多くの細菌は、ストレス環境から遠ざかるか、または栄養源、O2、光および他の陽性刺激に向かうかの何れかで、特定の方向に移動するために鞭毛を利用する。細菌鞭毛は、その構築のために30を超える遺伝子産物を必要とする複雑な細胞機構である。サルモネラ・エンテリカ(Salmonella enterica)の場合、現在、生物発生およびその鞭毛機能に関与する60を超える遺伝子がある。これらの遺伝子は、3種類のプロモータークラスの転写階層に構成される。鞭毛転写階層の最上部は、ヘテロ多量体、転写活性化複合体を形成するマスター制御因子タンパク質FlhDおよびFlhCをコードするflhDCオペロンである。FlhD4C2複合体は、σ70RNAポリメラーゼがクラス2鞭毛プロモーターから転写するように支配する。クラス2鞭毛遺伝子は、鞭毛組み立てにおけるキーとなる構造中間体である、フック基底小体(HBB)と呼ばれる回転モーターの構造および組み立てに必要とされるタンパク質をコードする。HBBは鞭毛III型分泌(T3S)系を含み、これは、組み立て中に鞭毛タンパク質を細胞質から成長中の構造まで搬出する。HBB遺伝子発現に加えて、鞭毛クラス2転写は、σ28(FliA)およびFlgMを生成させる。これらは、鞭毛クラス3プロモーターの転写をHBBの完成と連結させる制御タンパク質である。σ28タンパク質は、RNAポリメラーゼが鞭毛クラス3プロモーターから転写するように支配する鞭毛特異的な転写因子である。クラス3遺伝子は、鞭毛フィラメントの構造遺伝子および細胞外リガンド濃度の変化に従い鞭毛回転の方向を調節する化学感覚シグナル伝達系の遺伝子を含む。HBB完成前に、FlgMはσ28に結合し、鞭毛クラス3プロモーター転写を阻止する。HBB完成時に、鞭毛T3S基質特異性の変化の結果、FlgM分泌および鞭毛クラス3プロモーターからのσ28依存性転写開始が起こる。T3S基質に対する分泌シグナルの必要性は未だ不明であるが、全基質は、構造が乱れたN末端ペプチド分泌シグナルを利用し、II型の分泌とは異なり、分泌過程中には切断されない。基質分泌は、分泌装置に対するT3Sシャペロンによる送達により促進されることが多い。FlgMタンパク質は、97アミノ酸長であり、その分泌は、N末端分泌シグナルに依存する。FlgM分泌は、その分泌シャペロン、FlgMのC末端半分に結合するσ28により大きく促進される。
FlgM核酸配列と、切断部位と、対象核酸配列と、を含む核酸コンストラクトが開示される。コンストラクトは、精製タグをコードする核酸配列をさらに含んでよい。
開示のコンストラクトは、FlgM核酸配列を含む。FlgM核酸配列は、野生型FlgMであってよい。一部の態様において、FlgM核酸配列は、FlgMの突然変異体配列であってよい。FlgMの突然変異体配列は、野生型FlgMと比較して、1以上のヌクレオチド突然変異を有し得る。一部の態様において、この突然変異は、コードされるアミノ酸配列を変化させない。一部の態様において、FlgMの突然変異体核酸配列中の突然変異は、コードされるFlgMペプチドが対象核酸配列によりコードされるペプチドの分泌のためにベクターとして作用する能力に影響を及ぼさない。
開示のコンストラクトは、FlgM核酸配列と対象核酸配列との間に切断部位を含んでよい。この切断部位は、タバコエッチウイルス(TEV)プロテアーゼ切断部位またはエンテロキナーゼ(ETK)切断部位であってよい。当業者にとって公知の他の切断部位が使用され得る。切断部位はFlgM核酸配列と対象核酸配列との間にあるものの、切断部位は必ずしもこれらの配列と連続していなくてもよい。言い換えると、精製タグをコードする配列は、切断部位のすぐ前または後であってよい。
開示のコンストラクトは、対象核酸配列を含む。対象核酸配列は、本明細書中で提供されるFlgM系を用いて、発現させ、精製しようとする対象ペプチドをコードし得る。
開示のコンストラクトは、精製タグをコードする配列をさらに含んでよい。精製タグの例としては、ポリヒスチジン、グルタチオンS−トランスフェラーゼ(GST)、Myc、HA、FLAGおよびマルトース結合タンパク質(MBP)が挙げられるが限定されない。したがって、これらの精製タグをコードする核酸配列が開示される。精製タグおよびそれらをコードする核酸配列は当技術分野で周知である。
開示のコンストラクトは、転写調節エレメント(TCE)も含んでよい。TCEは、それらに操作可能に連結される核酸配列の発現を駆動可能なエレメントである。本明細書に開示されるコンストラクトは、少なくとも1つのTCEを含む。TCEは、場合によっては構成的または制御可能であってよい。例えば、ParaBADプロモーターを含むコンストラクトが本明細書に開示される。ParaBADプロモーターは、アラビノース誘導性プロモーターである。これにより、アラビノースの存在または非存在でプロモーターの下流の核酸配列の発現をオンまたはオフにすることが可能となる。
本明細書中の上記および他の部分で開示の核酸コンストラクトによりコードされるポリペプチドも本明細書に開示される。例えば、FlgMと、切断部位と、対象ペプチドと、を含むポリペプチドが本明細書に開示される。ポリペプチドは、精製タグをさらに含んでよい。
FlgMを含むポリペプチドが開示される。FlgMは、野生型FlgMであってよい。一部の態様において、FlgMは、突然変異体FlgMであってよい。突然変異体FlgMは、野生型FlgMと比較して1以上のアミノ酸突然変異を有し得る。一部の態様において、突然変異体FlgM中の突然変異は、対象ペプチドの分泌のためにベクターとして作用するFlgMの能力に影響を及ぼさない。
開示のポリペプチドは、FlgMと対象ペプチドとの間に切断部位を含んでよい。切断部位は、TEVプロテアーゼ切断部位またはETK切断部位であってよい。切断部位がFlgMと対象ペプチドとの間にあるものの、切断部位は、FlgMおよび対象ペプチドと必ずしも連続していなくてもよく、言い換えると、精製タグは、切断部位のすぐ前または後にあり得る。
開示のポリペプチドは、対象ペプチドを含む。対象ペプチドは、本明細書中で提供されるFlgM系を用いて発現させようとするペプチドであってよい。
開示のポリペプチドは、精製タグをさらに含んでよい。精製タグの例としては、ポリヒスチジン、グルタチオンS−トランスフェラーゼ(GST)、Myc、HA、FLAGおよびマルトース結合タンパク質(MBP)が挙げられるが限定されない。
表1および4で提供されるような組み換え細胞株が開示される。
FlgMと、切断部位と、対象ペプチドと、を含有する開示のポリペプチドの何れかを含む細胞株を培地中で培養することを含む、対象ペプチドを生成させる方法が開示される。本方法は、培地から対象ペプチドを精製することをさらに含んでよい。
開示の方法を行うことまたはそれを行うことを支援するのに有用なキットとして何らかの適切な組み合わせで、上述の材料ならびに他の材料を一緒にパッケージ化し得る。ある種のキット中のキット構成要素が設計され、開示の方法で一緒に使用するのに適合化される場合、これは有用である。例えば、対象ペプチドを生成させるためのキット、開示の組み換え細胞株のうち1つを含むキットが開示される。本キットは、培地も含有し得る。
実施例
本試験では、成熟生成物においてジスルフィド架橋を形成する高密度のシステイン残基を含有する、小型で極めて安定であり、薬理活性を有するポリペプチドの分泌のためにベクターとして鞭毛FlgMタンパク質を利用した。原理の証明として、サルモネラ・チフィムリウム(Salmonella typhimurium)におけるμ−コノトキシンSIIIAの組み換え発現のために、細菌分泌系を操作した。鞭毛III型分泌(T3S)装置を用いて、図1に示すように、組み換えコノトキシンを培地に選択的に分泌させた。
i.細菌株、プラスミドおよび培地
使用され得る代表的な細菌株を表1に記載する。Luria−Bertani(LB)培地中で細胞を培養し、必要に応じてアンピシリン(100μg/mL)またはテトラサイクリン(15μg/mL)を補給した。S.チフィムリウム(S.Typhimurium)P22 HT105/1 int−201の一般形質導入ファージを形質導入クロス(transductional crosses)において使用した。

SIIIA(QNCCNGGCSSKWCRDHARCCGR;配列番号2)をコードする全66塩基対をカバーする長いプライマーSIIIA_long_fw(CAGAACTGCTGCAACGGCGGCTGCAGCAGCAAATGGTGCCGCGATCATGCGCGCTGCTGCGGCCGC;配列番号1)を用いて、デ・ノボフィルインPCR反応においてSIIIAを増幅した。サルモネラ・チフィムリウム(Salmonella typhimurium)の最適コドン使用に従い、SIIIA配列を設計した。鋳型プライマーの3’末端からのフィルインを開始する短いリバースプライマー(SIlIA_short_rv:GCGGCCGCAGCAGCGCGCATG;配列番号3)を活用して、66bp配列を2本鎖化した。

SIIIAコノトキシン融合物を発現する株を新鮮な単一コロニーから採取し、10mLのLB中で一晩増殖させた。一晩培養物を1Lの新鮮培地に入れて1:100希釈し、6時間増殖させた。適切な場合、0.2%アラビノースの添加によって、最初の2時間後にSIIIA発現を誘導した。遠心(7,000rpm)によって細胞をペレット化し、残存細菌を除去するために、FlgM−SIIIAを含有する上清を0.22μmポリエーテルスルホンフィルター(Corning,NY,USA)−低タンパク質結合膜に通過させた。さらなる精製のために、3gのNi−IDA樹脂(Protino Ni−IDA,Machery−Nagel)を充填した重力流カラム(Bio−Rad)を使用し、250mMイミダゾールを含有する緩衝液を用いてpH7.5でネイティブ条件下でアフィニティータグ付加タンパク質を溶出した。
一晩培養物をLB中で1:100希釈し、0.2%L−アラビノースを添加することによって個々のFlgM−毒素融合物の発現を誘導する前に、37℃で2時間増殖させた。さらに4時間にわたり細胞を37℃で維持し、その間に融合タンパク質を発現させた。全部で6時間後、全ての株に対して600nmでの光学密度(OD)を測定した。
全細胞溶解液および培養上清の発現FlgM−毒素融合物をSDSポリアクリルアミドゲル電気泳動に供し、検出のためにポリクローナル抗FlgMおよびポリクローナル抗FliK抗体(ウサギ)を用いて免疫ブロットすることによって分析した。LI−COR Odysseyイメージングシステムを用いた化学発光または赤外線検出によって抗原−抗体複合体を可視化した。化学発光による発色の場合、ホースラディッシュペルオキシダーゼ(HRP)と複合体化した二次ヤギα−ウサギ抗体(Bio−Rad)およびECL検出キット(Amersham Biosciences)を使用した。赤外線検出の場合、二次抗ウサギ−IRDye690(LI−COR)を使用した。Mac OS X用のImageJ 1.45sを用いてFlgM−SIIIAおよびFliKバンドのデンシトメトリー測定を行った。
AcTEVプロテアーゼ(Invitrogen)を用いて、鞭毛分泌基質から、SIIIAを切り出した。450UのTEVプロテアーゼを、750mL培養物からの分泌型タンパク質に相当する15mLの溶出分画に添加した。室温で一晩、1mMジチオスレイトール(DTT)および0.5mM EDTAを含有する溶出緩衝液中で切断を行った。
1mM酸化グルタチオン(GSSG)および0.1mM EDTAを用いて0.1M Tris緩衝液、pH7.5中で組み換えSIIIAを折り畳ませた。折り畳み反応を一晩室温で進行させ、8%の最終濃度になるようにギ酸を添加することにより反応を停止させた。
C18固相抽出カラム(Supelclean LC−18,Supelco)を使用して、折り畳みおよび切断反応の他の構成要素から酸化ペプチドを除去した。次に、単離ペプチドを乾燥させ、二重波長吸収検出器およびVydac C18分析カラムを備えたWaters600クロマトグラフ上でHPLCによって精製した。全体にわたり0.1%TFAに維持しながら、4.5%〜33.5%(aq)のアセトニトリル(0.9%変化/分)の直線勾配で、HPLC分離を行った。これによって、適正な折り畳み生成物の精製が可能になり、マトリックス支援レーザー脱離/イオン化法飛行時間型(MALDI−TOF)質量分析によって質量を確認した。
NaV1.2を発現するアフリカツメガエル卵母細胞を用いて組み換えSIIIAの機能的活性を電気生理学的に評価した。簡潔に述べると、卵母細胞をND96で灌流され、2本の電極が固定された、30μLの記録チャンバー中に置いた。−80mVの保持電位を使用し、20秒ごとに50ms、段階的に電位を−10mVまで変化させることによって、ナトリウムチャネルの活性化を達成した。次のように、ペプチドを保護するために静止浴中で毒素への曝露を行った。毒素適用前に、浴槽の灌流を停止し、対照反応を記録した。次に、ペプチド(最終濃度の10倍で3μL)を浴槽に適用し、マイクロピペットを用いてこれを5秒間撹拌した。約20分間、ペプチドによる遮断の開始の記録を得、その後、ND96での灌流を再開して未結合のペプチドを除去し、一方で約20分間にわたり遮断からの回復速度を監視した。遮断の開始の時間経過を単一指数関数にフィットさせ、遮断の実測の速度定数、kobsを得た。ペプチド洗い流し中の遮断からの回復速度は、遅すぎて曲線フィッティングにより測定できなかったので、洗浄の20分後の回復レベルおよび遮断の指数関数的減衰を仮定してkoffを推定した。全記録を室温で得た。
蛍光顕微鏡分析を行った。
ウミイモガイの毒腺および電位依存性ナトリウムチャネル(VGSC)を含む標的イオンチャネルからコノトキシンを合成する。SIIIAは、カエルにおけるテトロドトキシン(TTX)−耐性VGSCおよびげっ歯類におけるTTX−感受性VGSCを阻害する、コヌス・ストリアツス(Conus striatus)からのμ−コノトキシンである。μ−コノトキシンSIIIAは、VGSCのα−サブユニットの部位1に結合し、それによってチャネル孔を塞ぎ、ナトリウム伝導性を遮断する。生理学的条件下で、2つの形態のSIIIA:前駆体型および成熟ペプチドが存在するが、この成熟ペプチドは、アミノ基に対してC末端グリシン−アルギニン残基をプロセシングし、N末端グルタミン残基をピログルタミン酸に変化させることによって修飾されている。本明細書中で、2つの形態のSIIIA、前駆体ペプチドおよび、生理学的成熟型(ピログルタミン酸の代わりにN末端グルタミン酸)と最も密接に類似するペプチドを組み換えにより発現させた。哺乳動物調製物において、μ−SIIIAは、NaV1.2および1.6などの神経性ナトリウムチャネルおよび骨格筋サブタイプNaV1.4を効果的に遮断する。SIIIAはVGSCを標的とし、マウスにおいて強力な鎮痛活性を有するので、医学研究および薬物探索のために使用され得る。T3SS細菌発現系を開発することにおける基本的な利点は、翻訳後、鞭毛構造において狭い分泌チャネルを通じてポリペプチドが移動させられ、それによって、封入体での凝集の問題が回避されることである。ポリペプチド分泌は、細胞質中での分子間凝集を防ぐ。ポリペプチド鎖が細菌細胞質の還元環境に存在し、酸化する細胞外環境に入るので、分子内ジスルフィド結合が形成される。
本試験において、組み換え小型ペプチド発現の封入体問題を回避する発現系を使用する新しいアプローチを開始した。これは、鞭毛分泌シグナル、例えばフックタンパク質FlgEまたはフラジェリンFliCに融合される場合、非鞭毛タンパク質を輸送することが示されているサルモネラ・エンテリカ(Salmonella enterica)血清型チフィムリウム(Typhimurium)(サルモネラ・チフィムリウム(Salmonella typhimurium))の鞭毛分泌系を活用する。鞭毛分泌系は、細胞質から外部媒体にタンパク質を選択的に分泌する細菌III型分泌系のファミリーのメンバーである。今までのところ特徴が調べられているほぼ全てのIII型分泌系は、植物および動物病原体の多くに対する毒性決定因子の分泌または細菌鞭毛の構造および機能のために必要とされるタンパク質分泌の何れかに必要とされる。細菌鞭毛は、細胞壁および膜内に埋め込まれた回転モーターに連結される、細胞表面から多くの体長を延長させる外部ヘリックスフィラメントから構成される。サルモネラの場合、化学感覚系は、鞭毛回転の時計回りおよび反時計回りの方向を調節し、これにより、細菌が、栄養などの誘引物質に向かって、または細菌にとって有害である細菌忌避物質から遠ざかるように移動することができるようになる。
i.細菌株および増殖条件
本試験に使用した株は全て、野生型株サルモネラ・エンテリカ(Salmonella enterica)血清型チフィムリウム(Typhimurium)であるLT2の誘導体であり、表4に記載する。一連のこの実験中に全株を構築した。株構築は、P22を介して行われる形質導入またはλRedを介した標的化突然変異誘発の何れかを利用した。そのネイティブプロモーターから発現されるflhDCオペロンがある株の場合、一晩静置した培地から新鮮LB培地(10g Bactoトリプトン、5g Bacto酵母抽出物および5g NaCl/L)への100倍希釈液によって鞭毛遺伝子発現が誘導された。トランスポゾンTn10のtetAプロモーターから発現されるflhDCオペロンがある株(ΔPflhDC8089::tetR−PtetA)の場合、テトラサイクリン類似体アンヒドロ−テトラサイクリン(1μg/mL)の付加によって鞭毛遺伝子発現を誘導した。この実験で使用した株の場合は全て、アラビノース利用オペロンaraBADをFlgM+遺伝子またはFlgM遺伝子融合物FlgM−6His−TEV−δ−SVIEまたはFlgM−6His−ETK−δ−SVIE(それぞれ、ΔaraBAD::FlgM+、ΔaraBAD::FlgM−6His−TEV−δ−SVIEまたはΔaraBAD::FlgM−6Hi−ETK−δ−SVIEI)で置換した。これにより、増殖培地へのL−アラビノースの添加によって、ParaBADプロモーターからのFlgM、FlgM−6His−TEV−δ−SVIEおよびFlgM−6His−ETK−δ−SVIE産生の誘導が可能となった。flhDCオペロンの誘導の2時間後に、アラビノースを0.2%最終濃度になるように添加した。さらに5時間後、培養物を10,000gで30分間遠心し、細胞をペレット化した。0.2μm低タンパク質結合フィルター(Acrodisk Syringe Filter,PALL Life Sciences)で上清をろ過し、残存細胞を除去した。TCA(トリクロロ酢酸)を10%最終濃度になるように添加することによって、ろ過上清中の分泌型タンパク質を沈殿させた。5mM PMSF(フッ化フェニルメチルスルホニル)を含有する冷PBS(リン酸緩衝塩水)中で細胞ペレットを再懸濁し、続いて超音波処理を行い、細胞縣濁液を溶解させた。細胞溶解液は、全細胞タンパク質について直接分析するか、または4℃で30分間遠心(15,000g)することによって、可溶性および不溶性分画分析のために分離した。FlgM分泌における様々な濃度のNaClおよびKClの影響を試験するために、LB培地をNaClなしで調製し、NaClまたはKClの何れかを所望の濃度になるように添加した。

分泌型の可溶性、不溶性または全細胞タンパク質のレベルをウエスタンブロットによって分析した。14%勾配ゲル(BIO−RAD)を用いて、全細胞溶解液および培養上清中の発現DnaK、FlgMおよびσ28レベルをSDS−PAGEにより調べた。ΔaraBAD::FlgM+コンストラクトを含有する株の分析、50単位および100OD単位の同等物をそれぞれ細胞および上清分画に対して載せた。FlgM−6His−TEV−δ−SVIEおよびFlgM−6H−ETK−δ−SVIE分泌について株を分析するために、50および300OD単位を細胞および上清分画に対してそれぞれ載せた。検出のために、抗DnaK(マウス)、抗σ28および抗FlgM抗体(ウサギ)を使用した。ローディング濃度に対して、および細胞溶解ゆえの上清中のタンパク質の存在に対して、タンパク質標準対照としてDnaKを使用した。抗原−抗体複合体を可視化するために、二次抗ウサギ−IRDye690および抗マウス−IRDye800抗体(LI−COR)を使用した。LI−COR Odyssey赤外線撮像システムソフトウェアを用いて、FlgM、σ28およびDnaKバンドのデンシトメトリー測定を行った。培養試料において3つ組で全アッセイを行った。
運動性アッセイは、軟寒天トリプトンプレート(1Lあたり:10g Bactoトリプトン、5g NaCl、3g Bactoアガー)を利用した。爪楊枝で細菌コロニーを拾い、軟寒天に突き刺し、続いて37℃で約5時間温置した。必要に応じて、ParaBADまたはflhDCオペロン誘導のために、それぞれ、アラビノース(0.2%)またはアンヒドロ−テトラサイクリン(1μg/mL)の何れかを添加した。
i.ParaBAD::FlgMから産生されるFlgMが分泌される。
FlgMは、外部の消費された増殖培地に完成したHBBを通じて分泌される。精製前に細胞を溶解させる必要がないタンパク質精製のために、FlgMのC末端への外来タンパク質の融合を使用し得ることが示されている。分泌シグナルとしてFlgMを用いたタンパク質精製のために鞭毛T3S系を発展させるために、FlgM産生および分泌の既知の態様は、この系を用いてタンパク質産生を最大化することを特徴とした。FlgM遺伝子は、flgA MNオペロンにおいてクラス2鞭毛プロモーターから転写される。この結果、最初のHBB組み立て中にFlgM産生が起こる。クラス2産生FlgMは、2つのfliAZ転写物のうち1つにおいてクラス2転写を介しても産生される、fliA遺伝子の産物であるσ28タンパク質と結合する。HBB完成時に、鞭毛T3S系の分泌基質特異性の変化の結果、FlgM分泌およびσ28依存性クラス3転写の開始が起こる。FlgMおよびσ28は、それぞれσ28−依存性FlgMNおよびfliAZ転写物から産生され続ける。約80%の定常状態FlgM転写は、そのクラス3プロモーター由来である。FlgMは抗σ28因子であるので、クラス3プロモーターを介したその産生は、自己阻害下にある。この例において、アラビノース−誘導性染色体araBADプロモーター(ParaBAD)から転写されるFlgM遺伝子があるコンストラクトを使用することによって、FlgM遺伝子転写をFlgM自己阻害から外した。染色体araBADオペロンの標的化欠失と、それに続くその場でのFlgM+遺伝子の挿入によってこれを完遂した。この結果、araBAD構造遺伝子の欠失ゆえにアラビノース誘導因子が分解されない株においてFlgM産生のアラビノース−依存性誘導が生じた。ParaBADから転写されたFlgMは、そのネイティブプロモーターから生成され、分泌されるFlgMより高いレベルで、アラビノースの存在下で生成され、分泌された(図4)。鞭毛構造を形成しないfliF欠失株は、ParaBAD−誘導性FlgMを分泌することができなかった(図4)。これらの結果から、FlgM内在性ペプチド分泌シグナルが高レベルFlgM分泌に十分であり、鞭毛−依存性T3S系をFlgM分泌のために使用し得ることが示される。
flhDCオペロンは、鞭毛転写階層の最上部にある。flhDCの制御は複雑であり、flhDCプロモーター領域内に6個の既知の転写開始部位がある。カノニカルなTATAAT配列(PflhDC7793対立遺伝子)に対する、flhDCプロモーター領域内のP1およびP4転写開始部位に対する−10配列の変化の結果、細胞あたりのHBB構造数が2倍になり、鞭毛フックタンパク質の産生および分泌が向上した。他の突然変異の結果、フックタンパク質分泌が増加し、細胞あたりのHBB構造が増加し、その結果、flhDCオペロン転写もしくは転写後調節の既知の阻害剤の発現低下または除去が生じた。ParaBADから転写されたFlgMの分泌におけるflhDC制御因子の突然変異の影響を試験した。この例に含まれるflhDC発現の既知の転写および転写後阻害剤は、EcnR、RscB、LrhA、FliT、DskAおよびYdiVであった。EcnRは、FlhDC介在性自己抑制に関与する。FlhD4C2複合体は、ecnRの転写を支配し、同様に、その結果、RcsBタンパク質に呼応してflhDC転写の抑制が起こる。膜および細胞壁の損傷に反応して莢膜多糖合成および遺伝子数を制御するRcsBは、flhDC転写のリプレッサーである。LrhAはまた、flhDCプロモーター領域内で結合し、flhDCオペロン転写を阻害することが示されているflhDCの制御因子でもある。FliTは、クラス2およびクラス3鞭毛プロモーターの両方から転写される。FliTは、FlhD4C2複合体に結合し、鞭毛クラス2転写の活性化を妨げる。また、FliTは、鞭毛フィラメントキャッピングタンパク質FliDに対する分泌シャペロンでもある。HBB完成後のFliDの分泌によって、HBB完成とFlhD4C2によるさらなるクラス2転写の阻害が連結される。DskAタンパク質は、flhDCの転写を阻害するためにppGppとともに作用する。また、DskAは、RpoS−介在機構を通じてflhDC転写を阻害し得るrpoS翻訳も刺激し得る。YdiVは、栄養増殖条件での変化に反応した分解に対してClpXPプロテアーゼにFlhD4C2を標的化する転写後制御因子である。対照として、flhDC mRNA安定性の正の制御因子であるCsrAを使用した。flhDCプロモーター調節領域をトランスポゾンTn10からのtetR−PtetAカセットで置換することによって、PflhDC8089対立遺伝子を構築した。この結果、テトラサイクリン類似体アンヒドロ−テトラサイクリンの添加により誘導され得る、tetAプロモーターの調節下でのflhDCオペロンの置換が生じた。フック産生および分泌を増加させる、flhDC P1およびP4カノニカルプロモーターアップ(up)変化も試験した(PflhDC7793対立遺伝子)。ParaBADから発現される分泌型FlgMタンパク質のレベルにおけるflhDC制御突然変異の影響を図5で示す。
HBBの完成は、ロッドフック型の分泌基質から後期またはフィラメント型分泌基質への鞭毛T3S基質−特異性の切り替えと一致する。後期分泌基質は、フック−フィラメント接合部タンパク質(FlgKおよびFlgL)、フィラメントキャップタンパク質(FliD)、あるいは発現フィラメントタンパク質(FliCまたはFljB)および抗σ28因子FlgMを含む。各後期分泌基質の効率的な分泌には、同系のT3Sシャペロンの助けが必要である。これらは、FlgN(FlgKおよびFlgLの場合)、FliT(FliDの場合)、FliS(FliCおよびFljBの場合)およびσ28(FlgMの場合)を含む。T3Sシャペロンは3種類のクラスに分けられる:i)分泌前にその系において基質に結合し、タンパク質分解から保護するもの、ii)基質分泌を促進するものおよびiii)分泌を安定化し、かつ促進するもの。σ28タンパク質は、後者のカテゴリーに入り、これは、細胞質においてFlgMをタンパク質分解から保護し、おそらく鞭毛の基部へのFlgMの局在化を促すことによってFlgMの分泌を促進する。−10および−35プロモーター配列(R91CおよびL207P)の認識を欠如させる2個のアミノ酸置換があるσ28の突然変異体は、FlgM分泌に対するそのT3Sシャペロン活性を保持する。このことから、σ28のT3Sシャペロン機能が、その転写活性とは個別の過程であったことが示された。
HBB完成時のFlgM分泌の最初の報告から、分泌型FlgMレベルがFlgMに対する分泌競合物候補であるフィラメントタンパク質を欠く株でより高かったことを示す定性的データが与えられた。分泌型FlgMレベルにおいて潜在的な後期分泌−基質競合物を除去する影響を試験して、それらの除去によって、分泌型FlgMの収率が向上するか否かを調べた。組み立てられた鞭毛における後期基質サブユニット数は、フック−フィラメント接合部タンパク質FlgKおよびFlgLに対してそれぞれ約11であり、フィラメントキャッピングタンパク質FliDに対して5であり、FliCまたはFljBに対しては、最長で20,000のフィラメント長に依存する。図7A,図7Bに示す結果は、FlgK、FlgLまたはFliDの除去の影響は、分泌型FlgMレベルにおいて殆どない一方で、フィラメント基質除去の結果、分泌型FlgMレベルが野生型よりも最大で1.9倍高くなったことを示す。この結果は、後期T3S基質レベルが鞭毛を通じたFlgMの分泌において重要な制限因子ではないことを示す。
ParaBADから発現されるFlgMの分泌型レベルにおける、後期分泌シャペロン、FlgN、FliSおよびFliTの除去の影響も試験し、これらが搬出のためのFlgMの鞭毛分泌系への送達に対してσ28と競合するか否かを調べた。また、T3Sシャペロンは、それらの同系分泌基質の非存在下での制御機能と関連付けられる。FlgKおよびFlgLに対するT3SシャペロンであるFlgNは、FlgM mRNA翻訳を阻害する。σ28は、鞭毛クラス3プロモーターに対する転写因子であり、FliTは抗FlhD4C2因子として作用する。FliSのみが、その同系分泌基質FliCおよびFljB非存在下で制御機能を有することを報告されていない。
flhDCオペロンは、鞭毛レギュロンおよびSpi1の遺伝子の両方のマスターオペロンである。Spi1レギュロンは、Spi1インジェクチソーム(injectisome)T3S系の構造および組み立てに必要とされる遺伝子をコードする。fliZ遺伝子はfliAZオペロンにおいて転写され、FliZは、その産物がSpi1転写を同様に活性化するhilDの転写を活性化する。HilD活性化遺伝子の一産物であるRtsBは、鞭毛−Spi1レギュロン全体に対するフィードバックループを提供するflhDC転写を抑制するように作用する。過剰発現FlgM(ParaBADから)の分泌型レベルにおけるSpi1およびSpi2サルモネラ病原性系の両方の欠失の影響を試験した。Spi1の欠失の結果、Spi1誘導条件下で細胞を増殖させなかったにもかかわらず、FlgM分泌型レベルが野生型の場合の55%に低下し、一方、Spi2の欠失は、FlgMの分泌型レベルに顕著な影響がなかった(図9A,図9B)。
細胞質からタンパク質収率を向上させるために使用される共通技術は、細胞性プロテアーゼを除去することによる。さらに、OmpTなど、外膜に存在するプロテアーゼは、細胞溶解後の分解によって、タンパク質収率を低下させ得る。ClpAおよびClpXタンパク質は、分解のためClpPセリンプロテアーゼへの送達のための様々な基質と相互作用する。DegPは、広い基質特異性を示す周辺質プロテアーゼである。DegPは、非折り畳み、誤局在化、ハイブリッドおよび、周辺質における過剰発現から不適切に折り畳まれている組み換えタンパク質を排他的に対象とする。
高浸透圧によって、S.エンテリカ(S.enterica)においてSpi1 invA遺伝子転写が上昇し、Spi1依存性III型分泌は、高塩分条件下で増殖させた細菌でのみ生じた。ParaBAD−FlgM+により産生されるFlgMの分泌型レベルにおける高NaClまたはKClの何れかの影響を試験した。NaCl濃度を200mMに上昇させ、次いで400および600mMの濃度で低下させた場合、FlgM分泌型レベルが上昇した。200mM NaClで、FlgM分泌型レベルは、標準的なLB培地中のNaCl濃度(86mM)と近い100mM NaClのレベルよりも3.7倍高い(図11A,図11B)。したがって、LB培地中のNaCl濃度は、FlgM分泌にとって最適ではない。KClは、分泌型FlgMレベルにおいて同様の影響を有し、200および400mM KClによって、分泌型FlgMの最大レベルが、100mM NaClの分泌型FlgMレベルと比較して3.2倍および3.6倍となった。これは、細胞質中でのFlgMの溶解度における影響によるものである。軟寒天での運動性はσ28−依存性鞭毛クラス3転写の阻害ゆえにFlgM誘導条件下で抑制されたにもかかわらず、イオン強度の上昇は、軟寒天での運動性においても正の効果を有した(図11C)。
ParaBAD::FlgM+によるFlgM過剰発現状態下でのこの収率を最大化した株および条件を得るために、分泌型FlgMの収率を向上させた上述の個々の結果を組み合わせた(図12A,図12B)。これらの株の一部(例えばfljBenx vh2およびclpXを含有する株、flhD8089を含有する株など)は、最大FlgM分泌レベルを与える。これらの全てが、野生型の約5倍の分泌型FlgMレベルを生じさせる。これらの株において、細胞内に蓄積したFlgMは微量であり、このことから、細胞FlgMの発現および安定性が限定的であったことが示される。
δ−SVIEタンパク質は、有毒ウミイモガイにより産生される小型ペプチドであり、脊椎動物神経筋系においてナトリウムチャネルを阻害する。この31アミノ酸ペプチド(DGCSSGGTFCGIHPGLCCSEFCFLWCITFID)は疎水性であり、3対の分子内ジスルフィド結合を形成する6個のシステイン残基を含有する。本ペプチドの疎水性の性質とδ−SVIEの活性立体構造を生成させるための複数のジスルフィド結合形成に対する要件とが、このペプチドが大腸菌(E.coli)で過剰発現される場合、適正な折り畳みを妨げる障害でとなる。したがって、FlgM介在分泌を介した活性型δ−SVIEの産生について試験した。細胞からの分泌はFlgMのN末端から開始するので、FlgM−δ−SVlE融合物のδ−SVIE配列は、N末端からC末端に、それがリボソームを出るように細胞から出る。これは、活性立体構造への、δ−SVIEの適正な折り畳みおよびジスルフィド形成を促進し得る。2つのタンパク質間で操作されたTEVまたはETKプロテアーゼ切断部位の何れかを有するヘキサ−His−タグ付加(6His)FlgMへのδ−SVIEのC末端融合物を染色体ParaBAD発現遺伝子座から発現させた。6Hisによって、Ni−アガロースアフィニティーカラムを用いた精製が容易になり、TEVおよびETK切断部位はそれぞれTEVプロテアーゼおよびエンテロキナーゼにより認識され、これによって、分泌型δ−SVIEがそのFlgM分泌シグナルから分離されるようになる。
鞭毛およびインジェクチソーム(injectisome)のIII型分泌(T3S)系は、T3Sシグナルに融合される組み換えタンパク質の産生および精製に対する経路を提供する。N末端が異常なペプチドシグナルおよび分泌燃料としてのプロトン駆動力への連結があり得る。全てではないが多くのT3S基質は、細胞質におけるそれらの安定性のために、および/またはT3S基質を細胞質膜における分泌装置に標的化するための分泌パイロットとして、同系T3Sシャペロンを利用し得る。T3S系の別の特性は、初期から後期分泌基質特異性への分泌−特異性切り替えを受ける能力である。鞭毛系において、これは、40nmを超える長さである細胞外フック成長の完成時に起こり、その結果、フィラメント型基質分泌および組み立てへの移行が起こる。FlgMタンパク質は、フック完成からフィラメント組み立てへの移行の不可欠な部分である。FlgMは、後期フィラメント型鞭毛分泌基質および抗σ28因子である。FlgMは、小型の97アミノ酸タンパク質である。FlgMのN末端半分はT3Sシグナルを含み、一方でC末端半分は抗σ28相互作用ドメインを含む。FlgMのC末端への組み換えタンパク質の融合によって、精製前に細胞を溶解させる必要なく、それらの分泌および精製が可能となることが示された。FlgM産生および分泌を促進する条件を調べた。次に、組み換えタンパク質FlgM−6His−TEV−δ−SVIEおよびFlgM−6His−ETK−δ−SVIEを作製し、鞭毛T3S系を介して細胞から−δ−SVIEタンパク質の分泌を支配するためのベクターとしてFlgMを用いて細胞溶解なくこれらのタンパク質を作製し、精製するために、これらの条件を適用した。
多数のジスルフィド結合を有するコノペプチドおよび、その不安定性のために細胞質中で検出不可能なジフテリア毒素のAサブユニットなど、以前生成困難であったタンパク質の分泌を促進するための分泌シグナルとしてのFlgMの使用から、タンパク質産生および、さもなくば産生されないタンパク質の精製に対するこの系の有用性が明らかとなる。
最適分泌株を同定し得る。flhDCプロモーターのプロモーターアップ(up)対立遺伝子であるflhD7793およびflhDCプロモーターの、アンヒドロテトラサイクリン(AnTc)誘導性tetAプロモーターでの置換の結果生じる対立遺伝子であるflhD8089によって、それぞれ、FlgM分泌が実質的に増加し、これは、flhDC転写の阻害剤:FliT、LrhA、DskA、RcsBまたはEcnRの除去により観察されるものよりも大きい(図5A)。AnTc−誘導性flhDCオペロン(flhD8089対立遺伝子)は注目すべきものであるが、それは、これが、全ての既知の阻害剤タンパク質の結合部位に対して欠失しており、発現に対してAnTcにのみ依存するからである。flhD8089対立遺伝子は、FlgMの分泌シャペロンのfliA(σ28)H14N対立遺伝子と組み合わせられ得るが、これは、FlgM分泌において最大の正の影響に対して示した(図6A)。clpXプロテアーゼ突然変異体対立遺伝子とのさらなる組み合わせによって、分泌が増加し得る(図10A)。FlgM鞭毛III型分泌シグナルに融合した対象タンパク質も発現する株においてこの遺伝子バックグラウンドを使用することによって、特に200mM NaClまたはKCl存在下で増殖させられる場合、対象タンパク質の産生が向上し得る(図11A)。
σ28タンパク質は、fliA遺伝子およびFlgMに対するIII型分泌シャペロンの産物である。σ28レベル上昇の結果、分泌型FlgM融合タンパク質基質のレベルが上昇する(図6A)。σ28タンパク質はまた、タンパク質精製系のためにも使用され得る。FlgM−σ28複合体の見かけのKdは、2x10-10Mである。この非常に高い親和性は、FlgMの精製の結果、σ28が同時に精製され得、その逆も同様であることを意味する。インテイン自己切断部位により挟まれたキチン結合ドメイン(CBD)に融合されたFlgMが構築されている。この融合キメラを発現する細胞からの抽出物をキチンカラム全体に注いだ場合、σ28付きの複合体中のFlgM−インテイン−CBDキメラがカラムに結合した。還元剤の添加によって、精製FlgM−σ28複合体を放出するインテイン自己切断が触媒された。キチンカラムにσ28−CBDを結合させるのは、この態様を使用し得る。発明者らの鞭毛分泌株から産生されるFlgM−インテイン−Xキメラ(Xは対象タンパク質である。)の消費増殖培地をこのようなカラム全体に注ぎ得る。インテイン自己切断の誘導により、純粋な形態のタンパク質Xが放出され得る。これは、低コストで高収率のタンパク質産生系を提供し得る。

- 対象ペプチドを作製する方法であって、培地中で請求項17〜31の何れかに記載のポリペプチドを発現する細胞株を培養することを含み、前記ポリペプチドが前記対象ペプチドを含む、方法。
