本発明は、米国エネルギー省がカルフォルニア大学の理事に対して与えた規約番号W−7405−ENG−36、および米国エネルギー省がロスアラモス国家安全保障有限責任会社に対して与えた規約番号DE−AC52−06NA25396に基づく支援によってなされた。政府は本発明の所定の権利を有する。
本明細書は、2008年8月29日に提出された米国仮特許番号第61/190,581号に対して優先権を主張するものである。
世界的に人口が増加し、使用可能な農地が失われるか、または使用不能になり続けるため、より効率的で、かつ、地球に優しい農業システムに対する必要性は、人類にとって興味のある最優先事項である。収量、タンパク質含有量、および植物の成長速度を向上させることは、目下の課題により効果的に対処し得る農業システムの開発において主な目的になっている。
本発明は、増強された成長速度、種子および果実の収量、ならびに総バイオマス収量を示すトランスジェニック植物、また、成長が増強されたトランスジェニック植物を産生する方法に関する。一実施形態においては、グルタミンフェニルピルビン酸トランスアミナーゼ(glutamine phenylpyruvate transaminase:GPT)を過剰に発現するために設計されたトランスジェニック植物が提供される。

本発明は、成長特性および他の農学的な性質が十分に向上している新規なトランスジェニック植物を提供する。十分に向上した農学的な性質は、促進された生育、増大した成熟植物の生体重および全現存量、より早期かつより豊富な量の開花、ならびに果実および種子におけるより多くの収量を含むがこれらに限定されない。本発明のトランスジェニック植物は、1つ以上の発現可能な遺伝的コンストラクトを植物に対して導入することにより生成される。この遺伝的コンストラクトは、グルタミンフェニルピルビン酸トランスアミナーゼ(glutamine phenylpyruvate transaminase:GPT)をコードする1つ以上のポリヌクレオチドの発現を促進することができる。本発明は、例えば、異種のシロイヌナズナのGPT遺伝子を保有し、かつ発現するトランスジェニックタバコ植物の生成によって実証されている(後述する実施例2)。また、同じ代謝経路、すなわちシグナル代謝産物2−ヒドロキシ−5−オキソプロリン(2-hydroxy-5-oxoproline)の生合成、において機能するGPTホモログは、全ての植物種に含まれていることが期待される。このように、本発明の実施において、GPTホモログまたはその機能的な変異体をコードする全ての植物遺伝子が、本発明のトランスジェニック植物の生成に有用であり得る。
本発明に係るトランスジェニック植物を生成するために、所望の導入遺伝子に関する遺伝子コード配列は、形質転換された植物細胞内で導入遺伝子配列の発現を導くことができる核酸コンストラクト(本明細書中において、(導入遺伝子)発現ベクター/発現ベクター、発現カセット、発現コンストラクトまたは遺伝的コンストラクトとしても交換可能に引用される)中に組み込まれなければならない。所定の導入遺伝子を保有する核酸コンストラクトは、多数の公知の方法を用いて、植物細胞または種々の細胞内に導入され得る。公知の方法は、限定されないが、エレクトロポレーション、DNAボンバードメントまたは遺伝子銃、マイクロインジェクション、ならびに種々のDNAベースのベクター(例えばアグロバクテリウム・ツメファシエンス(Agrobacterium tumefaciens)およびアグロバクテリウム・リゾゲネス(Agrobacterium rhizogenes)のベクター)の使用を介する方法を含む。形質転換植物細胞中にいったん導入されると、核酸コンストラクトは、一過的に、あるいは安定的に、組み込まれた導入遺伝子(すなわちGPT)の発現を導くであろう。安定的に発現させることが好ましく、安定的な発現は、核酸コンストラクトをクロモソームに組み込ませることができる植物の形質転換ベクターを利用することにより達成される。一度植物細胞への形質転換が成功すれば、これを栽培することによりトランスジェニック植物を再生し得る。
リウム形質転換システムに使用するバイナリーベクターは、典型的に、T−DNAのボーダー領域、マルチクローニングサイト、大腸菌およびアグロバクテリウム・ツメファシエンスのための複製機能、ならびに選択マーカーおよびレポーター遺伝子を含む。
植物内で機能する多数のプロモーターが公知である。GPT導入遺伝子コンストラクトを構築する際、選択されるプロモーターは、恒常的な非特異的プロモーターであってもよく、例えばカリフラワーモザイクウイルス35Sリボソーマルプロモーター(CaMV 35Sプロモーター)であってもよい。このプロモーターは、植物内における導入遺伝子の発現のために広く使用されている。他の強力な恒常的プロモーターの具体例は、限定されないが、イネのアクチン1プロモーター、CaMV 19Sプロモーター、Tiプラスミドのノパリン合成酵素プロモーター、アルコールデヒドロゲナーゼプロモーターおよびスクロース合成酵素プロモーターを含む。
本発明は、シグナル代謝産物である2−ヒドロキシ−5−オキソプロリンの合成に直接的に機能する、グルタミンフェニルピルビン酸トランスアミナーゼ(GPT)酵素を含む植物を最初に開示する。これまで、機能が明らかになった植物のトランスアミナーゼは全く記載されていない。出願人は、いくつかの植物種および動物種由来のGPTポリヌクレオチドコード配列を同定し、試験した。そして、この遺伝子を宿主植物に組み込み、生育がより速く、葉のタンパク質量が向上され、そしてCO2固定速度がより速い、生育特性が顕著に向上されている異種性のトランスジェニック植物とすることに成功した。
同一性、を考慮すれば(実施例22を参照)、他の非植物GPTホモログは、本発明に係るトランスジェニック植物の生成に使用するためのGPT導入遺伝子の調製に使用され得る。配列番号25に示すシロイヌナズナの成熟タンパク質配列と(BLASTアラインメントにより)個々に比較された際、以下の成熟GPTタンパク質配列について、以下の配列同一性および相同性(BLAST「陽性」、類似アミノ酸を含む)が得られた。
好ましい実施形態において、転写終結を導き、mRNA転写産物のポリアデニル化を正常にさせるために、3’転写終結配列が導入遺伝子の下流に組み込まれる。好適な転写終結因子は、植物において機能することが知られている転写終結因子であり、限定されないが、アグロバクテリウム・ツメファシエンスのノパリン合成酵素(nopaline synthase:NOS)およびオクトピン合成酵素(octopine synthase:OCS)遺伝子、オクトピン
合成酵素遺伝子由来のT7転写産物、ジャガイモまたはトマト由来のプロテアーゼインヒビターIまたはII遺伝子の3’末端、CaMV 35Sターミネーター、tmlターミネーターおよびエンドウマメのrbcS E9ターミネーターを含む。さらに、天然の遺伝子における転写終結因子が使用され得る。ある実施形態では、実施例として後述するが、ノパリン合成酵素の転写終結因子が使用される。
選択マーカーは、形質転換体を選抜する手段を可能にするために、導入遺伝子の発現ベクターに通常含まれる。種々のタイプのマーカーが使用可能であるが、種々のネガティブセレクションマーカーが通常利用される。ネガティブセレクションマーカーは、形質転換されていない細胞を阻害し、または殺す選択剤に対する耐性を付与するマーカーを含み、例えば、抗生物質(例えばカナマイシン、ゲンタマイシン、アナマイシン(anamycin)、ハイグロマイシンおよびハイグロマイシンB)に対する耐性、または除草剤(例えばスルホニル尿素、グルホシネート(gulfosinate)、ホスフィノトリシンおよびグリフォセート)に対する耐性を付与する遺伝子を含む。スクリーニング可能なマーカーは、例えば、β−グルクロニダーゼをコードする遺伝子(Jefferson, 1987, Plant Mol. Biol. Rep 5: 387-405)、ルシフェラーゼをコードする遺伝子(Ow et al., 1986, Science 234: 856-859)およびアントシアニン色素の生産もしくは制御に関わるタンパク質をコードする種々の遺伝子(例えば米国特許第6,573,432号を参照)を含む。大腸菌のグルクロニダーゼ遺伝子(gus,gusAまたはuidA)は、植物の遺伝子導入において広く使用されてきた選択マーカーである。この大きな理由は、グルクロニダーゼ酵素の安定性、高い感受性および検出の容易性(例えば、蛍光定量的方法、分光光度的方法、種々の組織化学的方法)にある。さらに、最も高等な植物種には、検出可能なグルクロニダーゼが基本的に存在しない。
本発明における導入遺伝子の発現ベクターコンストラクトを、植物または植物細胞に導入するための種々の方法は、当業者によく知られており、あらゆる形質転換可能な植物または植物細胞が標的として利用され得る。
トランスジェニック植物は、標準的な方法を用いて選抜され、スクリーニングされ、そして同定され得る。本発明の好ましいトランスジェニック植物は、向上された生育および/または他の所望の農学的性質を示す1つ以上の表現型の特徴を提示し得る。トランスジェニック植物は、通常、形質転換体を選抜するために、選択圧下で再生され、その後にトランスジェニック植物生成が行なわれる。さらに、使用される選択圧は、所望の導入遺伝子発現コンストラクトまたはカセットの存在を確認するために、T0世代を超えて使用され得る。
本発明の様々な態様を、いくつかの実施例によりさらに記載および説明するが、いずれも本発明の範囲を限定するものではない。
シグナル代謝産物である2−オキソグルタラメートの合成に直接的に関与する植物酵素を見出す試みとして、出願人らは、推定される植物酵素が、2−オキソグルタラメートの合成に関与していると特徴づけられているヒトのタンパク質と、ある程度の構造上の関連性を有しているであろうとの仮説を立てた。ヒトのパンパク質であるグルタミントランスアミナーゼK(E.C.2.6.1.64)(文献では、システイン共役β―リアーゼ、キヌレニンアミノトランスフェラーゼ、グルタミンフェニルピルビン酸トランスアミナーゼ、等ともいわれている)は、ハロゲン化した生体異物のシステイン共役の処理に関与していることがわかっている(Perry et al., 1995, FEBS Letters 360:277-280)。ヒトのシステイン共役β―リアーゼは、窒素同化に関与する活性を有するよりも、むしろヒトおよび動物において解毒活性を有する(上述のCooper and Meister, 1977)。とはいえ、2−オキソグルタラメートの合成におけるこのタンパク質の潜在的な関与は重要であった。

これらのプライマーは、後述のトランスジェニック植物を作成するための発現ベクターでのサブクローニングを促進するため、ClaI(ATCGAT)およびKpnI(GGTACC)の制限酵素部位を含むように設計した。TaKaRa ExTaq DNAポリメラーゼ酵素を用いて、次の条件により高性能のPCRを行なった:まず94℃で4分間変性し、94℃で30秒間の変性、55℃で30秒間のアニーリング、および72℃で90秒の伸長反応を30サイクル行ない、最後に72℃で7分間伸長反応させる。増幅産物はClaIおよびKpnI制限酵素で切断し、アガロースゲル電気泳動法で単離して、カリフラワーモザイクウイルス(CaMV:cauliflower mosaic virus)35Sの構成的プロモーターおよびノパリンシンターゼ(NOS:nopaline synthase)3’ターミネーターを含むベクターpMon316(Rogers, et. al. 1987 Methods in Enzymology 153:253-277)にライゲーションした。ライゲーション産物をDH5α細胞に形質転換し、挿入されたことを確認するために形質転換体の配列を決定した。
上述の実施例1に記載したように、単離したcDNAによりコードされたタンパク質が、2−オキソグルタラメートの合成を触媒することが可能か否かを調べるため、当該cDNAを大腸菌で発現させて、2−オキソグルタラメートの合成能を標準的な方法で検定した。
簡潔にいえば、精製したタンパク質の産物を、150mM Tris−HCl(pH8.5)、1mM β−メルカプトエタノール、200mM グルタミン、100mM グリオキシル酸および200μM ピリドキサル5’−リン酸を含む反応液に加えた。試験するタンパク質を加えていない反応液をコントロールとして用いた。試験反応液およびコントロール反応液を37℃で20時間インキュベートし、遠心分離によって浄化して沈殿物を取り除いた。化学的に合成した標準の2−オキソグルタラメートを参照として、13CNMRを用いて、2−オキソグルタラメートの存在と量を分析するために上清を試験した。反応産物は2−オキソグルタラメートおよびグリシンであり、一方、基質(グルタミンおよびグリオキシル酸)は存在比が減少した。サイクリック2−オキソグルタラメートは、オープンチェーンのグルタミン前駆体から容易に区別できる、まったく異なるシグナルを生じさせた。
GPT活性に関する代替的なアッセイは、Calderon et al., 1985, J Bacteriol 161(2): 807-809の変性に続いて、HPLCを用いて2−オキソグルタラメート産物を測定した。簡潔にいえば、25mM Tris−HCl(pH8.5)、1mM EDTA、20μM FAD、10mM システインおよび〜1.5%(v/v)メルカプトエタノールを含む、変性抽出バッファーを用いた。検査物質からの組織サンプル(すなわち、植物組織)を約1/3の比率(w/v)で抽出バッファーに加え、37℃で30分間インキュベートし、200μMの20%TCAによって停止させた。約5分後、アッセイ溶液を遠心分離し、その上清を用いてHPLC(0.01N H2SO4の移動相、約0.2ml/minの流速、40℃、でのION−300 7.8mm ID X 30cm L columnを用いる)によって2−オキソグルタラメートを定量した。注入量は約20μlであり、保持時間はおよそ38分から39分である。検出は210nmのUV光で行なう。
本実験は、試験タンパク質が2−オキソグルタラメートの合成を触媒することができるということを証明した。ゆえに、これらのデータは、単離したcDNAが、植物において2−オキソグルタラメートの合成に直接的に関与するグルタミンフェニルピルビン酸トランスアミナーゼをコードするということを示す。よって、試験タンパク質はシロイヌナズナのグルタミンフェニルピルビン酸トランスアミナーゼ、すなわちGPTといえる。
(植物の発現ベクターpMON−PJUの生成)
簡潔に、植物の発現ベクターpMon316−PJUを以下のように作製した。単離したシロイヌナズナのGPTをコードするcDNA(実験例1)をpMon316ベクターのClaI−KpnIポリリンカー部位に入れて、クローニングした。これにより、GPT遺伝子は、カリフラワーモザイクウイルス(CaMV)35Sプロモーターおよびノパリンシンターゼ(NOS)転写ターミネーターの制御下におかれる。選択マーカーを提供するためにカナマイシン耐性遺伝子を加えた。
pMON−PJUおよびコントロールベクターpMon316(挿入したDNAなし)は標準的なエレクトロポレーション法(McCormac et al., 1998, Molecular Biotechnology 9:155-159)を用いてアグロバクテリウム・ツメファシエンス株のpTiTT37ASEに形質転換し、続いて抗生物質スペクチノマイシン(100μg/ml)およびカナマイシン(50μg/ml)を含んだLBプレートに塗り広げた。アグロバクテリウムの抗生物質耐性コロニーをPCRによって試験し、プラスミドを含むことを確認した。
無菌の葉の切片を、Murashige&Skoog(M&S)培地上でカルスに発達させ、そこから形質転換小植物体が出現した。そして、これらの小植物体を発根可能な選択培地(選択試薬としてカナマイシンを含むM&S培地)に移植した。その後、正常で、発根した、形質転換したタバコの小植物体を土壌に移植し、成熟期まで生育させて、花が咲いた植物を自家受精させて、得られた種子を収穫した。成長期の間、植物の成長表現型を調べ、多くの若いトランスジェニック植物でCO2固定速度を測定した。
トランスジェニックタバコ植物のT0世代から収穫した種子を、カナマイシン(100mg/L)を含むM&S培地で発芽させて、導入遺伝子の質を高めた。種子のうち少なくとも1/4がこの培地で発芽せず(カナマイシンは、通常の遺伝子分離の結果もたらされ得る、耐性のない種子の発芽を阻害すると期待される)、残りの種子のうち半分以上がカナマイシンに対して感受性(ごく弱い)を示したため取り去られた。
収穫されたトランスジェニック植物(GPT形質転換体およびベクターコントロール形質転換体の両方)を根および葉のグルタミンシンセターゼ活性、乾燥させていない植物の総量、根および葉の総タンパク質ならびにCO2固定速度(Knight et al., 1988, PlantPhysiol. 88: 333)を分析した。形質転換していない野生型A.ツメファシエンス植物
もベースラインコントロールを設けるため、同じパラメータで分析した。
シロイヌナズナGPT遺伝子を持つトランスジェニックトマト(Lycopersicon esculentum)(マイクロトムトマト)植物を、実施例3で記載したベクターおよび方法を用いて作成した。T0トランスジェニックトマト植物を作成し、成熟期まで生育させた。トランスジェニックトマト植物の初期生育特性のデータを表2に示す。トランスジェニック植物は野生型のコントロール植物と比較して、成長速度、開花および種子の収穫量の有意な促進を示した。さらに、トランスジェニック植物は複数の主茎を発達させた、一方、野生型の植物は単数の主茎を発達させた。野生型の植物と比較したGPTトランスジェニックトマト植物の写真を図3に示す。
本実施例では、オオムギのGPTの推定コード配列を、植物体への一過性発現アッセイによって導入遺伝子コンストラクトから単離し、発現させた。生物学的に活性な組換えオオムギGPTが生成され、HPLCで確認したところ、2−オキソグルタラメートの増加した合成を触媒した。
OD650)の懸濁液を、急速に成長するタバコ葉へ注入することから成る。皮内注射は有意な量の葉の表面がアグロバクテリウムにさらされることを確実にするため、葉の表面にわたる格子内で行った。植物を3−5日間生育させて、組織を他の全ての組織抽出で説明したように抽出し、GPT活性を測定した。
本実施例では、イネのGPTの推定コード配列を単離し、大腸菌で発現させた。生物学的に活性な組換えイネGPTが作成され、HPLCで確認したところ、2−オキソグルタラメートの増加した合成を触媒した。
(イネGPTコード配列と大腸菌における発現)
イネ(Oryza sativa)のGPTコード配列を決定し、合成して、PET28ベクターに導入し、大腸菌で発現させた。つまり、大腸菌細胞を発現ベクターにより形質転換し、LB培養液で一晩培養した形質転換体を希釈し、OD0.4まで成長させて、イソプロピル−β−D−チオガラクトシド(0.4μM)で誘導して発現させ、3時間培養して集菌した。そして、合計25×106個の細胞の生物学的活性に関して、以下のNMRアッセイによって分析した。形質転換していない野生型の大腸菌細胞をコントロールとして分析した。追加のコントロールには、空のベクターで形質転換した大腸菌細胞を用いた。
Calderon et al., 1985, J Bacteriol 161(2): 807-809の変性に続いて、HPLCを用いてGPTを過剰発現している大腸菌細胞における2−オキソグルタラメートの産生量を測定した。つまり、25mM Tris−HCl(pH8.5)、1mM EDTA、20μM ピリドキサルリン酸、10mM システインおよび〜1.5%(v/v)メルカプトエタノールを含む変性抽出バッファーを用いた。サンプル(大腸菌細胞25×106個の溶菌液)を抽出バッファーに約1/3比率(w/v)で加え、37℃で30分間インキュベートし、200μMの20%TCAで停止した。約5分後、アッセイ溶液を遠心分離し、上清を用いて、0.01N H2SO4の移動相、流速約0.2ml/分、40℃においてION−300 7.8mm ID X 30cm L columnを用いたHPLCにより、2−オキソグルタラメートを定量した。注入量は約20μlであり、保持時間はおよそ38分から39分である。検出は210nmのUV光で行なった。
配列番号5のイネGPTコード配列の発現は、2−オキソグルタラメート合成触媒の生物学的活性を持つ組換えGPTタンパク質の過剰発現という結果になった。特に、1.72nmolの2−オキソグルタラメートの活性が組換えイネGPTを過剰発現している大腸菌細胞においてみられ、コントロールの大腸菌細胞における、わずか0.02nmolの2−オキソグルタラメートの活性に比べて、86倍の活性レベルの増加がみられた。
本実施例では、ダイズのGPTの推定コード配列を単離し、大腸菌で発現させた。生物学的に活性な組換えダイズGPTが生成され、HPLCで確認したところ、2−オキソグルタラメートの増加した合成を触媒した。
(ダイズGPTコード配列と大腸菌における発現)
ダイズ(Glycine max)のGPTコード配列を決定し、合成して、PET28ベクターに導入し、大腸菌で発現させた。つまり、大腸菌細胞を発現ベクターにより形質転換し、LB培養液で一晩培養した形質転換体を希釈し、OD0.4まで培養して、イソプロピル−β−D−チオガラクシド(0.4μM)で誘導して発現させ、3時間培養して、集菌した。合計25×106個の細胞により、下記のHPLCアッセイを用いて生物学的活性を分析した。形質転換していない野生型の大腸菌をコントロールとして分析した。追加のコントロールには、空のベクターを形質転換した大腸菌細胞を用いた。
上記の実施例20に記載したように、GPTを過剰発現している大腸菌細胞における2−オキソグルタラメートの生成を判定するためにHPLCを用いた。
配列番号7のダイズGPTコード配列の発現は2−オキソグルタラメート合成触媒の生物学的に活性な組換えGPTタンパク質の過剰発現という結果になった。特に、31.9nmolの2−オキソグルタラメートの活性が組換えダイズGPTを過剰発現している大腸菌細胞においてみられ、コントロールの大腸菌細胞における、わずか0.02nmolの2−オキソグルタラメートの活性に比べて、1600倍近くの活性レベルの増加がみられた。
本実施例では、ゼブラフィッシュのGPTの推定コード配列を単離し、大腸菌で発現させた。生物学的活性のある組換えゼブラフィッシュGPTが作成され、HPLCで確認したところ、2−オキソグルタラメートの増加した合成を触媒した。
(ゼブラフィッシュGPTコード配列および大腸菌における発現)
ゼブラフィッシュ(Danio rerio)のGPTコード配列を決定し、合成して、PET28ベクターに導入し、大腸菌で発現させた。つまり、大腸菌細胞を発現ベクターにより形質転換し、LB培養液で一晩培養した形質転換体を希釈し、OD0.4まで培養して、イソプロピル−β−D−チオガラクシド(0.4μM)で誘導して発現させ、3時間培養して、集菌した。合計25×106個の細胞により、下記のHPLCアッセイを用いて生物学的活性を分析した。形質転換していない野生型の大腸菌をコントロールとして分析した。追加のコントロールには、空のベクターで形質転換した大腸菌細胞を用いた。
上記の実施例6に記載したように、GPTを過剰発現している大腸菌細胞における2−オキソグルタラメートの生成を判定するためにHPLCを用いた。
配列番号16のゼブラフィッシュGPTコード配列の発現は2−オキソグルタラメート合成触媒の生物学的活性を持つ組換えGPTタンパク質の過剰発現という結果になった。特に、28.6nmolの2−オキソグルタラメートの活性が組換えゼブラフィッシュGPTを過剰発現している大腸菌細胞においてみられ、コントロールの大腸菌細胞における、わずか0.02nmolの2−オキソグルタラメートの活性に比べて、1400倍以上の活性レベルの増加がみられた。
この実施例では、シロイヌナズナのGPTの推定コード配列の異なる2つの断片を構築し、推定される葉緑体のシグナルペプチドが欠失しているまたはトランケートされたGPTタンパク質の活性を評価するために、大腸菌で発現させた。最初の30個のアミノ末端のアミノ酸残基または最初の45個のアミノ末端のアミノ酸残基を欠失するようにトランケートされた、配列番号2のシロイヌナズナGPTの全長アミノ酸配列に対応する組換え断片GPTタンパク質は、無事に発現し、HPLCで確認したところ、2−オキソグルタラメートの増加した合成を触媒する生物学的活性を示した。
(トランケートされたシロイヌナズナGPTコード配列と大腸菌における発現)
配列番号1のシロイヌナズナ(Arabidopsis thaliana)のGPTコード配列の断片のDNAコード配列を構築し、合成して、PET28ベクターに導入し、大腸菌で発現させた。本実施例で用いたトランケートされたシロイヌナズナGPTコード配列のDNA配列を配列番号15(−45AAコンストラクト)に示し、対応するトランケートされたGPTタンパク質のアミノ酸配列を配列番号16に示す。つまり、大腸菌細胞を発現ベクターにより形質転換し、LB培養液で一晩培養した形質転換体を希釈し、OD0.4まで培養して、イソプロピル−β−D−チオガラクシド(0.4μM)で誘導して発現させ、3時間培養して、集菌した。合計25×106個の細胞により、実施例20に記載したように、HPLCアッセイを用いて生物学的活性を分析した。形質転換していない野生型の大腸菌をコントロールとして分析した。追加のコントロールには、空のベクターで形質転換した大腸菌細胞を用いた。
Claims (22)
- 植物プロモーターに対して操作可能に結合されているGPT導入遺伝子を含んでいる、トランスジェニック植物。
- 上記GPT導入遺伝子は、
配列番号2、配列番号4、配列番号10、配列番号14、配列番号16、配列番号19、配列番号25、配列番号26、配列番号27、配列番号28、配列番号29、配列番号30および配列番号31のアミノ酸配列からなる群から選択されるアミノ酸配列を有しているポリペプチドをコードする、請求項1に記載のトランスジェニック植物。 - 上記GPT導入遺伝子は植物のゲノムに取り込まれている、請求項1に記載のトランスジェニック植物。
- 単子葉植物である、請求項3に記載のトランスジェニック植物。
- 双子葉植物である、請求項3に記載のトランスジェニック植物。
- 請求項3に記載のトランスジェニック植物の子孫であって、
上記GPT導入遺伝子を含んでいる、子孫。 - 請求項3に記載のトランスジェニック植物の種子であって、
上記GPT導入遺伝子を含んでいる、種子。 - 類似の野生型植物または非形質転換植物と比較して増強された成長速度、増大されたバ
イオマス収量、増大された種子の収量、増大された花または花芽の収量、増大された果実またはさやの収量、大型の葉、増大されたGPT活性、増大されたGS活性、増大された2−オキソグルタラメートのレベル、向上された窒素利用効率、および/または塩または塩分に対する増強された耐性を示す、請求項3に記載のトランスジェニック植物。 - 類似の野生型植物または非形質転換植物と比較して植物の成長特性を向上させる方法であって、
(a)植物にGPT導入遺伝子を導入する工程と、
(b)上記植物、または当該植物の子孫においてGPT導入遺伝子を発現させる工程と、
(c)GPT導入遺伝子を含まない同一種の植物と比較して増大された成長特性を有する植物を選抜する工程と、
を包含する方法。 - 上記増大された成長特性は、増大されたバイオマス、早期の開花、早期の発芽、増大された草高、増大された開花、増大された発芽、より大きな葉、増大された果実またはさやの収率、および増大された種子の収率からなる群から選択される、請求項9に記載の方法。
- 類似の野生型植物または非形質転換植物と比較して植物の窒素利用効率を向上させる方法であって、
(a)植物にGPT導入遺伝子を導入する工程と、
(b)上記植物、または当該植物の子孫においてGPT導入遺伝子を発現させる工程と、
(c)GPT導入遺伝子を含まない同一種の植物と比較して増大された窒素利用効率を有する植物を選抜する工程と、
を包含する方法。 - 類似の野生型植物または非形質転換植物によって産生された種子と比較して、塩または塩分を含んだ環境において植物種子の発芽または成長の耐性を向上させる方法であって、
(a)植物にGPT導入遺伝子を導入する工程と、
(b)植物または当該植物の子孫においてGPT導入遺伝子を発現させる工程と、
(c)GPT導入遺伝子を含まない同一種の植物と比較して増強された成長特性を有する植物を選抜する工程と、
(d)上記植物から種子を収穫し、塩分の高い環境において向上された発芽を示す種子を選抜する工程と、
を包含する方法。 - 選抜した種子から植物を繁殖させて、当該植物から種子を収穫する工程をさらに含む、請求項12に記載の方法。
- 配列番号2、配列番号4、配列番号10、配列番号14、配列番号16、配列番号19、配列番号25、配列番号26、配列番号27、配列番号28、配列番号29、配列番号30および配列番号31のアミノ酸配列からなる群から選択されるアミノ酸配列を有しているポリペプチドをコードする、単離されたポリヌクレオチドと操作可能に結合された植物プロモーターを含む、核酸コンストラクト。
