Description
クローニングおよびプラスミド構築。
pLT32Flag−COMPcc−SHe発現プラスミドの構築。Flag−COMPcc−SHeのコード配列(図1.B)を含有するプラスミドはGenscript(配列番号31)でオーダーした。Flag−COMPcc−SHeコード配列は、NdeI/NotIで開いたpLT32H細菌発現ベクターにおいてNdeI/NotI断片として連結させた(Mertensら、1995)。
pLT32SHe−tGCN4形質転換E.コリの30ml前培養物をLuriaブロス内で28℃において増殖させ、これを使用して1リットルの新たな培地を接種した。0.6−0.8のA600において、細胞を1mmのイソプロピル1−チオ−β−d−ガラクトピラノシドで処理し、さらに4時間インキュベートし、次いで遠心分離(6000×g、20分、4℃)によって回収した。細菌のペレットは20mlのトリス−HClバッファー(50mMのトリス−Hcl、50mMのNaClおよび1mMのEDTA)、pH8中に再縣濁し、超音波処理した。細菌の残骸は遠心分離(20,000×g、1時間、4℃)によってペレット状にした。上清を、50mMのNaClを含有するトリス−HClバッファー(バッファーA)で予め平衡化したDEAEセファロースカラムにかけた。洗浄後、0−40%バッファーB(50mMのトリス−Hcl、1MのNaCl)から40−100%バッファーBまでの2ステップ勾配により結合タンパク質を溶離した。SHe−tGCN4を含有する分画をプールし、25%硫酸アンモニウム飽和状態に調節し、25%硫酸アンモニウム、50mmのトリス−HCl、pH8で予め平衡化したフェニル−セファロースカラムにかけた。結合タンパク質は2ステップ勾配で溶離した。2ステップの溶離は、0−40%および40−100%の50mMトリス−HClバッファー、pH8(バッファーA)で実施した。SHe−tGCN4を含有する分画はSuperdex75カラムに充填した。ゲル濾過はリン酸緩衝生理食塩水(PBS)で実施し、SHe−tGCN4を含有する分画はプールし−70℃で保存した。
熱不安定性E.コリエンテロトキシンの解毒変異体、LTR192Gを鼻腔内(i.n.)投与に使用した。この調製物は寛大なことにDr.J.Clements(Department of Microbiology and Immunology、Tulane University Medical Center、New Orleans、LA、USA)によって提供された(Nortonら、2010)。
その中で天然由来のシステインがセリンに置換されN末端にシステインが付加された、SHe(cc4s)、化学合成したHPLC精製SHeペプチドを、Pepscan(Pepscan、Lelystad)でオーダーした。SHe(cc4s)ペプチドは、そのN末端システイン残基を介して、製造者の説明書に従いヘテロ二官能性スルホ−MBS(Pierce)を使用した化学結合により、HBc VLP表面上のmHBcの免疫優性ループ中のリシンと融合させた。わずか400μgのmHBcを溶かした200μlのPBSをスルホ−MBSと1時間、(1mg/mlの最終濃度で)インキュベートした。サイズ排除クロマトグラフィーによる非結合スルホ−MBS分子の除去後、スルホ−MBS結合mHBc VLPは2mlのH2Oに希釈した。後に、(100mlのPBSに溶かした)100μlのSHe(cc4s)ペプチドを加え、室温で1時間インキュベートして、mHBc VLPとペプチドの架橋を可能にした。最後に、遊離SHe(cc4s)ペプチドをサイズ排除クロマトグラフィーにより除去した。純度および架橋効率は、SDS−PAGE、次にクーマシー染色によって試験した。
Hep−2細胞(ATCC、CCL−23)、Vero細胞(ATCC、CCL−81)、HEK293T細胞(Dr M.Hallからの進物)およびA549細胞ATCC、CCL−185)を、10%熱不活化ウシ胎児血清(FCS)、1%ペニシリン、1%ストレプトマイシン、2mMのL−グルタミン、非必須アミノ酸(Invitrogen、Carlsbad、カリフォルニア)、および1mMのピルビン酸ナトリウムを補充したDMEM培地中で増殖させた。
特定病原体を含まない、メスのBALB/cマウスをCharles River(Charles River Wiga、Sulzfeld、ドイツ)から得た。動物は12時間光/暗サイクルで温度調節した環境中に収容し、食物および水は制限せず与えた。動物実験室内で1週間適応させた後、マウスは8週齢で免疫処置した。
鼻腔内免疫処置または感染用に、マウスにイソフルランによって軽く麻酔をかけた。ワクチン+アジュバントまたはウイルスの投与に使用した最終容積は50μlであった(鼻孔当たり25μl)。ワクチン+アジュバントはPBSで製剤化し、一方ウイルス接種原はHBSSで製剤化した。
攻撃後3日または4日でマウスを屠殺した。マウス肺は無菌状態で除去し、10%スクロースを含有する1mlのHBSSにおいて30秒間Heidolph RZR2020ホモジェナイザーを用いて均質にした。肺ホモジネートは4℃での遠心分離により後に浄化し、Hep−2細胞におけるウイルス滴定に使用した。単層のHep−2細胞には、96ウエルプレート中、ペニシリンとストレプトマイシンを補充した無血清OPTIMEM培地(Invitrogen)中で、50μlの連続1:3希釈肺ホモジネートを感染させた。4時間後、2%FCSを含有するDEMEM培地で2回細胞を洗浄し、50μlのオーバーレイ培地(1%FCS、0.5%アガロースを含有する完全DEMEM培地)において37℃で5日間インキュベートした。アガロースオーバーレイの上部に50ulの4%パラホルムアルデヒド溶液を加えることにより、細胞を固定した。4℃での一晩の固定後、オーバーレイ培地およびパラホルムアルデヒド溶液を除去し、細胞はPBSで2回洗浄し、1%BSAを含有するPBS(PBS/BSA)でブロッキングした。その後、ポリクローナルヤギ抗RSV血清(AB1128、Chemicon International)を加えた(1/4000)。PBS/BSAで3回洗浄した後、細胞はhrp結合抗ヤギIgG抗体(SC2020、Santa Cruz)と共に30分間インキュベートした。非結合抗体は、0.01%トリトン−X100を含有するPBS/BSAでの4回の洗浄、およびPBSでの1回の洗浄によって除去した。最後に、TrueBlueペルオキシダーゼ基質(KPL、Gaithersburg)の使用によって、プラークを目に見える状態にした。異なる希釈のプラークを計数し、それぞれの希釈に関して、肺(1ml)当たりのPFU数は、希釈中に存在したプラークの数×希釈×20(=合計1000μlの上清容積/希釈系列の第一ウエルの感染に使用した50μlの上清容積)として計算した。次いでPFU/肺の数を、異なる希釈に関して計算したPFU/肺の平均数として計算した。ホモジナイズした肺の各上清は二連で試験したので、最終的なPFU/肺の数はこれら二連の平均として計算した。
qRT−PCRによって肺RSV負荷を決定するため、前に記載したのと同様に肺ホモジネートを調製し浄化した。これらの肺ホモジネート由来の全RNAは、製造者の説明書に従い高純度RNA組織キット(Roche、Mannheim)の使用によって調製した。ヘキサマープライマーおよびTranscriptor First Strand cDNA合成キット(Roche、Mannheim)の使用によってcDNAを調製した。ゲノムRSV M cDNAの相対レベルは、RSV A2 M−遺伝子のゲノムRNAに特異的なプライマー(5’TCACGAAGGCTCCACATACA3’および5’GCAGGGTCATCGTCTTTTTC3’)、ならびに5’末端をフルオレセイン(FAM)および3’末端付近を黒色消光色素で標識したヌクレオチドプローブ(#150 Universal Probe Library、Roche)の使用によって、qRT−PCRにより決定した。GADPH mRNAの相対量は、マウスGADPHに特異的なプライマー(5’TGAAGCAGGCATCTGAGGG3’および5’CGAAGGTGGAAGAGTGGGAG3’、ならびにLightCycler480 SYBR Green I Maseter Mix(Roche)の使用により、qRT−PCRにより決定した。肺ホモジネート当たりのゲノムRSV RNAの相対量は、RSV M−遺伝子RNAの相対量とマウスGADPHm RNAの相対量の間の比として計算した。
各免疫処置後2週間で、血液サンプルを外側尾静脈から回収した。アベルチンで麻酔した動物の心臓穿刺により最終出血を実施した。血液を37℃で30分間凝固させ、その後の2回の遠心分離から上清を得ることにより血清を入手した。
Hek293T細胞を示した発現ベクターでトランスフェクトした。24時間後、酵素を含まない解離バッファー(Invitrogen、Carslbad、カリフォルニア)を使用して細胞を剥離し、PBSで1回洗浄し、1%BSAを含有するPBS(PBS/BSA)中で1時間インキュベートした。その後、示した濃度で示した血清または抗体と共に、細胞をインキュベートした。1時間後、細胞はPBS/BSAで3回洗浄し、抗マウスIgG alexa633二次抗体と共に30分間インキュベートした。PBS/BSAで4回およびPBSで1回細胞を洗浄した後、Becton Dickinson LSR IIフローサイトメーターを使用して細胞を分析した。一種のGFP発現細胞を、側方散乱シグナルのピーク表面、前方散乱シグナルのピーク表面およびピーク高、および緑色蛍光シグナルのピーク表面に基づいて選択した。最後に、これらのGFP陽性単細胞の中で、alexa633蛍光シグナルを測定した。
Vero細胞を擬似感染、または無血清培地の存在下において0.5MOIのRSV A2で感染させた。4時間後、遊離ウイルスは洗浄除去し、1%FCSを含有する増殖培地中で細胞をインキュベートした。16時間後、細胞をPBSで1回洗浄し、2%パラホルムアルデヒドで20分間固定した。その後、細胞をPBSで2回洗浄し、5分間0.2%トリトン−X100洗剤で透過処理した。PBSで1回洗浄した後、細胞はPBS/BSAでブロッキングした。1時間後、She特異的3G8モノクローナル抗体またはアイソタイプ対照抗体を5μg/mlの最終濃度で加えた。PBS/BSAで2回細胞を洗浄した後、ポリクローナル抗RSVヤギ血清を加えた。1時間後、細胞をPBS/BSAで3回洗浄した。細胞と示した抗体の結合は、それぞれalexa488とalexa568蛍光色素で標識した抗マウスと抗ヤギIgG抗体の使用により分析した。染色細胞の共焦点画像はZeiss共焦点顕微鏡で記録した。
SHe特異的モノクローナル抗体(mAb)を産生する安定的ハイブリドーマ細胞を、ハイブリドーマ技術(Kohler and Milstein 1975)によって作成した。簡単に言うと、SHe特異的ハイブリドーマを、オールハイドロゲルを補充した10μgのSHe−tGCN4ワクチン(Brenntag Biosector)を用いて3週間間隔で3回腹腔内免疫処置した個々のマウスから誘導した。融合3日前に、マウスを同じ製剤でもう1回追加抗原刺激し、脾臓細胞を単離し、次いでPEG1500(Roche Diagnostics GmbH、ドイツ)の存在下でSP2/0−Ag14ミエローマ細胞と融合させた。融合した細胞は、10%ウシ胎児血清、10%BMコンディムドH1(Roche Diagnostics GmbH、ドイツ)、2mMのL−グルタミン、および24μMのβ−メルカプトエタノールおよび1×HATサプリメント(Invitrogen、Carlsbad、カリフォルニア)を補充したRPMI1640培地中で増殖させた。SHe特異的抗体を分泌するハイブリッドはSHeペプチドElisaスクリーニングによって同定し、モノクローナル抗体産生ハイブリッドは、限界希釈手順による2ラウンドのサブクローニング後に得た。モノクローナル抗体は、プロテインA−セファロースカラム(electrical engineering biosciences)で精製した。
SHタンパク質は、RSVビリオンの表面、およびRSV感染細胞の原形質膜で、ペンタマーとして発現される。SHのペンタマー構造は、5パラレルα−ヘリックスのコイルドコイルとしてオリゴマー化するSH膜貫通型ドメインによって構造化される。その本来の立体配座を模倣するペンタマーとしてRSV AのC末端SH外部ドメイン(SHe)を提示するために、これも5パラレルα−ヘリックスで構成されるラット軟骨オリゴマー基質タンパク質の短いペンタマーコイルドコイルドメイン(COMPcc)と、SHeを遺伝学的に融合させた(Malashkevichら、1996;図1)。Flag−タグをCOMPのN末端と融合させ、Flag−COMPcc−SHeにした。Flag−COMPcc−SHeはpLT−32(Mertensら、1995)発現ベクターにおいてクローニングし、E.コリ中で発現させ精製した。ゲル濾過分析によりFlag−COMPcc−SHeが55−60kDa複合体として溶離したことを明らかにし、11kDaのFlag−COMPcc−SHeタンパク質は実際ペンタマーとしてオリゴマー化することを示した(図2)。
Flag−COMPcc−SHeを用いた予防接種が、RSV感染に対する防御を誘発し得るかどうか試験するために、本発明者らはBALB/cマウスRSV感染モデルを使用した。BALB/cマウスは、1μgのE.コリ熱不安定性エンテロトキシンLTR192Gアジュバントと組み合わせた25μgのFlag−COMPcc−SHeで3回鼻腔内免疫処置した。テトラマーGNC4足場と融合したPBSおよびインフルエンザA M2外部ドメインは(M2e−tGNC4)(De Filetteら、2008)は陰性対照として使用した。免疫処置は2週間毎に実施した。1つのRSV感染(5×105PFU)は陽性対照として使用した。各免疫処置後の第一週と第二週の間に、血液を回収してSHe特異的IgG抗体の誘導を調べた。SHe特異的抗体の存在は、最初にSHeペプチドELISAによって試験した。M2eペプチドELISAは陰性対照として使用した。図3は、SHeペプチド特異的IgG抗体を誘導し、それぞれFlag−COMPcc−SHeによる第二および第三免疫処置後に追加抗原刺激することを実証する。3連続のFlag−COMPcc−SHe/LTR192G免疫処置は高レベルのIgG2a SHe特異的抗体、ただしごく低レベルのIgG1SHe特異的抗体をもたらし、Th1指向型/誘導型免疫応答を示した。PBSまたはM2e−tGCN4/LTR192G予防接種マウスにおいて、SHe特異的IgG抗体を検出することはできなかった(図3A、BおよびC)。予想通り、Flag−COMPcc−SHe/LTR192GまたはPBS予防接種マウスデータの血清において、M2e特異的抗体を検出することはできなかった。以前の結果(De Filetteら、2008)に従い、M2e−tGCN4で免疫処置したマウスは高力価のM2e特異的IgG2a抗体を蓄積した。
B型肝炎ウイルスコアタンパク質(HBc)ウイルス様粒子(VLP)は、密なアレイとして抗原を提示することができる。このようにしてHBc−VLPは、提示抗原に対する強力な液性免疫応答を誘導することができる(Boisgeraultら、2002)。したがって、ペンタマーとしてのSHeの提示に対する代替として、mHBc−VLPの免疫優性領域ループにSH外部ドメインを提示した。HBc−SHe−VLPは、リシンがHBc免疫優性領域の上部に導入されたHBcの変異体であるmHBcとSHeペプチドの化学結合によって得られた(De Filetteら、2005)。化学結合を可能にするため、システイン残基をSHeのN末端に加えた。加えて、SHeペプチドの位置4に存在するシステイン残基をセリン残基に置換した。このペプチドをSHe−CC4Sと呼んだ。サイズ排除クロマトグラフィーによるmHBc−SHe−VLPの精製後、架橋の程度をSDS PAGEによって調べた。図7は、約50%のHBcタンパク質がSHe−CC4Sペプチドと化学結合したことを示す。ゆっくり移動したバンドは、2または3個のSHe(cc4s)ペプチドが結合したmHBcモノマーを表し得る。SHeCC4S結合mHBcタンパク質が依然予想される大きさ(30−34nm)のVLPに構築するかどうか試験するために、動的光散乱法による分析を、生成されたmHBc−SHe粒子および完全に機能的な参照物である1604M2e−HBc VLPに実施した。図8は、mHBc−SHe−CC4Sの大きさ分布は1604M2e−HBc対照のそれと重複し、30nmにおいて最大値であり、それはHBc VLPの報告された大きさ(Clarkeら、1987)に相当することを示す。
mHBc VLPの表面上でのSHeペプチドの提示に次いで、融合ペプチド(Refmarina GCN4)に対する強力な液性応答を誘導することが知られているtGCN4とも、SHeを融合させた。SHeとFlag−タグは、それぞれtGCN4コード配列の5’末端と3’末端で遺伝学的に結合させ、PLT32発現ベクターにクローニングした。E.コリにおける発現後、アニオン交換、疎水性相互作用およびゲル濾過クロマトグラフィーにより組換えSHe−tGCN4−Flagを精製した(図9)。
mHBc−SHe(CC4S)およびSHe−tGCN4を用いた予防接種が、RSV感染に対する防御を誘発し得るかどうか試験するために、1ugのLTR192Gアジュバントと組み合わせた10μgのmHBc−SHe(CC4S)およびSHe−tGCN4で、Balb/cマウスを3回鼻腔内予防接種した。PBSおよび空mHBcを、後者は1ugのLTR192Gと組み合わせて、陰性対照として使用した。免疫処置は3週間毎に実施した。1つのRSV感染(5.105PFU)は陽性対照として使用した。各免疫処置後の第二週と第三週の間に、血液を回収してSHe特異的IgG抗体の誘導を調べた。She特異的抗体の存在は、SHeペプチドELISAによって試験した。図10Aは、SHeペプチド特異的IgG抗体を誘導し、それぞれmHBc−SHe(CC4S)およびSHe−tGCN4による第二および第三免疫処置後に追加抗原刺激することを実証する。3連続のFlag−COMPcc−SHe/LTR192G免疫処置は高レベルのIgG2a SHe特異的抗体、および幾分低いレベルのIgG1SHe特異的抗体をもたらし、Th1指向型/誘導型免疫応答を示した(図10B)。
感染細胞と相互作用することができるSHe特異的抗体が、RSV感染に対する防御をもたらし得るかどうか調べるために、本発明者らは、SHe−TGCN4免疫処置マウスに基づきRSV SHe特異的モノクローナル抗体を開発した。ELISA中でSHeペプチドと効率よく結合する抗体を産生した1つのIgG1(3D11)と1つのIgG2a(3G8)亜型ハイブリドーマを選択し、サブクローニングし抗体産生に使用した。3D11および3G8はプロテインAアフィニティークロマトグラフィーによって精製し、ELISAによりSHeとの結合効率に関して試験した。図13は、3D11および3G8はコーティングSHeペプチドと結合することができ、それぞれIgG1およびIgG2a亜型であることを示す。
SHe特異的抗体がインビボでRSV複製を低減することができるかどうか試験するために、SHe特異的モノクローナル抗体でマウスを受動免疫処置した。SHe特異的3G8モノクローナル抗体、アイソタイプ対照抗体またはPBSを、RSV攻撃の1日前と1日後にマウスに鼻腔内投与した。RSV攻撃後3日で、血液を回収し、治療マウスの血清中のモノクローナル抗体の存在に関して試験した。RSV攻撃後4日で、マウスを屠殺し肺中ウイルス力価を決定した。ペプチドELISAは、各抗体で治療したマウス血清中の低濃度のSHe特異的およびアイソタイプ対照抗体の存在を実証した(データ示さず)。図16は、SHe特異的モノクローナル抗体を与えたマウスは、PBSまたはアイソタイプ対照モノクローナル抗体で治療したマウスと比較して、低い肺中RSV力価を有することを示す。これらのデータは、SHe特異的抗体の鼻腔内投与は、マウスにおけるRSV感染を低減することができることを示唆する。
SHeベースのワクチンも、代替アジュバントおよび異なる担体と共に、代替経路によりこのワクチンを投与するとき、RSV感染に対して防御することができるかどうか試験するために、担体としてのキーホールリンペットヘモシアニン(KLH)と共に腹腔内でワクチンを試験した。マレイミド活性化KLH(Pierce)を、RSV A SH外部ドメインに対応するペプチド(CGGGSNKLSEYNVFHNKTFELPRARVNT(配列番号50);RSV A SH外部ドメイン(She)に対応する配列に下線を引く)と化学結合させた。直接的化学結合を助長するため、CysGlyGlyGlySerリンカーをRSV A SHeペプチドのN末端に加えた。さらに、本来のRSV A SHe中に存在したシステイン残基をセリン残基に置換した。化学結合は製造者(Pierce)の説明書に従い実施した。架橋KLH−SHeタンパク質はサイズ排除クロマトグラフィーによって単離した。
SHeベースのワクチンを用いた腹腔内(I.P.)予防接種が、RSV感染に対する防御を誘発し得るかどうか試験するために、50μlのフロイント不完全アジュバント(Milipore)とそれぞれ組み合わせた20μgのKLH−SHeまたはKLHで、Balb/cマウス(グループ当たり6匹のマウス)を3回腹腔内予防接種した。アジュバントなしのPBS予防接種を追加の陰性対照として使用した。予防接種後の第二週と第三週の間に、血液を回収してSHe特異的IgG抗体の誘導を決定した。SHe特異的抗体の存在を決定しSHeペプチドELISAにより定量化した。図17(A−B)は、KLH−SHeを用いた3連続予防接種は、IgG1亜型とIgG2a亜型の両方の、高レベルのSHe特異的IgG抗体を誘導することを実証する。PBSまたはKLH予防接種マウス由来の血清において、SHe特異的IgG抗体を検出することはできなかった。さらにフローサイトメトリー分析は、KLH−SHeを腹腔内予防接種したマウス由来の血清は、それらの表面でRSV SHタンパク質を発現するHEK293T細胞と特異的に結合することができ、一方で免疫前血清はできなかったことを明らかにした。
KLH−SHeを用いた鼻腔内予防接種が、RSV感染に対する防御を誘発し得るかどうか試験するために、1μgのLTR192Gアジュバントとそれぞれ組み合わせた20μgのKLH−SHeまたはKLHで、Balb/cマウス(グループ当たり6匹のマウス)を3回鼻腔内予防接種した。アジュバントなしのPBS予防接種を追加の陰性対照として使用した。予防接種後の第二週と第三週の間に、血液を回収してSHe特異的IgG抗体の誘導を調べた。SHe特異的抗体の存在はSHeペプチドELISAにより試験した。図18(A−B)は、KLH−SHeを用いた3連続予防接種は、IgG1亜型とIgG2a亜型の両方のSHe特異的IgG抗体を誘導することを実証する。PBSまたはKLH予防接種マウス由来の血清において、SHe特異的IgG抗体を検出することはできなかった。さらにフローサイトメトリー分析は、免疫前血清ではなく、KLH−SHe血清を鼻腔内予防接種したマウス由来の血清が、それらの表面でRSV SHタンパク質を発現するHEK293T細胞と特異的に結合することができることを明らかにした。
SHeベースワクチンで予防接種したマウスにおけるRSV複製の低減が、RSV SHe特異的抗体により仲介され得るかどうかさらに調べるために、受動輸送の実験を実施した。いずれも75μlのフロイント不完全アジュバントと組み合わせたKLH−SHeまたはKLHのいずれか20μgで、Balb/cマウスを腹腔内予防接種した。追加の陰性対照として、アジュバントなしのPBSでマウスを予防接種した。SHeペプチドELISAは、KLH−SHeで予防接種した全てのマウスの血清が、高レベルのSHe特異的IgG抗体を含有することを示した。最後の出血後、各グループのマウスの血清をプールし、56℃で30分間熱失活させた。KLH−SHe血清はRSV感染を防御することができるかどうか試験するために、40μlのKLHまたはKLH−SHe血清を、RSV攻撃(2.105PFU)(第0日)1日前(第−1日)と1日後(第1日)にマウスに鼻腔内投与した。PBSで治療したマウスは追加の対照として含めた。全てのマウスの体重を毎日モニタリングした(図19C)。感染後5日で、マウスを屠殺し肺ホモジネートを調製した。プラークアッセイによる分析は、KLH−SHe血清で治療したマウスの肺ホモジネートは、KLH血清で治療したマウス由来の肺ホモジネートより、約40倍少ない複製ウイルス(ウイルス力価の平均の比)を含有していたことを実証した(図19B)。KLH血清で治療したマウスの肺中RSV力価が、PBSで治療したマウスの肺中RSV力価と違わなかったという観察結果は、対照血清の投与はマウスにおける肺中RSV複製に影響を与えないことを示す。
それらの亜型内で高度に保存されているにもかかわらず、RSV BウイルスのSHeアミノ酸配列は、RSV A亜型ウイルスのそれと異なる。したがって、RSV Bウイルスに対して防御するため、SHeベースワクチンはRSV BのSHeアミノ酸配列を含むことが最も必要とされ得る。
mHBc−SHeB VLPが免疫原性であるかどうか試験するために、50μlのTitermax(Sigma)と組み合わせた20μgのmHBc−SHeBで、1匹のBALB/cマウスを3回皮下免疫処置した。3回の免疫処置は2週間間隔で実施した。各免疫処置1日前および最終免疫処置後2週間で、出血を実施した。mHBc−SHeB免疫血清が、感染細胞の表面上で発現されるRSV B SHタンパク質を認識し得るかどうか試験するために、Vero細胞には、(親切なことにDr.Marc van Ranst、University of Leuven、Leuven、ベルギーにより提供された)RSV Bウイルスの臨床分離株を擬似感染させたか、または感染させた。感染後72時間で、細胞を固定し、0.2%トリトンX−100を使用して透過処理、または非透過処理した。フロイント不完全アジュバントと組み合わせたmHBc−SHeB免疫血清(1/100希釈)またはKLH(KLH血清)で予防接種したBALB/cマウス由来の対照免疫血清(1/100希釈)のいずれかで、細胞を次いで染色した。サンプルは免疫蛍光顕微鏡またはフローサイトメトリーによって分析した。図21AおよびBは、mHBc−SHeB免疫血清は、非感染細胞ではなく、透過処理と非透過処理RSV B感染細胞の両方と結合することができることを示す。対照的に、対照免疫血清はRSV B感染細胞と結合しなかった。これは、mHBc−SHeBによるマウスの予防接種は、おそらく大部分はRSV B感染細胞の表面上で発現されるRSV B SHタンパク質との結合によって、RSV B感染細胞を認識することができる血清抗体を誘導することを実証する。
mHBc−SHeBを用いた予防接種は、RSV B感染からマウスを防御することができるかどうか試験するために、6匹のマウスの2グループを、50μlのフロイント不完全アジュバントを補充したmHBcまたはmHBc−SHeB VLPで免疫処置した。追加の対照として、6匹のマウスをPBSで予防接種した。3週間間隔で3回、腹腔内予防接種した。各免疫処置後2週間で出血を実施した。SHe特異的抗体の誘導は、コーティングペプチドとしてSHeAまたはSHeBを使用したペプチドELISAにより決定した。この分析は、全てのマウスにおいて、3連続のmHBc−SHeB免疫処置が、IgG1亜型とIgG2a亜型の両方の高力価のRSV B She特異的IgG抗体を誘導することを実証した(図22A−C)。mHBc−SHeB免疫血清も、はるかに低い程度ではあるがSHeAペプチドと結合した(図22A、BおよびD)。
ペンタマーとしてSHeを提示するための代替のタンパク質足場として、本発明者らは、E.コリLPP−56リポタンパク質(Liuら、2004)由来のLiuらによって記載されたペンタマートリプトファン−ジッパー(LPP(5))を使用した。Mertensら、1995によって記載されたように、LPP(5)トリプトファン−ジッパーのコード配列はSHeコード配列と遺伝学的に融合させ、ヘキサヒスチジンペプチドおよびカスパーゼ切断部位を含有するE.コリ発現ベクター(pLH36)にクローニングした。この発現プラスミドはpLH36−HisDEVD−LPP−SHe(配列番号49)と名付けた。このプラスミドからの発現によってキメラLPP(5)−SHeタンパク質(配列番号52)が生じる(MHHHHHHPGGSDEVDAKWDQWSSDWQTWNAKWDQWSNDWNAWRSDWQAWKDDWARWNQRWDNWATGGNKLCEYNVFHNKTFELPRARVNT、ヒスタグ配列には下線を引き、リンカーはイタリック体であり、DEVDカスパーゼ切断部位はイタリック体および下線であり、ペンタマーLPPトリプトファン−ジッパーは太字であり、RSV A SH外部ドメインは太字およびイタリック体である)。E.コリにおける発現の誘導後、二次的ニッケルアフィニティー、アニオン交換およびゲル濾過クロマトグラフィーによりLPP(5)−SHeタンパク質を精製した。図23は、粗製細胞抽出物中で(23A)および精製タンパク質として(24B)、両方で、She特異的3G8モノクローナル抗体によりLPP(5)−SHeタンパク質を認識することができることを実証する。
独立した動物モデルにおけるワクチンの有効性を証明するため、コットンラットを使用する。コットンラット(サイグモンドンヒスピダス(Sigmondon hispidus))はRSV感染の影響を受けやすい(Princeら、1978)。それぞれ6匹のコットンラットの5グループを使用する。100μgのKLH(賦形剤対照)または100μgのKLH−SHe(すなわち、RSV−A由来のSHeペプチドと担体としてのKLHの化学結合体)で、2グループの動物を腹腔内(i.p.)免疫処置した。KLHおよびKLH−SHeワクチン抗原はフロイント不完全アジュバントで製剤化し、これらを使用して第0日、21日、および42日にコットンラットを免疫処置する。第三グループの動物には、アラムアジュバントの存在下においてホルマリン不活化RSV(FI−RSV)で筋肉内免疫処置する。後者のグループは、RSVによる二次攻撃により明らかとなるワクチン増大型疾患の誘導に関する陽性対照として働く。第四グループの動物には、第0日にコットンラット当たり2.04×105プラーク形成単位のRSV−Tracyを感染させ、二次攻撃に対する防御に関する陽性対照として働く。第五グループのコットンラットは攻撃の日まで未治療状態に保ち、RSV攻撃に関する対照として働く。予防接種のスケジュールは図24中に示す。

- 呼吸器合胞体ウイルスの低分子量疎水性タンパク質の外部ドメイン、および担体を含む免疫原性組成物。
- 外部ドメインがオリゴマーとして提示される、請求項1に記載の免疫原性組成物。
- 外部ドメインが配列番号1−3、またはそれらの変異体を含む、請求項1または2に記載の免疫原性組成物。
- 外部ドメインが担体と遺伝学的に結合した、請求項1から3のいずれかに記載の免疫原性組成物。
