及び薬学的に許容されるその塩で示される。
本研究には3つの目的があった。(1)オペラント再燃・再発モデルにおける状況関連の条件刺激としての音楽の効力を検証する、(2)in vivo微小透析を使用して、音楽に誘発される薬物探索の再発中にドパミン作動性の変化が起こるかどうかを判定する、及び(3)刺激に誘発される薬物探索行動を減じる18−MCの有効性を評価する。全ての研究は、自己投与、消去及び再発モデルを使用して実施し、研究においてラットは音楽の刺激の有無の下でコカインを求めてレバー押しをした(表1)。本研究の結果は、状況関連の刺激及びこれに関連付けされた薬物探索行動の根底にある機序に新たな見識をもたらし、複雑な状況関連の刺激の存在下でも、再燃の治療薬となり得る18−MCの効力を示してもいる。
動物
実験開始時に体重約250gのナイーブな雌のSprague−Dawleyラット(Taconic Germantown,ニューヨーク州)を、温度及び湿度を調整したコロニールームに一匹ずつ収容し、標準的な12:12昼/夜サイクルとした。餌と水を自由に与えた。「Guide for the Care and Use of Laboratory Animals」(1996)に従ってプロトコルをデザインして実行し、Albany Medical CollegeのInstitutional Animal Care and Use Committeeの承認を受けた。実験手順の前にラットを一週間順化させた。
コカイン塩酸塩(約0.4mg/kg/注入、Sigma−Aldrich,セントルイス,ミズーリ州)を、0.9%塩化ナトリウムに、2mg/mlの薬物対生理食塩水の比で溶解し、次いで、中性の生理的pHにしてから静脈内(i.v.)自己投与セッションで使用した。18−MC(40mg/kg,Obiter Research LLC.シャンペーン,イリノイ州)及び生理食塩水を、両方腹腔内(i.p.)に投与した。動物に、頚静脈孔と微小透析カニューレ挿入の両方の手術をするため、ナトリウムペントバルビタール(50mg/kg、i.p.)で麻酔をした。ナトリウムメトヘキシタール(10mg/kg)を使用してカテーテルの開存性を確認した。微小透析実験と一緒に使用した他の全ての試薬は、現地の供給業者から得られた、分析用のものであった。
マイルス・デイヴィスの「Four」(Prestige Blue Haze,1954)を、この実験で状況関連の刺激として使用する楽曲とした。マイルス・デイヴィスの曲目を選んだ理由は、発明者らの研究所において、過去の条件付け例で使用して成功したからである(Polston et al.,2011b)。この曲目が最初に選ばれた理由は、反復性のビートとメロディがあり、認知と識別を容易にする助けとなるからである。薬物トレーニング、自己投与、及び適用試験の各セッション中、「Four」を、65〜75デシベルの間にとどめたボリュームで連続ループでかけた。このデシベル範囲を選んだ理由は、ラット及び音楽に関与する過去の調査において使用されて成功したからである(Feduccia et al.,2008;Otsuka et al.,2009;Polston et al.,2011b)。
実験は、吸音発泡体を取り付けた防音の箱の中に設置したラットオペラント条件付けチャンバー(ENV−009,Med Associates,セント・オールバンズ,バーモント州)の中で実施した。オペラント箱を室内ファンで常に換気し、試験チャンバーの前壁に2つの格納可能なレバーを約20cmの距離を置いて備え、後壁に室内照明を据え付けた。オペラント試験チャンバーの下に設置した注入ポンプ(PHM−100VS,Med Associates,セント・オールバンズ,バーモント州)を、i.v.薬物送達のために、ポリエチレンチューブ及びInstech(375/22PS)スイベルと組み合わせて使用した。ステレオスピーカー(Orb Audio,ニューヨーク,ニューヨーク州)を、天井から取り付け、オペラント箱の中心の上方につり下げた。このスピーカーを、オペラント試験チャンバー内で、音響を調節するステレオ受信機(ソニー株式会社、東京、日本)と接続した。さらに、赤外線デジタルビデオカメラ(Clover Inc.,セアリトス,カリフォルニア州)をオペラント箱の天井から取り付け、試験チャンバーの床を、遮られることなく観察できるようにした。このカメラを、Any−Maze(商標)ビデオトラッキングソフトウェア(Stoelting Inc.,ウッド・デール,イリノイ州)と一緒に使用して、自発運動活性、及び装置内の予め定義した空間領域中で過ごす時間を分析した。試験チャンバーの床(30.5cm×31.8cm)を操作的に定義し、それを3つの空間ゾーンに分割することによって、プログラムが、各ゾーンで過ごした時間の詳細な読み取りを秒単位で、動物が移動した距離をメートル単位で自動生成するようにした。装置の「活性ゾーン」(15.25cm×15.9cm)を、活性薬物と組み合わせたレバー及び周囲空間領域を含む領域と定義した。「不活性ゾーン」(15.25cm×15.9cm)は不活性レバー及び周囲空間領域を含み、「後部ゾーン」(15.25cm×31.8cm)は、試験チャンバーの後半部からなるものとした。試験チャンバーをこのように操作的に定義することによって、このシステムでは、装置内で空間嗜好性を判定する方法を自動化した。ビデオも定期的に記録し、分析して、Anymazeが正しく機能していることを確認した。
レバー押し反応の初期形成中に、モジュラーペレットディスペンサー(ENV−203M,Med Associates,セント・オールバンズ,バーモント州)及び受け器をオペラント試験チャンバーに加え、スクロースのチョコレート風味のペレット45mg(Bio−Serv,フレンチタウン,ニュージャージー州)が送達されるようにした。絶食ラットを、終夜16時間セッション中に、定率1(FR1)強化スケジュールにて、レバー押しをしてスクロースペレットを得るようにトレーニングした。トレーニング中、両方の受け器のレバーが存在していたが、一方のみ(活性レバー)が報酬送達と関連付けされていた。他方のレバー(不活性レバー)に対する反応は、記録したが、プログラムされた帰結は何もないものとした。活性レバーに反応すると、餌ペレットが即時に送達され、次いで両方のレバーが格納され、20秒の休止期間となるようにした。休止の後、室内照明が0.5秒間瞬間的に光り、レバーが装置の前壁から再び出てくるようにした。16時間セッション中に200回の活性なレバー押しの完了に成功した場合、そのラットを「トレーニングされた」とみなした。
脳を−80℃で凍結し、クライオスタット(Microm HM500M,ヴァルドルフ,ドイツ)を利用して組織学的検査を実施した。プローブ配置をクライオスタットの切片から直接マッピングし、プローブがBLAの領域に特有の境界内に位置していなければ、データを分析から除外した(図3Bを参照)。
透析物試料を、電気化学検出高速液体クロマトグラフィーシステム(HPLC−EC)を利用して分析した。このシステムは、ESA540オートサンプラー(ESA,ノース・チェルムズフォード,マサチューセッツ州)、ESA溶媒送達ユニット、ESAカラム(MD−150/RP−C18、直径=3.0μm)、並びにESA5020保護セル及びESA5014B分析用セルを備えたESA Coulochem II電気化学検出器からなるものとした。ガラス炭素作用電極の電位を参照電極に対して300mVに設定した。10%アセトニトリル(pH=3.0)中、75mMのリン酸二水素ナトリウム一水和物、1.7mMの1−オクタンスルホン酸ナトリウム塩、100μl/lのトリエチルアミン、25μMのEDTAから構成されるMD−TM移動相(ESA,ノース・チェルムズフォード,マサチューセッツ州)を流速0.530ml/分でポンプ注入した。電気化学データをAgilent Technologies Chem Station Plusソフトウェア(Agilent Technologies,ウィルミントン,デラウェア州)で処理した。このソフトウェアで、クロマトグラフ、DA、DOPAC及びHVAの濃度(単位pmol)をy軸上にプロットし、対してイオン親和性についての時間の表示(単位分)をx軸に沿ってプロットした可視描画を生成した。
全てのデータを平均±SEMとして表す。実験1については、活性及び不活性レバー押しを、要因分散分析(ANOVA)を使用し、条件(音楽、NMCond又はNMTest)及びトライアルを独立変数として分析した。自発運動活性データ、並びに活性ゾーン、不活性ゾーン及び後部ゾーンで過ごした時間の長さを、一元配置ANOVAを使用し、条件を独立変数として、3時点(自己投与の最終日、消去の最終日及び再発試験日)で分析した。全ての有意な結果をNewman−Keuls事後検定によってさらに検討した。
実験1−音楽に誘発される再発
図1は、自己投与トライアル、消去セッション及び再発試験日中に、コカイン強化を求めて起こった反応の平均を表す。要因ANOVAにより、トライアルの有意な効果(F(20,336)=55.052、p<0.001)及び条件×トライアルの有意な相互作用(F(40,336)=1.643、p<0.01)が明らかになったが、条件単独での効果は見られなかった(F(2,336)=1.369、p=0.256)。事後分析により、消去の初日(Ext1)に有意差が見られ、自己投与中に音楽で条件付けされなかった動物(NMCond)の方が、音楽でトレーニングされた動物より有意に高い反応をした(p<0.05)ことが明らかになった。さらに、音楽条件付けをした動物は、再発試験日(試験)に、NMCond群(p<0.001)及びNMTest群(p<0.001)の動物と比較して、活性レバーに対して有意に高く反応した。しかし、再発試験中に、NMCond群とNMTest群の間には有意差がなかった(p=0.98)。
表2は、ドパミン、DOPAC及びHVAの基礎レベルの平均濃度を示す。ドパミン(F(5,20)=0.6371、p=0.674)にもその代謝物(DOPAC:F(5,20)=2.123、p=0.105;DOPAC:F(5,20)=1.637、p=0.196)にも基礎レベルに有意差はなかった。したがって、図3Aのデータは、微小透析トライアル中のドパミン作動性反応を、ベースラインに対する百分率として表す。グラフからわかるように、音楽刺激の開始(120分)後に、ベースラインと比較してドパミンの有意な放出(F(14,56)=5.204、p<0.001)があった(140分:p<0.01;160分:p<0.01)。DOPAC(F(14,56)=1.734、p=0.105)にもHVA(F(14,56)=1.259、p=0.262)にもレベルに有意な変化は見られなかった。微小透析をした動物と、トレーニング、自己投与セッション及び消去セッション中に同じ音楽条件付けを受けさせた他の群との間で行動比較をすると、活性又は不活性レバー押しにおいて群間で差異は見られず(F(6,878)=0.810、p=0.563)、群×トライアルの相互作用はなかった(F(38,878)=0.615、p=0.999)。再発試験セッション中の平均(±SEM)活性レバー押しは、14.6(±1.86)であり、再発試験セッション中の不活性レバー押しは2.40(±1.03)であった。
図4は、自己投与トライアル、消去セッション及び再発試験日中に、コカイン強化を求めて起こった反応の平均を表す。要因ANOVAにより、処置(F(2,251)=3.606、p<0.05)及びトライアル(F(40,502)=25.172、p<0.001)の有意な効果、並びに処置×トライアルの有意な相互作用(F(40,502)=1.726、p<0.01)が明らかになった。グラフからわかるように、事後検定により、自己投与セッション中も消去セッション中も、18−MC処置群と生理食塩水処置群との間に有意差はなかったことが示された。しかし、18−MCで処置した動物の方が、再発試験日に活性レバー反応が有意に少なかった(p<0.001)。図5Aは、各処置群の自発運動活性を表し、自己投与の最終日(F(1,12)=0.235、p=0.637)にも、消去の最終日(F(1,12)=0.141、p=0.714)にも、再発試験日(F(1 12)=2.454、p=0.143)にも群間で有意差はなかった。図5Bは、自己投与の最終日、消去の最終日、及び再発試験セッション日に活性ゾーンで過ごした時間の長さの平均を示す。ANOVAにより、自己投与の最終日(F(1,12)=0.018、p=0.896)にも消去の最終日(F(1,12)=0.900、p=0.362)にも、活性ゾーンで過ごした時間に、群間で有意差がないことが明らかになった。しかし、再発試験セッション前に18−MCで処置したラットでは、活性ゾーンで過ごした時間の長さが有意に減少した(F(1 12)=8.523、p<0.01)。自己投与の最終日(不活性ゾーン:F(1 12)=0.021、p=0.888;後部ゾーン:F(1 12)=0.001、p=0.981)にも、消去の最終日(不活性ゾーン:F(1 12)=0.007、p=0.933;後部ゾーン:F(1 12)=0.282、p=0.605)にも、再発試験セッション(不活性ゾーン:F(1 12)=0.511、p=0.488;後部ゾーン:F(1 12)=3.606、p=0.082)でも、オペラントチャンバーの不活性ゾーン又は後部ゾーンで過ごした時間に対する処置の効果はなかった。
条件刺激の影響は、目標指向行動に関して広範に調査されてきたが、複雑な環境刺激の強力な影響は、これに匹敵するほどには探求されていない。薬物と組み合わせた音楽のCSには、コカイン自己投与中に組合せを反復した後、薬物探索行動に大いに影響する能力があることが、動物再燃・再発モデルを使用して初めて示されることになる。ヒトの薬物体験中に起こる複雑な心理的プロセスを模倣するため、複雑な状況関連の刺激を利用して、渇望中に起こる連合学習プロセスを評価した。過去の研究(Polston et al.,2011a;Polston et al.,2011b)を補完して、本研究結果は、ラットには複雑な楽節を識別する能力があるという見解をさらに支持し、音楽を介入させる他の前臨床モデルにおいてラットを使用することができることを示す。

- 薬物使用再燃を防止する方法において、
薬物使用初期の後に有効量のα3β4ニコチン拮抗薬を哺乳動物に投与するステップと、
薬物使用再燃を防止するステップと、

- 請求項1に記載の方法において、手綱脚間経路及び扁桃体基底外側核中のα3β4ニコチン受容体を遮断することにより、ドパミン作動性中脳辺縁系経路を間接的に調節するステップをさらに含むことを特徴とする方法。
- 請求項1に記載の方法において、前記投与するステップが、前記α3β4ニコチン拮抗薬を腹腔内注射によって0.05mg/kg〜200mg/kgの用量で投与するステップとさらに定義されることを特徴とする方法。
- 請求項1に記載の方法において、前記薬物が、バルビツレート、モルヒネ、コデイン、ヘロイン、レボルファノール、メペリジン、メタドン、プロポキシフェン、アセチルメタドール(LAAM)、ペンタゾシン、ブトルファノール、ナルブフィン、ブプレノルフィン、デゾシン、フェンタニル、d−アンフェタミン、1−アンフェタミン、d1−アンフェタミン、メタンフェタミン、3,4−メチレンジオキシ−N−メチルアンフェタミン(MDMA)ベンズフェタミン、フェンテルミン、ジエチルプロピオン、フェンメトラジン、フェンジメトラジン、クロルフェンテルミン、クロルテルミン、マジンドール、フェニルプロパノールアミン、コカイン、メチルフェニデート、ニコチン、カチノン(カート植物)、メプロバメート、クロルジアゼポキシド、ジアゼパム、オキサゼパム、ロラゼパム、フルラゼパム、プラゼパム、クロラゼプ酸、アルプラゾラム、トリアゾラム、テマゼパム、ハラゼパム、クアダゼパム、ミダゾラム、エスタゾラム、エタノール、ペントバルビタール、フェノバルビタール、セコバルビタール、アモバルビタール、Δ9−テトラヒドロカンナビノール(THC)、これらの組合せ、これらの類似体、及びこれらの誘導体からなる群から選択されることを特徴とする方法。

- 請求項7に記載の方法において、前記投与するステップが、薬物使用後の時間に前記α3β4ニコチン拮抗薬を投与するステップとさらに定義されることを特徴とする方法。
- 請求項7に記載の方法において、前記防止するステップが、前記哺乳動物の条件付け場所嗜好性(CPP)を減少させるステップをさらに含むことを特徴とする方法。
