実施例1
ウシtoll様受容体9(TLR−9)の遺伝子クローニング
ウシTLR9メッセンジャーRNA(mRNA)源として、近所の食肉解体処理場から新鮮なウシ脾臓を得た。市販キットおよびその説明書(TRIZOL(登録商標),GIBCOBRL)を用いて、ChomczynskiおよびSacchi(1987)により概説されているのと実質的に同じ方法で、ウシ脾臓組織から総RNAを調製した。逆転写酵素(−Expand Reverse Transcriptase,Roche)の供給業者により記載されているのと実質的に同じ方法で、ウシ脾臓総RNAから第1鎖cDNAを合成した。開始コドン領域から終止コドンの下流の3’UTR領域までのウシTLR9(Genbank AY859726)のポリメラーゼ連鎖反応(PCR)増幅のためのプライマーを設計した(Bov−TLR9−forおよびBov−TLR9−rev,後記を参照されたい)。しかし、完全長産物(予想:〜3100bp)の増幅を目的としたウシ脾臓第1鎖cDNAを使用する初期PCR実験(Expand High Fidelity PCRキット,Roche)は、反復的に陰性であることが判明した。ウシTLR9遺伝子の更に詳細な精査は高いGC含量(〜64%)を示した。したがって、この特定の問題に関して最適化されたPCR系(Advantage(商標)GC2,Clontech)を試験することに決定した。対応PCR反応は、予想サイズ(〜3100bp)の弱いDNA断片を与えた。

pCR2.1−Topo(Invitrogen)内へのPCR断片のクローニングを行い、PCR無過誤形態(pCR2.1−Topo−ウシTLR9)を特定するために4個のクローンを配列決定した。プライマー導入KpnIおよびXbaI制限酵素部位を利用することにより、ウシTLR9インサートを切り出し、アガロースゲルで精製し、KpnI/XbaI切断哺乳類発現ベクターpcDNA3.1(neo)およびpcDNA3.1(hyg)(共にInvitrogen)内にサブクローニングして、それぞれpcDNA3.1(neo)−ウシTLR9およびpcDNA3.1(hyg)−ウシTLR9を得た。対応インサートを再配列決定した(後記を参照されたい)。

MEM、1×非必須アミノ酸、8%(v/v)iFCS内で維持されたメイディン・ダービー・イヌ腎臓(MDCK)細胞をATCCから得た。分泌性アルカリホスファターゼ活性の存在に関する消費増殖培地の試験は陰性であり、これはレポーター遺伝子アッセイにおけるこの細胞系の使用のための前提条件である。

突然変異誘発法の後、部位特異的突然変異誘発が成功し新たな突然変異が導入されなかったかどうかを調べるために、複数(5個)のクローンを配列決定した。プライマーにより導入されたEcoRIおよびEcoRV部位を使用して、pIRESpuro3(Clontech)内に融合構築物を再クローニングするために、正しいクローンを使用した。得られたベクター(pIRESpuro−bovTLR9−21)における融合構築物を再配列決定した(後記を参照されたい)。

突然変異誘発法の後、部位特異的突然変異誘発が成功したかどうかを調べるために、複数(8個)のクローンを配列決定した。1個のクローンは適切なヌクレオチドを含有していた。このクローンを使用して、pCRII−TOPO内に存在するプライマー導入EcoRIおよびEcoRV部位を用いてpIRESpuro3(Clontech)内に融合構築物を再クローニングした。得られたベクター(pIRESpuro−canTLR9−21)を、完全に配列決定した。配列は、アミノ酸配列に影響を及ぼさない2つのサイレント突然変異を特定した。したがって、このクローンが更なる用途に使用可能であると結論づけられた。
イヌTLR9−21融合構築物でのMDCK−pNifTyhyg−SEAPのトランスフェクション
a)クローンのトランスフェクションおよび選択
TLR9−21融合構築物の発現および検出のためのMDCK−pNifTyhyg−SEAK−クローン15の可能性を調べるために、このクローン細胞系をpIRES−puro−イヌTLR9−21でトランスフェクトした。300μg/ml ヒグロマイシンおよび8μg/ml ピューロマイシンで補足された培地での反復継代によりトランスフェクタントを選択した。標準的なオリゴヌクレオチドODN−2006−PTOでの得られたポリクローナル細胞系の試験は、SEAP分泌の誘導を示した。単細胞クローニングを行い、ODN−2006−PTOでの更なる試験のために54個のクローンを増殖させた(後記グラフを参照されたい)。大量のSEAPの誘導を示す多数のクローンが特定された。最良のシグナル対ノイズ比を有する4個のクローン(番号17、23、32および40)を増殖および凍結安定体の作製のために選択した。再試験後、クローン番号17を更なる実験のために選択した(図5を参照されたい)。
実験1:
ヒト用医薬からの標準的なオリゴヌクレオチド2006−PTOおよび2007−PTOと共に、一連のホスホロチオアートオリゴヌクレオチド(PTO−ODN,thio(チオ)5−4からthio5−10まで)を試験した。
ヒト用医薬からの標準的なオリゴヌクレオチド2006−PTOと共に、ニワトリTLR21(融合部分のドナー)に対して非常に強力であることが判明した一連のホスホジエステルオリゴヌクレオチド(10個のPDE−ODN)をイヌTLR9−21融合体に対して試験した。
公開されているオリゴヌクレオチド1668−PTO、2216−PTOおよび2395−PTOからの恐らく活性要素でありうるものに基づいて、「リード最適化」を試みた。これらの活性要素は親ODNにおいては下線および/またはイタリック体で示されており、mod1/2 ODNにおいては反復配列内に配置されている。
ここでは、文献に記載されている2個のPTO−ODN(ODN−17およびODN−Ling1)を試験した。また、thio5−8におけるCpG要素の完全(thio5−8pde2A)および部分(thio5−8pde2B)ホスホロチオアート結合の置換の影響を試験した。更に、免疫調節要素−ttcgtc−の多量体を試験した。
ここでは、5’−および3’dG連続(run)と組合された場合にニワトリTLR21に対して高活性であることが判明したPDEオリゴヌクレオチドのPTO形態の幾つかを試験した。しかし、PTO形態は、5’−および3’dG連続を欠いている(したがって、マイナスG,「mG」)。
ブタTLR9−21融合構築物でのMDCK−pNifTyhyg−SEAPのトランスフェクション
a)クローンのトランスフェクションおよび選択
MDCK−pNifTyhyg−SEAK−クローン15を、pIRES−puro−ブタTLR9−21でトランスフェクトした。300μg/ml ヒグロマイシンおよび8μg/ml ピューロマイシンで補足された培地での反復継代によりトランスフェクタントを選択した。標準的なオリゴヌクレオチドODN−2006−PTOでの得られたポリクローナル細胞系の試験は、SEAP分泌の誘導を示した。単細胞クローニングを行い、ODN−2006−PTOでの更なる試験のために、75個のクローンを増殖させた。SEAPの誘導を示す多数のクローンが特定された。最良のシグナル対ノイズ比を有する幾つかのクローンを、増殖および凍結安定体の作製のために選択した。再試験後、クローン番号20を更なる実験のために選択した(図19を参照されたい)。

実験計画
狂犬病に対するワクチン接種がされていない3〜4月齢の5頭のビーグル子イヌ(このうちの雌イヌは過去12カ月間に狂犬病に対するワクチン接種がされていない)の2群を使用した。子イヌに1mlのそれぞれのワクチン組成物をワクチン接種した。ワクチン接種の直前(T=0)ならびにワクチン接種後のT=2、T=4、T=6、T=8、T=12、T=16、T=20およびT=24週の時点で血液サンプルを採取し、狂犬病ウイルスに対する抗体価を決定した。

1ml当たり5μgのThio−tcg−8−PTO(TCGTCGTCGTCGTCGTCGTCGTCG)の添加を伴う(群2)または伴わない(群1)1mlのNobivac(登録商標)Rabiesワクチンを、子イヌの頸部の皮下(s.c.)にワクチン接種した。
ワクチン接種の直前(T=0)ならびにワクチン接種後のT=2、T=4、T=6、T=8、T=12、T=16、T=20およびT=24週の時点で血液サンプルを各子イヌから採取した。血液サンプルを2〜8℃で一晩にわたって凝固させた。遠心分離後、血清を適当な容器内に移し、分析まで−20℃で保存した。ウイルス中和試験である迅速蛍光フォーカス抑制試験(Rapid Fluorescent Focus Inhibition Test)(RFFIT)を用いて、血清における狂犬病に対する抗体価を決定した。
RFFITは、狂犬病中和抗体の存在を定量するための標準的なインビトロ試験として国際的に認識されている。検査すべき血清の系列3倍希釈物を調製し、標準的用量(30〜300フォーカス形成単位(FFU)の力価を有するWHO/PHEURによるもの)を含有する等体積の狂犬病ウイルス懸濁液と混合した。血清/ウイルス混合物を37℃および5% CO2において90分間インキュベートした。プレインキュベーション期間後の非中和ウイルスを増殖させるために、感受性細胞(BHK細胞)を混合物中に加え、37℃および5% CO2において24時間インキュベートして、単層を形成させた。インキュベーションおよび狂犬病ウイルス特異的免疫染色の後、顕微鏡検査により蛍光フォーカスに関して単層を観察し、ついで力価(単位:IU/ml)を計算した。
図32から認められうるとおり、Nobivac狂犬病ワクチンおよびCpG ODN Thio−tcg−8−PTO(TCGTCGTCGTCGTCGTCGTCGTCG)が投与されたイヌにおいて見出された抗狂犬病ウイルス力価は、このCpG ODNを含有しない同じ狂犬病ワクチンの場合の量の3倍である。

- 医薬としての使用のための、抗原成分または抗原成分をコードする遺伝情報の免疫学的量および医薬上許容される担体と組み合わされた、請求項1〜13のいずれか1項記載の免疫刺激性非メチル化オリゴデオキシヌクレオチド。
- 哺乳類種、好ましくはヒト、ブタ、ウシおよびイヌ種における感染症の予防または抑制における使用のための、抗原成分または抗原成分をコードする遺伝情報の免疫学的量および医薬上許容される担体と組み合わされた、請求項1〜13のいずれか1項記載の免疫刺激性非メチル化オリゴデオキシヌクレオチド。
- 家禽TLR21のToll−インターロイキンI受容体抵抗性(TIR)ドメインと哺乳類TLR9の細胞外リガンド結合性ドメインとを含むことを特徴とする、ハイブリッドtoll様受容体(ハイブリッドTLR)。
- 哺乳類TLR9の細胞外リガンド結合性ドメインが、ヒト、イヌ、ブタまたはウシTLR9である、請求項20記載のハイブリッドTLR。
- 請求項20または21記載のハイブリッドTLRを含む細胞。
- 細胞が、NF−κBレポーター遺伝子を含むプラスミドを含む、請求項22記載の細胞。
- プラスミドが細胞内で安定に維持されている、請求項22記載の細胞。
- レポーター遺伝子が、分泌性アルカリホスファターゼをコードする遺伝子である、請求項23または24記載の細胞。
