Abstract
Description
(i)セルロースナノ結晶を除く単一の多糖または少なくとも2つの異なる多糖に由来するナノ結晶、または
(ii)セルロースに由来するナノ結晶と組み合わせた、セルロース以外の単一の多糖または少なくとも2つの異なる多糖に由来するナノ結晶、
を含有する。
(i)セルロースナノ結晶を除く正に荷電した多糖ナノ結晶のみ、または、
(ii)少なくとも負に荷電した多糖に由来するナノ結晶と混合された、セルロース以外の正に荷電した多糖ナノ結晶、
から選択される。
a)セルロース以外の多糖のナノ結晶を親水性相中に取り込む工程と、
b)多糖ナノ結晶を含有する前記親水性相および内相(特に疎水性内相)の構成のための相を提供する操作と、
c)前記親水性相中に前記内相を分散させることによって前記組成物を形成する操作と、
を含むことを特徴とする。
(i)セルロースナノ結晶を除く正に荷電した多糖ナノ結晶のみ、または
(ii)少なくとも負に荷電した多糖に由来するナノ結晶と混合された、セルロース以外の正に荷電した多糖ナノ結晶、
から選択される。
c.1)少なくとも5/95の疎水性内相/親水性連続相の体積比を有する中間体の水中油エマルションを得る工程と、
c.2)50%より高い内相のパーセンテージを有するエマルション組成物を得る工程であって、
c.2.1)ある体積の疎水性相を工程c.1)において得られた中間体エマルションへ添加し、このようにして得られた混合物を撹拌する工程、および/または
c.2.2)少なくとも前記親水性相の一部の除去によって工程c.1)において得られた中間体エマルションを濃縮する工程
を含む工程と、

−MIPE/HIPEエマルションの高安定性を生じる不可逆的吸着(界面からのナノ結晶の脱離のために必要とされる解離エネルギーは、この脱離が自然に起こらないようなものであると思われる)、
−非毒性、
−生物分解性、
−生物学的源からのもの、
−後処理によって容易に修飾される。
したがって、本発明に記載の組成物は、親水性連続相中で分散された内相を含み、内相のパーセンテージは50%より高い。
−中間内相(MIPE)または高内相(HIPE)タイプのエマルション組成物(その中で前記内相は疎水性液相によって構成される)、または
−フォーム形態における組成物(その中で前記内相は気相によって構成される)
である。
本発明に記載の組成物は、有利には、エマルションまたはフォームである。

したがって3つのタイプのフォーム構造は以下のように定義される。
Φl*<Φlならば、泡沫は球であり接触はなく、泡立つ液体に対応し、
0.05<Φl<Φl*ならば、泡沫は接触し、潰れた球体の形であり、ウェットフォームに対応し、
Φll<0.05ならば、泡沫は接触し多面体の形であり、ドライフォームに対応する。
−水中油タイプのエマルションの分散された油相、または
−水性液体フォーム中で分散された気体
を指すことができる。
本発明によれば、「増加させた内相」を含む組成物は50%より高い(好ましくは55%より高い)内相のパーセンテージを有する。
いくつかの実施形態において、60/40より高い(より好ましくは80/20より高い)疎水性内相/親水性連続相の体積比を有するMIPEエマルションまたはHIPEエマルション組成物が形成される。
したがって組成物(特にエマルション組成物)は、少なくともそれらのうちのいくらかについてはセルロースのナノ結晶以外のナノ結晶である、多糖のナノ結晶によって安定化される。
(i)セルロース以外の多糖または多糖の混合物に由来するナノ結晶、または
(ii)セルロースに由来するナノ結晶とブレンドされた、セルロース以外の多糖または多糖の混合物に由来するナノ結晶
を含有し得る。
−グルカン(D−グルコースのポリマー)
−ガラクタン(D−ガラクトースのポリマー)
−キシラン(D−キシロースのポリマー)、および
−特にキチンを含む、D−グルコサミンのポリマー
から得られるナノ結晶から選択される。
−デンプン(α−1,4−グルカンおよびα−1,6−グルカン)
−アミロペクチン(α−1,4−グルカンおよびα−1,6−グルカン)
−アミロース(α−1,4−グルカン)
が含まれる。
−カードラン(β−1,3−グルカン)
−ラミナリン(β−1,3−グルカンおよびβ−1,6−グルカン)
−レンチナン(純粋なβ−1,6:β−1,3−グルカン)
−パミロン(pamylon)
−プレウラン(pleuran)(β−1,3−グルカンおよびβ−1,6−グルカン)

(ii)有利には負に荷電した少なくとも別の多糖(例えばセルロース)からのナノ結晶と混合された、有利には正に荷電したキチンナノ結晶
を含有し得る。
−他の乳化化合物または安定化化合物、
−または他の固形微粒子(前記固形微粒子は官能化または非官能化のいずれかである)
を添加せずに、キチンナノ結晶のみによって安定化される。
多糖のナノ結晶(特にキチン)は、高度に結晶性の固形微粒子である。
−カニ殻。240nm×15nm(Nair and Dufresne,Biomacromolecules 2003)もしくは260±80nm×23±3nm(Belamie et al.J.Phys.Chem.2004)、または
−エビ殻。200〜560nm×18〜40nm(Phonggying et al.Polymer 2007)。
組成物安定性を至適化するために、多糖ナノ結晶は、有利には、特に静電的な態様を考慮に入れて、それらの表面特徴に従って選択される。
組成物は、有利には、前記組成物の全重量に基づいて、重量で0.035%〜2%(より好ましくは重量で0.05%〜1%)の多糖(有利にはキチン)のナノ結晶を含む。

本出願人は、エマルション組成物安定性が規定された最小イオン強度の水相の使用によって増加できることも示した。
エマルション組成物の構造を定義するために、親水性相中で分散された疎水性相を構成する脂質小滴のサイズ分布に関する特徴を、特に使用することができる。

疎水性相は、植物油、動物油、鉱物油、合成油、疎水性有機溶媒および疎水性液体ポリマーから選択される。
「親水性相」または「水相」によって、疎水性相と不混和性の液体が意味される。好ましくは水と混和性の親水性相が使用される。親水性相は、実施例において示されるように水であり得る。
本発明は、上で説明された組成物を得るためのプロセスも高める。
a)セルロース以外の多糖(有利にはキチン)のナノ結晶を親水性相中に取り込む工程と、
b)(i)多糖ナノ結晶を含有する前記親水性相および(ii)内相の構成のための相(必要に応じて疎水性液相または気相)を提供する操作と、
c)前記親水性相中に前記内相を分散させることによって前記組成物を形成する操作と
を含む。
いくつかの多糖(特にキチン)は「非晶性」と呼ばれる1つの部分から成り、一方第2の部分は「結晶性」である。
「連続的な」または「2工程における」と呼ばれる実施形態によれば、親水性相中で疎水性相を分散させることによってエマルション組成物を形成するための操作c)は、以下の2つの連続的工程、
c.1)ピッカリングタイプの中間体の水中油エマルションを得るために、少なくとも5/95の疎水性相/親水性相の体積比で、前記疎水性相中で前記親水性相を分散させる工程、続いて
c.2)必要に応じて、MIPEタイプまたはHIPEタイプの50%より高い内相のパーセンテージを有する対象となるエマルション組成物を得る工程であって、
c.2.1)ある体積の疎水性相を工程c.1)において得られた中間体エマルション組成物へ添加し、このようにして得られた混合物を撹拌する工程、および/または
c.2.2)少なくとも前記親水性相の一部の除去によって工程c.1)において得られたエマルション組成物を濃縮する工程
を含む工程
から成る。
本発明に記載のMIPEエマルションまたはHIPEエマルションを得るために使用されるピッカリング中間体エマルションは、水相中で分散された疎水性相を含み、セルロース以外の多糖のナノ結晶を含む乳化粒子(言いかえれば「エマルション粒子」と呼ばれる)を含有する、エマルション形における組成物である。
ピッカリング中間体エマルション組成物を製造するプロセスは、有利には、以下の工程
(a)上で定義されるようなセルロース以外の多糖のナノ結晶(有利にはキチンのナノ結晶)を提供すること、および次いで
(b)前記ピッカリング中間体エマルション中で少なくとも50%(好ましくは少なくとも80%)の被覆率を生成することができる質量の量で前記組成物の水相中に前記ナノ結晶を取り込むことおよび前記中間体エマルションを安定化すること
を含む。
ピッカリング中間体エマルションの安定条件が満たされる(工程c.1))ならば、工程(複数可)を介してプロセスを継続して、MIPEエマルションまたはHIPEエマルションを形成することができる(工程c.2))。
c.2.1)ある体積の疎水性相を工程c.1)において得られた中間体エマルション組成物へ添加することによって、および/または
c.2.2)少なくとも前記親水性相の一部の除去によって工程c.1)において得られた中間体エマルション組成物を濃縮することによって
得ることができる。
内相小滴が可変的なサイズであること、および
内相小滴が膨潤し、次いでそれらが歪むこと
が可能である。
第1の実施形態によれば、疎水性相の添加によってプロセスを継続して、MIPEまたはHIPEエマルションを形成することができる(工程c.2.1))。
第2の実施形態によれば、プロセスはMIPEまたはHIPEエマルションを形成するために濃縮工程により継続することができる(工程c2.2))。
−クリーム分離/重力沈降、
−遠心分離(例えば2000g10分間)、
−濾過(有利には従来の多孔質膜システムまたは連続的な限外濾過)、
−浸透法、
−凍結濃縮法または乾燥(連続相のみが蒸発する条件において)
から例えば選択される適切な技法による親水性連続相の除去をもたらす。
−各々の構成要素(親水性相、疎水性相、ナノ結晶)の濃度、
−エマルションを得るプロセス(超音波、ローター−ステーターなど)および、
−使用した条件(速度、時間周期、温度、エネルギー、体積など)、
に依存する。
予想外に、本発明者は、適切な割合において疎水性相を親水性相(ナノ結晶を含有する)と直接混合することによって、キチンナノ結晶が高内相エマルションを提供することを観察した。
本記述中で既に以前に言及され、実施例において例証されるように、本発明に記載のMIPEエマルションまたはHIPEエマルションは、例えばMIPEエマルションまたはHIPEエマルションを単純に凍結乾燥することによってソリッドフォームまたはソリッドエマルションを調製して、得ることができる。
a)本記述中で定義されるようなMIPEエマルションまたはHIPEエマルション、好ましくは本記述中で規定されたプロセスに従って得られるMIPEエマルションまたはHIPEエマルションを提供する工程、
b)微粒子ソリッドフォームを得るために蒸発(好ましくは凍結乾燥)によって親水性相および前記MIPEエマルションまたはHIPEエマルションの疎水性相を除去する工程
を含む多糖のソリッドフォームを調製するプロセスを提供することである。
A.プロトコル
プロトコル1:キチンナノ結晶の調製
キチンナノ結晶を得るプロセスは、文書“In vitro chiral nematic ordering of chitin crystallites”,Revol,J.−F.;Marchessault,R.H.;Int.Journal of Biological Macromolecules,1993,15,329−335、および“Structure and chirality of the nematic phase in chitin suspensions”Belamie,E;Davidson,P;Giraud−Guille,M.M.Journal of Physical Chemistry B,2004,108(39),14991−15000中で記載される。
20μLのキチンナノ結晶の水性懸濁物(0.0025%重量/体積)を、電子顕微鏡法のためにカーボングリッド上に沈着させる。
このH/Eエマルションは2つの異なるプロトコル(すなわち2工程または単工程)に従って調製することができる。
第1の水中油のピッカリング・エマルションは、0.01mM HClおよび20mM NaCl中の既知の濃度のキチンナノ結晶を含有する水相の使用によって調製される。
水相および油相をチューブ中に直接置く。
約20μLのエマルションサンプルを1mLの蒸留水の中へ取り込む。
走査電子顕微鏡(SEM)による観察のためのエマルションサンプルを調製するために、スチレン/開始剤(st:AIBN(アゾビスイソブチロニトリル)100:1重量/重量の比)の150μLの混合物を事前に1〜2分間超音波へさらし、1.0〜1.5mLの0.3%溶液の20mM NaCl中の0.01mM HClサンプル溶液と混合する。混合物を10分間窒素により脱気する。
内相パーセンテージは、エマルションの全体積によって割ったエマルションの油体積に等しい。
キチンナノ結晶の脱アセチルプロセスはFan et al.,Carbohydrate polymers,2012,79,1046−1051中に記載される。
10mLチューブ中のエマルションを2000gで2〜5分間遠心分離する。
フォーム形成アッセイは、文書Alargova et al.“Foam superstabilization by polymer microrods”,Langmuir,vol.20(24),2004,10371−10374中に記載されるプロトコルに基づいて実行された。
B.1.ピッカリング・エマルションについての異なるパラメーターの関数としての平均直径の変遷
エマルションの安定性を査定する方法は、小滴直径を測定して合体についてチェックすることである。
得られた結果を以下の表1および図1中で示す。

本標準条件(20mM NaClおよび0.01mM HCl中の3g/Lのキチンナノ結晶)におけるサンプルを異なる温度で1週間保存した。

50mM NaClおよび0.01mM HCl中の5g/Lのキチンナノ結晶のエマルションサンプルを、50℃で8時間、次いで25℃で5日間(均一に乾燥するのに必要とされる時間)プラグを抜いたチューブ中で維持した。

エマルションは上述のプロトコルに従って調製され、24時間の待ち時間後に体積を測定して内相のパーセンテージを計算する(プロトコル6)。

キチンナノ結晶およびシクロヘキサンに基づいて調製されたエマルションは凍結され、次いで凍結乾燥された(プロトコル8)。
ナノ結晶キチン/セルロースの混合エマルションは「2工程」における上述のプロトコル3(a)に従って調製され、その結果、20mM NaClおよび0.01mM HClを含有する水相中の1.5g/Lのキチンナノ結晶および1.5g/Lのセルロースナノ結晶の組み合わせを得た。

ドライフォーム形成アッセイは上述のプロトコル9に従って実施された。
−2g/L〜13g/Lのキチンナノ結晶濃度、
−0〜5 mMのNaClの間のイオン強度、
−0.01mM HCL(pH5)、0.1mM HCl(pH4)および1mM HCl(pH3)のpH
が調べられた。
得られた結果を、全エマルション体積と比較したフォーム体積分率として、表11中で以下に示す。

2g/L〜13g/Lのキチン溶液(0.01mM HClおよび5mM NaCl)は、前述のプロトコル9に従って水平撹拌へさらされる。
Claims (16)
- 親水性連続相中で分散された内相を含む組成物であって、内相のパーセンテージが50%より高く、前記組成物が、前記内相と前記親水性連続相との間の界面に位置する、セルロース以外の多糖のナノ結晶を含有することを特徴とする組成物。
- 前記セルロース以外の多糖のナノ結晶が、正に荷電したナノ結晶から選択されることを特徴とする、請求項1に記載の組成物。
- 前記多糖のナノ結晶が、
(i)セルロースナノ結晶を除く正に荷電した多糖ナノ結晶のみ、または
(ii)少なくとも1つの負に荷電した多糖に由来するナノ結晶と混合された、セルロース以外の正に荷電した多糖ナノ結晶
から選択されることを特徴とする、請求項2に記載の組成物。 - 前記セルロース以外の多糖のナノ結晶が、キチンナノ結晶から選択されることを特徴とする、請求項2または3のいずれかに記載の組成物。
- エマルションまたはフォームであることを特徴とする、請求項1〜4のいずれかに記載の組成物。
- 前記組成物が、(i)75%より高い内相のパーセンテージを有する高内相もしくはHIPEタイプ、または(ii)50%〜75%の間の範囲の内相のパーセンテージを有する中間内相もしくはMIPEタイプのエマルションであることを特徴とする、請求項1〜5いずれかに記載の組成物。
- ドライエマルション、ソリッドフォーム、多孔質ポリマー材料またはポリマー材料ビーズから選択される、請求項1〜6のいずれかに記載の組成物から得られる産物。
- 親水性連続相中で分散された内相を含む組成物を得るプロセスであって、内相のパーセンテージが50%より高く、前記プロセスが、以下の操作
a)セルロース以外の多糖のナノ結晶を親水性相中に取り込む操作と、
b)多糖ナノ結晶を含有する前記親水性相および前記内相の構成のための相を提供する操作と、
c)前記親水性相中に前記内相を分散させることによって前記組成物を形成する操作と
を含むことを特徴とするプロセス。 - 操作a)において取り込まれる前記多糖ナノ結晶が、正に荷電したナノ結晶から選択されることを特徴とする、請求項8に記載のプロセス。
- 操作a)において取り込まれる前記多糖ナノ結晶が、
(i)セルロースナノ結晶を除く正に荷電した多糖ナノ結晶のみ、または
(ii)少なくとも1つの負に荷電した多糖に由来するナノ結晶と混合された、セルロース以外の正に荷電した多糖ナノ結晶
から選択されることを特徴とする、請求項9に記載のプロセス。 - 操作a)において取り込まれる前記多糖ナノ結晶が、キチンナノ結晶から選択されることを特徴とする、請求項9または10のいずれかに記載のプロセス。
- 前記得られた組成物が、操作c)の終了時にエマルションまたはフォームであることを特徴とする、請求項8〜11のいずれかに記載のプロセス。
- 前記組成物が、疎水性内相および親水性連続相を含むエマルション組成物であり、前記エマルションを形成する操作c)が
c.1)少なくとも5/95の疎水性内相/親水性連続相の体積比を有する中間体の水中油エマルションを得る工程と、
c.2)50%より高い内相のパーセンテージを有するエマルション組成物を得る工程であって、
c.2.1)ある体積の疎水性相を工程c.1)において得られた中間体エマルションへ添加し、このようにして得られた混合物を撹拌する工程、および/または
c.2.2)少なくとも前記親水性相の一部の除去によって工程c.1)において得られた中間体エマルションを濃縮する工程
を含む工程と、
を含むことを特徴とする、請求項8〜12のいずれかに記載のプロセス。 - 前記中間体の水中油エマルションが、工程c.1)において50%以下である内相のパーセンテージを有することを特徴とする、請求項13に記載のプロセス。
- 前記組成物が、疎水性内相および親水性連続相を含むエマルション組成物であり、前記エマルションを形成する操作c)が、多糖ナノ結晶含有親水性相および疎水性内相を混合して、50%より高い内相のパーセンテージを有する前記エマルションを直接得ることを特徴とする、請求項8〜12のいずれかに記載のプロセス。
- 前記形成されたエマルション組成物が、工程cの終了時に、(i)75%より高い内相のパーセンテージを有する高内相もしくはHIPEタイプ、または(ii)50%〜75%の間の範囲の内相のパーセンテージを有する中間内相もしくはMIPEタイプであることを特徴とする、請求項8〜15のいずれかに記載のプロセス。
