Abstract
Description
適合対象である細胞に対して適合性のあるハンセン溶解度パラメータ(HSP)を有する、液体。
(2)前記適合性のあるハンセン溶解度パラメータ(HSP)は、前記細胞に対する生体適合性レベルが確定された1又は2以上の液体とそれらのハンセン溶解度パラメータとに基づいて得られる生体適合性に関連付けられたハンセン溶解度パラメータの閾値情報に基づいて決定される、(1)に記載の液体。
(3)前記適合性のあるハンセン溶解度パラメータ(HSP)は、前記細胞に対する生体適合性を有する1又は2以上の液体のハンセン溶解度パラメータに基づくハンセン溶解度パラメータ(HSP)空間における所定の中心値(δD、δP、δH)及び所定の相互作用半径Rによって規定されるHSP球内にある、(1)又は(2)に記載の液体。
(4)前記適合性のあるハンセン溶解度パラメータ(HSP)は、前記細胞の1又は2)以上の細胞構成要素のハンセン溶解度パラメータに基づくハンセン溶解度パラメータ(HSP)空間における所定の中心値(δD、δP、δH)及び所定の相互作用半径によって規定されるHSP球外にある、(1)〜(3)のいずれかに記載の液体。
(5)前記液体は、前記細胞に対して適合性のあるモル体積を有する、(1)〜(4)のいずれかに記載の液体。
(6)前記適合性のあるモル体積は、前記細胞に対して生体適合性レベルが確定された1又は2以上の液体とそれらのモル体積とに基づいて得られる生体適合性に関連付けられた液体モル体積の閾値情報に基づいて決定される、(5)に記載の液体。
(7)モル体積は、330cm3/mol未満であり、
前記ハンセン溶解度パラメータ(HSP)が、前記中心値(δD、δP、δH)が(12.73, 2.33, 3.46) ([J/cm3]1/2)であり前記相互作用半径Rが3.4 ([J/cm3]1/2)以内のHSP球内である、(1)〜(3)のいずれかに記載の液体。
(8)液体モル体積は、330cm3/mol以上であり、
前記ハンセン溶解度パラメータ(HSP)が、前記中心値(δD、δP、δH)が(12.73, 2.33, 3.46) ([J/cm3]1/2)であり前記相互作用半径Rが9.0([J/cm3]1/2)以内のHSP球内である、(1)〜(3)のいずれかに記載の液体。
(9)液体モル体積は、125cm3/mol以上であり、
前記ハンセン溶解度パラメータ(HSP)が、DNA、コレステロール、水、ホスファチジルコリン、スフィンゴミエリン、ホスファチジルエタノールアミン及びホスファチジルセリンの全てのHSP球外にある、(1)、(2)及び(4)のいずれかに記載の液体。
(10)沸点が33℃超であり、融点が25℃未満である、(1)〜(9)のいずれかに記載の液体。
(11)以下の表から選択される1又は2以上の液体である、(1)〜(10)のいずれかに記載の液体。
(13)生体適合性液体を判定するためのHSP閾値情報を決定する方法であって、
1又は2以上の液体について、適合対象細胞に対するそれらの生体適合性とHSPとを取得する工程と、
前記生体適合性及び前記HSPに基づいて、所定の生体適合性に関連付けたHSP空間における中心値(δD、δP、δH)及び相互作用半径Rによって前記HSP閾値情報としてのHSP球を規定する工程と、
を備える、方法。
(14)前記1又は2以上の液体について、さらにモル体積を取得する工程を備え、
前記モル体積に応じて前記HSP球を規定する、(13)記載の方法。
(15)生体適合性液体を判定するためのHSP閾値情報を決定する方法であって、
1又は2以上の液体について、適合対象細胞の細胞構成要素のHSPを取得する工程と、前記HSPに基づいて、HSP空間における前記細胞構成要素についての中心値(δD、δP、δH)及び相互作用半径Rによって前記HSP閾値情報としてのHSP球を規定する工程と、を備える、方法。
(16)生体適合性液体のスクリーニング方法であって、
被験液体が、適合対象細胞に対して適合性であるHSPを有しているか否かを判定する工程及び/又は液体が、適合対象細胞に対して適合性であるモル体積を有しているか否かを判定する工程を備える、方法。
本明細書において、HSP(ハンセン溶解度パラメータ)は、以下のように定義される。

なお、HSPに加えて、液体のモル体積が生体適合性に影響を及ぼしうる。液体モル体積は、分子間の相互作用や速度論的な現象(拡散等)に関連するからである。モル体積が大きいほど、生体を構成する細胞膜成分への溶解性が低くなり、浸透性や拡散性も低くなり生体適合性が増大する傾向がある。一方、モル体積が小さいほど、細胞への浸透性や拡散性が高くなり、生体適合性が低下する傾向がある。なお、混合溶媒のモル体積は、各単一溶媒のモル体積の加重平均として表すことができる。
本明細書において、細胞とは、真核細胞及び原核細胞を意味している。したがって、動物細胞、植物細胞のほか、微生物細胞等を含みうる。
本明細書において、生体適合性とは、細胞と外界を隔てる細胞膜より外部構成要素を備えないnaked細胞に対する適合性を意味している。naked細胞としては、生きている細胞であって細胞膜のみで外界で隔てられた状態の細胞であり、例えば、粘膜や角質、細胞壁、外膜などの細胞外要素を備えない細胞を意味している。また、naked細胞は、単細胞であっても集合体であってもよいし、naked細胞である限り、生体の組織や器官であってもよい。
本開示の生体適合性液体は、適合対象である細胞に対して適合性のあるHSPを有することができる。この適合性のあるHSPは、適合対象たる細胞に対して予め生体適合性レベルが確定された複数の液体とその液体のHSPとによって得られる生体適合性に関連付けられたHSP閾値情報に基づいて決定することができる。
(第1の形態)
本開示によれば、HSP閾値情報を、HSP空間における所定の中心値(δD、δP、δH)及び所定の相互作用半径Rによって規定されるHSP球とし、当該HSP球の内部を適合性あるHSPとすることができる。なお、HSP球の内部にあるというとき、HSP球の外縁を規定する座標であるHSPに一致するか又は当該HSPよりもHSP球内部にある場合を意味するものとする。
HSP閾値情報としてのHSP球は、液体のモル体積閾値情報に関連付けて規定してもよい。液体のモル体積に応じて、適合対象細胞に対する生体適合性が異なりうる。上述したように、モル体積が大きいほど、適合対象細胞に対する生体適合性が向上する傾向があり、モル体積が小さいほど、適合対象細胞に対する生体適合性が低下する傾向がある。
こうしたHSP閾値情報やモル体積閾値情報は、例えば、以下のようにして取得できる。すなわち、1又は2以上の被験液体の適合対象細胞であるnaked細胞に対する生体適合性を評価し、意図する生体適合性とHSPとを関連付けて、意図する生体適合性を肯定するためのHSP中心値及び相互作用半径を求めることで、HSP閾値情報を得ることができる。また、意図する生体適合性とモル体積とを関連付けて、意図する生体適合性を肯定するためのモル体積の閾値を求めることで、モル体積閾値情報を得ることができる。
液体のモル体積が、330cm3/mol以上のとき、好ましくは、以下のHSP球をHSP閾値情報とし、当該HSP球内部にあるHSPを適合性のあるHSPとすることができる。
HSP球1:
中心値(δD、δP、δH):(12.73, 2.33, 3.46) ([J/cm3]1/2)
相互作用半径R:9.0([J/cm3]1/2)
液体の生体適合性は、HSP及びモル体積のほか、沸点及び/又は融点を考慮することができる。沸点及び融点は、生体適合性液体の液体としての利便性や操作性に影響する。沸点は33℃を超えることが好ましい。33℃未満であると、生体の生存に一般的に適した通常の作業環境(約10〜30℃)において、液体として取扱難いからである。また、融点は、25℃未満であることが好ましい。融点が25℃以上であると、通常の作業環境(約10〜30℃)において、液体として取扱難いからである。
HSP球2:
中心値(δD、δP、δH):(12.73, 2.33, 3.46) ([J/cm3]1/2)
相互作用半径R:3.4 ([J/cm3]1/2)
また、HSP閾値情報を、HSP空間における所定の中心値(δD、δP、δH)及び所定の相互作用半径によって規定されるHSP球とし、当該HSP球外部を適合性あるHSPとすることができる。なお、HSP球外部にあるというとき、HSP球の外縁を規定する座標であるHSPに一致するか又は当該HSP球よりも外側にある場合を意味するものとする。
本開示は、生体適合性液体を判定するための指標を決定する方法を提供することができる。本方法は、1又は2以上の液体について、適合対象細胞に対するそれらの生体適合性とHSPとを取得する工程と、
前記生体適合性及び前記HSPに基づいて、所定の生体適合性に関連付けたHSP空間における中心値(δD、δP、δH)及び相互作用半径Rによって前記HSP閾値情報としてのHSP球を規定する工程と、
を備えることができる。
本明細書の開示によれば、生体適合性液体のスクリーニング方法が提供される。本スクリーニング方法は、被験液体が、適合対象細胞に対して適合性であるHSPを有しているか否かを判定する工程及び/又は液体が、適合対象細胞に対して適合性であるモル体積を有しているか否かを判定する工程を備えることができる。なお、ここでいう適合性であるHSPに有しているか否かについては、既に説明した態様を適用して判定することができる。
被験液体のモル体積と規格化細胞生存率との関係を図2に示す。図2に示すように、モル体積の大きさと、規格化細胞生存率との間には、一定の関係があった。すなわち、モル体積が125cm3/mol以下の場合には、規格化細胞生存率が極めて低く(生体適合性がほとんどなく)、モル体積が125cm3/mol超330cm3/mol未満のときには、規格化細胞生存率には低いものから高いものまで分布があり、モル体積が330cm3/mol以上のときには、規格化細胞生存率は、おしなべて高かった。以上のことから、液体のモル体積自体に応じて、液体の生体適合性は影響され、125cm3/mol以下の場合にはそれ自体で強毒性(低生体適合性)となり、同330cm3/mol以上の場合には、低毒性(高生体適合性)となり、125cm3/mol超330cm3/mol未満のときには、HSPに大きく依存して細胞毒性(生体適合性)が呈されることがわかった。
上記(1)によれば、生体適合性とHSPとの関係については、モル体積が330cm3/mol未満であるとき、HSPが規格化細胞生存率(生体適合性)に影響することがわかった。したがって、このモル体積範囲を充足する被験液体から規格化細胞生存率が0.7以上となる被験液体を選択し、これらのHSPからハンセン球法によりHSP中心値を求めた。その結果を、表11の一部に示す。また、表11には、得られたHSP中心値からの被験液体及び比較液体のHSPの距離Dも示す。さらに、図3に、モル体積が330cm3/mol未満であって規格化細胞生存率が0.7以上の被験液体の各HSP及びこれらに基づくHSP中心値、及び細胞毒性液体のHSPを示す。
モル体積が330cm3/mol未満の被験液体につき、規格化細胞生存率とHSP中心値からの距離Dとのプロットを図4に示す。図4に示すように、HSP距離Dが3.4([J/cm3]1/2)のとき、規格化細胞生存率が急激に低下することがわかった。また、当該距離Dは、生体適合性を規定した規格化細胞生存率が0.7に相当するHSP距離Dでもあった。すなわち、このHSP距離Dは、HSP中心値からの相互作用半径Rを規定できる閾値とすることができる。
上記で規定した2つのHSP球で規定できる生体適合性液体は、表2〜4に示すとおりであった。すなわち、以下のHSP球1で規定される生体適合性液体(候補)は、表2に示すように、39種類選択することができた。また、以下のHSP球2で規定される生体適合性液体(候補)は、表3A、表3B及び表4A、表4Bに示すように、181種の生体適合性液体を選択することができた。

表11の被験液体から細胞適合性液体(S5〜S9, S11〜S14, S18〜21)のみを抜き出し、それらのHSPと表12に示す細胞構成要素のHSP球との間の相対エネルギー差(RED)を算出したものを表13に示す。また、生体適合性液体のHSPと細胞構成要素の各HSP球との関係を図6に示す。
上記で規定した生体適合性HSP空間内に存在するHSPを有する生体適合性液体(候補)としては、表7〜9に示すように、合計241種選択できた。なお、液体の選択にあたっては、融点が25℃未満で沸点が33℃超を条件とした。

- 生体適合性液体であって、
適合対象である細胞に対して適合性のあるハンセン溶解度パラメータ(HSP)を有する、液体。 - 前記適合性のあるハンセン溶解度パラメータ(HSP)は、前記細胞に対する生体適合性レベルが確定された1又は2以上の液体とそれらのハンセン溶解度パラメータとに基づいて得られる生体適合性に関連付けられたハンセン溶解度パラメータの閾値情報に基づいて決定される、請求項1に記載の液体。
- 前記適合性あるハンセン溶解度パラメータ(HSP)は、前記細胞に対する生体適合性を有する1又は2以上の液体のハンセン溶解度パラメータに基づくハンセン溶解度パラメータ(HSP)空間における所定の中心値(δD、δP、δH)及び所定の相互作用半径Rによって規定されるHSP球内にある、請求項1又は2に記載の液体。
- 前記適合性あるハンセン溶解度パラメータ(HSP)は、前記細胞の1又は2以上の細胞構成要素のハンセン溶解度パラメータに基づくハンセン溶解度パラメータ(HSP)空間における所定の中心値(δD、δP、δH)及び所定の相互作用半径によって規定されるHSP球外にある、請求項1〜3のいずれかに記載の液体。
- 前記液体は、前記細胞に対して適合性のあるモル体積を有する、請求項1〜4のいずれかに記載の液体。
- 前記適合性あるモル体積は、前記細胞に対して生体適合性レベルが確定された1又は2以上の液体とそれらのモル体積とに基づいて得られる生体適合性に関連付けられた液体モル体積の閾値情報に基づいて決定される、請求項5に記載の液体。
- モル体積は、330cm3/mol未満であり、
前記ハンセン溶解度パラメータ(HSP)が、前記中心値(δD、δP、δH)が(12.73, 2.33, 3.46) ([J/cm3]1/2)であり前記相互作用半径Rが3.4 ([J/cm3]1/2)以内のHSP球内である、請求項1〜3のいずれかに記載の液体。 - 液体モル体積は、330cm3/mol以上であり、
前記ハンセン溶解度パラメータ(HSP)が、前記中心値(δD、δP、δH)が(12.73, 2.33, 3.46) ([J/cm3]1/2)であり前記相互作用半径Rが9.0([J/cm3]1/2)以内のHSP球内である、請求項1〜3のいずれかに記載の液体。 - 液体モル体積は、125cm3/mol以上であり、
前記ハンセン溶解度パラメータ(HSP)が、DNA、コレステロール、水、ホスファチジルコリン、スフィンゴミエリン、ホスファチジルエタノールアミン及びホスファチジルセリンの全てのHSP球外にある、請求項1、2及び4のいずれかに記載の液体。 - 沸点が33℃超であり、融点が25℃未満である、請求項1〜9のいずれかに記載の液体。
- 以下の群から選択される1又は2以上の液体である、請求項1〜10のいずれかに記載の液体。
- 細胞外要素を備えないnaked細胞に対する生体適合性を備える、請求項1〜11のいずれかに記載の液体。
- 生体適合性液体を判定するためのHSP閾値情報を決定する方法であって、
1又は2以上の液体について、適合対象細胞に対するそれらの生体適合性とHSPとを取得する工程と、
前記生体適合性及び前記HSPに基づいて、所定の生体適合性に関連付けたHSP空間における中心値(δD、δP、δH)及び相互作用半径Rによって前記HSP閾値情報としてのHSP球を規定する工程と、
を備える、方法。 - 前記1又は2以上の液体について、さらにモル体積を取得する工程を備え、
前記モル体積に応じて前記HSP球を規定する、請求項13記載の方法。 - 生体適合性液体を判定するためのHSP閾値情報を決定する方法であって、
1又は2以上の液体について、適合対象細胞の細胞構成要素のHSPを取得する工程と、前記HSPに基づいて、HSP空間における前記細胞構成要素についての中心値(δD、δP、δH)及び相互作用半径Rによって前記HSP閾値情報としてのHSP球を規定する工程と、を備える、方法。 - 生体適合性液体のスクリーニング方法であって、
被験液体が、適合対象細胞に対して適合性であるHSPを有しているか否かを判定する工程及び/又は液体が、適合対象細胞に対して適合性であるモル体積を有しているか否かを判定する工程を備える、方法。
