本発明は、不適切な構造、毒性、不溶性、新規構成要素若しくは不十分な量により宿主細胞系で製造することがそうでなければ困難でありうる組換え巨大分子の製造方法である。以下で詳細に記述される該方法は、所望の組換え巨大分子を製造することが可能な宿主細胞を膜透過処理剤で処理して宿主細胞の膜の完全性を破壊すること、次いで所望の組換え巨大分子の製造に十分な条件に該宿主細胞を曝露することを必要とする。
本明細書に記述される方法により、選択された巨大分子が発現される宿主細胞の細胞膜
を、該巨大分子が大量にかつ正しいコンホメーションで発現されることを可能にするように計画的に操作する。
をコードする第一の核酸配列;およびきっちり合うように調節されるプロモーターを含んでなる第二の制御配列の機能的制御下の該巨大分子をコードする第二の核酸配列を含有し、該第二の制御配列は宿主細胞中での該巨大分子の発現を制御する。
本明細で別の方法で定義されない限り、本明細書で使用される技術および科学用語は、本発明が属する技術分野の当業者によりおよび公表された本文への参照により普遍的に理解されると同一の意味するところを有する。本明細若しくは請求の範囲の多様な態様が多様な環境下で「含んでなる」言語を使用して提示される一方、関連する一態様は「よりなる」若しくは「より本質的になる」言語を使用してもまた記述されうる。「ある(a若しくはan)」という用語が1つ若しくはそれ以上を指し、例えば「ある化合物(a compound)」は1種若しくはそれ以上の化合物を表すことが理解されることに注目されるべきである。であるから、「ある(a)」(若しくは「ある(an)」)、「1つ若しくはそれ以上」および「最低1つ」という用語は本明細書で互換性に使用される。
「宿主細胞」という用語により、組換えベクターの受容体として使用され得る若しくは使用された原核微生物若しくは単細胞の実体としての培養された真核生物細胞株由来の細胞を意味している。該用語はトランスフェクトされた元の細胞の子孫を包含する。単一親細胞の子孫は、偶発的若しくは計画的突然変異により、必ずしも形態が完全に同一またはゲノム若しくは全DNAが元の親に相補的でないかもしれないことが理解される。所望の生合成酵素をコードするヌクレオチド配列の存在のような関連する特性により特徴付けられるために親に十分に類似である親細胞の派生物/子孫は、該定義に包含されかつ上の用語により包括される。適切な宿主細胞は当業者により容易に選択されうる。本方法で有用な宿主細胞は細菌細胞、酵母細胞、哺乳動物細胞および昆虫細胞を包含する。

A.巨大分子の発現のための核酸分子を含有する宿主細胞の構築
Giacaloneら、2006、Biotechniques、40(3):355−63に記述されるとおり構築されたpRHAプラスミドは、成功裏にトランスフェクトされた大腸菌(E.coli)宿主の同定を見込む選択可能なマーカーampr(アンピシリン耐性のため)を含有する。巨大分子すなわちタンパク質FGF−20をコードするDNA配列をpRHAプラスミドにrhaTプロモーターの後にクローン化する。場合によっては、該タンパク質を宿主細胞からの後の精製のためGST若しくはヒスチジンタグでタグをつける。rhaTプロモーターは、L−ラムノースの添加により誘導可能かつD−グルコースの存在により抑制される厳しく調節されるプロモーターである。標的巨大分子をコードする核酸配列を含んでなるこのプラスミドをコンピテントな大腸菌(E.coli)細胞に形質転換する。形質転換された大腸菌(E.coli)宿主細胞を50μg/mlのアンピシリンを含有するLB寒天上で画線し、そして該プラスミドを含有する細胞について選択するため37℃で一夜増殖させる。単一コロニーを選択しかつアンピシリンを含有する15mLのLB培地中37℃で15時間増殖させる。細胞をペレットにしかつ次の段階のため使用する。
該大腸菌(E.coli)を、巨大分子の発現を抑制するため0.2%D−グルコースを補充されているLB培地中で培養する。宿主細胞が、巨大分子の産生を最大化するために一般に選択される細胞密度である1.0のOD595という選択された細胞密度に達する場合に、大腸菌(E.coli)宿主細胞を収集しかつ培地を捨てる。
細胞をその後、細胞膜の完全な溶解を伴わずに細胞膜の完全性を破壊するのに十分な環境条件下で外的に添加された透過処理剤コリシンYに曝露する。本実験において、収集された大腸菌(E.coli)をその後中性緩衝液に再懸濁し、そして2μg/mLの透過処理剤コリシンYで約5分間処理する。この処理が、宿主細胞を完全に溶解することを伴わずに細胞膜の完全性を破壊する。細胞膜のこの破壊がそれにより小分子量化合物例えば約50ないし2000ダルトンの小分子の膜を通る輸送を可能にする。

コンピテントな大腸菌(E.coli)宿主細胞を実施例1に記述されたところのプラスミドpDT−E148Sでトランスフェクトする。タンパク質5−α還元酵素のバリアントを得かつ実施例1に記述されたとおりpRHAプラスミドにrhaTプロモーターの後に個別にクローン化して、それによりコードされるタンパク質5−α還元酵素バリアント中でのみ異なるpRHAプラスミドのライブラリーを提供する。タンパク質バリアントコーディング配列を含有する各プラスミドを、pDT−E148Sを含有する大腸菌(E.coli)細胞に形質転換する。形質転換された細胞を、双方のプラスミドを含有する細胞について選択するためアンピシリンおよびカナマイシンを含有するLB寒天上で画線しかつ37℃で一夜増殖させる。各バリアントを運搬する宿主細胞の単一コロニーを選択しかつアンピシリンおよびカナマイシンを含有する15mLのLB培地中37℃で15時間増殖させる。
バリアント大腸菌(E.coli)宿主細胞を96ウェルプレートに蒔き、各プレートは異なるバリアントを含有する。ウェル中の各大腸菌(E.coli)を、巨大分子バリアントの発現を抑制するがしかし宿主細胞の周辺質に蓄積するDT−E148Sの発現を許容するため0.2%D−グルコースを補充されているLB培地中で培養する。宿主細胞が1.0のOD595という選択された細胞密度に達する場合に、大腸菌(E.coli)宿主細胞を収集しかつ培地を捨てる。

上の実施例のいずれについても、以下の既知かつ公表されている手順は、細胞透過処理剤の適用若しくは発現後に細胞膜の透過性の程度を試験するのに使用しうる一方法の一例である。これらの手順はとりわけO’KeefeとCollier、PNAS、86:343−6(1989)により詳細に記述されている。当業者は本明細書に記述される方法の最適化においてこれら若しくは同様の手順を使用しうる。
形質転換された大腸菌(E.coli)細胞を1.0のOD595までL培地中で増殖させる。細胞を遠心沈降しかつ1mlあたり3×109細胞の濃度まで2mMトリス・HCl/50mM NaCl、pH7.0に再懸濁する。各実験点について、細胞を50mM NaClを含有する多様なpH値の5mM緩衝液に1:10希釈する。この点から細胞を温度調節された磁気的に攪拌されるキュベット中で37℃に保つ。以下の緩衝液すなわちPipes(pH7.0および6.5)、Mes(pH6.0)ならびにクエン酸ナトリウム/コハク酸ナトリウム(pH5.5および5.0)を個々のキュベットに添加する。細胞(pH特異的緩衝液を含有する)を1minインキュベートし、そしてその後100mMトリス−HCl(pH7.5)にもたらす。1分後にヨウ化3,3’−ジプロピルチアジカルボシアニン(Molecular Probes、オレゴン州ユージーン)を1μg/mlの最終濃度まで添加する。シグナルが安定化した場合に測定される蛍光をpH7.0での蛍光により除算して相対蛍光を得る。蛍光は645nmでの励起および668nmでの測定を用いSLM AMINCO(イリノイ州アーバナ)SPF−500C蛍光分光計で測定する。双方のスリットを5mmに設定する。
無カリウムM9培地(リン酸ナトリウムをリン酸カリウムの代わりに用いる)中で一夜増殖させた細胞を同一培地で1:100希釈する。5hr増殖させた後に該細胞を1.0のOD595まで増殖培地に再懸濁する。0.1mLの細胞を、4.8μCi(1Ci=37GBq)のL−[2,3,4,5−3H]プロリンを含有する多様なpH値(上と同一)の1.9mlの20mM緩衝液に添加する。37℃で10min後に該細胞をMillipore HAフィルターで濾過し、そして5mlの無カリウムM9塩で洗浄する。フィルターを乾燥しかつOCS(Amersham)に溶解し、そして放射活性をシンチレーションカウンターで測定する。
細胞を輸送アッセイについてのとおり増殖させる。1.0のOD595まで再懸濁した後に該細胞を86RbCl(20μCi/ml)の存在下37℃で1hrインキュベートする。0.1mlの細胞をその後多様なpH値の1.9mlの20mM緩衝液(上と同一)に添加しそして37℃で10minインキュベートする。該細胞をその後、洗浄緩衝液に前浸漬されたMillipore HAフィルターで濾過する。フィルターを10mM
RbClを補充された5mlのM9塩で洗浄しかつ乾燥し、そして放射活性をガンマカウンターで測定する。
上のアッセイのいずれかでの95%超の活性の低下は、本明細書に記述される方法により、細胞膜が、該膜の完全な溶解を伴わずに小分子量化合物の輸送に十分に透過処理されたことを示す。
Claims (35)
- a)i.宿主細胞中での膜透過処理剤の発現を制御する第一の制御配列の機能的制御下の該剤をコードする第一の核酸配列;および
ii.誘導可能なプロモーターを含む第二の制御配列の機能的制御下の巨大分子をコードする第二の核酸配列であって、該制御配列が宿主細胞中での該巨大分子の発現を制御する、該核酸配列
を含んでなる宿主細胞を、該巨大分子の所望の産生レベルを満たすのに十分な選択された細胞密度まで培養する工程;
b)細胞膜の完全な溶解を伴わずに細胞膜の完全性を破壊するように該剤を誘導する環境条件に宿主細胞を曝露して、それにより小分子量化合物の該膜を通る輸送を可能にする工程;ならびに
c)破壊された細胞膜を通り輸送し得る成分を含んでなる栄養素カクテルの存在下で該宿主細胞を培養する工程であって、該成分が、第二の制御配列のプロモーターを誘導する誘導剤および膜が破壊された宿主細胞中での該巨大分子の発現を可能にする代謝要件を含む、工程;を含んでなり、かつ、該巨大分子が宿主細胞中で発現される、
宿主細胞中での巨大分子の組換え発現方法。 - a)膜の完全な溶解を伴わずに細胞膜の完全性を破壊しそして小分子量化合物の該膜を通る輸送を可能にする膜透過処理剤と宿主細胞を接触させる工程であって、該宿主細胞が誘導可能なプロモーターを含んでなる制御配列の機能的制御下に巨大分子をコードする核酸配列を含有し、該制御配列が該宿主細胞中での該巨大分子の発現を制御する、工程;
b)破壊された細胞膜を通って輸送し得る成分を含んでなる栄養素カクテルの存在下で該宿主細胞を培養する工程であって、該成分が膜を破壊された宿主細胞中での該巨大分子の高められた発現を可能にする代謝要件を含む、工程;
を含んでなる、宿主細胞中での巨大分子の組換え発現を高める方法。 - 宿主細胞が、細菌細胞、酵母細胞、哺乳動物細胞および昆虫細胞よりなる群から選択される、請求項1若しくは2に記載の方法。
- 膜透過処理剤が、細菌タンパク質毒素、そのフラグメントおよびそのバリアントよりなる群から選択される、請求項1に記載の方法。
- 細菌タンパク質毒素が、ジフテリア毒素、炭疽菌保護抗原、緑膿菌(Pseudomonas aeruginosa)外毒素A、ボツリヌス毒素、破傷風毒素、コレラ毒素、黄色ブドウ球菌(S.aureus)α毒素、β−バレル膜孔形成毒素、大腸菌(E.coli)溶血素、ウェルシュ菌(Clostridium perfringens)ι毒素、リステリオリシンOおよび細胞溶解素よりなる群から選択される、請求項4に記載の方法。
- 膜透過処理剤が植物タンパク質毒素、そのフラグメントおよびそのバリアントよりなる群から選択される、請求項1に記載の方法。
- 植物毒素がリシン、アブリンおよびモデシンよりなる群から選択される、請求項6に記載の方法。
- 膜透過処理剤がチャンネルタンパク質である、請求項1に記載の方法。
- チャンネルタンパク質が、イオンチャンネルタンパク質、電位制御チャンネルタンパク質、化学制御チャンネルタンパク質および非制御チャンネルタンパク質よりなる群から選
択される、請求項8に記載の方法。 - 膜透過処理剤が、受動輸送タンパク質、酵素、ギャップ結合タンパク質、核孔複合体、膜孔形成タンパク質、ホリン、コリシン、プロテグリン、補体および補体関連タンパク質、膜融合ウイルスタンパク質、それらのフラグメントならびにそれらのバリアントよりなる群から選択される、請求項1に記載の方法。
- 膜孔形成タンパク質がナイスタチン、アムホテリシンB、グラミシジンA、アラメシシンおよびポリンよりなる群から選択される、請求項10に記載の方法。
- 酵素がリパーゼである、請求項10に記載の方法。
- コリシンがコリシンE1、コリシンE3、コリシンA、コリシンIaおよびコリシンYである、請求項10に記載の方法。
- 補体がパーフォリンである、請求項10に記載の方法。
- 膜透過処理剤がヘマグルチニンである、請求項10に記載の方法。
- 膜透過処理剤が界面活性剤、イオノフォア、若しくは浸透圧ストレスを誘導する剤である、請求項2に記載の方法。
- 巨大分子がタンパク質、DNA若しくはRNAである、請求項1若しくは2に記載の方法。
- 核酸配列、第一の核酸配列若しくは第二の核酸配列が、プラスミド、バクテリオファージ若しくはウイルスである輸送ベクター中に独立して存在する、請求項1若しくは2に記載の方法。
- 第一の制御配列が構成的プロモーターを含んでなる、請求項1に記載の方法。
- 第一の制御配列が誘導可能なプロモーターを含んでなる、請求項1に記載の方法。
- 第二の制御配列が第一の制御配列のものと異なるプロモーターを含んでなる、請求項1に記載の方法。
- 誘導可能なプロモーターがきっちり合うように調節されるプロモーターである、請求項1若しくは2に記載の方法。
- 第二の制御配列が、第一の制御配列のものと異なる誘導可能なプロモーターを含んでなる、請求項22に記載の方法。
- 工程(b)の環境条件が、pHを変えること、塩濃度を変えること、浸透圧を変えること、温度を変えること、宿主細胞を光に曝露すること、および宿主細胞を作用剤、生体分子、化学物質若しくはリガンドに曝露することの最低1種を含んでなる、請求項1に記載の方法。
- pHを変えることがpHを約4ないし6.5に低下させることを含んでなる、請求項24に記載の方法。
- 作用剤、生体分子、化学物質若しくはリガンドが、還元剤、酸化剤、酸、塩基、塩、カオトロープ、界面活性剤、および第一の制御配列の誘導可能なプロモーターを誘導する作用剤よりなる群から選択される、請求項24に記載の方法。
- 小分子量化合物が、大きさが約50ないし約2000ダルトンの間の範囲にある、請求項1若しくは2に記載の方法。
- 栄養素カクテルが、巨大分子の発現を制御するプロモーターを誘導するための誘導剤、アデノシン三リン酸(ATP)、アミノ酸、リボヌクレオチド、補助因子、デオキシリボヌクレオチド、非天然のアミノ酸、安定化剤、浸透圧調節物質、酸化還元を変える剤、および巨大分子を産生するのに必要な環境因子よりなる群から選択される1種若しくはそれ以上の成分を含んでなる、請求項1若しくは2に記載の方法。
- 発現された巨大分子を宿主細胞から回収することをさらに含んでなる、請求項1若しくは2に記載の方法。
- 栄養素カクテルの存在下で細胞を培養する前に宿主細胞を濃縮若しくは収集することをさらに含んでなる、請求項1若しくは2に記載の方法。
- a)i.宿主細胞中での膜透過処理剤の発現を制御する第一の制御配列の機能的制御下の該剤をコードする第一の核酸配列;および
ii.誘導可能なプロモーターを含んでなる第二の制御配列の機能的制御下の巨大分子をコードする第二の核酸配列であって、該制御配列が該宿主細胞中での該巨大分子の発現を制御する、該核酸配列
を含有する宿主細胞を、ミニウェルプレートの各ウェル中で;
該巨大分子の所望の産生レベルを満たすのに十分な選択された細胞密度まで培養する工程であって;ここで各ウェルの宿主細胞中の該第二の核酸配列が異なる巨大分子をコードする、工程;
b)細胞膜の完全な溶解を伴わずに該膜の完全性を破壊するように該処理剤を誘導するのに適する環境条件に該宿主細胞を曝露しそれにより小分子量化合物の該膜を通る輸送を可能にする工程;
c)破壊された細胞膜を通って輸送し得る成分(該成分は各ウェル中の細胞の第二の制御配列のプロモーターを誘導する誘導剤を含んでなり);および各ウェルの膜を破壊された宿主細胞中での該巨大分子の発現を可能にする代謝要件(ここで該巨大分子は各ウェルの宿主細胞中で発現される)を含んでなる栄養素カクテルの存在下で該宿主細胞を培養する工程;
d)各ウェルの宿主細胞を試験試薬で処理する工程;ならびに
e)試験試薬に応答した巨大分子の正体を決定する工程
を含んでなる、イン シトウ(in situ)薬物スクリーニング方法。 - a)i.宿主細胞中での膜透過処理剤の発現を制御する第一の制御配列の機能的制御下の該剤をコードする第一の核酸配列;および
ii.誘導可能なプロモーターを含んでなる第二の制御配列の機能的制御下の巨大分子をコードする第二の核酸配列であって、該制御配列が宿主細胞中での該巨大分子の発現を制御する、該核酸配列;
を含有する宿主細胞をミニウェルプレートの各ウェル中で、
該処理剤の蓄積を可能にする選択された細胞密度まで培養する工程;
b)細胞膜の完全な溶解を伴わずに該膜の完全性を破壊するように該剤を誘導するのに適する環境条件に該宿主細胞を曝露し、それにより小分子量化合物の該膜を通る輸送を可能にする工程;
c)破壊された細胞膜を通って輸送し得る成分を含んでなる栄養素カクテル(該成分は各ウェル中の該細胞の第二の制御配列のプロモーターを誘導する誘導剤を含んでなり)および各ウェルの膜を破壊された宿主細胞中での該巨大分子の発現を可能にする代謝要件の存在下で、該宿主細胞を培養する工程であって;ここで該巨大分子が各ウェルの宿主細胞中で発現される、工程;
d)各ウェルの宿主細胞を異なる試験試薬で処理する工程;ならびに
e)巨大分子での応答を誘導した試験試薬の本質を決定する工程、
を含んでなる、イン シトウ(in situ)薬物スクリーニング方法。 - 試験試薬に対する細胞の応答が結合若しくはシグナル事象の誘発である、請求項31若しくは32に記載の方法。
- 試験試薬で細胞を処理する前に細胞を溶解する工程をさらに含んでなる、請求項31若しくは32に記載の方法。
- 試験試薬が破壊された細胞膜を通り輸送する小分子である、請求項31若しくは32に記載の方法。
