Abstract
Description

ヒトGDNF遺伝子を有するプラスミドDNAの本質的に単一の分子から成るCK30PEG10kナノ粒子をCopernicus Therapeutics社(クリーブランド、オハイオ州)から入手した(図1、2、および10を参照)。Copernicus Therapeutics社は、この調査のために2つの発現プラスミドを提供してくれた:pUGG(ポリユビキチンCプロモーターによる転写制御の下、高感度緑色蛍光タンパク質(eGFP)に結合されたhGDNFバリアントlb)およびpGDNF−1b(同プロモーターの下、hGDNF−1b)(同書)。pUGGはGFP−GDNF融合タンパク質を生成し、脳内の形質移入および発現を検出することができる。GFP配列を欠損するpGDNF−1bを、6−OHDA神経保護の試験に使用した。実験に使用されるSD系ラットは、Taconic Farms、ジャーマンタウン、ニューヨーク州から入手した。
PEG化ポリリジンDNAナノ粒子の電子顕微鏡写真は、ナノ粒子が溶解素について対イオン酢酸塩を使用して形成した場合に、ロッド様形状を有することを示し(図10;スケールバー=200nm)、Fink TL、Klepcyk PJ、Oette SM、Gedeon CR、Hyatt SL、Kowalczyk TH、Moen RC、and Cooper、MJ.(2006)Gene Ther、13:1048−1051を参照されたい。
ラットに、5mmの長さのポリエチレン管を備えた10マイクロメーターのハミルトン・シリンジを使用して、pUGGナノ粒子または裸のプラスミドを鼻腔内投与した(88μgのDNA;両側交互に2.5μlずつ増加して20μl)。7日後、動物を屠殺し、脳を冠状の切片に切り分けた。脳切片のそれぞれの発現はGFP−ELISAによって確認された。捕捉抗体はマウスの抗GFP抗体であった(SigmaG6539、1:4000)。eGFP標準および脳のホモジネートを加え、プレートを室温で2時間インキュベートした。ウサギ抗GFP抗体(Abcam290,1:4,000)およびHRPに結合された抗ウサギ第二抗体(GE NA0340、1:4,000)を使用してeGFPの検出を行った。発色させるためにSureBlueTMB基質(KPL#52−00−01)を加えた。Gen5ソフトウェアを備えたBioTek ELx800プレートリーダーに光学濃度を読んだ。鼻腔内投与から7日後、pUGGナノ粒子を与えられたラットの脳の吻側-尾側軸の全てに沿ってGFPが検出され、鼻腔のちょうど背後にある前頭皮質および嗅球で値が最も高かった(図3Aおよび3B)。この結果は、pUGGナノ粒子が鼻腔内送達の後に、首尾よく脳内の細胞を形質移入することを示し、この形質移入がコードされたタンパク質を発現させる。
ラット6−OHDAモデルでパーキンソン病の試験を行った。ラットにpGDNF−1bナノ粒子ナノ粒子(NP)、若しくは裸のプラスミド(88μgのDNA;両側交互に2.5μlずつ増加させて20μl、n=7〜8/群)、またはリン酸緩衝塩類溶液のいずれかを鼻腔内投与した。7日後、GDNFが発現しているときに、ラットに麻酔をかけ、6−OHDA(2mg/mlの4μl;総用量8μg)を、左内側前脳束に外科的注入した。以下の定位座標に注入した:−1.2mm側部、ラムダ縫合に対して+0.44mm前部、および頭蓋表面に対して−8.3mm腹部。
アンフェタミン曝露の完了後、ラットに深く麻酔をかけ、4%のパラホルムアルデヒドの経心腔的灌流によって、屠殺した。チロシンヒドロキシラーゼ(TH)免疫組織化学(IHC)のために、各ラットの線条体および黒質(SN)から切片を回収した。THはドーパミンニューロンのためのマーカーである。代表的なSN切片を図7に示す。BIOQUANT画像分析ソフトウェアを使用して、%病変を計算するために100から減算されたパーセンテージで、各ラットの損傷を受けたSNと損傷を受けていないSNの両方のTH免疫染色密度を測定した。各動物について、当該値は、吻側に沿った6つの切片からSNの尾側の軸までの平均の染色を示す。
Claims (53)
- 脳内に核酸を送達し発現させる方法であって、核酸ナノ粒子を鼻腔内投与することを含み、投与されるナノ粒子は、1つのナノ粒子あたり約1つの比率で核酸分子を含む、前記方法。
- 核酸配列は治療的タンパク質をコードする請求項1に記載の方法。
- 核酸配列は治療的アンチセンス部分をコードする請求項1に記載の方法。
- 核酸配列は治療的アンチセンス部分を含む請求項1に記載の方法。
- 核酸ナノ粒子はポリカチオンをさらに含む請求項1に記載の方法。
- ポリカチオンはポリリジンである請求項5に記載の方法。
- 核酸ナノ粒子はプラスミドDNAを含む請求項1に記載の方法。
- 核酸配列は配列番号1の核酸配列を含む請求項1に記載の方法。
- 核酸配列はGDNF活性を有するポリペプチドをコードする請求項8に記載の方法。
- ポリペプチドは配列番号2のアミノ酸配列を有する請求項9に記載の方法。
- 対象のニューロンを細胞死から保護する方法であって、有効量の治療的核酸ナノ粒子を対象に鼻腔内投与することを含み、該ナノ粒子は細胞損傷からニューロンを保護し、または細胞損傷からの回復を促進する生成物を発現しまたは含む、前記方法。
- 生成物はポリペプチドである請求項11に記載の方法。
- ポリペプチドはGDNF活性を有するポリペプチドである請求項12に記載の方法。
- ポリペプチドは配列番号2のアミノ酸配列を有する請求項12に記載の方法。
- ポリペプチドは、GDNF、ニュールツリン、パーセフィン、アルテミン、BDNF、SDF−1、神経成長因子(NGF)、活性依存性神経栄養因子(ADNP)、インスリン、インスリン様成長因子1(IGF−1)、オキシトシン、βインターフェロン、ニューロテンシン、コレシストキニン、神経ペプチドY、黄体形成ホルモン放出ホルモン、成長ホルモン、アルギニンバソプレシン、インターフェロン、IL−1、IL−2、IL−4、IL−6、IL−12、IL−17、TNF、およびTGF、抗−VEGF、サイトカインまたはオートクリン因子から成る群から選択される活性を有するポリペプチドである、請求項12に記載の方法。
- 対象のニューロンはドーパミン作動性神経である請求項11に記載の方法。
- 対象のニューロンは黒質に位置する請求項16に記載の方法。
- 対象のニューロンは海馬に位置する請求項11に記載の方法。
- 対象のニューロンは大脳皮質に位置する請求項11に記載の方法。

- 有効量のナノ粒子を鼻腔内投与することによって、ハンチントン病、アルツハイマー病、痴呆、バッテン病、テイ・サックス病、多発性硬化症、うつ病、アルコール依存症、物質依存症、自閉症圏障害、心的外傷後ストレス症候群(PTSD)、外傷性脳損傷(TBI)、慢性外傷性脳症(CTE)、中枢神経系ループス、中枢神経系の自己免疫疾患、てんかん、脳卒中、筋萎縮性側索硬化症、疼痛性障害、および神経筋疾患から成る群から選択される疾患に罹患している対象を治療する、請求項11に記載の方法。
- 治療的核酸ナノ粒子の有効量は0.1ng〜1.0μgである請求項11に記載の方法。
- 治療的核酸ナノ粒子の有効量は1.0μg〜1mgである請求項11に記載の方法。
- 治療的核酸ナノ粒子の有効量は1.0mg〜1gmである請求項11に記載の方法。
- 治療的核酸ナノ粒子の投与量が、液滴として、スプレーミストとしてエアロゾル化され、または数分から数時間で送達するためにポンプを用いて、鼻腔内投与される、請求項11に記載の方法。
- 治療的核酸ナノ粒子の投与量が、定められた時間経過にわたって複数回、鼻腔内投与される、請求項11に記載の方法。
- 各ナノ粒子は、配列番号1の核酸配列を有するプラスミドを含む、請求項11に記載の方法。
- 配列番号1は、配列番号2のアミノ酸配列を有するポリペプチドを発現する請求項11に記載の方法。
- ポリペプチドはニューロンを細胞死から保護し、またはその回復および再生を促進する、請求項1または11のいずれかに記載の方法。
- 生成物はアンチセンスRNAである請求項11に記載の方法。
- 脳または脳幹障害を罹患している対象を治療する方法であって、有効量の圧縮された核酸ナノ粒子を対象に鼻腔内投与することを含み、投与されるナノ粒子は、1つのナノ粒子あたり約1つの比率で核酸分子を含み、当該核酸は治療的部分をコードしまたは含み、ここでナノ粒子は脳または脳幹障害を治療する、前記方法。
- ナノ粒子は生成物をコードする核酸を含む請求項35に記載の方法。
- 生成物はポリペプチドである請求項36に記載の方法。

- 治療的核酸ナノ粒子の有効量は1.0ng〜1mgである請求項35に記載の方法。
- 治療的核酸ナノ粒子の有効量は1.0mg〜1gmである請求項35に記載の方法。
