するノビレチンの作用を評価した。抗DNP IgEで感作したRBL-2H3細胞及びBMMCをそれぞれノビレチン(0〜70μM)で処理し、抗原(DNP-HSA)で刺激することにより脱顆粒を誘導
した。脱顆粒のマーカーとして、β-ヘキソサミニダーゼを使用した。結果を図1(A)
及び(B)に示す。図1(A)に示すように、RBL-2H3細胞の脱顆粒はノビレチンにより
用量依存的に抑制された。30μM 以上のノビレチンで対照(コントロール)に対する有意差が認められた。また図1(B)に示すように、ノビレチン(50μM)はBMMCの脱顆粒を
著しく抑制した。抑制されたβ-ヘキソサミニダーゼ放出率は、対照の約55%であり、こ
の割合は、RBL-2H3細胞の脱顆粒に対する抑制率とよく似ていた(図1(A))。これら
の結果は、ノビレチンが好塩基球細胞及びBMMCのIgEを介した脱顆粒反応を抑制すること
を示している。

示す。図6に示すように、RBL-2H3細胞の脱顆粒はβ-LGにより用量依存的に抑制された。対照(コントロール)に対する有意差はタンパク質濃度1800μg/mL 以上のβ-LGで認められた。この結果は、β-LGが好塩基球細胞のIgEを介した脱顆粒反応を抑制することを示している。

:0〜2148.6μg/mL)、ノビレチン単独(0〜50μM)、またはβ-LGとノビレチンの両方で処理し、抗原(DNP-HSA)により脱顆粒を誘導した。β-ヘキソサミニダーゼの放出を脱顆粒のマーカーとして使用した。結果を図10に示す。図10に示すように、RBL-2H3細胞
の脱顆粒はβ-LG単独、及びノビレチン単独よりも、両者を併用することでより一層強く
抑制されることが判明した。またその抑制効果は、β-LG及びノビレチンのいずれも用量
依存的であった。この結果から、β-LGとノビレチンとを併用することで、好塩基球細胞
の脱顆粒によるアレルギー症状がより一層抑制できることを示している。

ヨーグルト(めざましプルーンヨーグルト、四国乳業株式会社製)を遠心分離に供し乳清を回収した。またノビレチンはDMSOに溶解した。斯くして調製したヨーグルト及びノビレチンを使用して、(2-1)に記載する方法に従い、RBL-2H3細胞の脱顆粒に及ぼすヨーグルト又は/及びノビレチンの作用を確認した。具体的には、抗DNP IgE抗体で感作したRBL-2H3細胞を、図11に記載する濃度のヨーグルト乳清(タンパク質濃度2.64〜10.57μg/mL)又は/及びノビレチン(5μM及び10μM)で10分間処理して、次いで抗原(DNP-HSA)で30分間処理して感作した。脱顆粒のマーカーとしてβ-ヘキソサミニダーゼを使用して、RBL-2H3細胞の脱顆粒に対する抑制効果を評価した。

レチンの両者を組み合わせて、図12に示す割合でそれぞれ雌のBALB/cマウスに経口投与し、in vivoでの抗アレルギー作用を評価した。結果を図12に示す。PCA応答は、30mg/kg体重のノビレチン単独、及び20mg/kg体重のβ-LG単独の経口投与によっても顕著に抑制されたが、さらに両者を併用することでより一層強く抑制された。この結果は、生体内でのアレルギー反応の抑制に、β-LGとノビレチンの両方を経口投与することがより有効であることを示す。
