Abstract
Description
The present invention relates to a method for fractionating polychlorinated biphenyls, and more particularly to a method for fractionating polychlorinated biphenyls contained in an aliphatic hydrocarbon solvent solution of polychlorinated biphenyls.
The European Union (EU) Food Regulation Standard (COMMISSION REGULATION (EU) No 1259/2011) is a polychlorinated biphenyl class as a target for analysis of harmful substances in meat such as beef and pork and animal food such as eggs. And dioxins.
There are 209 types of polychlorinated biphenyls (PCBs), which are classified into dioxin-like polychlorinated biphenyls (DL-PCBs) and non-dioxin-like polychlorinated biphenyls (non-DL-PCBs). DL-PCBs is a general term for PCBs that exhibit the same toxicity as polychlorinated dibenzopararadioxins (PCDDs) and polychlorinated dibenzofurans (PCDFs), which are dioxins. Is used to include the DL-PCBs. On the other hand, non-DL-PCBs are PCBs other than DL-PCBs.
Thus, since PCBs are partly classified as dioxins, it is convenient to analyze them together with dioxins.
When analyzing PCBs and dioxins contained in foods (hereinafter sometimes collectively referred to as “PCBs etc.”), it is necessary to first extract PCBs etc. from food samples and secure an analytical sample. . When the sample is a solid material, for example, PCBs and the like are extracted from the solid material by a Soxhlet extraction method. In addition, when the sample is in a liquid form such as a beverage or edible oil, for example, PCBs from a liquid food or from a solution obtained by transferring PCBs contained in the liquid food into a solvent using a collector such as a filter. After collecting and collecting, PCBs collected in the collector are extracted by washing the collector or applying the Soxhlet extraction method to the collector. The extracted liquid such as PCBs thus obtained is quantitatively analyzed as an analytical sample using an analyzer such as a gas chromatograph mass spectrometer (GC / MS).
The above extract is a polychlorinated polycyclic ring such as polychlorinated diphenyl ether (PCDE), which has similar chemical structure and chemical behavior to various contaminant components that may affect the analysis results such as PCBs, for example, PCBs. Since it contains aromatic hydrocarbons, it is usually concentrated after being purified and applied to an analyzer. As a method for purifying the extract, Patent Document 1 discloses a chromatograph equipped with a first column packed with silica gel sulfate and silver nitrate silica gel as a purification agent, and a second column packed with activated carbon-containing silica gel or graphite carbon as an adsorbent. A method using a graph column is described. In this method, activated carbon silica gel or graphite carbon can be selectively used as the adsorbent for the latter column, and when both are used in combination, they should be used in a stacked or mixed state. Can do.
In the purification method using this chromatographic column, first, an extract such as PCBs is injected into the former column, and then a hydrocarbon solvent is supplied to the former column. This hydrocarbon solvent dissolves PCBs and the like in the injected extract and passes through the front column and the rear column. At this time, PCBs and the like dissolved in the hydrocarbon solvent pass through the purification agent in the former column and are adsorbed by the adsorbent in the latter column. On the other hand, the contaminating components contained in the extract are dissolved in a hydrocarbon solvent together with PCBs and the like, and partly decomposed and partly adsorbed when passing through the purification agent in the preceding column. Of the contaminating components or decomposition products thereof, those that are not adsorbed by the purification agent pass through the adsorbent of the subsequent column in a state dissolved in the hydrocarbon solvent, and are discharged from the column.
Next, the front column and the rear column are separated, and alkylbenzene capable of dissolving PCBs and the like is supplied to the rear column. And if the alkylbenzene which passes a back | latter stage column is ensured, alkylbenzene solutions, such as PCBs from which the contaminant component was removed, will be obtained. This alkylbenzene solution can be used as an analytical sample such as PCBs, and after being concentrated as appropriate, it is analyzed by an analyzer such as GC / MS.
In such a purification treatment method, all kinds of PCBs and dioxins contained in the extract are adsorbed by the adsorbent in the subsequent column and extracted with alkylbenzene. Therefore, in the analyzer, all kinds of PCBs, PCDDs and PCDFs contained in the alkylbenzene solution are analyzed simultaneously.
However, when analyzing all types of PCBs and alkylbenzene solutions containing PCDDs and PCDFs simultaneously, the results may be unreliable. For example, when analyzing an alkylbenzene solution by high resolution GC / MS, it is known that mono-ortho PCBs affects the quantitative analysis results of PCDDs and PCDFs, and conversely, PCDDs and PCDFs affect the quantitative analysis results of mono-ortho PCBs. Yes.
Therefore, in the analysis of PCBs and the like, attempts have been made to prepare analytical samples by fractionating PCBs and the like into several types. For example, Patent Document 2 describes a method in which graphite-like carbon or a mixture of graphite-like carbon and other materials such as silica gel, activated carbon-containing silica gel, activated carbon, alumina, or zeolite is used as an adsorbent such as PCBs.
In this method, an extraction liquid such as PCBs purified in advance is supplied to a column packed with an adsorbent, and the adsorbent adsorbs PCBs or the like. Then, several kinds of solvents are sequentially supplied to the column to prepare several kinds of solutions such as PCBs. According to Patent Document 2, for example, three types of solutions can be prepared, such as a solution containing non-DL-PCBs, a solution containing mono-ortho PCBs, and a solution containing non-ortho PCBs, PCDDs, and PCDFs.
However, since this method adsorbs all kinds of PCBs and the like to the adsorbent as in the method described in Patent Document 1, it is difficult to accurately fractionate PCBs and the like. For example, a solution containing mono-ortho PCBs may contain some non-ortho PCBs, PCDDs and PCDFs, and a solution containing non-ortho PCBs, PCDDs and PCDFs may contain some mono-ortho PCBs. is there.
The present invention is intended to enable PCBs to be fractionated with high accuracy into a fraction of non-ortho PCBs and a fraction containing mono-ortho PCBs and non-DL-PCBs.
The method for fractionating polychlorinated biphenyls according to the present invention includes a step of passing an aliphatic hydrocarbon solvent solution of polychlorinated biphenyls through an activated carbon-containing silica gel layer and a graphite-containing silica gel layer in this order.
In this fractionation method, when the aliphatic hydrocarbon solvent solution of polychlorinated biphenyls passes through the activated carbon-containing silica gel layer and the graphite-containing silica gel layer in this order, the non-ortho PCBs of the polychlorinated biphenyls are activated carbon-containing silica gel layers. Or it is adsorbed on the graphite-containing silica gel layer. On the other hand, monoortho PCBs and non-DL-PCBs of polychlorinated biphenyls remain in the aliphatic hydrocarbon solvent solution and pass through the activated carbon-containing silica gel layer and the graphite-containing silica gel layer. As a result, the polychlorinated biphenyls in the aliphatic hydrocarbon solvent solution are non-ortho PCBs adsorbed on the activated carbon-containing silica gel layer or the graphite-containing silica gel layer, and the mono-ortho PCBs remaining in the aliphatic hydrocarbon solvent solution and the non-DL- Fractionated into PCBs.
In one embodiment of the fractionation method of the present invention, the aliphatic hydrocarbon solvent solution that has passed through the graphite-containing silica gel layer is further passed through the alumina layer.
In this case, when the aliphatic hydrocarbon solvent solution that has passed through the activated carbon-containing silica gel layer and the graphite-containing silica gel layer passes through the alumina layer, the remaining mono-ortho PCBs and non-DL-PCBs are adsorbed on the alumina layer. As a result, the polychlorinated biphenyls in the aliphatic hydrocarbon solvent solution are fractionated into non-ortho PCBs adsorbed on the activated carbon-containing silica gel layer or graphite-containing silica gel layer, and mono-ortho PCBs and non-DL-PCBs adsorbed on the alumina layer. Is done.
A fractionation method of this form is provided by supplying a solvent capable of dissolving polychlorinated biphenyls to an activated carbon-containing silica gel layer and a graphite-containing silica gel layer through which an aliphatic hydrocarbon solvent solution has passed, and the activated carbon-containing silica gel layer and the graphite-containing silica gel Securing the solvent that has passed through the layer, and supplying a solvent capable of dissolving polychlorinated biphenyls to the alumina layer through which the aliphatic hydrocarbon solvent solution has passed, and securing the solvent that has passed through the alumina layer And may further be included.
When these steps are included, the non-ortho PCBs adsorbed on the activated carbon-containing silica gel layer or the graphite-containing silica gel layer, and the mono-ortho PCBs and non-DL-PCBs adsorbed on the alumina layer are extracted by dissolving in the supplied solvent, respectively. Obtained as an extract.
The present invention according to another aspect relates to a method for preparing a sample for analyzing polychlorinated biphenyls contained in a polychlorinated biphenyl solution, and the preparation method includes a silver nitrate silica gel layer and a sulfate silica gel layer. A step of adding a polychlorinated biphenyls solution to the purification layer, a step of supplying an aliphatic hydrocarbon solvent to the purification layer to which the polychlorinated biphenyls solution has been added, and an aliphatic hydrocarbon solvent having passed through the purification layer. A step of passing the activated carbon-containing silica gel layer and the graphite-containing silica gel layer in this order; a step of passing the aliphatic hydrocarbon solvent that has passed through the graphite-containing silica gel layer through the alumina layer; and an alumina layer through which the aliphatic hydrocarbon solvent has passed. On the other hand, a solvent capable of dissolving polychlorinated biphenyls is supplied, and the solvent that has passed through the alumina layer is used as the first analytical sample. And supplying a solvent capable of dissolving polychlorinated biphenyls to the activated carbon-containing silica gel layer and the graphite-containing silica gel layer through which the aliphatic hydrocarbon solvent has passed, and passing through the activated carbon-containing silica gel layer and the graphite-containing silica gel layer Securing the solvent as a second analytical sample.
In this preparation method, when the aliphatic hydrocarbon solvent is supplied to the purified layer to which the polychlorinated biphenyls solution is added, the aliphatic hydrocarbon solvent passes through the purified layer. At this time, the polychlorinated biphenyls and impurities contained in the polychlorinated biphenyl solution are dissolved in the aliphatic hydrocarbon solvent. A part of the contaminating components reacts with the silver nitrate silica gel layer or the sulfuric acid silica gel layer of the purification layer and decomposes. Further, a part of the contaminating components and decomposition products are adsorbed on the silver nitrate silica gel layer or the sulfate silica gel layer. On the other hand, polychlorinated biphenyls pass through the purification layer in a state dissolved in an aliphatic hydrocarbon solvent. As a result, polychlorinated biphenyls are separated from some of the contaminating components.
When the aliphatic hydrocarbon solvent in which the polychlorinated biphenyls are dissolved, which has passed through the purification layer, passes through the activated carbon-containing silica gel layer and the graphite-containing silica gel layer in this order, the non-ortho PCBs of the polychlorinated biphenyls are activated carbon-containing silica gel. The monoortho PCBs and non-DL-PCBs of the polychlorinated biphenyls are adsorbed to the alumina layer when adsorbed on the layer or the graphite-containing silica gel layer and pass through the alumina layer. Therefore, the first analysis sample is a monoortho PCBs and non-DL-PCBs analysis sample, and the second analysis sample is a non-ortho PCBs analysis sample. That is, according to this preparation method, the sample for analysis of mono-ortho PCBs and non-DL-PCBs and the sample for analysis of non-ortho PCBs can be prepared separately.
In this preparation method, it is usually preferable to add a polychlorinated biphenyl solution to the silver nitrate silica gel layer of the purification layer.

The purified
The silver nitrate

The sulfated

The first active
The second active

The activated carbon-containing

The graphite-containing

The method for preparing a sample for analysis mainly includes the following fractionation step and extraction step.
<Fractionation process of polychlorinated biphenyls>
After setting to the initial state, a polychlorinated biphenyl solution is injected into the
The polychlorinated biphenyl solution to be injected here is, for example, an environmental sample such as air or soil, an electrical insulating oil of electrical equipment such as a transformer or a capacitor, and a sample from a sample that may contain polychlorinated biphenyls such as a food sample. However, it may also be an electrically insulating oil or fish oil (fish oil) that may contain polychlorinated biphenyls. Such a polychlorinated biphenyl solution has a similar chemical structure and chemical behavior as the polychlorinated biphenyls, and may affect the analysis results of the polychlorinated biphenyls, such as PCDDs, PCDFs, and PCDEs. In many cases, aromatic hydrocarbons are included as contaminant components.
Moreover, in the case of the extract from a soil sample, this extract often contains paraffins (linear hydrocarbon compounds) that are contained in a large amount in the soil as impurities. Paraffins are easily adsorbed on carbon-based adsorbents together with non-ortho PCBs, PCDDs and PCDFs, and are easily extracted together with non-ortho PCBs and PCDDs and PCDFs from the adsorbents. In the case of analysis by GC-HRMS method), it is known as a causative substance of rock mass fluctuation that affects analysis accuracy.
The extraction liquid of polychlorinated biphenyls can be directly injected into the
The amount of the polychlorinated biphenyl solution injected into the
When the polychlorinated biphenyl solution is an oily oil such as electrical insulating oil or fish oil, the polychlorinated biphenyl solution is fractionated together with an aliphatic hydrocarbon solvent that can dissolve the polychlorinated biphenyl solution or as a solution previously dissolved in the solvent. The

Next, the solvent is supplied from the

By the above extraction process, the analysis sample of mono-ortho PCBs and non-DL-PCBs and the analysis sample of non-ortho PCBs and PCDDs and PCDFs are obtained separately.
The two types of analytical samples prepared in this way are applied to the analysis separately. As an analysis method, GC-HRMS, GC-MSMS, GC-QMS, ion trap GC / MS, or the like is usually used depending on the type of solvent used for extracting polychlorinated biphenyls and PCDDs and PCDFs from the
When analyzing samples for analysis of mono-ortho PCBs and non-DL-PCBs, since the sample for analysis is substantially free of non-ortho PCBs and PCDDs and PCDFs, mono-ortho PCBs and non-DL-PCBs are not affected by them. It is possible to quantify with high accuracy. For this reason, predetermined non-DL-PCBs (IUPAC numbers are # 28, # 52, # 101, # 101) which are subject to analysis in the European Union (EU) Food Regulation Standard (COMMISSION REGULATION (EU) No 1259/2011) 138, # 153 and # 180, 6 types of PCBs having 3 to 7 chlorine atoms) can be quantified by analysis of this analytical sample.
On the other hand, in the analysis of analytical samples of non-ortho PCBs and PCDDs and PCDFs, since the analytical sample does not substantially contain mono-ortho PCBs and non-DL-PCBs, non-ortho PCBs and PCDDs are not affected by these PCBs. And PCDFs can be quantified with high accuracy.
Note that GC-TOFMS can also be used as the GC / MS method, and in this case, two types of analytical samples can be mixed and analyzed simultaneously.

Second Embodiment Referring to FIG. 3, a second example of an apparatus capable of performing the analytical sample preparation method according to the present invention will be described. In FIG. 3, the

And after heating an adsorption layer to 90 degreeC, 1.5 mL of toluene was supplied from the lower layer side of the adsorption layer, and the toluene which passed the adsorption layer was collect | recovered through the 1st branch path.
Table 2 shows the results of quantitative analysis of polychlorinated biphenyls and the like contained in the collected toluene by the HRGC / HRMS method and calculation of the individual recovery rates of polychlorinated biphenyls and the like. The recovery rate of polychlorinated biphenyls refers to the ratio (%) of polychlorinated biphenyls, etc. contained in the solvent from which the polychlorinated biphenyls are extracted relative to the initial amount of polychlorinated biphenyls added to the sample. .
According to Table 2, the recovered toluene contains non-ortho PCBs and PCDDs and PCDFs at a high recovery rate (more than 50% defined in various regulatory standards for polychlorinated biphenyls and dioxins), and mono-ortho PCBs and non-DL- Substantially free of PCBs. This result shows that in Examples 1 to 6, the group of non-ortho PCBs and the group of PCDDs and PCDFs could be separated from the group of mono-ortho PCBs and DL-PCBs with high accuracy.
Further, since no substantial interference peak due to PCDE was found in the quantification by the HRGC / HRMS method, it was found that in Examples 1 to 6, non-ortho PCBs, PCDDs and PCDFs were effectively separated from PCDE. did.
Comparative Examples 1-5
In the sample preparation device for analysis of polychlorinated biphenyls, only the specification of the adsorption layer of the fractionating device was changed, and the polychlorinated biphenyls, PCDDs and PCDFs contained in the fish oil sample were extracted in the same manner as in Examples 1-6. did. The extract was quantitatively analyzed by the HRGC / HRMS method, and the recovery rate of polychlorinated biphenyls and the like was calculated. The specifications of the adsorption layer are as shown in Table 3. The results are shown in Table 4.
According to Table 4, the extract has a low recovery rate of some of the PCDDs and PCDFs with a large number of chlorine, and also contains a large amount of mono-ortho PCBs. This result shows that in Comparative Examples 1 to 5, the group of non-ortho PCBs and PCDDs and PCDFs could not be separated from the group of mono-ortho PCBs and non-DL-PCBs.
Further, in Comparative Examples 2 to 5, since interference peaks due to PCDE were observed in the measurement results by the HRGC / HRMS method, it was found that PCDE was mixed in the group of non-ortho PCBs and PCDDs and PCDFs.
Examples 7-8
In the same manner as in Examples 1 to 6, polychlorinated biphenyls and the like contained in the liquid paraffin sample were extracted with toluene. At this time, in the adsorption layer, the ratio of the activated carbon contained in the activated carbon-containing silica gel, the ratio of the graphite contained in the graphite-containing silica gel, and the lamination ratio (volume ratio) between the activated carbon-containing silica gel layer and the graphite-containing silica gel layer are as shown in Table 5. Set to.
Table 6 shows the results of quantitative analysis of polychlorinated biphenyls and the like contained in the toluene extract by the HRGC / HRMS method and calculation of the recovery rate of polychlorinated biphenyls and the like. Here, in order to confirm that non-ortho PCBs and PCDDs and PCDFs can be extracted from the adsorbed layer without being affected by liquid paraffin, only non-ortho PCBs, PCDDs and PCDFs were quantitatively analyzed, and their recovery rates were calculated. The rock mass fluctuation was also confirmed.
According to Table 6, the recoveries of non-ortho PCBs and PCDDs and PCDFs are high. In addition, no substantial change in rock mass was observed. This result shows that liquid paraffin is not adsorbed by the adsorbing layer but passes through the adsorbing layer and is discarded, and does not hinder the extraction and quantification of these polychlorinated biphenyls.

The ratio of activated carbon contained in the activated carbon-containing silica gel, the ratio of graphite contained in the graphite-containing silica gel, and the lamination ratio (volume ratio) between the activated carbon-containing silica gel layer and the graphite-containing silica gel layer were set as shown in Table 7.
In the extraction operation of polychlorinated biphenyls and the like, about 4 mL of a fish oil sample solution was added to the silver nitrate silica gel layer of the purified layer, and the purified layer was heated to 60 ° C. And 85 mL of n-hexane was gradually supplied to the purification layer, and this n-hexane was passed through the purification layer and the adsorption layer. After n-hexane passed through the adsorption layer, the adsorption layer was dried by passing compressed air. Then, 1.5 mL of an n-hexane mixed solvent containing 50% by weight of dichloromethane was supplied from the activated carbon-containing silica gel layer side of the adsorption layer while maintaining the alumina layer of the adsorption layer at room temperature (25 ° C.). By collecting the mixed solvent that passed through, a first sample for analysis was obtained. According to Table 8, the second sample for analysis contains non-ortho PCBs and PCDDs and PCDFs at a high recovery rate. On the other hand, the first sample for analysis contains mono-ortho PCBs and non-DL-PCBs at a high recovery rate. This result shows that in Examples 9 to 12, the group of non-ortho PCBs, PCDDs and PCDFs, and the group of mono-ortho PCBs and non-DL-PCBs could be fractionated with high accuracy.
Table 8 also relates to the first analytical samples obtained in Examples 9 to 12, with non-DL-PCBs subject to EU food regulation, with a recovery rate (60 to 120%) in an appropriate range under the same regulation.
