本発明は、miR−29a〜cの作動薬又は拮抗薬を含む、医薬品組成物も提供する。作動薬は、miR−29a〜cをコードする核酸セグメントを含む発現ベクター、又は成熟miR−29a〜c配列もしくはその有効部分を含むポリヌクレオチドであってもよい。作動薬は、脂質送達ビヒクルに含まれていてもよい。作動薬は、miR−29a〜c又はその標的とハイブリダイズするポリヌクレオチドであってもよい。

本発明の特定の実施形態は、miR−29a、miR−29b、及び/又はmiR−29cの機能的対立遺伝子の一方又は両方を欠くトランスジェニック動物を提供する。また、誘導性の組織選択的又は構成的プロモーターの制御下でmiR−29a〜cを発現するトランスジェニック動物、そのような動物から得られる組換え細胞系、及びトランスジェニック胚は、miR−29a〜cが線維症の制御でと病的な心臓肥大及び心不全の発症で演ずる、正確な役割を決定するのに役立てることができる。さらに、これらのトランスジェニック動物は心臓の発生に洞察を提供するかもしれない。核酸をコードする誘導性の、あるいは抑制可能なmiR−29a〜cの使用は、過剰に調節された、あるいは調節されていない発現のモデルを提供する。また、miR−29a〜cに関して「ノックアウト」されたトランスジェニック動物も、一方又は両方の対立遺伝子において、企図される。また、1つ又は両方のクラスターに関する一方又は両方の対立遺伝子において、miR−29a〜cに関して「ノックアウト」されたトランスジェニック動物も、企図される。
本明細書で使用される「心不全」という用語は、心臓が血液を送出する能力が低下する、任意の状態を意味するのに広く使用される。その結果、うっ血及び浮腫が組織に発症する。最も頻繁に、心不全は、冠血流量の減少から生じる心筋の収縮性が低下することによって引き起こされ、しかし、心臓弁の損傷、ビタミンの欠乏、及び原発性心筋疾患を含めた多くのその他の要因が、心不全をもたらす可能性がある。心不全の正確な生理学的機構は、全体が理解されていないが、心不全は一般に、交感神経、副交感神経、及び圧受容器応答を含めた心臓の自律的な性質のいくつかの障害に関わると考えられる。「心不全の徴候」という文言は、心不全に関連した実験所見を含む、息切れ、圧痕水腫、肥大した圧痛肝、うっ血頚静脈、及び肺ラ音などの心不全に関連する続発症の全てを包含するのに広く使用される。
ノーザンブロット分析。心不全でない、あるいは心不全と診断された、無記名のヒトの左心室の心臓組織試料を、Gilead Colorado(Westminster、CO)から入手した。心筋梗塞に罹っていると診断された匿名の人物の、境界領域の心臓組織試料を得た。トータルRNAを、トリゾール試薬(Gibco/BRL)を使用することによって、細胞、マウス、ラット、ヒト心臓組織試料、又は単離した筋細胞から単離した。臭化エチジウムでノーザンゲルを染色することにより、同等の負荷であることを確認した。マイクロRNAを検出するノーザンブロットを、前述のように行った(van rooijら、2006)。U6プローブはローディングコントロールの役目を果たした。α−MHCの発現を検出するために、成体の野生型及びmiR−208突然変異動物の両方の心臓組織由来のRNA10μgを含むノーザンブロットを、5’UTR領域及び第1エキソンの一部を包含する、α−MHCのcDNA断片を用いて解析した。
ストレス応答性miRNAによる心臓肥大及び心不全の調節
その細胞表現型のモデュレーションへの関与に照らし、本発明者らは、miRNAが、遺伝子発現の転写的及び翻訳的変化をもたらすことが知られている心臓ストレスに対する心臓の応答を調節する役割を演ずると仮定した。心臓肥大でのmiRNAの潜在的関与を調査するために、186の異なるmiRNAを示すマイクロアレイを使用して、心臓肥大の2つの確立されたマウスモデルで、並列miRNAマイクロアレイ分析を行った(Babakら、2004)。心臓に対する後負荷を増大させることによって肥大を誘発させる、胸部大動脈瘤絞扼術(TAB)を施したマウスを、擬似手術を行った動物と比較した。第2のモデルでは、心臓で活性化カルシニュリン(CnA)を発現するトランスジェニックマウスは、重篤な、十分に特徴付けられた形の肥大をもたらすものであり(Molkentinら、1998)、このマウスを野生型同腹子と比較した(図14)。TABにかけたマウスの心臓から単離されたRNAは、擬似手術した対照に比べて27miRNAで高い発現を示し、CnA Tgマウスは、非トランスジェニック同腹子対照に比べ、33miRNAで高い発現を示し、即ちその21が両方のモデルで上方制御されたものであった。同様に、TAB及びCnA誘発性肥大は、それぞれ15及び14のmiRNAという低い発現を伴い、そのうち7miRNAは、一般に下方制御されたものであった(図14B)。これらのmiRNAのノーザン分析(公表されていないデータ)及び先のマイクロアレイ分析(Baradら、2004; Sempereら、2004; Shingaraら、2005; Liuら、2004)は、これらが広範な組織で発現することを示す。それらの相対的な発現レベルと、ヒト、ラット、及びマウス配列の保存と、肥大中の発現レベルとに基づき、本発明者らは、11の上方及び5の下方制御されたmiRNAに焦点を当てた(図14C)。
miR−208による調節のための下流標的としてのmiR−29ファミリーの発見
本発明者らは、miR−208の動作を媒介し得る下流miRNAを特定する試みにおいて、野生型及びmiR−208−ヌルマウスからの心臓でmiRNAマイクロアレイを行った(図16)。本発明者らは、miR−29ファミリーの多数のメンバーが、miR−208−ヌルマウスで上方制御されることを発見した(図17)。標的の予測は、miR−29ファミリーメンバーが、多数のコラーゲン及び細胞外基質のその他の成分をコードするmRNAを標的とすることを示した(図18)。このように、miR−208−ヌルマウスでのmiR−29ファミリーメンバーの上方制御は、これら動物で見られる線維症を遮断する原因である可能性が高い(図19)。
miR−29a〜cは繊維化遺伝子の発現を調節する。
MI後に心臓でのmiR−29a〜cに関する可能性ある機能を定義しようとするため、本発明者らは、可能性あるmiR−29a〜c標的を特定するのにコンピュータによる予測を利用した。Targetscan予測ウェブサイトは、miR−29a〜cの可能性ある標的として、コラーゲン、メタロペプチド、及びインテグリンをコードする予期せぬ多数の線維症関連mRNAを示した(targetscan.orgの世界的なウェブ)。miR−29a〜cの下方制御が心臓線維症を調節し得るか否かを決定するために、本発明者らは、心臓でのECM産生に関与することが予測される標的に焦点を当てた。エラスチン(ELN)、フィブリリン1(FBN1)、コラーゲンI型、α1及びα2(COL1A1、COL1A2)、及びコラーゲンIII型、α1(COL3A1)は全て、miR−29a〜cに関する1つ又は複数の保存された潜在的なシード配列を含有する(実施例20A)。
心臓線維芽細胞でのmiR−29a〜cの調節
心臓線維症は、欠陥を有する心臓に典型的に見られるリモデリングプロセスの主な態様である。線維芽細胞の増殖及びECM成分の多量の沈着は、心筋の硬直及び拡張期不全をもたらす。形質転換増殖因子β(TGFβ)は、心臓でのコラーゲンの産生及び沈着で主要な役割を演じることが示され、線維芽細胞から筋線維芽細胞への形質転換を誘発する(Border及びNoble、1994)。TGFβに曝された心臓線維芽細胞に関するリアルタイムPCR分析は、miR−29a〜c発現の低下を明らかにし、MI後のmiR−29a〜cの減少はTGFβ調節され得ることを示唆している(図21A)。興味深いことに、B型ナトリウム利尿ペプチド(BNP)のようなナトリウム利尿ペプチドは、線維症及び筋線維芽細胞の変換に関連したTGFβ調節遺伝子発現を阻害することが示されている(Kapounら、2004)。これに関して、本発明者らは、心臓特異的miRNA miR−208に欠けるマウスが心臓線維症及びリモデリングに対して耐性があり、ベースラインでBNPの高い発現を示すことを、先に報告した(van Rooijら、2007)。BNPはTGFβの作用に拮抗することが知られているので、本発明者らは、これらマウスにおける高いBNPレベルによって、miR−29a〜cの発現が増大し得ると推測した。確かに、ノーザン分析は、miR−208を除去した後に、miR−29a〜c発現で用量依存的な増加を示したが、これはBNPの高い発現レベルと一致するものであった(図21B)。これらのデータは、TGFβが、心筋細胞によって分泌されたBNPによって阻害することができる少なくとも部分的にはmiR−29a〜cのレベルの低下を通して、線維芽細胞でコラーゲン関連遺伝子の発現を誘発することを示す。
miR−29a〜cのin vivoノックダウンは、線維症及びコラーゲン遺伝子の発現を誘発する。
コラーゲン発現の負のレギュレーターとしての、miR−29a〜cの潜在的な役割をさらに探求するために、本発明者らは、miR−29bの成熟miRNA配列に相補的なコレステロール修飾オリゴヌクレオチドを使用して、miR−29bをin vivoでノックダウンし(抗miR-29b)、負の対照として生理食塩液又は4塩基ミスマッチ含有するオリゴヌクレオチドを用いた(mm miR-29b)(図22A)。抗miR−29bを尾静脈に1回注射(80mg/kg)してから3日後、本発明者らは、試験をした全ての組織でmiR−29b発現の劇的な減少を観察した(図22B)。対照的に、同等の用量のmm miR−29bアンチセンスオリゴヌクレオチドは、生理食塩液の対照に比べ、miR−29bの発現レベルに対して効果を発揮しなかった。抗miR−29bによるノックダウンは、pre−miRNAのレベルが抗miRとmm処理動物との間で同等のままであるので、成熟miRNAに特異的であるように見えた。肝臓及び腎臓でのノックダウンは完了したように見えたが、低レベルのmiR−29bが、心臓及び肺では検出可能のままであった(図22B)。
miR−29a〜c模倣体によるコラーゲン発現の下方制御
miR−29a〜cの過発現によってコラーゲン発現を低下させることが可能か否かを決定するために、本発明者らは、線維芽細胞をmiR−29b模倣体に曝した。線維芽細胞培養物中のmiR−29b発現のレベルは、miR−29b模倣体に曝してから3日後に、400倍程度に増加した(図22F)。miR−29a発現は影響を受けず、miR−29c発現は、miR−29b模倣体によってごくわずかしか増加しなかった(図22F)。リアルタイムPCR分析は、コラーゲン遺伝子の発現が、miR−29b模倣体に応答して減少することを示した(図22G)。しかし、コラーゲン発現の減少の大きさは、miR−29bの発現の増加に比べると中程度であり、miR−29a〜cレベルはコラーゲンレベルの唯一の決定因子ではないことが示される。

Claims (88)
- (a)心臓線維症、心臓肥大、又は心不全を有する対象を特定する工程と、
(b)前記対象にmiR−29a〜cの発現又は機能の作動薬を投与する工程と
を含む、その必要がある対象の心臓線維症、心臓肥大、又は心不全を治療する方法。 - miR−29a〜cの作動薬が、miR−29a、miR−29b、miR−29c、又はこれらの組合せの成熟配列を含むポリヌクレオチドである、請求項1に記載の方法。
- ポリヌクレオチドが、配列番号18、配列番号19、又は配列番号20の配列を含む、請求項2に記載の方法。
- miR−29a〜cの作動薬が、非経口投与又は直接注射によって心臓組織に投与される、請求項2に記載の方法。
- 非経口投与が静脈内又は皮下投与である、請求項4に記載の方法。
- miR−29a〜cの作動薬が、経口、経皮、持続放出、制御放出、遅延放出、坐薬、カテーテル又は舌下投与によって投与される、請求項2に記載の方法。
- 第2の治療薬を前記対象に投与する工程をさらに含む、請求項1に記載の方法。
- 前記第2の治療薬が、β遮断薬、イオノトロープ、利尿薬、ACE−I、AII拮抗薬、BNP、Ca++遮断薬、エンドセリン受容体拮抗薬、及びHDAC阻害薬からなる群から選択される、請求項7に記載の方法。
- 前記第2の治療薬が、miR−29a〜cの作動薬と同時に投与される、請求項7に記載の方法。
- 前記第2の治療薬が、miR−29a〜cの作動薬の前又は後に投与される、請求項7に記載の方法。
- 心臓線維症、心臓肥大又は心不全の1つ又は複数の症状が、miR−29a〜cの作動薬の投与の後に対象内で改善される、請求項1に記載の方法。
- 前記1つ又は複数の改善された症状が、高い運動能力、高い心臓駆出容積、低い左心室拡張末期圧、低い肺毛細血管楔入圧、高い心拍出量、高い心係数、低い肺動脈圧、低い左心室収縮末期及び拡張末期径、低下した心臓線維症、心筋での少ないコラーゲン沈着、低い左心室及び右心室壁応力、低い壁張力、高い生活の質、及び低い疾患関連罹患率又は死亡率、又はこれらの組合せである、請求項11に記載の方法。

- (a)組織線維症の危険性を有する、又は危険性がある対象を特定する工程と、
(b)前記対象の骨格筋又は線維芽細胞でのmiR−29a〜cの発現及び/又は活性を増大させる工程と
