Claims (30)
- さらに干渉物質を含むことがある試料中のグルコース量を定量するためのグルコースセンサーであって、
グルコースに結合するための第1の受容体及び該第1の受容体に附随する第1の蛍光体を含み、該第1の受容体は、グルコースとの会合定数KG1及び該干渉物質との会合定数KM1を有する、第1の指標系、及び
グルコースに結合するための第2の受容体及び該第2の受容体に附随する第2の蛍光体を含み、該第2の受容体は、グルコースとの会合定数KG2及び該干渉物質との会合定数KM2を有し、KG2/KM2はKG1/KM1と異なる、第2の指標系
を含む感知領域と、
該感知領域に光を導入するための光導波路と
を備える、グルコースセンサー。 - 前記第1の蛍光体及び前記第2の蛍光体は、少なくとも5nm異なるピーク発光波長を有する、請求項1に記載のグルコースセンサー。
- [KG2/KM2]/[KG1/KM1]は、2より大きいか又は0.5未満である、請求項1又は2に記載のグルコースセンサー。
- 前記第1の受容体及び前記第2の受容体は、ボロン酸受容体である、請求項1から3のいずれか一項に記載のグルコースセンサー。
- 前記第1の受容体は、2つのボロン酸基を含み、前記第2の受容体は、1つのボロン酸基を含む、請求項4に記載のグルコースセンサー。
- 前記干渉物質は、(a)cis−ジオール基を含む干渉物質、及び(b)アミンから選択される、請求項1から5のいずれか一項に記載のグルコースセンサー。
- cis−ジオール基を含む前記干渉物質は、糖アルコール及び糖類から選択される、請求項6に記載のグルコースセンサー。
- 前記干渉物質は、マンニトール、ソルビトール、ガラクチトール、イノシトール、フルクトース、ガラクトース、アラビノース、グルタミン及びカテコールアミンからなる群から選択される、請求項1から7のいずれか一項に記載のグルコースセンサー。
- 前記干渉物質は、マンニトールである、請求項8に記載のグルコースセンサー。
- さらに干渉物質を含むことがある試料中のグルコース量を定量する方法であって、
グルコースに結合するための第1の受容体及び該第1の受容体に附随する第1の蛍光体を含む第1の指標系を少なくとも含む感知領域と、該感知領域に光を導入するための光導波路とを備えるグルコースセンサーを、該試料に挿入し、
該センサーの該感知領域に入射光を提供して、該第1の蛍光体の発光を検出することを含む第1の測定法により、グルコース量の第1の測定値を得、
該試料中の該干渉物質の量に対する感度の点で該第1の測定法と異なる、第2の測定法によりグルコース量の第2の測定値を得、
該第1の測定値と該第2の測定値を比較し、それによって、該干渉物質が該試料中に存在するかどうかを決定すること
を含み、該干渉物質は、グルコースの該第1の受容体への結合に干渉することができる物質である、方法。 - グルコースに結合するための第1の受容体及び該第1の受容体に附随する第1の蛍光体を含み、該第1の受容体は、グルコースとの会合定数KG1及び前記干渉物質との会合定数KM1を有する第1の指標系、及びグルコースに結合するための第2の受容体及び該第2の受容体に附随する第2の蛍光体を含み、該第2の受容体は、グルコースとの会合定数KG2及び前記干渉物質との会合定数KM2を有し、KG2/KM2はKG1/KM1と異なる第2の指標系を含む感知領域と、
該感知領域に光を導入するための光導波路と
を備えるグルコースセンサーを前記試料に挿入し、
該センサーの該感知領域に入射光を提供し、
該第1の蛍光体の発光を検出することを含む第1の測定法により、グルコース量の第1の測定値を得、
該第2の蛍光体の発光を検出することを含む第2の測定法により、グルコース量の第2の測定値を得、
該第1の測定値と該第2の測定値を比較し、それによって、前記干渉物質が前記試料中に存在するかどうかを決定し、前記試料中の前記干渉物質の存在に対して修正すること
を含む、請求項10に記載の方法。 - 前記第1の測定値及び前記第2の測定値は、単一の発光検出工程で得られる、請求項11に記載の方法。
- 前記第1の蛍光体及び前記第2の蛍光体は、少なくとも5nm異なるピーク発光波長を有する、請求項11又は12に記載の方法。
- [KG2/KM2]/[KG1/KM1]は、2より大きいか又は0.5未満である、請求項11から13のいずれか一項に記載の方法。
- 前記第1の受容体及び前記第2の受容体は、ボロン酸受容体である、請求項11から14のいずれか一項に記載の方法。
- 前記第1の受容体は、2つのボロン酸基を含み、前記第2の受容体は、1つのボロン酸基を含む、請求項15に記載の方法。
- さらに干渉物質を含むことがある前記試料は、in vivo試料である、請求項10から16のいずれか一項に記載の方法。
- さらに干渉物質を含むことがある前記試料は、in vivo試料であり、前記方法は、
(i)時点ttestで前記第1の測定値を得、
(ii)前記第2の測定法により前記第2の測定値を得、ここで、前記第2の測定法は、該時点ttestで、前記試料中のグルコース、又は該干渉物質の量を電気化学的に測定することを含み、
(iii)前記第1の測定値と前記第2の測定値を比較し、それによって前記第1の測定値が、前記試料中の該干渉物質からの寄与を含むかどうかを決定すること
を含む、請求項10に記載の方法。 - 前記方法は、ヒト又は動物対象に実施され、前記試料は、該対象のin vivo体液であり、ステップ(i)は、時間ttestで該体液にin vivo測定を実施することを含み、ステップ(ii)は、時間ttestで該対象から該体液の一部を取り出し、それに対してin vitro電気化学的測定を実施することを含む、請求項18に記載の方法。
- 前記第1の測定値と前記第2の測定値を比較する前記ステップ(iii)において、前記第1の測定値が前記干渉物質からの寄与を実質的に含まない、又は低減された寄与を含むことが決定されるまで、一連のステップ(i)〜(iii)を1回又は複数回それぞれ異なる時点で実施することを含む、請求項18又は19に記載の方法。
- 前記第1の測定値と前記第2の測定値を比較する前記ステップ(iii)において、ttestで前記試料中の前記干渉物質の量の概算値が得られ、
(iv)ttestで前記試料中に存在する前記干渉物質にとって十分な時間を遅らせることで、前記試料を実質的に浄化し、
(v)前記センサーの前記感知領域に入射光を提供し、前記第1の蛍光体の発光を検出することで、前記試料中のグルコース量の第3の測定値を得ること(該第3の測定値は、前記干渉物質からの寄与が実質的にない又は低減されている)
をさらに含む、請求項18又は19に記載の方法。 - 前記干渉物質は、(a)cis−ジオール基を含む干渉物質、及び(b)アミンから選択される、請求項10から21のいずれか一項に記載の方法。
- cis−ジオール基を含む前記干渉物質は、糖アルコール及び糖類から選択される、請求項22に記載の方法。
- 前記干渉物質は、マンニトール、ソルビトール、ガラクチトール、イノシトール、フルクトース、ガラクトース、アラビノース、グルタミン及びカテコールアミンからなる群から選択される、請求項10から23までのいずれか一項に記載の方法。
- 前記干渉物質は、マンニトールである、請求項24に記載の方法。
- さらに干渉物質を含むことがある試料中のグルコース量を定量するためのグルコースセンサーであって、
グルコースに結合するための2つのボロン酸基を含む第1の受容体、及び該第1の受容体に附随する第1の蛍光体を含む第1の指標系、及び
グルコースに結合するための1つのボロン酸基を含む第2の受容体、及び該第2の受容体に附随する第2の蛍光体を含む第2の指標系を含む、感知領域と、
該感知領域に光を導入するための光導波路と
を備えるグルコースセンサー。 - 前記糖類は、cis−ジオール基を含む、請求項26に記載のグルコースセンサー。
- 前記糖類は、マンニトール、ソルビトール、ガラクチトール、イノシトール、フルクトース、ガラクトース及びアラビノースからなる群から選択される、請求項26に記載のグルコースセンサー。
- 前記糖類はマンニトールである、請求項26に記載のグルコースセンサー。
- 前記第1の受容体は、グルコースとの会合定数KG1及び前記干渉物質との会合定数KM1を有し、前記第2の受容体は、グルコースとの会合定数KG2及び前記干渉物質との会合定数KM2を有し、KG2/KM2がKG1/KM1と異なる、請求項26に記載のグルコースセンサー。
