細胞ベースのアッセイおよび分析化学
1 細胞生存性
1.1 Celltiter−Blue細胞生存性アッセイ(Promega)
このアッセイは、生存性の尺度として細胞がレサズリン(resazurin)を還元してレゾルフィン(resorufin)にする能力に基づく。TREx(商標)−293 Nox4細胞をT−225フラスコ内で培養し、トリプシン処理により採集し、細胞培地に再懸濁した。90μl中20,000個の細胞を96ウェルの細胞培養プレート(透明な底を備えた黒色のもの)に播種した。90μlの細胞培地のみを含むバックグラウンドプレートも1つ用意した。
このアッセイは、膜の統合性の尺度としての周囲の細胞培地における乳酸デヒドロゲナーゼ(LDH)活性に基づく。膜の統合性はアポトーシス、ネクローシスまたは化学物質による影響を受ける可能性がある。TREx(商標)−293 Nox4細胞をT−225フラスコ内で培養し、トリプシン処理により採集し、HBSSに再懸濁して細胞100,000個/mlにした。90μlの細胞懸濁液をV底ポリプロピレン製96ウェルプレートの各ウェルに添加した。HBSSのみを含むバックグラウンドプレートを1つ用意した。化合物をHBSS中に最終濃度の10倍に希釈し、ウェル当たり10μlを添加した。化合物を10μMの最終濃度で二重に試験した。
[(実験量-自然量)/(総量-自然量)]*100%
上記2種類の細胞生存性アッセイで試験した場合、本発明化合物はいずれも有意の細胞毒性を示さなかった。
Amplex(登録商標)Redベースのアッセイによる用量応答測定を下記に従って実施した:化合物の系列希釈を、リキッドハンドラーJanus(登録商標)(Perkin Elmer)およびスケジューリングソフトウェアOverlord(Process Analysis and Automation)に基づくシステムを用いて実施した。
マウス(10〜12週)、野生型C57BI6、6匹の動物を用いた:
対照(n=3)発作誘発マウスをビヒクルで2時間および12時間処理した;
試験(n=3)発作誘発マウスを本発明化合物M4(N2−(3,4−ジメチルフェニル)−6−((4−(3−メトキシフェニル)ピペラジン−1−イル)メチル)−1,3,5−トリアジン−2,4−ジアミン)で腹腔内処理した。
M4の注射量:投与量は3.2mg/kgであり、それぞれの動物につきそれぞれの投与事象(2時間および12時間)で計算した。
1. フィラメントで1時間ブロックすることによる一過性の中大脳動脈閉塞(transient middle cerebral artery occlusion)(tMCAO);
2. 1時間後にフィラメントを取り除くことにより再潅流;
3. 2時間後にM4の腹腔内注射、および12時間後に再び腹腔内注射;
4. 24時間後に動物をと殺し、トリフェニルテトラゾリウムクロリド(TTC)染色により梗塞サイズを測定する−したがって中大脳動脈閉塞(MCAO)の手順は1時間の閉塞および23時間の再潅流となる;
5. 発作分析を先の記載に従って実施した(Experimental Neurology 200 (2006), pp. 480-485; Circulation May 1, 2007, pp. 2323-2330)。梗塞サイズを測定するために、マウスをtMCAO、pMCAO、または皮質光血栓作成(cortical photothrombosis)の24時間後にと殺した。マウス脳スライスマトリックス(Harvard Apparatus)を用いて脳を2−mm厚さの冠状断面に切断した。切片を2% TTC(Sigma−Aldrich)で染色して梗塞部を視覚化した。病変体積を計算する面積測定(planimetric measurement)(ImageJ software,United States National Institutes of Health)を、先の記載に従って脳浮腫に対して補正した(Ann Neurol 2003;54:330-342)。
動物試験はRegierung von Unterfranken(ウンターフランケン行政府)により承認され、最近発表されたメカニズム推進による基礎発作試験(mechanism-driven basic stroke studies)における試験についての推奨に従って実施された。成体雄C57/BL6マウス(20〜25g)をCharles River(ドイツ、ズルツフェルド)から購入した。他の箇所に詳述したように、焦点性脳虚血を誘発するためにtMCAOモデルを用いた。簡単に言えば、マウスを70% N2O/30% O2混合物中の2%イソフルラン(isoflurane)で麻酔した。外科処置期間全体を通して、サーボ制御式加温ブランケットを用いて体心温度を37℃付近に維持した。正中線頚部切開を行なった後、規格化シリコンゴムコート6.0ナイロンモノフィラメント(60−1720RE,Doccol,カリフォルニア州レッドランズ)を右総頚動脈に挿入し、内頚動脈を経由して前進させて、MCA基部を閉塞した。1時間後、マウスを再麻酔し、閉塞フィラメントを取り除いて再潅流させた。梗塞の変動性を低減するために、すべての動物を同じ手術者が手術した;動物当たりの手術時間は15分を超えなかった。
マウスをtMCAOの24時間後にと殺した。脳を速やかに摘出し、マウス脳スライスマトリックスを用いて脳を2−mm厚さの冠状断面に切断した。切片をPBS中の2% 2,3,5−トリフェニルテトラゾリウムクロリド(TTC;Sigma−Aldrich,ドイツ、ゼールツェ)で染色して梗塞部を視覚化した。面積測定(ImageJ software,National Institutes of Health,メリーランド州ベテスダ)は処置グループを知らされていない試験員により実施され、それを用いて病変体積を計算し、他の箇所の記載に従って脳浮腫に対して補正した。TTC染色前に、この場合も2−mm厚さの冠状脳断面に切断した後(前記を参照)、脳全体について脳内出血(ICH)の発生を肉眼評価した。ICHを示す脳を梗塞体積の評価から除外した。それぞれ、M4で処置した3匹のマウスおよび3匹の対照マウスについての結果を表2に示す。図2は、それぞれ、M4で処置したグループおよび対照グループについての平均梗塞体積を示す棒グラフである。
