上述したイオン排除クロマトグラフィ―は、イオン交換樹脂を用いるクロマトグラフィーの一つの手法であって、海水で代表される塩化ナトリウム溶液などの高濃度共存試料が分析試料である場合であっても、固定相での交換基として強酸性アニオン基を用いており塩化物イオンが固定相に結合しないため、分離能が低下しないと考えられる。しかしながら、イオン排除クロマトグラフィ―によっても、後述の実施例から明らかになるように、溶離液として希硝酸を使用する通常の手法ではセレンの価数別分離を実現することができない。そこで、本発明に基づく方法では、溶離液として、硝酸と硫酸塩とを含む水溶液を溶離液として使用し、セレンの価数別の分離と定量を可能にしている。
(1)島津製作所製のShim−pack(登録商標) SCRシリーズ(例えば、SCR−102H:長さ300mm、内径8mm、粒子径7μm、固定相がスルホ基)、
(2)Shodex社製のSH1011及びSH1821、(例えば、SUGAR SH1011:サイズ8mm×300mm、粒径6μm、官能基スルホ基)、
(3)Shodex社製のKC−811(例えば、RSpak KC−811:サイズ8mm×300mm、粒径6μm、官能基スルホ基)、
(4)東ソー社製のTSKgel(登録商標) OApak−A及びOApak−P(例えば、TSKgel(登録商標) OApak−A:粒子径5μm、官能基カルボキシメチル基、基材ポリマー(メタクリレート系))、
(5)東ソー社製のTSKgel(登録商標) SCX(粒子径5μm、官能基スルホ基、基材ポリマー(スチレン系))、
(6)Transgenomic社製のICSepシリーズ(例えば、ICSep ION−300:耐圧7MPa、粒子径7μm、官能基スルホ基、対イオンH+、架橋率6%)、
(7)サーモフィッシャー社製のDionex IonPacイオン排除カラムシリーズ(例えば、Dionex IonPac ICE−AS1:サイズ9×250mm、あるいはDionex IonPac ICE−AS6:サイズ9×250mm、官能基スルホ基及びカルボキシ基)
などが挙げられる。特に、強酸性カチオン交換樹脂を固定相として有するものを用いることが好ましく、強酸性カチオン交換樹脂はイオン形が水素(H)形であることが好ましい。図2に示したものでは、イオン排除カラムが前段のガードカラム36と後段の分離カラム37とに分かれているが、必ずしもガードカラムを設ける必要はない。上記に例示した各イオン排除カラム製品を分離カラム37として用いる場合には、当該イオン排除カラム製品に適合したガードカラムが指定されていることが多いから、そのように指定されたカラムをガードカラム36として用いればよい。一般には、ガードカラム36と分離カラム37とは相互に同種類の固定相が充填される。

セレンとヒ素とを含む試料液を対象として、試料液中のセレンとヒ素とを同時に価数別に定量する場合には、上記と同様のイオン排除カラム(ガードカラム36及び分離カラム37)及び溶離液を使用し、試料液をガードカラム36に導入し、分離カラム37の出口からの液をICP−MS装置38に導き、ICP−MS装置の質量分析部においてセレンの原子イオン(例えば78Se+)及びヒ素の原子イオン(一般に75As+)を検出してカウントする。そしてインジェクタ33において試料液を導入した時点からの経過時間に応じてICP−MS装置でのカウント値をセレンとヒ素とに分けて集計し、クロマトグラムを得ることにより、試料液中のセレン及びヒ素のそれぞれの価数別の定量が行われたことになる。
試料液として、セレン原子換算でSe(IV)を50μg/L、Se(VI)を50μg/L含み、ヒ素原子換算でAs(III)を50μg/L、As(V)を50μg/L含むものを使用し、溶離液中に含ませる硫酸塩として硫酸ナトリウム(Na2SO4)を使用し、溶離液中の硫酸ナトリウム濃度を変化させ、変化させた硫酸ナトリウム濃度ごとにセレン及びヒ素についてのクロマトグラムを得た。溶離液中の硝酸濃度は10mmol/Lで一定とした。結果を図3に示す。なお図3に示す範囲で溶離液中の硫酸ナトリウム濃度を変化させても、溶離液のpHは2.1で一定であった。
実施例1と同じ組成の試料液を使用し、溶離液中に含ませる硫酸塩として硫酸ナトリウムを使用し、溶離液中の硫酸ナトリウム濃度は0.7mmol/Lに固定し、硝酸濃度を変化させることによって溶離液のpHを変化させ、pHごとにセレン及びヒ素についてのクロマトグラムを得た。結果を図4に示す。
実施例1と同じ組成の試料液を使用し、溶離液に加える硫酸塩の種類を異ならせたときにセレン及びヒ素のクロマトグラフがどのように異なるかを調べた。硫酸塩としては、硫酸ナトリウム、硫酸アンモニウム((NH4)2SO4)、硫酸カルシウム(CaSO4)を使用した。また、比較例として、硫酸塩の代わりに塩化ナトリウム(NaCl)を溶離液に添加した場合も調べた。溶離液における硝酸濃度は10mmol/Lで一定とし、また、硫酸塩の濃度も硫酸イオン(SO4 2-)に換算して0.5mmol/Lで統一した。塩化ナトリウムを加える場合には、その濃度を0.5mmol/Lとした。結果を図5に示す。
実施例1と同じ組成の試料液を使用し、溶離液に加える硫酸塩を硫酸アンモニウムとし、その濃度を0.7mmol/L及び70mmol/Lとして、セレンについてこれらの濃度でのクロマトグラフを得た。溶離液での硝酸濃度は10mmol/Lとした。結果を図6に示す。図6から分かるように、硫酸塩濃度が極端に高い場合には、Se(IV)とSe(VI)の各ピークが拡がって両者が相互に重なり合うようになる。ピークの重なりが大きいと分離しての定量が難しくなるから、溶離液中の硫酸塩の濃度は1mmol/L以下程度にすることが望ましいことが分かる。
実施例1での溶離液中の硫酸ナトリウム濃度を0.7mmol/Lとした場合に得たセレンのクロマトグラムとヒ素のクロマトグラムとを重ねて表示したものを図7に示す。同時にセレンとヒ素とのそれぞれについて価数別の定量を行えることが分かる。
セレン化合物及びヒ素化合物の濃度が異なる試料液を用意し、セレン及びヒ素の両方について同時に価数別の定量を行い、検量線を得た。同時に繰返し精度も求めた。溶離液としては、硝酸10mmol/L及び硫酸ナトリウム0.7mmol/Lを含むものを用いた。セレンについての検量線を図8(a)に、ヒ素についての検量線を図8(b)に示す。繰返し精度を示す変動係数(CV)は、Se(IV)に対し4.3%、Se(VI)に対し4.6%、As(III)に対し3.5%、As(V)に対し0.8%であった。その結果、本明細書で述べた測定の測定下限は、Se(IV)に対し5μg/L、Se(VI)に対し5μg/L、As(III)に対し1μg/L、As(V)に対し1μg/Lであると見積もられる。また本発明の方法による再現性がよいことが確かめられた。
実試料について、セレンとヒ素とに対して同時に価数別の定量分析を行った。試料としては、日本分析化学会が頒布する河川水認証標準物質JSAC 0302−3(総セレンの認証値が5.0±0.2μg/L、総ヒ素の認証値が5.2±0.1μg/L)、2種類の工場排水(工場排水A、工場排水B)を用いた。工場排水A、工場排水Bの水質を表1に示す。表1中に示した総ヒ素の濃度は、誘導結合プラズマ−原子発光分光分析(ICP−AES(atomic emission spectrometry))法によるものである。
Claims (6)
- 試料液に含まれるセレンを価数別に定量する方法であって、
アニオン性交換基を有するイオン排除カラムに硝酸と硫酸塩とを含む水溶液を溶離液として通液しつつ前記イオン排除カラムに前記試料液を注入し、前記試料液中のセレン化合物を価数別に分離する段階と、
前記イオン排除カラムから流出する液を誘導結合プラズマ質量分析装置に導入してセレンを定量する段階と、
を有する方法。 - 前記溶離液のpHが1以上5以下である、請求項1に記載の方法。
- 前記硫酸塩が、アルカリ金属の硫酸塩、アルカリ土類金属の硫酸塩、及び硫酸アンモニウムからなる群から選ばれた1以上の化合物である、請求項1または2に記載の方法。
- 前記溶離液における前記硫酸塩の濃度が、0.002mmol/L以上100mmol/Lである、請求項1乃至3のいずれか1項に記載の方法。
- セレンのほかにヒ素を含む前記試料液を前記イオン排除カラムに注入して前記試料液中に含まれるヒ素化合物をセレン化合物と同時に価数別に分離し、
前記誘導結合プラズマ質量分析装置においてセレンとヒ素とを同時に定量する、請求項1乃至4のいずれか1項に記載の方法。 - 前記イオン排除カラムは、強酸性カチオン交換樹脂を固定相として含む、請求項1乃至5のいずれか1項に記載の方法。
