Abstract
Description
CH2Cl2(98mL)にPEG350モノ(メチルエーテル)(3.438g、9.82mmol)を溶解した溶液にデス・マーチン・ペルヨージナン(5.00g、11.79mmol)を添加し、得られた溶液を室温で20時間攪拌した。反応中、微細な白色沈殿物が形成し、これを反応終了時にセライトパッドで濾過することにより除去した。白色固体を黄色油状物に懸濁させたまま真空で濾液から溶剤を除去した。固体をジエチルエーテルで研和し、セライトパッドで濾過することにより固体を除去した。濾液から溶剤を真空で除去すると、生成物PEG350モノ(メチルエーテル)アセトアルデヒド(3.42g、100%)が黄色油状物として残った。

テトラヒドロフラン(53mL)にPEG350モノ(メチルエーテル)アセトアルデヒド(3.71g、10.7mmol)を溶解した溶液に亜リン酸ジエチル(1.77g、12.8mmol)を添加した。溶液を0℃に冷却し、1,8−ジアザビシクロ[5.4.0]ウンデカ−7−エン(1.94g、12.8mmol)を添加した。0℃で10分間攪拌後、反応を室温まで温め、さらに24時間攪拌した。溶剤を真空で除去すると、暗黄色油状物が残り、これをカラムクロマトグラフィ(100%CH2Cl2〜15%MeOH/85%CH2Cl2)により精製し、3.30g(64%)の所望のジエチルα−ヒドロキシPEG350モノ(メチルエーテル)ホスホネート生成物を黄色油状物として得た。

2Lのジャケット反応容器を窒素で浄化し、エピクロロヒドリン(222.9mL、2.843mol)、続いてH2O(11.37mL、0.631mol)を入れた。室温で10分間攪拌後、NaOH(113.7g、2.843mol)を添加し、得られた反応スラリーを72℃に加熱した。PEG1900モノ(メチルエーテル)を反応に添加して、得られた融解物を72℃で3時間攪拌した。その後、反応を35℃に冷却し、H2O(750mL)を添加した。反応溶液が室温に達したら、水性反応溶液をCH2Cl2(750mLで2回)で抽出した。その後、合わせた有機層から真空で溶剤を除去して黄褐色/茶色固体を得た。この固体をCH2Cl2(750mL)に溶解し、得られた濁った溶液をセライトパッドに通した。得られた透明なオレンジ色濾液から溶剤を真空で除去すると、ワックス状黄褐色の固体が残った。回収した固体を2:1(THF:ヘキサン)(粗生成物1gあたり1.3mLのTHF)から2回再結晶して439.7g(85%)の所望の生成物をオフホワイト固体として得た。

実施例5:PEG30000コンジュゲートの合成
PEG30000モノ(メチルエーテル)(30g、1mmol)を85℃に加熱し、加熱中、水(0.040mL)、エピクロロヒドリン(0.89mL、11.35mmol)及びNaOH(0.454g、11.35mmol)、続いてトリエチルアンモニウムハイドロクロライド(0.013g、0.091mmol)を添加した。得られた融解物/懸濁物をその温度で10分間攪拌し、95℃で30分間、105℃で2.5時間、その後、95℃でさらに1時間攪拌した。その後、反応を室温まで冷却した。0.5MのH2SO4(100mL)を添加し、反応を室温でさらに18時間攪拌した。生成物をCH2Cl2(150mLで2回)で抽出した。CH2Cl2を真空で(乾燥までではなく、濃い油状物になるまで)除去し、得られた油状物をTHF(150mL)/ヘキサン(75mL)(THFを添加し、加熱し、その後、ヘキサンを添加し、濁りが澄むまで約30秒間ぐるぐる混ぜた)から再結晶させて17.91g(理論的質量の60%)の所望の生成物を白色固体として得た。この材料をこれ以上精製せずに次の工程に進めた。
mPEG30k−ジオール(17.0g、0.565mmol)を水(50mL)に溶解し、室温で約30分間攪拌した。この濁った溶液に、NaIO4(0.147g、0.69mmol)を水(10mL)に予め溶解した溶液を一度に添加した。酸化剤添加の約45分間後に、濁りが澄んだ。その後、反応を18時間攪拌し、プロピレングリコール(0.0165mL)の添加によりクエンチした。この反応混合物をさらに1時間攪拌した。次に、水性反応混合物をCH2Cl2(100mLで2回)で抽出した。有機層を合わせ、CH2Cl2を真空で(乾燥までではなく、濃い油状物になるまで)除去し、得られた黄色油状物をTHF/ヘキサン(150mL:75mL)(THFを添加し、加熱し、その後、ヘキサンを添加し、濁りが澄むまで約30秒間ぐるぐる混ぜた)から再結晶して15.08g(理論的質量の89%)の所望の生成物を白色固体として得た。この材料をこれ以上精製せずに次の工程に進めた。
mPEG30k−アルデヒド(14.27g、0.475mmol)をCH2Cl2(50mL)に溶解し、トリエチルアミン(0.132mL、0.95mmol)を添加し、約30分間攪拌させた。その後、亜リン酸ジベンジル(0.249g、0.95mmol)をゆっくり添加した。室温で72時間攪拌後、溶剤を真空で(乾燥までではなく、濃い油状物になるまで)除去し、得られた油状物をTHF/ヘキサン(200mL:100mL)(THFを添加し、加熱し、その後、ヘキサンを添加し、濁りが澄むまで約30秒間ぐるぐる混ぜた)から再結晶して14.03g(理論的質量の97%)の所望の生成物を白色固体として得た。この材料をこれ以上精製せずに次の工程に進めた。
ジベンジル−αHmPP30k(10.6g、0.35mmol)を無水エタノール(100mL)に懸濁させ、完全に溶解するまで40℃に加熱した。10%のPd/炭素(0.4g)をゆっくり添加した。その後、H2の雰囲気(バルーン圧)下40℃で取り込みが完了するまで反応を攪拌した。Pd/炭素をセライトパッドでの濾過により除去し、濾液から溶剤を真空で(乾燥までではなく、濃い油状物になるまで)除去して9.22g(理論的質量の88%)の所望の生成物を白色粉末として得た。
9.177gのαHmPP5kリガンドを磁気撹拌子を含む1Lの三角フラスコに量り入れた。8.955mlのNaOH溶液(0.2M)を104mlの脱イオン水及び無水EtOH(200mL)とともに添加した。フラスコを時計皿で覆い、磁気撹拌で攪拌し、無色透明溶液が得られるまで穏やかに加熱した。上記の溶液を機械式攪拌器、テフロン(登録商標)アンカー型アジテーター、熱電対、窒素入口、バブラー及び還流冷却器を備えた2Lのジャケット付き反応器に添加した。フラスコを無水EtOH(2x25mL)ですすぎ、すすいだ水を反応器に添加した。無色透明溶液を100rpmで攪拌した。330.6mlの酸化鉄ナノ粒子コアのベンジルアルコール溶液(6.05g Fe/ml)を反応混合物に、塩基で洗浄し、DI水ですすぎ、オーブン乾燥したメスシリンダーで粉末漏斗を通して添加した。メスシリンダーを無水EtOH(25mLで2回)すすぎ、すすぎ液を反応器に添加した。単相の暗赤褐色の溶液が認められた。攪拌速度を200rpmに上昇させ、反応器を停止し、窒素下で攪拌しながら18時間50℃(内部温度)で加熱した。18時間加熱した後、反応器を25℃に冷却した。EtOAc(660mL)及び脱イオン水(320mL)を反応器に添加し、バッフルを挿入して混合を向上させた。反応混合物を500rpmで10分間攪拌した。攪拌を停止し、相を分離させた。濁った混合物は10分間未満で相分離し始め、約30分間後にはっきりした相分離が認められた。247gの暗赤褐色の溶液からなる下側水性相(SPIO粒子を含有)を反応器から2L24/40の丸底フラスコに注いだ。溶液を800mLに水で希釈(理論的に約2.5mg Fe/mL)し、短時間ロータリーエバポレーターにかけて残留揮発性有機物を除去した。その後、溶液を除菌濾過(0.22μm)し、50K PES Millipore 0.1m2メンブレンを用いてタンジェンシャルフロー濾過(TFF)により精製した。生成物を24Lの脱イオン水で約3時間かけて約10psiで保留液溜めに2.5Lの体積を維持しながら洗浄した。透過液を1リットル捕集するごとに透過液及び保留液サンプル(各約5mL)を取った。洗浄が完了したら、保留液を約120g(約16mg Fe/mL)に濃縮した。最終粒子の150mMの塩化ナトリウム溶液中で動的光散乱法により測定した流体力学的半径が19.2nmであった。粒子のサイズは、40℃でインキュベートした150mMの塩化ナトリウム溶液中で2日間後も変化しなかった。
タンジェンシャルフロー濾過の結果として得られたコロイド懸濁液を安定性及びゼータ電位について評価した。
動物を含むすべての手順はGE Global Research Institutional Animal Care and Use Committeeによって承認されたプロトコル下で完結した。酸化鉄ナノ粒子試験剤の注射前、注射後の様々な時点(例えば、1、2、5、7、21、28日・・・)で画像データを集めた。試験剤は、約150gの雌のLewisラット(米国マサチューセッツ州Wilmington所在Charles River Laboratories)の外側尾静脈に位置させたカテーテルを通して注射され、体重1kgあたり5mg Feで投与し、例外として、PEG30000α−ヒドロキシホスホネート被覆剤は材料に制限があるため3mg Fe/kgで投与した。カテーテルは注射後、生理食塩水で洗い流した。
Claims (42)
- α−ヒドロキシホスホネート部分が付着したナノ粒子を含む組成物であって、少なくとも1つのα−ヒドロキシホスホネート部分が次の式Iの構造を有する組成物。
- Sが直接結合、置換又は非置換の脂肪族又は脂環式基又はヘテロ脂肪族基である、請求項1記載の組成物。
- Sが炭素原子数1〜10の鎖長の直鎖アルキル基である、請求項1記載の組成物。
- Lが直接結合、カルボニル基、エーテル基、アミン基、アミド基、エステル基、カルバメート基、尿素基又は第4級アミン基である、請求項1乃至請求項3のいずれか1項記載の組成物。
- α−ヒドロキシホスホネート部分が次の式IIの構造を有する、請求項1記載の組成物。
- ナノ粒子が遷移金属又は遷移金属化合物を含む、請求項1乃至請求項5のいずれか1項記載の組成物。
- ナノ粒子が常磁性材料を含む、請求項1乃至請求項6のいずれか1項記載の組成物。
- 遷移金属又は遷移金属化合物が、鉄、マンガン、銅、コバルト、ニッケル及びこれらの組合せからなる群から選択される、請求項6記載の組成物。
- ナノ粒子が超常磁性酸化鉄を含む、請求項1乃至請求項5のいずれか1項記載の組成物。
- 前記酸化鉄が別の金属でドープされている、請求項9記載の組成物。
- ナノ粒子が、動的光散乱法によって150mM NaCl水溶液中で測定して、15nm〜100nmの平均流体力学的半径(DH)を有する、請求項1乃至請求項10のいずれか1項記載の組成物。
- ナノ粒子が、動的光散乱法によって150mM NaCl水溶液中で測定して、15nm〜50nmの平均流体力学的半径(DH)を有する、請求項11記載の組成物。
- ナノ粒子が、動的光散乱法によって150mM NaCl水溶液中で測定して、約20nm〜約30nmの平均流体力学的半径(DH)を有する、請求項11記載の組成物。
- 前記ナノ粒子が、タンジェンシャルフロー濾過して室温で1週間保存した後、動的光散乱法によって150mM NaCl水溶液中で求めた流体力学的半径(DH)に実質的変化のない安定な水性コロイド懸濁液を形成するのに十分に親水性である、請求項1乃至請求項13のいずれか1項記載の組成物。
- 哺乳類被験体への投与に適した組成物であって、
α−ヒドロキシホスホネート部分が付着したナノ粒子であって、α−ヒドロキシホスホネート部分が以下の式Iの構造を有するナノ粒子と、
薬学的に許容されるキャリア又は賦形剤と
を含む組成物。
- α−ヒドロキシホスホネート部分が式IIの構造を有する、請求項15記載の組成物。
- Sが直接結合、置換又は非置換の脂肪族又は脂環式基又はヘテロ脂肪族基又はこれらの組合せである、請求項15又は請求項16記載の組成物。
- Lが直接結合、カルボニル基、エーテル基、アミド基、エステル基、カルバメート基、尿素基又はこれらの組合せである、請求項15乃至請求項17のいずれか1項記載の組成物。
- ナノ粒子がMRI造影剤としての使用に適している、請求項15乃至請求項18のいずれか1項記載の組成物。
- ナノ粒子が超常磁性酸化鉄を含む、請求項15乃至請求項19のいずれか1項記載の組成物。
- ナノ粒子が、動的光散乱法によって150mM NaCl水溶液中で測定して、15nm〜100nmの平均流体力学的半径(DH)を有する、請求項15乃至請求項20のいずれか1項記載の組成物。
- ナノ粒子が、動的光散乱法によって150mM NaCl水溶液中で測定して、15nm〜50nmの平均流体力学的半径(DH)を有する、請求項21記載の組成物。
- ナノ粒子が約20nm〜約30nmの平均流体力学的半径(DH)を有する、請求項21記載の組成物。
- ナノ粒子が、タンジェンシャルフロー濾過して室温で1週間保存した後、動的光散乱法によって150mM NaCl水溶液中で求めた流体力学的半径(DH)に実質的変化のない安定な水性コロイド懸濁液を形成するのに十分に親水性である、請求項15乃至請求項23のいずれか1項記載の組成物。
- キャリアが等張性水媒体である、請求項15乃至請求項24のいずれか1項記載の組成物。
- キャリアが生理的pHにある、請求項15乃至請求項25のいずれか1項記載の組成物。
- PEG置換α−ヒドロキシホスホネート被覆ナノ粒子を用いて画像診断する方法であって、
以下の式Iの構造を有するα−ヒドロキシホスホネート部分が付着した酸化鉄ナノ粒子を含むナノ粒子組成物が予め投与されている個体をイメージングに付す工程
を含む方法。
- α−ヒドロキシホスホネート部分が式IIの構造を有する、請求項27記載の方法。
- 被験体に予め投与されているナノ粒子組成物が造影剤として作用する、請求項27又は請求項28記載の方法。
- 前記ナノ粒子が吸入又は経口摂取によって投与される、請求項27乃至請求項29のいずれか1項記載の方法。
- 前記ナノ粒子が非経口的に投与される、請求項27乃至請求項29のいずれか1項記載の方法。
- 前記ナノ粒子が注射又は注入によって投与される、請求項27乃至請求項29のいずれか1項記載の方法。
- 前記ナノ粒子が被験体に埋め込まれる、請求項27乃至請求項29のいずれか1項記載の方法。
- イメージングが磁気共鳴(MR)によるものである、請求項27乃至請求項33のいずれか1項記載の方法。
- PEG置換α−ヒドロキシホスホネート被覆ナノ粒子を用いて鉄欠乏症を処置する方法であって、
以下の式Iの構造を有するα−ヒドロキシホスホネート部分が付着した酸化鉄コアを有するナノ粒子を含むナノ粒子組成物を個体に投与する工程
を含む方法。
- α−ヒドロキシホスホネート部分が式IIの構造を有する、請求項35記載の方法。
- 被験体に予め投与されているナノ粒子組成物が生体利用可能な鉄源として作用する、請求項35又は請求項36記載の方法。
