Description
材料
Arenicola marina(M101):
本研究では、100mg/ml及び178mg/mlの濃度の2つのバッチを用いた。M101を、−80℃で保存し、研究に際しては4℃で解凍する。
HT29ヒト結腸腺癌細胞(ATCC、HTB−38)を、20%FBS、1%L−グルタミン、及び1%抗生物質(ペニシリン及びストレプトマイシン)を補ったDMEM中、37℃で、水飽和雰囲気下、5%CO2/95%空気中で、増殖させた。
腫瘍組織の酸素供給の動態
GLUT−1マーカーの免疫組織学的定量化により皮下HT29腫瘍における低酸素症の程度の軽減を評価することにより、M101の酸素供給能をインビボで決定した(下記表1)。
組織低酸素症の程度は、以下のスケールにより決定する。
「+++」=非常に強いGlut−1標識、非常に高い組織低酸素症
「++/−」=強いGlut−1標識、高い組織低酸素症
「+」、「+/−」=中程度のGlut−1標識、軽減された組織低酸素症
「+/−」=低いGlut−1標識、低い組織低酸素症

- 癌細胞における低酸素症を軽減するための、請求項1記載の分子。
- 癌を処置するために同時、別々、又は連続して使用するための組み合わせ製剤としての、抗癌剤、及び細胞外環形動物ヘモグロビン、グロビン、又はグロビンプロトマーからなる製品。
- 細胞外環形動物ヘモグロビンが、多毛類環形動物の細胞外ヘモグロビンから選択されることを特徴とする、請求項1若しくは2記載の分子、又は請求項3記載の製品。
- 細胞外環形動物ヘモグロビンが、ツリミミズ科(family Lumbricidae)の細胞外ヘモグロビン、クロムシ科(family Arenicolidae)の細胞外ヘモグロビン、及びゴカイ科(family Nereididae)の細胞外ヘモグロビンから選択されることを特徴とする、請求項1〜3のいずれか一項記載の分子、又は請求項3若しくは4記載の製品。
- 細胞外環形動物ヘモグロビンが、Lumbricus terrestrisの細胞外ヘモグロビン、Arenicola spの細胞外ヘモグロビン、及びNereis spの細胞外ヘモグロビンから選択されることを特徴とする、請求項1〜5のいずれか一項記載の分子、又は請求項3〜5のいずれか一項記載の製品。
- 細胞外環形動物ヘモグロビンが、Arenicola marinaの細胞外ヘモグロビン、及びNereis virensの細胞外ヘモグロビンから選択されることを特徴とする、請求項1〜6のいずれか一項記載の分子、又は請求項3〜6のいずれか一項記載の製品。
- 癌が、固形腫瘍であることを特徴とする、請求項1〜7のいずれか一項記載の分子、又は請求項3〜7のいずれか一項記載の製品。
