これらの3つの5-アルギニンペプチド(配列番号18〜20)は、また、大腸菌(エシェリキア・コリ)およびネズミチフス菌(サルモネラ・チフィムリウム)を含む他のグラム陰性生物に対しても強力な活性を示した。これらの新規に合成されたペプチドの高められた能力に起因して、当該ペプチドが哺乳動物細胞において細胞毒性を有するかどうかについて検討された。当該ペプチドは、乳酸デヒドロゲナーゼ細胞毒性検出キット(Lactic Dehydrogenase Cytotoxicity Detection kit)(ロシュ ダイアゴノスティック コーポレーション、インディアナポリス、インディアナ州(Roche Diagnostics Corp., Indianapolis, IN))を使用して評価され、当該ペプチドは、試験された最高濃度(75 μΜ)でも最小限の細胞毒性活性しか有していないことが判った。4時間のインキュベーションの後、3つのペプチドのすべてが、<4% 細胞毒性を示した。24時間のインキュベーションの後、細胞毒性のレベルにわずかな増加があり、ペプチド20-44ser26 (配列番号 19)は、23%、ペプチド20-44ser42 (配列番号20)は5%、そしてペプチド20-44cys (配列番号18)は10%の活性をそれぞれ示した。
3つペプチド化合物、BCC01-5RMP (配列番号 21)、BCC02-5RMP (配列番号 22)、および BCC03-5RMP (配列番号 23)は、緑膿菌(シュードモナス・アエルギノーザ)、大腸菌(エシェリキア・コリ)、ネズミチフス菌(サルモネラ・チフィムリウム)およびアシネトバクター・バウマニイに対するインビトロ殺菌活性に関して測定された。すべての実験に関して開始の接種量は、1 x 106コロニー形成単位(CFU)/mlであった。60〜80分間のインキュベーション後に、残存するコロニー形成単位が数えられ、そしてデータはCFU/mlにおける対数減少としてプロットされた。ペプチドは、シュードモナス属やアシネトバクター属に対して最も強い活性を示したが、サルモネラ種や大腸菌に対してはより低い活性であった。第3表に示すインビトロ殺菌活性は、BCC01-5RMPがこの評価で他の2つのペプチドよりもわずかに優性を有することを示している。
図1はC57/BLマウスの細菌性角膜炎モデルにおけるインビボでのBCC02-5RMP (配列番号 22)の使用の結果を示すものである。円形の創傷(2mm)は上皮を除去することにより、マウスの角膜上で作成され、105コロニー形成単位(CFU)のシュードモナス・アエルギノーザ(ATCC(登録商標) 27853(商品名)(アメリカン・タイプ・カルチャー・コレクション、マナッサス、バージニア州(American Type Culture Collection, Manassas, VA))で感染させた。感染した創傷は、最初の日において、2時間の間15分毎に、次いで3時間の間30分毎に、生理食塩水(0.9%塩化ナトリウム、ビヒクル対照)またはペプチドの示された濃度を含む生理食塩水で処置された。感染した創傷は、2日目に2回処置され、そして3日目に1回処置された。マウスは、感染後48時間で屠殺し、CFU/目を定量化した。平均値がプロットされ、この平均値は、各グループごとに5匹のマウスの代表するものである。マンホイットニー検定を、生理食塩水対照群と比較して、グループごとに行った。ペプチド化合物は、効果的にすべての細菌を殺し、そして0.25〜0.75 mg/mlの濃度の範囲内での感染を治癒した。
BCC01-5RMP (配列番号 21)、BCC02-5RMP (配列番号 22)、および BCC03-5RMP (配列番号 23)が、カブトガニ血球抽出成分(Limulus Amebocyte Lysate (LAL))を用いて、これらのリポ多糖(LPS)を結合する能力に関して試験された。これらの3つのすべてのペプチドは、シュードモナスLPSに結合した。LALを用いてのLPSを結合するこれらの能力に有意差はなかった。これらのペプチドのLPSを中和する能力が、LPS刺激化RAW264.7 細胞 (マウスのマクロファージ細胞系統)からの腫瘍壊死因子-α(TNF-α)の放出を減衰させる能力を測定することによってすることで評価された。これらの3つのすべてのペプチドは、TNF-αの放出を減衰させることが観測された。サイトカインの放出よって測定された、これらのペプチドのLPSを中和する能力に有意差はなかった。
この合成の目的は、BCC03-5R (配列番号 20) を合成し、そしてこれに2つのmini-PEG(商品名)分子(AEEA; ペプチド インターナショナル、インコーポレーテッド、ルイビル、ケンタッキー州(Peptides International, Inc., Louisville, KY))をカップリングさせることで、BCC03-5RMP (配列番号23): (AEEA)-(AEEA)-RRRRNQGRHFCGGALIHARFVMTAASSFQRを製造することである。合成後、当該ペプチド化合物は純化され凍結乾燥された。
本願明細書に開示され、請求の範囲に要求された本発明概念(群)の他の実施形態においては、ペプチド化合物群は、CAP37タンパク質のその他のアミノ酸部位(特に限定されるものではないが、例えば、ペプチド23-42、95-122、102-122および120-146が包含される。)の誘導体から構成されるものであり、そしてこれらは前記式(I)や第1表やそれに伴う記載のような本明細書の別の場所において述べたように、置換(例えば、131位および/または132位)で、および/またはN−もしくはC−末端アルギニン残基およびmini-PEG(商品名)分子で、誘導体化されているものである。CAP37タンパク質のこのようなペプチド23-42、95-122、102-122、および120-146は、例えば、米国特許第5,107,460号、米国特許第7,354,900号および米国特許第7,893,027号において記載されており、これらの文献の内容はその完全性をもってその関連により本願明細書に明示的に組み込まれる。

95-122に基づいたぺプチド化合物群と120-144に基づいたぺプチド化合物群を用いた皮膚創傷治癒研究
雄ラットへの単回静脈内投与後の 14C-BCC03-5RMP(配列番号: 23)の薬物動態、分布および排泄
ヒト角膜上皮細胞 (HCEC)の単層が、「傷付けられ」、次いでCAP37 で処置され、創傷閉鎖がは時間をかけて記録された。角膜創傷治癒のインビボモデルにおいて、2 mm径の傷がマウス角膜に形成された。創傷はCAP37で処置され、創傷閉鎖はフルオレセイン染色による16および24時間目に観察された。CAP37処置は、角膜創傷治癒を促進することが示された。関連CAP37配列は、米国特許第 7,354,900号に開示されており、その内容はその関連により完全に本願明細書に参照により明示的に組み込まれる。

Claims (56)

(A) 式(1)で表される第1および第2のペプチド化合物を有し、第1のペプチド化合物のPepが配列番号1〜14および25のうちのいずれか1つであり、かつ第2のペプチド化合物のPepが配列番号26〜31のうちのいずれか1つである;
(B) 式(1)で表される第1および第2のペプチド化合物を有し、第1のペプチド化合物のPepが配列番号26〜27のうちのいずれか1つであり、かつ第2のペプチド化合物のPepが配列番号28〜31のうちのいずれか1つである;および
(C)それぞれ独立して配列番号21〜23および32〜46のうちのいずれかの配列を有するものとして定義される、2つのペプチド化合物を有する。 - 請求項1〜8のいずれかに記載された少なくとも2つの組成物、および製薬上許容される担体を含むことを特徴とする製薬的組成物。
- 次の(A)〜(C)のうちのいずれかを有するものとしてさらに定義される請求項11の製薬的組成物:
(A) 式(1)で表される第1および第2のペプチド化合物を有し、第1のペプチド化合物のPepが配列番号1〜14および25のうちのいずれか1つであり、かつ第2のペプチド化合物のPepが配列番号26〜31のうちのいずれか1つである;
(B) 式(1)で表される第1および第2のペプチド化合物を有し、第1のペプチド化合物のPepが配列番号26〜27のうちのいずれか1つであり、かつ第2のペプチド化合物のPepが配列番号28〜31のうちのいずれか1つである;および
(C)それぞれ独立して配列番号21〜23および32〜46のうちのいずれかの配列を有するものとして定義される、2つのペプチド化合物を有する。 - 処置を必要とする被験体において細菌感染および真菌感染の少なくとも1つを処置するおよび/または阻害する方法であって、
以下の式(I)で表されるペプチド化合物を含む組成物を被験体に投与することを特徴とする方法:
SM-XR-Pep-YR (I)
(式中、Pepは配列番号l〜14、25〜31および47のアミノ酸配列のいずれか1つであり、
XRおよびYRは、(XR+YR)が4、5、または6つのアルギニン残基であるという前提の下に、それぞれ独立して0、1、2、3、4、5、または6つのアルギニン残基であり、そして
SMは、次の(l)〜(3)のうちのいずれか1つの可溶化部位であって、ここで
(1)はAEEAkである(式中k=1〜5)、
(2) は AEEEAkである(式中k=1〜5)、
(3)は AEEAmサブユニットとAEEEAnサブユニットとの組み合わせである(式中mおよびnは、(m+n)が2、3、4、5、6、7、8、9、または10であるという前提の下に、m=1〜9であり、n=1〜9であり、そしてこれらのサブユニットは任意の順序で配列され得るものである。)。)。 - (a) 前記細菌感染が、緑膿菌(シュードモナス・アエルギノーザ)、大腸菌(エシェリキア・コリ)、ネズミチフス菌(サルモネラ・チフィムリウム)およびアシネトバクター・バウマニイからなる群から選択されたいずれかのグラム陰性菌によって引き起こされたものであり、および/または
(b) 前記真菌感染が、カンジダ属菌、出芽酵母(サッカロマイセス・セレビシエ)、ヒトプラスマ種、ヒストプラスマ・カプスラーツム、アスペルギルス種、アスペルギルス・フミガーツスおよびクリプトコッカス・ネオフォルマンスからなる群から選択されてなるいずれかの真菌生物によって引き起こされたもの
である請求項13に記載の方法。 - 前記組成物が少なくとも1つの追加の治療的薬剤をさらに有するものである請求項13に記載の方法。
- 少なくとも1つの追加の治療的薬剤が、ゲンタマイシン、アミカシン、カナマイシン、トブラマイシン、ネオマイシン、エルタペネム、ドリピネム、イミペネム/シラスタチン、メロペネム、セフタジジム、セフェピム、セフタロリン、セフトビプロール、アズトレオナム、ピペラシリン、ポリミキシン B、コリスチン、シプロフロキサシン、レボフロキサシン、モキシフロキサシン、ガチフロキサシン、チゲサイクリン並びにこれらの組み合わせおよびこれらの誘導体からなる群から選択されたいずれかの抗生物質である請求項13に記載の方法。
- 少なくとも1つの追加の治療的薬剤が、配列番号26〜31のいずれか1つのペプチドである請求項15に記載の方法。
- (a) 配列番号21〜23のうちのいずれか1つのペプチド化合物と、配列番号26〜28のいずれか1つのペプチドからなる少なくとも1つの追加の治療的薬剤を含む、または
(b) 配列番号32〜35のうちのいずれか1つのペプチド化合物と、配列番号26〜27のいずれか1つのペプチドからなる少なくとも1つの追加の治療的薬剤を含む、
ことでさらに定義づけられるものである請求項17に記載の方法。 - 少なくとも1つの追加の治療的薬剤を前記組成物と同時におよび/または完全にもしくは部分的に順番に被験体に与える投与段階を有するものである請求項13に記載の方法。
- 少なくとも1つの追加の治療的薬剤が、ゲンタマイシン、アミカシン、カナマイシン、トブラマイシン、ネオマイシン、エルタペネム、ドリピネム、イミペネム/シラスタチン、メロペネム、セフタジジム、セフェピム、セフタロリン、セフトビプロール、アズトレオナム、ピペラシリン、ポリミキシン B、コリスチン、シプロフロキサシン、レボフロキサシン、モキシフロキサシン、ガチフロキサシン、チゲサイクリン並びにこれらの組み合わせおよびこれらの誘導体からなる群から選択されたいずれかの抗生物質である請求項19に記載の方法。
- 少なくとも1つの追加の治療的薬剤が、配列番号26〜31のいずれか1つのペプチドである請求項19に記載の方法。
- (a) 配列番号21〜23のうちのいずれか1つのペプチド化合物と、配列番号26〜28のいずれか1つのペプチドからなる少なくとも1つの追加の治療的薬剤を含む、または
(b) 配列番号32〜35のうちのいずれか1つのペプチド化合物と、配列番号26〜27のいずれか1つのペプチドからなる少なくとも1つの追加の治療的薬剤を含む、
ことでさらに定義づけられるものである請求項21に記載の方法。 - 処置を必要とする被験体において創傷の治癒および/または移植片の受け入れを促進するために創傷および移植片の少なくとも1つを処置する方法であって、
以下の式(I)で表されるペプチド化合物を含む組成物を被験体に投与することを特徴とする方法:

- 少なくとも1つの追加の治療的薬剤が、配列番号26〜31のいずれか1つのペプチドである請求項25に記載の方法。
- (a) 配列番号21〜23のうちのいずれか1つのペプチド化合物と、配列番号26〜28のいずれか1つのペプチドからなる少なくとも1つの追加の治療的薬剤を含む、または
(b) 配列番号32〜35のうちのいずれか1つのペプチド化合物と、配列番号26〜27のいずれか1つのペプチドからなる少なくとも1つの追加の治療的薬剤を含む、
ことでさらに定義づけられるものである請求項27に記載の方法。 - 少なくとも1つの追加の治療的薬剤を前記組成物と同時におよび/または完全にもしくは部分的に順番に被験体に与える投与段階を有するものである請求項23に記載の方法。

- 少なくとも1つの追加の治療的薬剤が、配列番号26〜31のいずれか1つのペプチドである請求項29に記載の方法。
- (a) 配列番号21〜23のうちのいずれか1つのペプチド化合物と、配列番号26〜28のいずれか1つのペプチドからなる少なくとも1つの追加の治療的薬剤を含む、または
(b) 配列番号32〜35のうちのいずれか1つのペプチド化合物と、配列番号26〜27のいずれか1つのペプチドからなる少なくとも1つの追加の治療的薬剤を含む、
ことでさらに定義づけられるものである請求項31に記載の方法。 - 細菌感染の処置において抗生物質の効能を高める方法であって、
以下の式(I)で表されるペプチド化合物を含む組成物を被験体に投与すること;
SM-XR-Pep-YR (I)
(式中、Pepは配列番号l〜14、25〜31および47のアミノ酸配列のいずれか1つであり、
XRおよびYRは、(XR+YR)が4、5、または6つのアルギニン残基であるという前提の下に、それぞれ独立して0、1、2、3、4、5、または6つのアルギニン残基であり、そして
SMは、次の(l)〜(3)のうちのいずれか1つの可溶化部位であって、ここで
(1)はAEEAkである(式中k=1〜5)、
(2) は AEEEAkである(式中k=1〜5)、

抗生物質を被験体に投与すること;
の工程を有し、
前記抗生物質が、(i)単独で投与した場合には最適以下の活性であるまたは細菌に対して有効でない、および(ii)当該ペプチド化合物との組み合わせにおいて投与された場合には細菌に対して有効である、量において投与されるものである
ことを特徴とする方法。 - 前記抗生物質が、ゲンタマイシン、アミカシン、カナマイシン、トブラマイシン、ネオマイシン、エルタペネム、ドリピネム、イミペネム/シラスタチン、メロペネム、セフタジジム、セフェピム、セフタロリン、セフトビプロール、アズトレオナム、ピペラシリン、ポリミキシン B、コリスチン、シプロフロキサシン、レボフロキサシン、モキシフロキサシン、ガチフロキサシン、チゲサイクリン並びにこれらの組み合わせおよびこれらの誘導体からなる群から選択されたいずれかの抗生物質である請求項33に記載の方法。
- 前記抗生物質が、前記組成物と同時に投与されるものである請求項33に記載の方法。
- 前記組成物が、前記抗生物質と組み合わせた前記ペプチド化合物からなるものであり、これによって前記ペプチド化合物と抗生物質が同時に投与されるものである請求項34に記載の方法。
- 前記抗生物質は、前記組成物と完全にもしくは部分的に順番に投与されるものである請求項33に記載の方法。
- 処置を必要とする被験体において創傷の治癒および/または移植片の受け入れを促進するために創傷および移植片の少なくとも1つを処置するにおいて抗生物質の効能を高める方法であって、
以下の式(I)で表されるペプチド化合物を含む組成物を被験体に投与すること;
SM-XR-Pep-YR (I)
(式中、Pepは配列番号l〜14、25〜31および47のアミノ酸配列のいずれか1つであり、
XRおよびYRは、(XR+YR)が4、5、または6つのアルギニン残基であるという前提の下に、それぞれ独立して0、1、2、3、4、5、または6つのアルギニン残基であり、そして
SMは、次の(l)〜(3)のうちのいずれか1つの可溶化部位であって、ここで

ことを特徴とする方法。 - 前記抗生物質が、ゲンタマイシン、アミカシン、カナマイシン、トブラマイシン、ネオマイシン、エルタペネム、ドリピネム、イミペネム/シラスタチン、メロペネム、セフタジジム、セフェピム、セフタロリン、セフトビプロール、アズトレオナム、ピペラシリン、ポリミキシン B、コリスチン、シプロフロキサシン、レボフロキサシン、モキシフロキサシン、ガチフロキサシン、チゲサイクリン、クリンダマイシン、クラリスロマイシン、バンコマイシン、アジスロマイシン、セフィキシム、セフトリアキソン、セファマンドール、セフォタキシム、セフジニル、バシトラシン、スルファセタミド、ドキシサイクリン並びにこれらの組み合わせおよびこれらの誘導体からなる群から選択されたいずれかの抗生物質である請求項38に記載の方法。
- 抗生物質が、前記組成物と同時に投与されるものである請求項38に記載の方法。
- 前記組成物が、前記抗生物質と組み合わせた前記ペプチド化合物からなるものであり、これによって前記ペプチド化合物と抗生物質が同時に投与されるものである請求項40に記載の方法。
- 前記抗生物質は、前記組成物と完全にもしくは部分的に順番に投与されるものである請求項33に記載の方法。
- 前記ペプチド化合物のXRは4つのアルギニン残基からなるものであり、YRは1つのアルギニン残基からなるものである請求項13、23、33および38のいずれか1つに記載の方法。
- 前記ペプチド化合物のSM基は2つのAEEA部分からなるものである請求項13、23、33および38のいずれか1つに記載の方法。
- 前記ペプチド化合物が、配列番号21〜23および32〜46のうちのいずれか1つの配列を有するものとしてさらに定義づけられるものである請求項13、23、33および38のいずれか1つに記載の方法。
- 前記組成物が、式(I)で表されるペプチド化合物を少なくとも2つ含むものである、請求項13、23、33および38のいずれか1つに記載の方法。
- 次の(A)〜(C)のうちのいずれかを有するものとしてさらに定義される請求項13、23、33および38のいずれか1つに記載の方法:
(A) 式(1)で表される第1および第2のペプチド化合物を有し、第1のペプチド化合物のPepが配列番号1〜14および25のうちのいずれか1つであり、かつ第2のペプチド化合物のPepが配列番号26〜31のうちのいずれか1つである;
(B) 式(1)で表される第1および第2のペプチド化合物を有し、第1のペプチド化合物のPepが配列番号26〜27のうちのいずれか1つであり、かつ第2のペプチド化合物のPepが配列番号28〜31のうちのいずれか1つである;および
(C)それぞれ独立して配列番号21〜23および32〜46のうちのいずれかの配列を有するものとして定義される、2つのペプチド化合物を有する。 - 配列番号22 [(AEEA)-(AEEA)-RRRRNQGRHFSGGALIHARFVMTAASCFQR]
(式中、AEEAは、8-アミノ-3,6-ジオキサオクタン酸である。)のペプチドを有してなる組成物であって、当該ペプチド化合物は、抗生物質と組み合わせて被験体に投与された場合に当該被験体において前記抗生物質の治療的効果を高める上で有効であることを特徴とする組成物。 - ゲンタマイシン、アミカシン、カナマイシン、トブラマイシン、ネオマイシン、エルタペネム、ドリピネム、イミペネム/シラスタチン、メロペネム、セフタジジム、セフェピム、セフタロリン、セフトビプロール、アズトレオナム、ピペラシリン、ポリミキシン B、コリスチン、シプロフロキサシン、レボフロキサシン、モキシフロキサシン、ガチフロキサシン、チゲサイクリン並びにこれらの組み合わせおよびこれらの誘導体からなる群から選択されたいずれかの抗生物質をさらに有してなる請求項48記載の組成物。
- 配列番号32 [(AEEA)-(AEEA)-RRRRGTRCQVAGWGSQRSGGRLSRFPRFVNVR]
(式中、AEEAは、8-アミノ-3,6-ジオキサオクタン酸である。)のペプチドを有してなる組成物であって、当該ペプチド化合物は、治療を必要とする被験者において創傷治癒および抗菌活性を有するものであることをを特徴とする組成物。 - ゲンタマイシン、アミカシン、カナマイシン、トブラマイシン、ネオマイシン、エルタペネム、ドリピネム、イミペネム/シラスタチン、メロペネム、セフタジジム、セフェピム、セフタロリン、セフトビプロール、アズトレオナム、ピペラシリン、ポリミキシン B、コリスチン、シプロフロキサシン、レボフロキサシン、モキシフロキサシン、ガチフロキサシン、チゲサイクリン、クリンダマイシン、クラリスロマイシン、バンコマイシン、アジスロマイシン、セフィキシム、セフトリアキソン、セファマンドール、セフォタキシム、セフジニル、バシトラシン、スルファセタミド、ドキシサイクリン並びにこれらの組み合わせおよびこれらの誘導体からなる群から選択されたいずれかの抗生物質をさらに有してなる請求項50に記載の組成物。
- 請求項1〜10および48〜51のいずれかに記載された組成物、および製薬上許容される担体を含むことを特徴とする製薬的組成物。
- 処置を必要とする被験体において細菌感染および真菌感染の少なくとも1つを処置するおよび/または阻害する方法において使用するための、請求項1〜10および48〜49のいずれかに記載された組成物であって、
前記方法が、被験体に請求項1〜10および48〜51のいずれかに記載された組成物を投与する工程を有し、ここにおいて細菌および/または真菌感染が阻止されるものであることを特徴とする組成物。 - (a) 前記細菌感染が、緑膿菌(シュードモナス・アエルギノーザ)、大腸菌(エシェリキア・コリ)、ネズミチフス菌(サルモネラ・チフィムリウム)およびアシネトバクター・バウマニイからなる群から選択されたいずれかのグラム陰性菌によって引き起こされたものであり、および/または

前記方法が、被験体に請求項1〜10および50〜51のいずれかに記載された組成物を投与する工程を有し、ここにおいて創傷の治癒および/または移植片の受け入れが促進すされるものであることを特徴とする組成物。 - (a) 前記創傷が、表面創傷、裂傷、擦傷、剥離、切傷、切断創、脚部ないし足部の糖尿病性潰瘍、褥瘡、床擦れ、末梢血管疾患による傷、術後傷、火傷、眼潰瘍、ドライアイおよび眼創傷創傷からなる群から選ばれてなるいずれかのものであり、および/または
