L. H. et al. Cell 1982, 30, 697-705)。SSEA−4は初期胚の細胞表面に発現する糖脂質であり、ヒトESおよびEC(embryonic carcinoma)細胞の表面に選択的に提示されるが、その理由は分かっていない(Henderson, J. K. et al. Stem Cells 2002, 20, 329-337)。ヒト幹細胞の他のマーカーの例としては、Oct3/4およびNanogが挙げられ、これらは幹細胞の未分化状態を維持するために必要な転写因子であり、分化の際に下方制御される(Rosner, M. H. et al. Nature 1990, 345, 686-692, and Mitsui, K. et al. Cell 2003, 113, 631-642)。これらに対する抗体は多能性細胞の検出において極めて有用であるが、不安定なタンパク質ツールであるため、高額であり、また細胞の固定や透過処理といった注意を要する処理が必要である。ヒト幹細胞の他のルーチンマーカーとしては、アルカリフォスファターゼが挙げられる(Thomson, J. A. et al. Proc. Natl. Acad. Sci. USA 1995, 92, 7844-7848)。その酵素活性を利用したアッセイは幹細胞を検出するための簡便かつ極めて有用な方法であるが、多能性幹細胞に対する特異性に関しては大きな懸念がある。なぜなら、この酵素はハウスキーピング酵素であり、他の多くの種類の細胞に広く発現しているためである。

ES細胞は、ヒトやマウスなどの哺乳動物の初期胚(例えば胚盤胞)の内部細胞塊から樹立された、多能性と自己複製による増殖能とを有する幹細胞である。
ES細胞は、受精卵の8細胞期、桑実胚期後の胚である、胚盤胞の内部細胞塊に由来する胚由来幹細胞であり、成体を構成するあらゆる細胞に分化する能力、いわゆる分化多能性と、自己複製による増殖能とを有している。ES細胞は、マウスで1981年に発見され(M. J. Evans and M. H. Kaufman (1981), Nature 292:154-156)、その後、ヒト、サルなどの霊長類でもES細胞株が樹立された(J. A. Thomson et al. (1998), Science 282:1145-1147;J. A. Thomson et al. (1995), Proc. Natl. Acad. Sci. USA, 92:7844-7848;J. A. Thomson et al. (1996), Biol. Reprod., 55:254-259;および J. A. Thomso
n and V. S. Marshall (1998), Curr. Top. Dev. Biol., 38:133-165)。
ES細胞は、対象動物の受精卵の胚盤胞から内部細胞塊を取出し、内部細胞塊をフィーダー細胞である線維芽細胞の上で培養することによって樹立することができる。また、継代培養による細胞の維持は、白血病抑制因子(LIF)、塩基性線維芽細胞成長因子(bFGF)などの物質を添加した培地を用いて行うことができる。ヒトおよびサルのES細胞の樹立と維持の方法については、例えば、H. Suemori et al. (2006), Biochem. Biophys. Res. Commun., 345:926-932;M. Ueno et al. (2006), Proc. Natl. Acad. Sci. USA, 103:9554-9559;H. Suemori et al. (2001), Dev. Dyn., 222:273-279;およびH. Kawasaki et al. (2002), Proc. Natl. Acad. Sci. USA, 99:1580-1585などに記載されている。
ヒトES細胞は、例えば0.1mM 2−メルカプトエタノール、0.1mM 非必須アミノ酸、2mM L−グルタミン酸、20% KSR及び4ng/ml bFGFを補充したDMEM/F−12培地を使用し、37℃、5% CO2湿潤雰囲気下で維持することができる。また、ES細胞は、3〜4日おきに継代する必要があり、このとき、継代は、例えば1mM CaCl2及び20% KSRを含有するPBS中の0.25% トリプシン及び0.1mg/ml コラゲナーゼIVを用いて行うことができる。
ES細胞の選択は、一般に、アルカリホスファターゼ、Oct−3/4、Nanogなどの遺伝子マーカーの発現を指標にして行うことができる。特に、ヒトES細胞の選択では、OCT−3/4、NANOG、などの遺伝子マーカーの発現をReal−Time PCR法で検出したり、細胞表面抗原であるSSEA−3、SSEA−4、TRA−1−60、TRA−1−81を免疫染色法にて検出することで行うことができる(Klimanskaya I, et al. (2006), Nature. 444:481-485)。
ヒトES細胞株である例えばKhES−1、KhES−2及びKhES−3は、京都大学再生医科学研究所(京都、日本)から入手可能である。
生殖幹細胞は、精巣由来の多能性幹細胞であり、精子形成のための起源となる細胞である。この細胞は、ES細胞と同様に、種々の系列の細胞に分化誘導可能であり、例えばマウス胚盤胞に移植するとキメラマウスを作出できる性質をもつ(M. Kanatsu-Shinohara et al. (2003) Biol. Reprod., 69:612-616;K. Shinohara et al. (2004), Cell, 119:1001-1012)。生殖幹細胞は、神経膠細胞系由来神経栄養因子(GDNF)を含む培地で自己複製可能であるし、またES細胞と同様の培養条件下で継代を繰り返すことによって、生殖幹細胞を得ることができる(竹林正則ら(2008),実験医学,26巻,5号(増刊),41〜46頁,羊土社(東京、日本))。
胚性生殖細胞は、胎生期の始原生殖細胞から樹立される、ES細胞と同様な多能性をもつ細胞であり、LIF、bFGF、幹細胞因子などの物質の存在下で始原生殖細胞を培養することによって樹立しうる(Y. Matsui et al. (1992), Cell, 70:841-847;J.L. Resnick et al. (1992), Nature, 359:550-551)。
人工多能性幹(iPS)細胞は、ある特定の核初期化物質を、DNAまたはタンパク質の形態で体細胞に導入することまたは薬剤によって当該核初期化物質の内在性のmRNAおよびタンパク質の発現を上昇させることによって作製することができる、ES細胞とほぼ同等の特性、例えば分化多能性と自己複製による増殖能、を有する体細胞由来の人工の幹細胞である(K. Takahashi and S. Yamanaka (2006) Cell, 126: 663-676;K. Takahashi et al. (2007) Cell, 131: 861-872;J. Yu et al. (2007) Science, 318: 1917-1920;M. Nakagawa et al. (2008) Nat. Biotechnol., 26: 101-106;WO 2007/069666;およびWO 2010/068955)。核初期化物質は、ES細胞に特異的に発現している遺伝子またはE
S細胞の未分化維持に重要な役割を果たす遺伝子もしくはそれらの遺伝子産物であればよく、特に限定されないが、例えば、Oct3/4、Klf4、Klf1、Klf2、Klf5、Sox2、Sox1、Sox3、Sox15、Sox17、Sox18、c−Myc、L−Myc、N−Myc、TERT、SV40 Large T antigen、HPV16 E6、HPV16 E7、Bmil、Lin28、Lin28b、Nanog、Esrrb、Esrrg、およびGlis1が例示される。これらの初期化物質は、iPS細胞樹立の際には、組み合わされて使用されてもよい。例えば、上記初期化物質を、少なくとも1つ、2つもしくは3つ含む組み合わせであり、好ましくは4つを含む組み合わせである。

る核初期化物質をコードする遺伝子を共に切除するために、それらの前後にLoxP配列を有してもよい。別の好ましい一実施態様においては、トランスポゾンを用いて染色体に導入遺伝子を組み込んだ後に、プラスミドベクターもしくはアデノウイルスベクターを用いて細胞に転移酵素を作用させ、導入遺伝子を完全に染色体から除去する方法が用いられ得る。好ましいトランスポゾンとしては、例えば、鱗翅目昆虫由来のトランスポゾンであるpiggyBac等が挙げられる(Kaji, K. et al., (2009), Nature, 458: 771-775;Woltjen et al., (2009), Nature, 458: 766-770;WO 2010/012077)。さらに、ベクターには、染色体への組み込みがなくとも複製されて、エピソーマルに存在するように、リンパ指向性ヘルペスウイルス(lymphotrophic herpes virus)、BKウイルスまたは牛乳頭腫ウイルス(Bovine papillomavirus)の起点とそれらの複製に係る配列を含んでいてもよい。例えば、EBNA−1およびoriP配列、またはLarge TおよびSV40ori配列を含むことが挙げられる(WO 2009/115295、WO 2009/157201、およびWO 2009/149233)。また、複数の核初期化物質を同時に導入するために、ポリシストロニックに発現させる発現ベクターを用いてもよい。ポリシストロニックに発現させるためには、遺伝子をコードする配列の間は、IRESまたは口蹄病ウイルス(FMDV)2Aコード領域により結合されていてもよい(Science, 322:949-953, 2008およびWO 2009/092042 2009/152529)。
kinase−3阻害剤(PLoS Biology, 6(10), 2237-2247 (2008))、miR−291−3p、miR−294、miR−295などのmiRNA(R.L. Judson et al., Nat. Biotech., 27:459-461 (2009))、等を使用することができる。
MEM、DMEM/F12またはDME培地(これらの培地にはさらに、LIF、ペニシリン/ストレプトマイシン、ピューロマイシン、L−グルタミン、非必須アミノ酸類、β−メルカプトエタノールなどを適宜含むことができる。)、(2)bFGFまたはSCFを含有するES細胞用培地、例えばマウスES細胞用培地(例えばTX−WES培地、トロンボX社)または霊長類ES細胞用培地(例えば霊長類(ヒト&サル)ES細胞用培地(リプロセル、京都、日本)、mTeSR−1)、などが含まれる。
あるいは、その代替培養法として、体細胞は、フィーダー細胞(たとえば、マイトマイシンC処理STO細胞、SNL細胞等)上で10%FBS含有DMEM培地(これにはさらに、LIF、ペニシリン/ストレプトマイシン、ピューロマイシン、L−グルタミン、非必須アミノ酸類、β−メルカプトエタノールなどを適宜含むことができる。)で培養し、培養から約25〜約30日またはそれ以上ののちにES様コロニーを生じさせることができる。
本発明において、体細胞を採取する由来となる哺乳動物個体は特に制限されないが、好ましくはヒトである。
nt ES細胞は、核移植技術によって作製されたクローン胚由来のES細胞であり、受精卵由来のES細胞とほぼ同じ特性を有している(T. Wakayama et al. (2001), Science, 292:740-743;S. Wakayama et al. (2005), Biol. Reprod., 72:932-936;J. Byrne et al. (2007), Nature, 450:497-502)。すなわち、未受精卵の核を体細胞の核と置換することによって得られたクローン胚由来の胚盤胞の内部細胞塊から樹立されたES細胞がnt ES(nuclear transfer ES)細胞である。nt ES細胞の作製のためには、核移植技術(J.B. Cibelli et al. (1998), Nat. Biotechnol., 16:642-646)とES細胞作製技術(上記)との組み合わせが利用される(若山清香ら(2008),実験医学,26巻,5号(増刊),47〜52頁)。核移植においては、哺乳動物の除核した未受精卵に、体細胞の核を注入し、数時間培養することで再プログラム化することができる。
体細胞と卵子もしくはES細胞とを融合させることにより、融合させたES細胞と同様な多能性を有し、さらに体細胞に特有の遺伝子も有する幹細胞である(Tada M et al. Curr Biol. 11:1553-8, 2001;Cowan CA et al. Science. 2005 Aug 26;309(5739):1369-73)。
本発明はまた、体細胞への分化を誘導された多能性細胞を含む細胞試料から多能性細胞を選別する場合にも用いることができる。
ここで、多能性細胞は、特定の臓器細胞およびその前駆細胞へ分化を誘導されるのみでなく、内胚葉細胞、中胚葉細胞および外胚葉細胞等の広範な細胞型を含む細胞集団へと分化を誘導されてもよい。本発明の標的となる臓器の例としては、皮膚、血管、角膜、腎臓、心臓、肝臓、臍帯、腸、神経、肺、胎盤、膵臓、脳、四肢末梢、および網膜等が挙げられるが、これらに限定されない。当業者に自明の任意の分化誘導法を用いることができる。例としては、特開2002-291469に記載される神経幹細胞への分化誘導法、特開2004-121165に記載される膵幹様細胞への分化誘導法、および特表2003-505006に記載される造血細胞への分化誘導法が挙げられる。さらに、胚様体の形成による分化誘導法の例として、特表2003-523766等に記載される方法等が挙げられる。
他の態様において、本発明の方法は、前記化合物を蓄積した細胞を除去することにより分化した細胞を選択するために使用することができる。
他の態様において、多能性細胞を選別するためのキットが提供される。該キットは上述の化合物、培養液等を含んでいてよい。該キットはさらに選別のための手順書または説明書を含んでいてよい。
シクロスポリンAを和光純薬工業株式会社より購入した。マウスモノクローナル抗MDR1抗体(MRK16)を協和メデックス株式会社より購入した。蛍光標識二次抗体を、Molecular Probesより購入した。牛胎児血清(FBS)をInvitrogenより購入した。Dulbecco’s Modified Eagle’s Medium(DMEM)をナカライテスク株式会社より購入した。
ヒトiPS細胞(クローン#201B7)(Takahashi, K. et al. Cell 2007, 131, 861-872)を、6穴プレート中、SNLフィーダー細胞上に2×105細胞/ウェルの密度でプレーティングした(McMahon, A.P. and Bradley, A., Cell, 62, 1073-1085, 1990)。プレーティングの6日後、細胞を2μM 化合物1(表2)とともに3時間インキュベートした(この時点で蛍光顕微鏡像を取得した)。細胞をAccutase(Invitrogen)で単一細胞に分離し、a−SSEA−4−Alexa 647で30分間室温にて染色した。洗浄後、FACS(FACS Aria II)分析を行った。
ヒトiPS細胞(クローン#201B7)を96穴プレート中のSNLフィーダー細胞上にプレーティングした。プレーティングの5日後、iPS細胞のドーナツ型コロニーが偶然いくつか得られた。これらの細胞を、4μM 化合物1とともに、4.5時間37℃でインキュベートした。蛍光顕微鏡像をCarl Zeiss Axioskopを使用して取得した。
hES細胞株、KhES−1を、先に記載の通り維持した(Suemori, H. et al. Biochem. Biophys. Res. Commun. 2006, 345, 926-932)。hESCの分化を誘導するため、細胞をMatrigel被覆プレート上に播種し、10%FBSを加えたDMEM中で500nMのall−transレチノイン酸(Sigma R2625)とともに4日間培養した。1μMの化合物1で2時間染色後、細胞をPBSでリンスし、新鮮な(色素を含まない)培地中に保持した。DP72を使用し、Olympus IX71による蛍光顕微鏡観察を行った。
ヒトMDR1を安定に発現しているKB/MDR1、およびKB3−1宿主細胞(Ueda, K et al., PNAS 84, 3004-3008, 1987)を、37℃の加湿インキュベーター(5%CO2)中において、10%FBSを添加したDMEM中で維持した。
細胞を、35mm/ガラスベースディッシュ(IWAKI)中で、10%FBSを添加したDMEM中、ディッシュあたり5×105細胞の密度で24〜48時間継代培養した。その後、細胞を、10%FBSおよび前記化合物を添加したDMEM中、37℃にて2時間インキュベートし、リン酸緩衝生理食塩水(PBS)で洗浄し、共焦点顕微鏡(LSM 700;Carl Zeiss)で観察した。
細胞をカバーガラス上で増殖させ、PBS+(0.1mg/ml CaCl2およびMgCl2・6H2Oを含むPBS)中の4%パラホルムアルデヒドで20分間室温にて固定した。PBS+で希釈した10%ヤギ血清で1時間ブロッキングを行った後、細胞を5μg/ml MRK16とともにインキュベートし、次いで蛍光標識二次抗体とともにインキュベートした。
RNAを5種類のヒトES(hES)細胞株(KhES−1、KhES−2、KhES−3、KhES−4、KhES−5)および3種類のヒトiPS(hiPS)細胞株(IMR90−1、IMR90−4、201B7)から調製した。ファーストストランドcDNAを逆転写酵素(Applied Biosystems)を用いて合成した。遺伝子
発現プロファイルを、44種類のABCトランスポーターと4種類のハウスキーピング遺伝子(GAPDH、18S、HPRT1、GUSB)とをカバーするTaqMan Array Gene Signature 96−Well Plate,Human ABC Transporters(Applied Biosystems)を用いた定量的リアルタイムPCR(RT−PCR)により分析した。発現量をGAPDHに対して標準化した。
細胞を上記のように調製した。1μMの化合物1で1時間染色した後、細胞をPBSでリンスし、さらに24時間培養した。DP72を使用し、Olympus IX71による蛍光顕微鏡観察を行った。化合物1による染色中およびその後の分析を行うまでの培養中、CsAを10μMの濃度で添加した。FACS分析のため、細胞を氷冷PBSで二回洗浄した後、0.25%トリプシン−EDTAを用いて単一細胞に分離し、直接分析のための懸濁液を得た。未処理の未分化または分化したhESCのベースラインを参照することにより、化合物1を含む細胞の割合を、FACSCaliburフローサイトメーター(Becton Dickenson,CA,USA)上で試験した。
Young−Tae Changのグループより蛍光化合物ライブラリーの提供を受けた(Ahn, Y-H., Lee, J-S. and Chang, Y-T. J. Am. Chem. Soc. 2007, 129, 4510-4511)。ヒトiPS細胞(クローン#201B2)を96穴プレート内のSNLフィーダー細胞上にプレーティングした。プレーティングして5日後、蛍光化合物を最終濃度4μMで添加した。一晩インキュベートしたあと、蛍光顕微鏡像をCarl Zeiss Axioskopを用いて取得した。
ヒトiPS細胞を化合物1のクロロアセチル誘導体(化合物22)(1μM)で4時間処理し、その後PBSで2回洗浄した。CTK溶液(Reprocell)処理によってSNLフィーダー細胞を除去した後、ヒトiPS細胞をAccutase処理により回収した。ProteoExtractR Cytosol/Mitochondria Fractionation kitを添付の説明書に従って使用し、細胞ペレットからミトコンドリア分画を得た。Ampholine(pH3.5−10,Phamalyte
3−10 for IEF,GE Healthcare)を用いて等電点電気泳動(IEF)を行った。その後、IEFストリップを10%SDS−PAGEゲル上で分離し、二次元解析を行った。二次元蛍光イメージをTyphoon 9400スキャナー(GE healthcares)を用いて532nm/580nmの励起/発光波長にて取得した。蛍光標識スポットをゲルから切り出し、質量分析を行った。
ヒトiPS細胞(クローン#201B7)を24穴プレート内のSNLフィーダー細胞上にプレーティングした。プレーティングから1日後、細胞を1μMの化合物1とともに4時間37℃にてインキュベートした。化合物1をPBSによる洗浄で除いた後、細胞を9.6nM MitoRedとともに30分間37℃にてインキュベートした。細胞をPBSでリンスし、新鮮な(色素を含まない)培地で保持した。蛍光顕微鏡像をCarl Zeiss Axioinvert 100Mを用いて取得した。
1,10−ジクロロデカン(8.73g、41.3mmol)およびトリエチルアミン(6mL)を、DMF(40mL)中のm−アミノフェノール(3.85g、35.3mmol)溶液に添加した。該混合物を100℃で3日間撹拌した。飽和NaHCO3水溶液で反応をクエンチし、EtOAcで抽出を行った。有機層を合わせて食塩水で洗浄し、無水MgSO4で乾燥し、濾過し、減圧下で濃縮した。残渣をフラッシュカラムクロマトグラフィーで精製し(シリカゲル、ヘキサン:EtOAc=9:1〜1:1)、表題化合物を得た(1.92g、6.76mmol、収率19%)。褐色油;Rf=0.57(33%EtOAc−hexane);1H NMR(600MHz,DMSO−d6)δ8.84(s,1H),6.80(dd,J=7.6,8.3Hz,1H),5.97(d,J=7.6Hz,1H),5.94(s,1H),5.92(d,J=8.2Hz,1H),5.36(s,1H),3.61(m,2H),2.89(m,2H),1.69(m,2H),1.49(m,2H),1.35−1.25(m,12H);13C NMR(150MHz,DMSO−d6)δ158.3,150.6,129.5,103.7,103.0,98.9,45.6,43.1,32.2,29.2,29.1×2,28.9,28.4,26.9,26.4;FABMS m/z 283[M+];C16H26NOClに対するHRMS計算値[M+]283.1703,測定値283.1708。
フタルイミドカリウム(1.88g、10.1mmol)をDMF(20mL)中の化合物23(1.92g、6.76mmol)の溶液に加えた。該混合物を100℃で一晩撹拌した。飽和NaHCO3水溶液で反応をクエンチし、EtOAcで抽出を行った。有機層を合わせて食塩水で洗浄し、無水MgSO4で乾燥し、濾過し、減圧下で濃縮した。残渣をフラッシュカラムクロマトグラフィーで精製し(シリカゲル、ヘキサン:EtOAc=5:1〜1:1)、表題化合物を得た(1.83g、4.64mmol、収率69%)。褐色油;Rf=0.51(33%EtOAc−ヘキサン);1H NMR(600MHz,DMSO−d6)δ8.83(s,1H),7.83(m,2H),7.81(m,2H),6.79(dd,J=8.3,7.6Hz,1H),5.97(d,J=8.2Hz,1H),5.94(s,1H),5.91(d,J=7.5Hz,1H),5.

4−フルオロベンズアルデヒド(242g、1.95mmol)およびm−アミノフェノール(213g、1.95mmol)を60% H2SO4(10mL)中の化合物24(770g、1.95mmol)の溶液に加えた。該混合物を130℃で一晩撹拌した。反応液を氷水で希釈し、濾過した。沈殿物を蒸留水(DW)で洗浄し、メタノールに溶解し、減圧下で濃縮した。残渣をHPLC(Inertsil ODS−3、methanol:0.1% TFA−DW=0:1〜1:0)で精製し、表題化合物を得た(34g、0.059mmol、収率3%)。紫色油;1H NMR(600MHz,酢酸−d4)δ7.51(dd,J=8.9,5.5Hz,2H),7.40(dd,J=8.9,8.9Hz,2H),7.31(d,J=8.9Hz,2H),6.98−6.93(m,2H),3.40(m,2H),3.04(m,2H),1.74−1.67(m,4H),1.44−1.32(m,12H);13C NMR(150MHz,DMSO−d6)δ164.0,162.4,158.6,158.3,158.1,157.9,132.2,132.08,132.02,128.4,118.2,117.0,116.2,116.1,113.2,113.0,97.3,94.1,43.0,39.0,29.1,29.0×2,28.9,28.7,28.2,27.2,26.6,26.0;FABMS m/z 460[M+];C29H35FN3Oに対するHRMS計算値[M+]460.2764,実測値460.2765。
塩化クロロアセチル(30μL、0.39mmol)およびトリエチルアミン(100μL)をDMF(1.5mL)中の化合物25(82.5mg、0.12mmol)の溶液に加えた。該混合物を37℃で3日間撹拌した。反応液をメタノールで希釈し、濾過した。ろ液をHPLC(Inertsil ODS−3、methanol:0.1%TFA−DW=0:1〜1:0)で精製し、表題化合物を得た(21mg,0.032mmol、収率27%)。紫色油;FABMS m/z 536[M+];C31H36ClFN3O2に対するHRMS計算値[M+]536.2480,実測値536.2488。
N−メチルアミノフェノールを先の論文の通り合成した(Charng-Sheng Tsai, et al.,
Chem.Comm., 46, 5575-5577, 2010)。簡単に述べると、DMF中のm−アミノフェノール(1.51g、13.84mmol)、CH3I(2.16g、15.22mmol)およびK2CO3(2.10g、15.19mmol)を室温で19時間撹拌した。該混合物を蒸留水で希釈し、EtOAcで抽出を行った。有機層を合わせて食塩水で洗浄し、無水Na2SO4で乾燥し、減圧下で濃縮した。残渣をフラッシュカラムクロマトグラフィーで精製し(シリカゲル、ヘキサン:EtOAc=1:0〜3:7)、表題化合物を得た(褐色油、658mg、5.34mmol、収率39%)。
m−アミノフェノール(1.31g、12mmol)をメタンスルホン酸(12mL)中の2−フルオロベンズアルデヒド(502mg、4.04mmol)の溶液に加えた。該混合物を120℃で24時間撹拌した。反応液を蒸留水で希釈し、K2CO3で中和した。沈殿物を濾過し、メタノールに溶解した。得られた溶液をセライト濾過し、ろ液をHPLC(Inertsil ODS−3、メタノール:0.1% TFA−DW=0:1〜1:0)で精製し、表題化合物を得た(10mg、0.025mmol、収率0.6%)。紫色油;1H NMR(300MHz,CD3OD)δ7.76−7.69(m,1H),7.51−7.40(m,3H),7.24(d,J=9.0Hz,2H),6.86(d,J=9.0Hz,2H),6.82(s,2H)。
ヒト多能性幹細胞に対して選択的な蛍光プローブを見いだすため、326種の蛍光化合物の化学ライブラリーをスクリーニングした。ヒトiPS細胞をフィーダー細胞よりも強く染色する31種のローダミン系分子(表2に示す化合物1〜21および26〜34)が、画像ベースのスクリーニングにより単離された(図1A)。さらに、表2に示す合成化
合物35および36により、ヒトiPS細胞をフィーダー細胞よりも強く染色することができた(図1A)。
た細胞において高度に発現していた(それぞれ29倍および24倍)。これらの結果は、ABCB1(MDR1)が化合物1の排除に重要な役割を担っている可能性を示している。
KP−1による、ヒトES細胞由来造血細胞の染色が試験された(Takayama et al., Blood 111, 5298-5306, 2008 または Takayama et al., J Exp Med 207, 2817-2830, 2010)(図6A)。FACS分析の結果、KP−1によってSSEA−4陽性ヒトES細胞と、CD45、CD235、CD41aまたはCD43を発現するヒト初期造血細胞とが区別されることが示され、KP−1は初期造血をモニタリングするのに有用であることが示唆された。
次に、臨床的に重要な別の分化過程である心筋形成について、KP−1によるモニタリングが可能かどうか調べた。先の研究(Wang et al., Bba-Biomembranes 1461, 177-200,
2011)に記載の方法に変更を加えて心筋分化を行った。簡単に述べると、ヒトiPS細胞をヒトラミニン211(BioLamina,Sweden)でコートした3.5cmの培養皿上で培養した。心筋コロニーの生成を促進するため、WNTシグナル伝達阻害剤を心筋分化3〜9日目に添加した。心筋コロニーを15日目に回収し、浮遊培養で7〜10日間培養した。フローサイトメトリーで確認したところ、KP−1によってヒトiPS細胞とヒトiPS細胞由来心筋細胞とを区別することができた(図6B)。ヒトiPS細胞に由来する心筋細胞における44種のヒトABCトランスポーターの発現パターンをRT−PCRによって調べた(図7A)。驚くべき事に、ABCG2もABCB1も心筋形成中に誘導されなかった。代わりに、2種の細胞表面膜ABCトランスポーター、ABCA1およびABCC5が分化中に誘導された。ABCD3も誘導されたが、このペルオキシゾームABCタンパク質はKP−1の選択性には関与していないと思われた。ABCA1およびABCC5がKP−1の排出を引き起こす能力を、HEK293細胞で各トランスポーターを過剰発現させることによって調べた。その結果、KP−1はABCC5の優れた基質である(図7B)が、ABCA1に対してはそうではないことが示された(図示せず)。KP−1は分化によって誘導される広範なABCトランスポーターの基質として作用する可能性がある。
Claims (15)
- 以下の工程を含む、多能性細胞を選別する方法:
(i)細胞試料を、MDR1トランスポーターを介して多能性細胞から排除される化合物と接触させる工程;および
(ii)前記化合物を蓄積した細胞を、多能性細胞として検出する工程。 - 前記化合物が式(I)

の構造を有する、請求項1記載の方法。 - R2およびR3がそれぞれ水素、メチル、エチル、ジメチルアミノエチルおよび10−クロロアセチルアミノデシルからなる群より選択されるか、またはR2およびR3が1−ピペリジニルの環状構造を形成し;
R4がメチルであり;
R5が水素、フッ素、メトキシ、フェノキシ、メチルチオ、メチル、プロピルおよびフェニルからなる群より選択され;
R6およびR7がそれぞれ水素、フッ素、塩素、メトキシおよびメチルからなる群より選択され;
R8がフッ素であり;
R9がメトキシであり;そして
XがO、Sおよびメチルアミノからなる群より選択される;
請求項2記載の方法。 - 前記細胞試料が初期化因子を導入した体細胞である、請求項1記載の方法。
- 前記初期化因子がOctファミリー遺伝子またはその遺伝子産物、Soxファミリー遺伝子またはその遺伝子産物、Klf4ファミリー遺伝子またはその遺伝子産物、およびMycファミリー遺伝子またはその遺伝子産物、からなる群より選択される少なくとも1つの因子である、請求項6記載の方法。
- 前記細胞試料が、体細胞への分化を誘導された多能性細胞からなる、請求項1記載の方法。
- 請求項1記載の方法により多能性細胞を選別し、選別された前記細胞を選択する工程を含む、多能性細胞を選択する方法。
- 請求項1記載の方法により多能性細胞を選別し、選別された前記細胞を細胞試料から排除する工程を含む、分化した細胞を選択する方法。
- MDR1トランスポーターを介して多能性細胞から排除される化合物を含む、多能性細胞を選別するためのキット。
- 前記化合物が式(I):
