その後、0.1MのPB(pH7.4)中4%のPFA、100mlにより30分間潅流した。胎仔については、4%PFAで一晩、次いで30%スクロースで一晩、脳を固定した。低温保持装置にて厚さ40μmの矢状断面の切片を作り、免疫染色用に連続切片を24穴組織培養ディッシュの異なるウェルに移した。NeuN(1:1000、Chemicon、CA、USA)、Mfsd2a(1:500)、GFAP(1:1000、Chemicon、CA、USA)、パルブアルブミン(1:500、Swant、Switzerland)、Glut1(1:500、Abcam)、PDGFR−β(1:150、eBioscience)に対する抗体を用いた免疫細胞化学の手順のために脳切片を処理し、4',6−ジアミノジノ−2−フェニルインドール(DAPI,1:5000)にて5分間インキュベートした後、洗浄し、標本にした。ZeissLSM710倒立蛍光共焦点顕微鏡(Carl Zeiss,Pte.Ltd.,Singapore)にて画像を取得した。カニクイザルの海馬切片で同じ局在決定手順を行った。NeuN免疫染色された海馬におけるニューロン断面及びパルブアルブミン染色された小脳におけるプルキンエ細胞を数え、平均値±SEMで密度(平方ミリメートル当たりの免疫陽性ニューロンの数(数/mm2)及びミリメートル当たりの免疫陽性細胞の数(数/mm))として示した。スチューデントのt検定を用いて統計的な有意性を評価した。
Y字型迷路による自発的交替試験:互いに120°の角度で3つのアームがあるY字型の迷路の中央にマウスを置いた。動物が3つのアームすべてを自由に探索できるようにする。マウスがインタクトな空間作業記憶を有していれば、直近に訪れたアームに入る傾向が減ることになる。Topscanプログラム(Cleversys Inc.,Reston,VA)を用いてアームへの侵入の数をスコア化し、交替の数を決定した。四肢すべてがアーム内にある場合、マウスはアームに入ったと見なされる。
対応のないスチューデントのt検定を用いて、遺伝子型間のDHA及びAAのレベル、及び組織学的な解析の有意差を算出した。二元配置ANOVAを用いてモック対WT及び変異体の間でのDPMとして表されたLPC−[14C]DHA、LPC−[3H]パルミチン酸、LPC−[14C]オレイン酸のシグナルの統計的な解析を計算した。p<0.05を有意であると見なした。
Mfsd2aの免疫学的局在決定は、それがBBBを構成する内皮で専ら見いだされる脳の微細血管にてそれが高度に濃縮される(図1a、b、c及び図5a、b)が、内皮を包む周皮細胞では発現されない(Armulik,A.ら.Pericytes regulate the blood−brain barrier.Nature,468,557−561,doi:10.1038/nature09522(2010);Bell,R.D.ら.Pericytes control key neurovascular functions and neuronal phenotype in the adult brain and during brain aging.Neuron,68,409−427,doi:10.1016/j.neuron.2010.09.043(2010))(図5c、d)ことを示しているということは、Mfsd2aのmRNAはBBBで見いだされた最高に濃縮された転写物の1つであることを示す以前の報告(Daneman,R.ら.The mouse blood−brain barrier transcriptome:a new resource for understanding the development and function of brain endothelial cells.PloS One,5,e13741,doi:10.1371/journal.pone.0013741(2010))を裏付けている。BBBにおけるこの局在化パターンはサルの小脳及び海馬でも言及された(図6)。Mfsd2aは胎齢e15.5日でのBBBにて発現されることが見いだされた(図7a)。BBBの内皮へのMfsd2aの局在化はBBBにおける輸送機能を示唆している。
オス及びメスのMfsd2a遺伝子欠損(KO)マウスはメンデル比率で生まれたが、生後間もなく有意に高い出生後死亡率を有した(Berger,J.H.,Charron,M.J.及びSilver,D.L. Major facilitator superfamily domain−containing protein 2a (MFSD2A)has roles in body growth,motor function,and lipid metabolism.PLoS One,7,e50629,doi:10.1371/journal.pone.0050629(2012))。組織の生理的な及び生化学的な測定値及び全身の健康状態は、独立して生成されたMfsd2aのKOマウスモデルにて平凡であることが報告された(Tang,T.ら.A mouse knockout library for secreted and transmembrane proteins.Nature Biotechnology,28,749−755,doi:10.1038/nbt.1644(2010))。一貫して、胎齢E18.5日でのKOマウスの胎仔及び胎盤の重量はWT同腹仔に類似した(図8a)。加えて、KOマウスは離乳後、運動機能不全を示し(Berger,J.H.,Charron,M.J.及びSilver,D.L.Major facilitator superfamily domain−containing protein 2a(MFSD2A)has roles in body growth,motor function,and lipid metabolism.PLoS One,7,e50629,doi:10.1371/journal.pone.0050629(2012))、尾懸垂の間、前足把持を伴った(図8b)。KOマウスの脳のサイズ及び重量はWT同腹仔よりも有意に小さかった(図9a、b)。しかしながら、WTとKOの脳の肉眼的解剖では目に見える差異はなかった(図9c、d)。興味深いことに、KOマウスの小脳はプルキンエ細胞の有意な喪失を示した(図1d、e)。さらに、海馬、特にCA1及びCA3の領域でニューロン細胞の密度に有意な低下があったが、KOマウスの歯状回(DG)では正常な細胞密度だった(図1f、g)。これらのデータはKOマウスが学習及び記憶に欠陥を有し得ることを示唆した。実際、行動試験は、KOマウスが学習、短期と長期の記憶に重度の欠陥を有し、同様に重度の不安を有することを示した(図10)。
ω−3脂肪酸の欠乏は齧歯類のモデルにて認知機能不全及び不安に関連付けされてきた(Lafourcade,M.ら.Nutritional omega−3 deficiency abolishes endocannabinoid−mediated neuronal functions.Nature Neuroscience,14,345−350,doi:10.1038/nn.2736(2011);Carrie,I.,Clement,M.,de Javel,D.,Frances,H.及びBourre,J.M. Phospholipid supplementation reverses behavioral and biochemical alterations induced by n−3 polyunsaturated fatty acid deficiency in mice.Journal of Lipid Research,41,473−480(2000))。これらのKO表現型は、Mfsd2aのKOマウスはω−3脂肪酸の脳でのレベルが低下し、他の脂質種で代替し得ることを我々に示唆した。我々は、WTマウス及びKOマウスにて質量分光分析によって脳、肝臓及び心臓の包括的なリピドミクス解析を行った。際立って、KOマウスの脳のPE、PC、PI及びPSの主要なリン脂質種のうちで他のω−3脂肪酸ではなくDHAがWTマウスに比べて有意に低下することを我々は見いだした(図2a及び図11には詳述)。DHAはPE、PC、PI及びPSにおけるリン脂質種38:6及び40:6として主に見いだされる(Kim,H.Y.Novel metabolism of docosahexaenoic acid in neural cells.The Journal of Biological Chemistry,282,18661−18665,doi:10.1074/jbc.R700015200(2007))(図2a及び図11)。DHAを含有する種の総レベルはWTマウスに比べてKOマウスの肝臓及び心臓では有意差はない(図2d、f及び図11)。しかしながら、KOマウスの脳におけるDHAの総レベルは、他の脂肪酸種における軽微な変化を伴っておよそ58.8%低下した(図2b及び図11)。KOマウスの脳はリン脂質のうちでアラキドン酸が33.8%増加した(図2c及び図11)が、それはDHA欠乏の齧歯類モデルで共通して増加する(Simopoulos,A.P.The importance of the omega−6/omega−3 fatty acid ratio in cardiovascular disease and other chronic diseases.Exp.Biol.Med.(Maywood)233,674−688,doi:10.3181/0711−MR−311(2008))。脳のDHAレベルを維持することにおいてMfsd2aの生理的な役割を強調するDHAが十分な餌でKOマウスが成長したことは注目に値する。KO胚の脳の発生において解剖上の変化が欠如しているにもかかわらず、脳のリン脂質における生化学的な変化はリン脂質のうちでDHAのレベルの有意な低下を伴って、胎齢e18.5日で明らかだった(図7c)ということは、胚発生の間でDHAレベルを維持することにおいてMfsd2aが本質的な役割を担うことを示している。
脳は多量の血漿由来のDHAを蓄積する。血漿では、血漿DHAの交換可能なブールがエステル化されていない脂肪酸としてまたはLPC17としてアルブミンで見いだされる。細胞系アッセイを用いて、我々は、Mfsd2aがエステル化されていないDHAまたは他のエステル化されていない脂肪酸を輸送しない(図12、表1)ことを見いだしたので、脂肪酸輸送体としてのMfsd2aを除外した。次に我々はMfsd2aがLPC形態でのDHCを輸送することができるかどうかを調べた。顕著なことに、Mfsd2aを発現している細胞は、対照細胞に比べてLPC−[14C]DHAの増強された濃度依存性の取り込みを示した(図3a)ということは、Mfsd2aが実際にLPC−DHAの輸送体であることを示している。MFSタンパク質におけるナトリウム結合に決定的である系統発生上保存された残基アスパラギン酸92と96(Granell,M.,Leon,X.,Leblanc,G.,Padros,E.及びLorenz−Fonfria,V.A.Structural insights into the activation mechanism of melibiose permease by sodium binding.Proceedings of the National Academy of Sciences of the United States of America,107,22078−22083,doi:10.1073/pnas.1008649107(2010))のアラニン変異誘発(D92A)及び(D96A)はそれぞれ、輸送の低下及び輸送の非存在を生じた(図3a)。D92A及びD96Aは野生型と類似の発現を有した(図13)。我々は次に血漿で見られる最も一般的なLPCであるLPC−オレイン酸及びLPC−パルミチン酸についてのMfsd2aの輸送特異性を調べた。野生型Mfsd2aを発現している細胞はLPC−[14C]オレイン酸及びLPC−[3H]パルミチン酸の濃度依存性の取り込みを示した(図3b、c)。ヒトMfsd2aも同様にLPCを輸送した(図14)。LPC−オレイン酸、LPC−パルミチン酸及びLPC−DHAのMfsd2aが介在する取り込みの比較は、Mfsd2aはLPC−DHAを輸送する最高の能力を有し、LPC−オレイン酸及びLPC−パルミチン酸がそれに続くことを示した(図3d)。さらに、野生型及び部分的に活性のあるD92A変異体を発現している細胞は経時的に飽和動態を示した(図15a)。重要なことに、LPC−[14C]DHA及びLPC−[14C]オレイン酸のMfsd2a依存性の輸送は、輸送されたLPCのPCへの迅速な変換を生じた(図3e、f、g、h)。さらに、Mfsd2aの発現がPC質量の増加を生じるということは、Mfsd2aの輸送活性が細胞へのLPCの正味の取り込みを生じることを示している(図15b)。
我々は次に、Mfsd2aによる輸送に必要とされるLPCリガンドのリガンド特異性及び化学的特徴を確定しようとした。この目標を実行するために、我々は、Mfsd2aを発現している細胞またはモック対照細胞を用いた競合アッセイを設定し、10倍過剰の非標識の競合相手の存在下または非存在下でそれらを25μMのLPC−[3H]パルミチン酸で処理した。我々は、14炭素の最小アシル鎖長を持つLPCが取り込みに対して効果的に競合することを見いだした(図17a、b、c)。リゾリン脂質LPE及びLPSが弱い競合を示した一方で、LPAはLPC−[3H]パルミチン酸の輸送に対して非競合性だった(図17c)。我々は、蛍光LPEを用いてMfsd2aが直接LPEを輸送できることを確認した(図18)。これらの結果は、ホスファチジルコリン頭基の両性イオンの電荷がリガンドの輸送に決定的であることを示している。さらに、短鎖脂肪酸及びグリセロホスファチジルコリンが単独では競合相手ではなかった(図17b、c)ということは、LPCの長いアシル鎖がリガンドの輸送に不可欠であるという結論を支持している。リゾプラスマローゲン、リゾ血小板活性化因子、及び血小板活性化因子が強力な競合相手であるということは、LPCのアシル鎖のカルボニル基は輸送に必要とされないことを示している(図17d、e、f)。リゾスフィンゴミエリンもLPC−[3H]パルミチン酸の輸送について競合したということは、グリセロール主鎖はリガンド機能に必要とされないことを示している(図17f)。LPAに類似して、スフィンゴシン−1−リン酸が競合相手ではない(図17f)ということは、ホスホコリン頭基のコリン部分がリガンドの輸送に必須であるという結論をさらに支持する。リゾ脂質様の界面活性剤を用いて、我々は、グリセロール主鎖及びカルボニル基ではなく、14炭素の最小のアシル側鎖及びホスホコリン頭基がLPCリガンドの必須の化学的特徴であること(図17g、h)を確認した。
我々は次に、脳へのLPC−DHAの輸送にMfsd2aが必要とされるかどうかを判定しようとした。Mfsd2aのKO及び野生型の同腹仔マウスにLPC−[14C]DHAを静脈注射した。意外なことに、Mfsd2aのKOマウスは野生型対照と比べて90%を超えてLPC−[14C]DHAの脳への取り込みを低下させた(図4a)。対照的に、KOマウスの末梢組織におけるLPC−[14C]DHAの取り込みは野生型対照に比べて低下しなかった(図4a、b)。エステル化されていないDHAは拡散を介して脳によって取り込まれ得る(Rapoport,S.I.,Chang,M.C.及びSpector,A.A. Delivery and turnover of plasma−derived essential PUFAs in mammalian brain.J.Lipid Res.42,678−685(2001))ので、DHA取り込みの拡散経路がMfsd2aのKOマウスでは変化するのかどうかを調べた。エステル化されていない[14C]DHAの脳による取り込みはLPC−[14C]DHAの取り込みに比べて有意に低かったが、野生型マウスに比べてKOマウスで低下したわけではなかった(図19)。総合して、これらの知見はMfsd2aのKOマウスにおける脳の低いDHAレベルへの因果関係を表しており、Mfsd2aは脳におけるLPC−DHAの生理的輸送体であるという結論を支持している。
我々は、餌DHA処理によってKOマウスをレスキューすることができるかどうかを調べた。妊娠前から授乳までのMfsd2aヘテロ接合体マウスの餌DHA処理、及び8週齢まで継続した離乳仔のDHA処理は、たとえば、脳のサイズ及び不安のようなKOの表現型をレスキューしなかった(図21a〜e)。重要なことに、胎齢e18.5日でのKO胎仔による餌DHAの脳による取り込みが野生型胎仔に比べて劇的に低下した(図21)ことは、KO胎仔における脳の低下したDHAレベル(図7b)に一致し、レスキューの欠如は、胚形成の間でのDHAの脳への輸送に関するMfsd2aの本質的な役割が原因で生じる可能性があることを示している。
本明細書で開示される知見は、ω−3脂肪酸が脳によって取り込まれる主要な経路はLPCへのその自然なコンジュゲーション及びMfsd2aによる脳への輸送を介することを示している。ω−3脂肪酸を含有するトリグリセリド及びコンジュゲートされていない脂肪酸はこの主要な経路によって輸送されないので、主として肝臓及び他の臓器によって取り込まれる。さらに、Mfsd2a欠損マウスは小さな脳及び神経学的欠損を有し、脳のDHAが欠乏する。我々は、LPC−DHA及び他の一般的なLPC−脂肪酸のようなLPC−脂肪酸の脳への取り込みは正常な脳の成長と機能に必須であると結論付けることができる。重要なことに、小児PNのケアの標準はLPC−脂肪酸及びLPC−DHAを提供しないが、それらは正常な脳の成長と機能に必要とされる。
本明細書で議論されるように、DHAは正常な脳の成長と認知機能に必須のω−3脂肪酸である(Kidd,P.M.(2007),Omega−3 DHA and EPA for cognition,behavior,and mood:clinical findings and structural−functional synergies with cell membrane phospholipids.Alternative medicine review:a journal of clinical therapeutic,12,207−227;Horrocks,L.A.,及びYeo,Y.K.(1999).Health benefits of docosahexaenoic acid (DHA).Pharmacological research:the official journal of the Italian Pharmacological Society 40,211−225.;Mozaffarian,D.,及びWu,J.H.(2011).Omega−3 fatty acids and cardiovascular disease: effects on risk factors,molecular pathways,and clinical events.Journal of the American College of Cardiology,58,2047−2067;Connor,W.E.(2000).Importance of n−3 fatty acids in health and disease.Am.J.Clin.Nutr.71,171S−175S)。脳におけるその重要性に一致して、DHAは脳のリン脂質で高度に濃縮される(Breckenridge,W.C.,Gombos,G.,及びMorgan,I.G.(1972).The lipid composition of adult rat brain synaptosomal plasma membranes.Biochim.Biophys.Acta.266,695−707;Innis,S.M.(2007).Dietary(n−3) fatty acids and brain development.The Journal of Nutrition,137,855−859))。脳のリン脂質における豊富な脂肪酸にもかかわらず、DHAは脳ではデノボ合成され得ず、血液脳関門(BBB)を越えて移入されねばならないが、脳におけるDHA取り込みのメカニズムは謎のままだった。本明細書で示されるように、我々は、我々が胎仔及び成人の脳へのDHAの取り込みのための主要な輸送体として微細血管の血液脳関門の内皮で発現されることを示した、主要ファシリテータスーパーファミリーのメンバーである以前オーファンだった輸送体Mfsd2aを特定した(Nguyenら.Nature,2014;509:503−6)。リピドミクス解析は、Mfsd2a欠損マウスが海馬や小脳におけるニューロン細胞の喪失及び神経学的な障害や重度の行動障害及び重要な脳サイズの低下を伴って脳におけるDHAの劇的に低下したレベルを有することを示した。驚くべきことに、細胞系の試験は、Mfsd2aがナトリウム依存性の方法でDHAをリゾホスファチジルコリン(LPC)の形態で輸送するが、エステル化されていない脂肪酸を輸送しないことを示した。特に、Mfsd2aは、たとえば、LPC−オレイン酸及びLPC−パルミチン酸のような長鎖脂肪酸を運ぶ一般的な血漿LPCを輸送した。重要なことに、KOマウスはLPC−DHA及び血漿に由来する他のLPCの脳への取り込みを劇的に低下させたということは、Mfsd2aがDHAの脳への取り込みに必要とされることを実証している。さらに、我々の知見は脳の成長と機能における血漿由来のLPCの予想外の本質的な生理的役割を明らかにしている。
この実施例では、我々は実施例9の結果をさらに詳しく述べた。我々は主として劣性の神経発達疾患の3396人の患者のコホートでエクソームの配列決定を行った。リゾホスファチジルコリン(LPC)−脂質について患者及びノックアウトマウスの血清を評価した。野生型または変異Mfsd2aをコードするcDNA構築物で細胞に形質移入し、脂質の取り込みをモニターした。
[試験の管理]
試験はヘルシンキ宣言の規定を順守して実施された。サンディエゴ大学の施設内倫理委員会及びエジプト及びトリポリの子供病院の倫理委員会が試験プロトコールを認可した。人員確保は、疾患の遺伝的基盤を特定するためにエクソームの配列決定が行われた3396人の家族から成る神経発達疾患の大きな試験の一部だった。試験の参加者または指名者すべてから書面でのインフォームドコンセントを得た。
我々は、小頭症、乏しい頭部制御と躯幹筋緊張低下を伴った痙性四肢麻痺、発達遅延、知的障害、及び脳室拡大、と同様に脳梁と脳幹の低形成を類似して提示する2つの家族を特定した(表5)。
各家族における両親と冒されたメンバー1人を含む6人のDNA試料が遺伝的検討で利用可能だった。これらの試料は、配列解析の比較のために民族的に一致した個体として使用された1349人のエジプト人と93人のリビア人を含む遺伝子研究を受けた3396人の患者が関与するさらに大きな取り組みの一部だった。各家族からの1人が比較ゲノムハイブリッド形成(CGH)解析及び核型分析を受けたが、構造的な染色体異常は検出されなかった。
我々は各家族からの冒されたメンバーにて全エクソーム配列決定(WES)を行った。家族1825では、我々はさらに両親のWESを行い、結果の特異性を高めた。ゲノムDNAをAgilent Human All Exon 50Mbキットのライブラリ調製、次いでIllumina Hiseq2000機器における両末端配列決定(2×150bp)に供した。各患者の試料について>90%のエクソームが>30×でカバーされた。変異体の特定にはGATKを用いた(DePristo,MA.Banks,E.Poplin,Rら.A framework for variation discovery and genotyping using next−generation DNA sequencing data.Nat.Genet.2011;43:491−8)。我々はXHMMを用いて分離している稀な構造的変異体について調べた(Fromer,M.Moran,JL.Chambert,Kら.Discovery and statistical genotyping of copy−number variation from whole−exome sequencing depth.American Journal of Human Genetics,2012;91:597−607)。次いで我々は、カスタムPythonスクリプトを用いてホモ接合性変異体についてフィルターをかけ、集団にて0.1%を超える頻度でホモ接合体区間に存在しない、またはタンパク質機能への損傷の可能性について高いスコアではない対立遺伝子を取り除いた。家族1422及び1825にて新規の変異はMfsd2a遺伝子において特定された。コホートの他のメンバーは推定上の有害な変異体を提示しなかった。

我々はインビトロの実験はすべて4つ組で行った。データは平均値及び標準誤差として表す。我々はスチューデントのt検定(両側)を用いて群間の比較を行った。複数の比較については、我々はGraphPadPrismソフトウエアを用いた分散分析と併せてターキーの検定を行った。注射されたmRNAに関わりなく、受精後1、2及び3日のMO注射の後の生存についてKaplan−Meier曲線を算出した。GraphPadPrismソフトウエアを用いたロングランク(Mantel−Cox)検定を用いて生存曲線を比較した。p値<0.05が有意差を示すと見なした。p値はすべて両側性で調べた。
[試験集団]
試験に動員された3396人の患者は、進行性の神経学的経過に対する固定した神経学的経過、癲癇の存在、自閉症または知的障害、肉眼的な醜形特徴、または、たとえば、脳MRI、EEG若しくは血液の化学的分析のような診断試験における顕著な知見に基づいて大部分は識別できる神経発達障害の多数の個々に稀な形態を表した。2つの家族が、思い返せば、小頭症、混合型筋緊張低下/痙性四肢麻痺、頭部制御の不在、癲癇性発作及び大きく拡張した脳室を含む多数の類似の特徴を示したものの、日常検査の際の普通でないまたは特徴的な臨床知見の非存在を考えると、彼らの臨床所見をコホートの残りから識別するのは困難だった。
我々は、変異型(5000人の我々の研究室内エクソームデータまたは4000人のNHLBIのデータベースにおける>0.1%の対立遺伝子頻度)及び両親にてヘテロ接合性ではない変異型を取り除くことによって冒されたメンバーにおけるWESからデータをフィルターにかけた(Tennessen,JA.Bigham,AW.O'Connor,TDら.Evolution and functional impact of rare coding variation from deep sequencing of human exomes.Science,2012;337:64−9)。我々は各家族のメンバーにて稀なタンパク質を変化させる変異型を特定した。これらの変異型の中で、家族1825にてたった4つが遺伝の様式に一致し(表7)、そのうちのたった1つが分離解析をパスし、それは、Mfsd2a遺伝子におけるchr1:40431005C>T変異型であり、c.476C>Tヌクレオチド及びp.T159Mタンパク質の変化につながった。この特定の後、我々はデータベースを検索し、家族1422が、c.497C>Tヌクレオチド及びp.S166Lタンパク質の変化につながる、同一遺伝子におけるchr1:40431162C>T変異型を抱くことを見いだした(図23C)。2つの家族の表現型の比較はそれらが正確に一致することを示した。双方の変異型は、標準のプログラムを用いて高い損傷予測を示し(表7、表8)、ホモ接合性のブロックに存在し(図26)、脊椎動物の進化の全体を通して完全に保存され、タンパク質の第4膜貫通ドメインに位置するアミノ酸残基(図23D、E)の中にあった。双方の変異は、構成的にスプライスされるエクソンの中にあり、遺伝の厳格な劣性様式に従って各家族で分離した(図27)。これらの変異型は、ほぼ10,000の染色体の我々の内部データセット、または公的に利用できるデータベースには存在しなかった。
本明細書で議論されるように、Mfsd2aは、マウスにてBBBの形成及び機能に最近関わるとされ、DHAの輸送に必要とされる12回膜貫通型タンパク質をコードする(Nguyen,LN.Ma,D.Shui,Gら.Mfsd2a is a transporter for the essential omega−3 fatty acid docosahexaenoic acid.Nature,2014;509:503−6;Ben−Zvi,A.Lacoste,B.Kur,Eら.Mfsd2a is critical for the formation and function of the blood−brain barrier.Nature,2014;509:507−11)。HEK293細胞におけるMfsd2aの発現は、残基N217及びN227におけるグリコシル化のためにSDS−PAGE上で約55kDaと約70kDaの2つのアイソフォームに分割される(Berger,JH.Charron,MJ.Silver,DL.Major facilitator superfamily domain−containing protein 2a(MFSD2A)has roles in body growth,motor function,and lipid metabolism.PloS One,2012;7:e50629)。HEK293細胞にて発現されたヒトのMfsd2aタンパク質にp.T159M及びp.S166Lの変異を導入し、ウエスタンブロット及び免疫蛍光法で調べた。双方とも類似のレベルで発現され、WTと同一の翻訳後修飾を示し、グリコヒドロラーゼPNGaseF処理に続いて低分子量の実体に分割した(図24A)。さらに、双方の変異体タンパク質は安定して発現され、WTと類似の方法(図24B)で原形質膜に部分的に局在化した。不活化している変異のために分子基盤を提供するために、我々は、MFSファミリーの大腸菌のナトリウム−メリビオース輸送体部分であり、Mfsd2aと高い配列類似性を共有するMelBの構造情報を利用した。p.T159及びp. S166は双方とも膜貫通ドメイン4に存在し、それぞれナトリウム結合及びリガンド結合を伝達する(Ethayathulla,AS.Yousef,MS.Amin,A.Leblanc,G.Kaback,HR.Guan,L.Structure−based mechanism for Na(+)/melibiose symport by MelB.Nat.Commun.2014;5:3009;Cordat,E.Leblanc,G.Mus−Veteau,I.Evidence for a role of helix IV in connecting cation− and sugar−binding sites of Escherichia coli melibiose permease.Biochemistry,2000;39:4493−9)。ヒトの残基p.T159はMelBの残基p.T221に保存され、p.T159M変異はナトリウム結合の相互作用を破壊させると予測される一方で、p.S166変異はMelB(p.W228)にて保存され、p.S166L変異はLPCの結合を妨害すると予測される(図24C)。
我々は、BBBでのMfsd2aによる血漿脂質の取り込みが血漿LPCのレベルに影響を与えるので、Msfd2a欠乏は高い血漿LPCレベルを生じるはずであるという仮説を立てた。実際、我々は、Msfd2aのKOマウスが対照に比べて40%高い血漿総LPCレベルを示すことを見いだした(図25A)。Msfd2aのKOマウスはまた対照に比べて個々のLPCの高いレベルも有した(図25B)。LPC−DHAのような他の豊富ではない血漿LPC種はMsfd2aのKOマウスでは高いレベルに向かう傾向を示した(図25B)。Msfd2aのKOマウスにおける高い血漿LPCの定常状態レベル、及びMsfd2aのKOマウスにおけるLPCの脳による取り込みがLPC種に応じて85〜90%の間で低下したという知見に一致して、静脈内注射したLPC−[14C]オレイン酸の追跡試験は、注射後2時間でのMsfd2aのKOマウスにおける血漿LPC−[14C]オレイン酸の高いレベルを示した(図25C)。これらの結果を考えると、我々は患者がヘテロ接合性の両親及び健常な年齢の一致した対照と比べてLPCの高い血漿レベルを有するかどうかを調べた。リピドミクス解析は、ヘテロ接合性の両親及び対照と比べて総血漿LPCは発端者で増加することを示した(図25D)。Msfd2aのKOマウスにおける知見に類似して、16:0、18:0、18:1及び18:2の長さの脂肪酸を含有する一般的な血漿LPC種はMsfd2a変異体の患者の血清で増加したということは、LPC取り込みの欠損を示唆している(図25E)。
我々は、不活化変異p.T159M及びp.S166Lのための分子基盤を提供するためにMelBの詳細な構造情報を利用した。MFSファミリーの輸送の全体的なメカニズムは大腸菌由来のグリセロール−3−リン酸輸送体GlpTのX線構造から最初に推論され、他のMFSファミリーメンバーの及びさらに最近、Mfsd2aの密接なオルソログであるMelBを含む構造によって確認された(Ethayathulla,AS.Yousef,MS.Amin,A.Leblanc,G.Kaback,HR.Guan,L. (b)ヒトの野生型のMfsd2aのcDNAを含む細胞及びヒトの変異Mfsd2aのcDNAを含む細胞またはモックで形質移入した細胞にて、試験化合物の存在下及び非存在下で、LPC−パルミチン酸、LPC−オレイン酸、LPC−ステアリン酸、LPC−リノール酸、LPC−リノレン酸、LPC−アラキドン酸、LPC−ドコサヘキサエン酸、または誘導体の取り込みを測定することとを含み、
試験化合物の非存在下に比べて試験化合物の存在下で、ヒトの野生型のMfsd2aのcDNAを含む細胞へのLPC−パルミチン酸、LPC−オレイン酸、LPC−ステアリン酸、LPC−リノール酸、LPC−リノレン酸、LPC−アラキドン酸、LPC−ドコサヘキサエン酸、または誘導体の取り込みの増加したレベルまたは低下したレベルが、Mfsd2aタンパク質による輸送の調節因子としての化合物を示す、前記スクリーニング方法。 - 細胞がHEK293である、請求項19に記載の方法。
- ヒトの変異Mfsd2aのcDNAがヒトのMfsd2aタンパク質配列におけるD93またはD97に相当する位置で変異を含む、請求項19に記載の方法。
- 試験化合物がMfsd2aタンパク質により直接輸送される、請求項19に記載の方法。
- 臓器の画像化法であって、標識した足場またはコンジュゲートを対象に投与することと、対象臓器にて前記標識した足場またはコンジュゲートのMfsd2aタンパク質による取り込みまたはMfsd2aタンパク質との相互作用を測定することとを含む、前記画像化法。
- 足場がLPCである、請求項23に記載の方法。
- 標識が蛍光である、請求項23に記載の方法。
- 標識がフッ素化される、請求項23に記載の方法。
- 臓器が脳または眼である、請求項23に記載の方法。
- 標識された足場がTop−Fluor−LPCまたはNBD−LPCである、請求項23に記載の方法。
- Mfsd2aタンパク質による化合物の輸送方法であって、足場またはコンジュゲートを取り込ませるのに十分な条件下で対象に前記足場またはコンジュゲートを提供することを含む、前記輸送方法。
- 足場がLPCである、請求項29に記載の方法。
- 化合物が、LPCのω炭素を介してLPCにコンジュゲートされる、請求項30に記載の方法。
- 足場またはコンジュゲートがBBBを越えるまたはBBBで蓄積する、請求項29に記載の方法。
- 足場またはコンジュゲートが脳または眼にて蓄積する、請求項29に記載の方法。
- 化合物にコンジュゲートされた足場を含む組成物。
- 足場がLPCである、請求項34に記載の組成物。
- 化合物がLPCのω炭素を介してLPCにコンジュゲートされる、請求項35に記載の組成物。
- 化合物が医薬剤である、請求項34に記載の組成物。
- 化合物が造影剤である、請求項34に記載の組成物。
- 対象における神経学的欠損に対する増加した易罹患性の評価方法であって、
(a)Mfsd2a遺伝子の全部または一部を含む、対象に由来する生物試料を提供することと;
(b)試料におけるMfsd2a遺伝子またはその一部にて変異または多型の存在を検出することと、
(c)Mfsd2a遺伝子またはその一部における変異または多型の存在に基づいて、対象が神経学的欠損に対する増加した易罹患性を有することを評価することとを含む、前記評価方法。 - 変異がThr159MetまたはSer166Leuである、請求項39に記載の方法。
- 多型が表4、6または7にて列記される単一ヌクレオチド多型の1以上である、請求項39に記載の方法。
- 変異が機能の喪失を生じる、請求項39に記載の方法。
- 変異がMfsd2aの低次形態対立遺伝子である、請求項39に記載の方法。
- 神経学的欠損が記憶及び学習における欠損または不安である、請求項39に記載の方法。
- 対象が受胎前のまたは妊娠中の女性である、請求項39に記載の方法。
- Mfsd2a遺伝子またはその一部において変異または多型が存在するならば、高DHA食を投与すること、またはLPCを用いた静脈内処置若しくは腸内処置を施与することをさらに含む、請求項45に記載の方法。
- LPCがLPC−DHAを含む、請求項46に記載の方法。
- 対象は、認知機能に関する問題があると診断された小児または成人である、請求項39に記載の方法。
- 認知機能が学習障害または不安である、請求項48に記載の方法。
- Mfsd2a遺伝子またはその一部において変異または多型が存在するならば、高DHA食を投与すること、またはLPCを用いた静脈内処置若しくは腸内処置を施与することをさらに含む、請求項48に記載の方法。
- LPCがLPC−DHAを含む、請求項50に記載の方法。
- 検出することが、Mfsd2a遺伝子またはその一部と優先的にハイブリッド形成するオリゴヌクレオチドプローブに試料を接触させることを含む、請求項39に記載の方法。
- 検出することが、Mfsd2a遺伝子またはその一部をPCRによって増幅することを含む、請求項39に記載の方法。
- 検出することが、Mfsd2a遺伝子、その一部、または相当するMfsd2aのcDNAもしくはその一部の配列を決定することを含む、請求項39に記載の方法。
- 対象に由来するMfsd2aタンパク質の輸送機能の評価方法であって、
(a)第1の細胞にて試験Mfsd2aのcDNAを、第2の細胞にて野生型Mfsd2aのcDNAを発現させることと、
(b)試験Mfsd2aのcDNAを発現する第1の細胞及び野生型Mfsd2aのcDNAを発現する第2の細胞をLPC−DHAまたはLPC−ω3脂肪酸に接触させることと、
(c)試験Mfsd2aのcDNAを発現する第1の細胞及び野生型Mfsd2aのcDNAを発現する第2の細胞へのLPC−DHAまたはLPC−ω3脂肪酸の取り込みを測定することとを含み、
野生型Mfsd2aのcDNAを発現する第2の細胞に比べて試験Mfsd2aのcDNAを発現する第1の細胞へのLPC−DHAまたはLPC−ω3脂肪酸の取り込みの低下したレベルは、試験Mfsd2aのcDNAが輸送に不十分なタンパク質をコードすることを示す、前記評価方法。 - 試験Mfsd2aのcDNAが、Thr159MetまたはSer166Leuの変異をコードする、請求項55に記載の方法。
- 試験Mfsd2aのcDNAが、表4、6または7に列記される多型の1以上をコードする、請求項55に記載の方法。
