Abstract
Description
「核酸」とは、4成分からなる一本鎖の形態または4成分からなる二本鎖へリックスの形態で、リボヌクレオシド(アデノシン、グアノシン、ウリジンまたはシチジン;「RNA分子」)の、またはデオキシリボヌクレオシド(デオキシアデノシン、デオキシグアノシン、デオキシチミジンまたはデオキシシチジン;「DNA分子」)のリン酸エステルで構成される高分子体を意味する。可能性のある4成分からなる二本鎖ヘリックスとしては、DNA-DNA、DNA-RNA、およびRNA-RNAが挙げられる。核酸、具体的にはDNA分子またはRNA分子という用語は単に分子の一次構造または二次構造のみを指し、特定の三次形態に限定されない。したがって、この用語は、4成分からなる二本鎖のDNAを含み、このようなDNAは特に、直鎖または環状DNA分子(例えば制限断片)、ウイルス、プラスミド、および染色体で見出される。本明細書では、特定の4成分からなる二本鎖DNA分子の構造について述べる場合、その配列は、DNAの非転写鎖(すなわちmRNAに相同な配列を有する鎖)に沿って5'から3'の方向でのみ配列を示す一般的な慣例に従って記載することができる。

ベクターCA-LTR-SHIVKU2は、サルのゲノムと、SIVのLTRを切り出しCAEVのLTRで置き換えたヒト免疫不全ウイルス(SHIV)とを含む。SHIVは、SIV-mac239のゲノムからなるキメラゲノムを含み、SIVのtat、env、およびrev遺伝子が切り出され、HIV-1のvpu、tat、env、およびrev遺伝子で置き換えられている。それゆえにこのベクターは、CAEVの5'および3'におけるLTRの転写制御下に、SIVのvpr、vpx、gag、pol、vif、およびnef遺伝子と、HIV-1のtat、rev、vpu、およびenv遺伝子とを有する。pol遺伝子は、インテグラーゼ(in)をコードする配列から切り出された。インテグラーゼをコードする配列から切り出されていない接種用ベクターpCA-LTR-SHIVKU2は、配列番号16の配列(図15)からなる。インテグラーゼをコードする配列から切り出されたベクターpCA-LTR-SHIVKU2-IN-は、配列番号17(図16)からなる。
プラスミドpSHIVKU2およびpΔ4SHIVKU2は、対照として用いられるプラスミドである。その構築は多くの出版物で説明されている(Liu ZQら、2006、Ramakrisna Hegdeら、2005)。
1.2.1.細菌培養
上記プラスミドを含む大腸菌(E.coli)K12(JM109)細菌を0.05mg/mlのカナマイシンを含むBL培地5ml中で予備培養し、続いて150rpmで撹拌しながら30℃で一晩インキュベートする。予備培養物からの細菌懸濁液をBL培地で1:100,000に希釈し、続いてこの希釈液50μlをペトリ皿に入れた寒天/BL/カナマイシンの表面上に塗り広げ、続いてこれを32℃で一晩インキュベートした。ペトリ皿の寒天上に生じた孤立したコロニーを0.05mg/mlのカナマイシンを含む5mlのBL液状培地に植え付け、150rpmで撹拌しながら、30℃で一晩培養する。少量の培養液(1ml)をMacherey-NagelまたはQiagenのミニプレップ(Mini-prep)キットを用いたDNAの迅速抽出に推奨された手順に従って使用し、続いて1%アガロースゲルで抽出されたDNAを分離してその品質をチェックする。成功と推定されるDNAに対応する細菌を用いて、1Lの培養液に植え付け、これを初期の条件と同じ条件下で培養して、DNAをマキシプレップ(maxi-preparation)で単離する。
30℃で一晩撹拌しながら(150rpm)培養された細菌を遠心分離(4000g、4℃、15分)により堆積物として回収し、この堆積物を8mlの再懸濁緩衝液(50mMのトリス-HCl、pH8、10mMのEDTA)に再懸濁する。次に8mlのアルカリ性溶解緩衝液(200mMのNaOH、1%SDS)を添加することによって細胞を溶解させて、プラスミドDNAを放出させる。8mlの中和緩衝液(3Mの酢酸カリウム、pH5.5)を添加することによって溶解産物を中和する。続いてこの混合物を氷中で5分インキュベートし、15,000g、4℃で15分遠心分離する。このDNAを含む溶液を前もって平衡化したカラムに移し、そのままプラスミドDNAを保持させる。カラムを洗浄緩衝液で3回洗浄し、DNAを溶出させて、イソプロパノールで沈殿させる。遠心分離(30分、15,000g、4℃)により沈殿したDNA堆積物を得る。続いてDNAを2mlの70%エタノールで洗浄し、4℃で、15,000gで10分遠心分離することにより過量の不純物と塩を除去し、続いて堆積物を乾燥させ、適切な体積の超純水に再懸濁する。
0.5μgのプラスミドのアリコート画分を、2ユニットの酵素EcoR1、BamH1またはSph1で、各酵素に適した1×緩衝液中で、20μlの最終容量で、37℃で60分消化することによって処理する。エチジウムブロマイド(ETB)とUV下でのゲル観察により明らかにしたTAE1×緩衝液中の1%アガロースゲルでの電気泳動の移動度により、消化のプロファイルをチェックした。
1.3.1.細胞モデルおよび培養条件
細胞系を、アメリカ合衆国のNational Institute of Health AIDS Research and Reference Reagent Programから得た。その細胞を10%ジメチルスルホキシド(DMSO)中で液体窒素中-170℃で凍結保存する。それらを解凍し、培養フラスコで培養する。
プラスミドpCA-LTR-SHIVKU2-IN-またはpSHIVKU2でのHEK293Tのトランスフェクション
用いられるトランスフェクション法は、ExGen500を用いた方法である。ExGen500(Euromedex、France)は、直鎖ポリエチレンイミンをベースとするカチオン性ポリマーからなる。このポリマーは、イオン結合を介してDNAと複合体を形成させる極めて大きいカチオン電荷密度を有する。次にこのExGen500/DNA複合体は、通常はアニオン性の細胞の細胞質膜との相互作用(硫酸プロテオグリカンを介した相互作用)が可能になる。続いて細胞による複合体のエンドサイトーシスが起こり、加えてそれらはエンドソーム/リソソームに輸送される。ExGen500は、その酸性pHでのプロトン付加能力により酸性小胞の培地を緩衝化し、それによってトランスフェクションされたDNAの分解を防ぐことができる。またこの特性は、DNAを細胞の細胞質に放出させる浸透圧性ショックも引き起こす。続いてExGen500は細胞核へのDNAの輸送を促進し、細胞質ヌクレアーゼによるその分解を回避する。
pCA-LTR-SHIVKU2-IN-またはpSHIVKU2のDNAでトランスフェクションされたHEK-293TをCEMx174と共に培養したところ、その48時後に、CEMx174は、CEMx174が統合して融合細胞が形成されることに起因するECP形成によって示される感染の徴候を示す。感染したCEMx174を、ウイルスを増幅するための新しいCEMx174を含む新しいフラスコに移し、増幅されたウイルスは上清で回収する。
48時間後に注射器を用いてウイルスストックを回収し、続いてそれらを直径0.22μmのフィルターに通過させて死細胞片を除去し、その後、チューブに入れて-80℃で保存する。
細胞変性の開発により、またはマーカー遺伝子発現の検出によりウイルスの感染力(infectiousity)を評価するために、ウイルス上清のアリコート画分(10〜100μl)を用いて標的細胞に植え付ける。
ウイルスを含む上清を、10回の工程で培地で希釈することにより(連続希釈)10-1〜10-6希釈液を得て、これを用いて、24-ウェルプレートで、0.5〜1mlのRPMI培地を含む1ウェルあたり1×105個の細胞を含む4連のウェルに植え付ける。このようにして植え付けられた細胞を37℃で5%CO2と共にインキュベートし、培地を3日毎に交換しながら維持する。この細胞を定期的にECPの発生について観察する。
接種用のゲノムpCA-LTR-SHIVKU2-IN-によって生産されたタンパク質がウイルス粒子に集合したかどうかを試験する目的で、本発明者らは、電子顕微鏡検査(EM)による形態学的な研究を行った。
1.5.1.NOD/SCIDマウスのヒト化および接種
6週齢のマウスに、ガンマ線を120センチグレイの線量で50秒照射する。
クエン酸ナトリウム上の全ての血液サンプルを遠心分離し(2000g、10分、20℃)、血漿と赤血球との間の白血球の層を回収する。細胞をPBS/EDTAで3倍に希釈し、慎重にフィコールのクッション(リンパ球を分離するための培地)上に載せ、続いて20℃で、2,000gで45分遠心分離する。PBMCを回収し、PBS/EDTAで数回洗浄し、PBS×1に再懸濁し、続いて各マウスに0.1ml中50×106個のPBMCを腹膜内経路で注入する。
ヒト化の48〜72時間後に、SCIDヒト化細胞を筋肉内経路(IM)でpCA-LTR-SHIVKU2-IN-、pSHIVKU2またはpΔ4SHIVKU2のDNA(50μg)と共に注入する。6〜8週齢のBALB/cマウスにそれぞれのDNA(100μg)をIM注射することによって直接免疫化する。
アブノバ(Abnova)サンドイッチELISA試験:
この試験は、96-ウェルプレートのウェルの底に結合させた、ウイルス抗原に対する抗体の検出に基づく。試験される血清(免疫化後の異なる時間で回収される)、陽性対照、および陰性対照をウェルに入れる。抗HIV Ac(抗体)が存在する場合、それがウイルス抗原に結合することができる。過量の非特異的な結合を除くためにウェルを数回洗浄した後、HPRO酵素に連結したストレプトアビジンと相互作用するビオチン分子を有する第二の検出のためにAcを添加する。続いて、着色反応を引き起こす酵素の基質TMBを添加することによって形成された抗原/Ac複合体を検出する。着色は、ELISAリーダー光度計での読み出しによる光学密度で表す。
この技術は、ウイルス感受性細胞の感染を阻害するAc(セロ-N(sero-N))を血清中和する能力に基づく。血清中のセロ-N Acの検出と評価のために、一定量の感染性ウイルスを試験される血清の連続希釈液と接触させ、続いてこの混合物をマイクロプレート上で許容細胞の培養物に植え付け、3〜5日インキュベートする。ほとんどの場合、ウイルスは細胞変性株であるため、ECPの非存在またはECP数の減少は、試験される血清中にAcが存在することを示す。
この試験の目的は、抗原性刺激に特異的な応答としてIFN-γを分泌するTリンパ球(TL)の比率を検出し評価することである。
1.6.1.末梢単核細胞の単離
ヒト末梢血単核細胞を上記で示したように調製する。培養とサイトメトリー試験のために、上述の手順に従って単離したマウス脾臓の脾細胞も、血清を含まないAIM V培地に再懸濁した。
抗原特異的細胞が、増殖してサイトカインと溶解分子を生産することができるかどうかを試験するために、脾細胞とPBMCを、37℃で10分、CFSE(1μg/ml)で標識し、続いて過量分を取り除くために細胞を1×PBSで洗浄する。標識した細胞を、96-ウェルプレートのディープウェルに、1mlのAIM V培地中、2×106/ウェルの量で撒き、続いて、抗CD49および抗CD28同時刺激Acの存在下で、ペプチドの様々なプール(Gag、Env、およびTat+Rev+Nef)で2μg/mlの量で刺激する。陰性対照として、ペプチドを含まない細胞を用い、陽性対照として、フィトヘムアグルチニン(PHA)を2μg/mlで添加された細胞を添加する。細胞を5日間培養し(37℃、加湿)、続いて同じペプチドのプールで6時間再度刺激した後、それらを標識する。細胞を遠心分離(2,000G、5分、4℃)で回収し、100μlのPBSに再懸濁し、まず、表面Ac(CD3、CD4、およびCD8)[パシフィックブルー(Pacific Blue)抗ヒトCD3(5μl)、PE抗ヒトCD4(10μl)、およびAPC/Cy7抗ヒトCD8(10μl)]で、室温で30分標識する。続いて細胞を遠心分離し、1×PBSで洗浄し、続いて100μlのBDのサイトフィックスサイトパーム(Cytofix Cytoperm)中に固定して透過性にする。続いて細胞質の標識のために、細胞を、「抗ヒトIFN-γ PE-Cy7」および「アレクサフルオロ(Alexa Fluor)647抗ヒトグランザイムA(Granzyme A)」Ac(5μl)と共に4℃で20分インキュベートする。最後に細胞をPBSで洗浄し、4%PFAで固定し、その後フローサイトメーターでの獲得および解析を行う。
BDのFACSDiva6ソフトウェアパッケージに連結したBDのLSRIIフローサイトメーターを使用した。この機器は、最大13種の蛍光パラメーターと2種の物理的パラメーター、すなわちFSCのサイズ(前方散乱)および複雑さ;または粒子のSSC(側方散乱)の測定を可能にする。この機器は、3つのレーザーを備えている。488nmで放射される青色のレーザーは、数種の蛍光色素(FITC、PE、PE-Cy7)を独立して励起することができる。633nmで放射される赤色のレーザーは、APCおよびAPC-Cy7蛍光色素を励起することができ、最後に405nmで放射される紫色のレーザーは、パシフィックブルー蛍光色素を励起することができる。
この研究には、合計12匹のマカク(カニクイザル)を用いる。対照グループは6匹のマカクで構成され、接種グループは残りの6匹のマカクで構成される。筋肉内経路を介した1回のDNAの二重注射(動物1匹あたり4mg)およびエレクトロポレーション(EP)で(動物1匹あたり1mg)動物を免疫化した。
2.1.pCA-LTR-SHIVKU2-IN-プラスミド構築の定性的および定量的チェック
2.1.1.pCA-LTR-SHIVKU2-IN-プラスミドの存在
大量のBL液状培地に撒くために用いられるBL培地での予備培養物から得られたポリクローナル細菌培養からプラスミドDNAを単離したら、約2000bpのエピソームがかなりの割合で観察される。また電気泳動プロファイルからも、プラスミドに相当する約14,000bpの単一のDNAバンドの存在(論理上は13,739bp)が示される。
プラスミドのEcoR1での消化プロファイルから、両端にEcoRI部位を有する断片からの約5,000bpと7,500bpの2つのバンド、両端にBamH1部位を有する断片から生じた2400bp、4,900bp、および7,400bpのBamH1による3つのバンド、および1つのSph1部位を有する断片から生じた10,000bpに位置するSph1によるバンドの存在が明らかになる。さらにBamH1消化による追加の2,400bpのバンドも観察されたが、これはエピソームに起因するものと思われる。
HEK293T細胞を、GFPを発現する対照プラスミドpCG-GFP、プラスミドpSHIVKU2、さらにプラスミドpCA-LTR-SHIVKU2-IN-でトランスフェクションした。プラスミドpCG-GFPでトランスフェクションされた細胞から、GFP+細胞の数を評価することによりトランスフェクション効率の推測が可能になった。
次いでHEK293T細胞中で、接種用pCA-LTR-SHIVKU2-IN-ゲノムによって生産されたタンパク質が適切にウイルス粒子に集合したかどうかを決定した。プラスミドpCA-LTR-SHIVKU2-IN-でトランスフェクションされたHEK293T細胞の形態をEMで試験した。
2.4.1.pCA-LTR-SHIVKU2-IN-が接種されたSCIDヒト化マウスにおける体液性応答の評価
ウイルスの抗原に特異的に結合するAcの存在について、免疫化の約1ヶ月後に採取された免疫化マウスの血清サンプルを市販のELISA試験を用いて試験する。Table 1(表1)(以下)にこの解析の結果を要約する。これらの結果から、接種用pCA-LTR-SHIVKU2-IN-DNAで免疫化されたマウスからのサンプルのうち約半分では、SHIVKU2のDNAで免疫化されたマウスも同様に、陽性の比率は低く(それぞれ44%および37%)、これは、pΔ4SHIVKU2で免疫化されたマウスの陽性の比率(50%)とは異なることが示される。これらの結果から、NOD/SCIDヒト化マウスにおいて接種用pCA-LTR-SHIVKU2-IN-DNAの体液性応答を誘導する能力が実証される。pΔ4SHIVKU2で免疫化されたマウスの3つの血清サンプルのうち1つのOD値は、pCA-LTR-SHIVKU2-IN-で免疫化されたマウスおよびpSHIVKU2で免疫化されたマウスの血清サンプルで得られたOD値よりも大きい。このプラスミドは複製能がなく、ウイルスのタンパク質はトランスフェクションされた細胞の膜に結合したままであるため、Acをほとんど誘導しないと予想される。
選択されたプラスミドpCA-LTR-SHIVKU2-IN-またはSHIVKU2が接種されたSCIDヒト化マウスの血清は、そのODがELISAにより陽性であることが見出されたものである。血清が少量であるため、ELISA試験で強いOD値を示した所定のサンプルを血清中和で試験することができなかった。試験の信頼性を保証するために、陰性であることが判明したpCA-LTR-SHIVKU2-IN-が接種されたSCIDヒト化マウスの血清サンプルもELISA試験に用いた。
BALB/cマウスにおけるpCA-LTR-SHIVKU2-IN-ワクチンの免疫原性を研究するために、動物に筋肉内経路を介して100μgのDNAを単回投与で注入した。ELISPOTで抗原特異的脾臓細胞(脾細胞)の比率を試験した。この研究結果から、実際に用いられたDNAが研究される全ての抗原に対して特異的な免疫反応を誘導する能力が示される(図12)。プラスミドpΔ4SHIVKU2によるT細胞応答は、プラスミドpCA-LTR-SHIVKU2-IN-を用いた場合よりも2倍高い。この両者のDNA間の小さい効率の差は、pΔ4SHIVKU2のDNAの品質がpCA-LTRSHIVKU2-IN-のDNAの品質よりも優れていることに関連する可能性がある。それでもこれらの結果から、正常なマウスにおいてこれらのワクチンにより誘導された基底レベルの免疫反応を決定することができ、さらにSCIDヒト化マウスで得られた免疫反応との比較を行うことができる。
50μgのCA-LTR-SHIVKU2-IN-、Δ4SHIVKU2またはSHIVKU2のDNAを単回投与で筋肉内注射することによりNOD/SCIDヒト化マウスを免疫化し、続いて脾細胞を用いてELISPOTにより免疫反応を試験し、抗原特異的細胞とIFN-γ産生細胞との比率を評価した。図13で示されるように、Gag、EnvまたはTat+Rev+Nef抗原での刺激に応答してヒトINF-γをSIVペプチドまたはHIVペプチドの形態で生産する相当数のT細胞が見出される。興味深いことに、pCA-LTR-SHIVKU2-IN-での免疫化後に得られた応答は、pSHIVKU2で得られた応答にある程度類似しており、これらの応答はいずれも、pΔ4SHIVKU2での免疫化後に得られた応答よりも明らかに大きいことに注目すべきである。GagおよびTat+Rev+Nef抗原に対してpCA-LTR-SHIVKU2により誘導された応答が優勢である。
2.5.1.ゼロ日目に行われる(マウス脾臓を回収する日に行われる)単一標識
選択された抗体が、実際にそれらの標的を検出するかどうかを試験するために、一方でSCIDヒト化マウスの脾臓におけるヒト細胞の存在と比率を評価するために、これらの単一標識が用いられる。
ウイルス抗原特異的T細胞の表現型および機能を試験するために、CFSEで標識した細胞を(Gag、EnvおよびTat+Rev+Nef)ペプチドと共に5日間インキュベートし、続いて同じペプチドで再度、6時間刺激する。続いて上記で示したように細胞をAcで標識して試験する。
2.6.1.ELISPOT IFN-λ試験によるT細胞の免疫反応の解析
採取された血液サンプルから単離された単核細胞画分を用い、市販のキットを用いてウイルスGag、Pol、Env、およびTat+Rev+Nef抗原による刺激に応答してIFN-λを分泌する細胞の比率を評価する。table 3(表3)に最初の20週間の解析結果を示す。この表から明らかに、接種用ベクターCAL-SHIV-IN-の1回の投与後、全ての動物が、IFN-λを分泌する抗原特異的細胞を特徴とする細胞性免疫反応を起こしたことが示される。これらの応答は、互いに異なるCMH-1ハプロタイプを有する動物ごとに異なる。免疫反応は、免疫化後(PI)2〜4週間で第一の一次応答ピークの存在と、PI後8〜10週間でそれに続く応答とを特徴とし、ここでは第二の免疫化は行われない。大変興味深いことに、第二のピークの強度はしばしば、特にIFN-λを分泌する細胞の数が3倍のBX80動物で、第一のピークの強度よりも大きいことに注目すべきである。
マルチパラメータのフローサイトメトリーによる解析の結果から、全ての動物が増殖したT細胞による応答を起こし、この応答は接種用ベクターによって発現された全ての抗原に特異的であることを解明することにより、ELISPOTにより得られた結果が裏付けられる(Table 4(表4))。これらの応答は動物間で異なっており、免疫後約8週まで及ぶ最初の一次応答期、続いて収縮期(2〜4週間)、続いて再出現期があることがわかる。52週目に試験ウイルスSIVmac251を用いた毒性試験を行うまで、このマルチパラメータのフローサイトメトリーによる免疫反応の長期追跡を継続した。
HIV-1の抗Env抗体を検出することができる市販のELISAキットを用いてSHIVの抗抗原抗体の検出を行った。各血液サンプリング時に回収された血清の長期試験によれば、抗Env抗体の存在は、BX80動物では免疫後20週目から、BX73動物では8週目から示された。SHIVのタンパク質に対するウェスタンブロットによって陽性血清中の抗体の存在を確認したところ、Gag-p27タンパク質に対する強いシグナル、加えてgp160/gp120糖タンパク質に対するシグナルが示された。
これらの結果から明らかに、1回の接種用DNA(CAL-SHIV-IN-)の注入で、T細胞と体液性(抗体)免疫反応を誘導できることが実証される。T細胞応答は、接種用ベクターによって発現される全てのウイルス抗原に対する。この応答は持続的であり、従来の増殖、収縮、およびメモリースキームに従っている。表現型解析により、セントラルタイプのメモリーT細胞とメモリーエフェクター細胞の存在を確認した。
ベクターCAL-SHIVKU2-IN-のゲノムから、Nar1酵素とSph1酵素で二重に消化することによりSIVのgag、pol、vif、vpx、およびvpr遺伝子を切り出し、Nru1酵素で部分的に消化しNot1酵素で全体を消化することによりSIVのnef遺伝子を切り出した。次に残りの断片を精製した。この断片は、CAEVのRTLに挟まれており、プラスミドpETにより輸送されるHIVのtat、rev、およびenv遺伝子が結合している断片であるが、これを使用して、HIV-1のgag、pol、vif、vpx、およびvpr遺伝子が結合している5kbの断片とHIV-1のnef遺伝子が結合している620bpの断片とを導入し、ベクターCAL-HIV-IN-を作製した(図17)。インテグラーゼをコードする配列から切り出されたCAL-HIV-IN-ベクターは、配列番号18の配列からなる。
このベクターのDNAを、ExGenと製造元推奨の手順を用いてトランスフェクションすることによってGHOST-CXCR4およびHEK-293T細胞に導入した。その後トランスフェクションされたGHOST-CXR4細胞は蛍光を放つため、接種用ベクターによるHIV-1のウイルスタンパク質の発現、より具体的にはHIVのRTLの制御下でGFP(緑色蛍光タンパク質)遺伝子の発現を促進するTatタンパク質の発現が確認された。
6週齢のBALB/cマウスの3つのグループ(1グループあたり6匹)を使用した。2つのグループを免疫化に使用し、3つめのグループを対照グループとした。2つの免疫化マウスのグループに、マウス1匹あたり100μgのベクターCAL-HIV-IN-のDNAを筋肉内経路で注入した。一つのグループの動物を2週間後に殺し、他のグループは免疫化(PI)の3週間後に殺した。対照マウスは免疫化の2週間後に殺した。それぞれのマウスの脾臓を取り出し、脾細胞を単離し、続いてこれを上述のようなELISPOT試験またはマルチパラメータのフローサイトメトリーでの解析のいずれかに用いた。Table 6(表6)にELISPOTによる解析の結果を示す。それによれば、IFN-λを分泌する全ての抗原に特異的な細胞の存在が示される。これらの細胞の大部分は、GagおよびTat+Rev+Nef抗原に特異的である。免疫化の2週間後および3週間後における応答は、実質的に類似している。

- 前記キメラレトロウイルスゲノムが、前記第二のレトロウイルスのgag、pol、vif、vpx、vpr、nef遺伝子と、前記第三のレトロウイルスのtat、rev、vpuおよびenv遺伝子とを含むことを特徴とする、請求項1、2、4および5のいずれか一項に記載の核酸。
- 前記第二のレトロウイルスが、オンコウイルス、レンチウイルスまたはスプーマウイルスから選択されることを特徴とする、請求項1から3のいずれか一項に記載の核酸。
- 前記第二のレトロウイルスおよび前記第三のレトロウイルスがそれぞれ、オンコウイルス、レンチウイルスまたはスプーマウイルスから選択されることを特徴とする、請求項1、2および4から6のいずれか一項に記載の核酸。
- 前記第二のレトロウイルスおよび前記第三のレトロウイルスがそれぞれレンチウイルスであることを特徴とする、請求項1、2および4から8のいずれか一項に記載の核酸。
- 前記レンチウイルスが、ヒト免疫不全ウイルスI型(HIV-1)、HIV-2、サル免疫不全ウイルス(SIV)、ネコ免疫不全ウイルス(FIV)またはウマ感染性貧血ウイルス(EIAV)から選択されることを特徴とする、請求項7から9のいずれか一項に記載の核酸。
- 前記レンチウイルスのキメラゲノムが、SIVのgag、pol、vif、vpx、vpr遺伝子と、HIV-1のnef、tat、rev、vpuおよびenv遺伝子とを含むことを特徴とする、請求項1、2および4から10のいずれか一項に記載の核酸。
- 前記レンチウイルスのキメラゲノムが、HIV-1のgag、pol、vif、vpx、vpr、nef、tat、rev、vpuおよびenv遺伝子、またはHIV-2のgag、pol、vif、vpx、vpr、nef、tat、rev、vpuおよびenv遺伝子を含むことを特徴とする、請求項1から11のいずれか一項に記載の核酸。
- 前記pol遺伝子が存在する場合、前記pol遺伝子が、インテグラーゼ(in)をコードする配列から切り出されたpol遺伝子であることを特徴とする、請求項1から12のいずれか一項に記載の核酸。
- 前記非組み込み型キメラレトロウイルスゲノムが、配列番号1、配列番号2または配列番号6の配列を含むことを特徴とする、請求項13に記載の核酸。
- 請求項1から14のいずれか一項に記載の核酸を含む組換えベクター。
- 請求項1から14のいずれか一項に記載の核酸、または請求項15に記載のベクターを含む、免疫原性組成物または接種用組成物。
- レトロウイルスによる感染の予防または治療に使用するための、請求項1から14のいずれか一項に記載の核酸、請求項15に記載のベクター、または請求項16に記載の接種用組成物。
- 前記レトロウイルスが、オンコウイルス、レンチウイルスまたはスプーマウイルスである、請求項17に記載の使用のための核酸、ベクターまたは接種用組成物。
- 前記レトロウイルスが、HIV-1、HIV-2またはFIVである、請求項17または18に記載の使用のための核酸、ベクターまたは接種用組成物。
