【解決手段】(1)[3H]および[14C]、または、[3H]および[35S]のいずれか1つの組み合わせとなるように放射標識されたソフトトラップ剤およびハードトラップ剤の存在下で、被験化合物の代謝反応を行う反応工程、(2)反応工程後に未反応のトラップ剤を取り除く前処理工程、および(3)前処理工程後に2種の付加体の放射線量を同時に測定する定量工程を含む反応性代謝物の検出方法。
【効果】2種の放射線量を分別同時定量が可能となり、測定や解析にかかる時間を短縮できるため、特に医薬品の探索初期段階において、反応性代謝物を検出する方法として有用である。

Description
たとえば、放射標識された[35S]システインおよび[14C]シアニドをトラップ剤として用い、肝臓ミクロゾームによって代謝された反応性代謝物とトラップ剤の付加体を検出する方法が開示されている(非特許文献3)。
また、複数のトラップ剤を用いたトラッピング試験は、複数のトラップ剤の分別同時定量が不可能であるため、測定および解析が長時間にわたり、特に探索初期段階に必要とされるスループット性が損なわれる。
被験化合物の放射標識体を用いず、定量性に優れ、ハイスループットで実施可能な反応性代謝物を検出する方法が求められている。
すなわち、本発明は、(1)[3H]および[14C]、または、[3H]および[35S]のいずれか1つの組み合わせとなるように放射標識されたソフトトラップ剤およびハードトラップ剤の存在下で、被験化合物の代謝反応を行う反応工程、(2)反応工程後に未反応のトラップ剤を取り除く前処理工程、および(3)前処理工程後に2種の付加体の放射線量を同時に測定する定量工程を含むことを特徴とする、ハイスループットで実施可能な反応性代謝物の検出方法を提供するものである。
また、医薬品開発の探索初期において、反応性代謝物生成を回避した化合物を取得するには反応部位を推定することが好ましいため、本発明のようなトラッピング試験に引き続いて付加体の構造解析が行われる場合がある。本発明は、性質の異なる2種のトラップ剤との付加体を分別して検出することが可能なため、分別せずに検出する方法と比較して、構造解析工程に移行しやすい。さらに、放射線量測定の際に必要な液体シンチレーションカクテルの使用量が削減できる。
反応性代謝物とは、被験化合物が体内で代謝されることによって生じる、反応性を有する代謝物を意味する。
トラップ剤とは、反応性代謝物の求電子部位との結合に適した求核性を有する低分子化合物を意味する。
ソフトおよびハードとは、Hard and soft acids and bases則におけるソフトおよびハードを指し、ルイス酸およびルイス塩基について、ソフトな酸はソフトな塩基と、ハードな酸はハードな塩基と反応しやすいことを示す原理に基づく用語である。
本発明の方法は、(1)[3H]および[14C]、または、[3H]および[35S]のいずれか1つの組み合わせとなるように放射標識されたソフトトラップ剤およびハードトラップ剤の存在下で、被験化合物の代謝反応を行う反応工程、(2)反応工程後に未反応のトラップ剤を取り除く前処理工程、および(3)前処理工程後に2種の付加体の放射線量を同時に測定する定量工程、を含む。
本発明で用いられる2種のトラップ剤は、一方はソフトトラップ剤、他方はハードトラップ剤である。
ソフトトラップ剤としては、グルタチオン、N−アセチルシステインまたはシステインが挙げられ、グルタチオンが好ましい。
ハードトラップ剤としては、シアニド、セミカルバジド、メトキシアミンまたはリシンが挙げられ、シアニドが好ましい。
トラップ剤としては、たとえば、グルタチオン、N−アセチルシステイン、システイン、セミカルバジド、メトキシルアミンもしくはリシンなどの[3H]で放射標識可能な化合物;グルタチオン、N−アセチルシステイン、システイン、シアニド、セミカルバジド、メトキシルアミンもしくはリシンなどの[14C]で放射標識可能な化合物;またはグルタチオン、N−アセチルシステインもしくはシステインなどの[35S]で放射標識可能な化合物が挙げられる。
分別同時定量を可能にするため、2種のトラップ剤の放射核種の組み合わせは、[3H]および[14C]、または、[3H]および[35S]のいずれか1つの組み合わせであり、[3H]および[14C]の組み合わせが好ましい。
放射標識された2種のトラップ剤としては、[3H]グルタチオンおよび[14C]シアニドの組み合わせが好ましい。
緩衝液としては、たとえば、リン酸緩衝液またはトリス塩酸緩衝液などが挙げられる。
薬物代謝酵素としては、たとえば、シトクロムP450、エステラーゼ、アルデヒドオキシダーゼ、キサンチンオキシダーゼ、エポキシド加水分解酵素、グルクロン酸転移酵素、硫酸転移酵素、グルタチオン転移酵素、アセチル転移酵素またはメチル転移酵素などが挙げられる。
代謝酵素を含む生体抽出物としては、たとえば、肝臓ミクロゾーム、肝臓サイトゾル、肝臓S9または特定の酵素を発現させた昆虫細胞膜画分などが挙げられる。
反応液に添加される試薬としては、たとえば、塩化マグネシウムまたはアラメチシンなどが挙げられる。
代謝反応の時間に特に制限はなく、試験の目的に応じて適宜決定すればよい。
所望の反応時間が経過した後、反応を停止させるが、反応を停止させる方法としては、特に制限はなく、たとえば、有機溶媒の添加、pHの変更または反応温度の変更を挙げることができる。
洗浄液の種類は、適宜決定すればよく、たとえば、精製水または非放射標識トラップ剤水溶液などを挙げることができる。
洗浄液の量は、測定時に十分なシグナル・ノイズ比が確保でき、かつ、2種の放射核種のうち、一方の放射線量が他方の放射線量の測定に影響を及ぼさない範囲内におさめることができるように、適宜決定すればよい。
測定時間に特に制限はなく、適宜決定すればよいが、たとえば、5〜30分間測定すればよく、5〜10分間が好ましい。
(1)反応工程
ヒト肝臓ミクロゾーム(1mg/mL)、β-NADPH(1mmol/L)、塩化マグネシウム(3.3mmol/L)、[3H]グルタチオン(約40kBq/100nmol/mL)および[14C]シアン化カリウム(約4kBq/40nmol/mL)を含有する0.1mol/Lリン酸緩衝液(pH7.4)を予め37℃に加温した。この溶液に、ジメチルスルホキシドに溶解した被験化合物を添加し、反応を開始した。反応液容量は0.15mLとし、反応液中の被験化合物の濃度は50μmol/L、ジメチルスルホキシドの濃度は1%とした。被験化合物として、ベンズブロマロン、カルバマゼピン、クロザピン、フルタミド、ネファゾドン、ネビラピン、チクロピジン、チエニル酸、トログリタゾン、アセトアミノフェン、アトルバスタチン、ジクロフェナク、エリスロマイシン、イミプラミン、インドメタシン、プロカインアミド、プロプラノロール、タクリン、タモキシフェン、ベラパミル、アムロジピン、デキサメタゾン、エチニルエストラジオール、レボフロキサシン、オランザピン、ピオグリタゾン、プラバスタチンおよびロシグリタゾンを用いた。反応開始1時間後、予め氷冷した0.1mol/Lリン酸緩衝液(pH7.4)0.4mLを添加し、氷上に静置することで反応を停止した。
(2)前処理工程
次に、予めメタノール2mLおよび精製水2mLでコンディショニングした固相抽出カラム(Oasis・HLB:粒子径60μm、担体量60mg、Waters社製)に上記(1)で得た液を添加し、液が完全にカラムを通過するまで自然に滴下させた。続いて精製水9mLをカラムに通過させ、未反応のトラップ剤を除去した後にメタノール/アセトニトリル等量混液1.5mLをカラムに通過させ、付加体を溶出した。
(3)定量工程
上記(2)で得た溶出液全量と液体シンチレーションカクテルHionic fluor(PerkinElmer社製)約8mLを混合し、液体シンチレーションカウンターTri-Carb・3110TRの[3H][14C]同時測定モードで、10分間、放射線量を測定した。
この結果は、本発明の測定結果から算出された付加体の総和が、反応性代謝物による毒性リスクを評価する上で有用な指標となり得ることを示している。
また、本発明は、共有結合試験とは異なり、放射標識した被験化合物を必要としないことから、探索初期より実施可能である。
Claims (4)
- (1)[3H]および[14C]、または、[3H]および[35S]のいずれか1つの組み合わせとなるように放射標識されたソフトトラップ剤およびハードトラップ剤の存在下で、被験化合物の代謝反応を行う反応工程、
(2)反応工程後に未反応のトラップ剤を取り除く前処理工程、および
(3)前処理工程後に2種の付加体の放射線量を同時に測定する定量工程を含む反応性代謝物の検出方法。 - ソフトトラップ剤およびハードトラップ剤が[3H]および[14C]の組み合わせとなるように放射標識された、請求項1に記載の反応性代謝物の検出方法。
- ソフトトラップ剤が[3H]グルタチオンである請求項1または2に記載の反応性代謝物の検出方法。
- ハードトラップ剤が[14C]シアニドである請求項1〜3のいずれか一項に記載の反応性代謝物の検出方法。
