本願は、本明細書中に参考として援用される、2010年4月9日に出願された米国仮出願第61/322,800号の米国特許法第119条の下での優先権を主張する。
分野
本発明は、ブチロフィリン様タンパク質ならびにそのフラグメント、バリアント、および誘導体、かかるタンパク質をコードする核酸、これらのタンパク質に結合する抗体、ならびに、これらのタンパク質のアゴニストおよびアンタゴニストに関する。かかる分子を含む薬学的組成物およびかかる分子またはかかる分子を含む組成物の使用もまた意図する。
免疫応答または炎症応答の調整は、種々の治療状況で有益であり得る。免疫応答または炎症応答の下方調整は、各種自己免疫疾患または炎症性疾患の処置で望ましい場合もある。任意の免疫応答の上方調整は、例えば、特定の抗原(例えば、ワクチン中に含まれる抗原または癌細胞もしくは線維性疾患を媒介する細胞上で優先的に発現する抗原)に対する応答を増幅するのに有益であり得る。したがって、免疫応答または炎症応答を調整することができる分子は、潜在的に種々の治療状況で治療的に有益である。本発明は、不適切なおよび/または異常な炎症および/または免疫応答によって特徴づけられる疾患の診断および処置のための治療薬を提供する。これらの薬剤のいくつかは免疫応答を刺激することができる。他の薬剤は、炎症応答および/または免疫応答を阻害することができる。
本発明は、BTNL9タンパク質、BTNL9タンパク質をコードする核酸、およびBTNL9タンパク質に結合する抗体を提供する。より具体的には、本明細書中に記載のBTNL9タンパク質は、多量体タンパク質または融合タンパク質であり、これらは単離タンパク質および/または可溶性タンパク質であり得る。BTNL9タンパク質ならびにBTNL9に結合するアンタゴニスト抗体およびアゴニスト抗体の使用も提供する。

本発明は、BTNL9タンパク質またはBTNL9タンパク質のインヒビターもしくはアゴニスト(抗BTNL9抗体および/またはBTNL9タンパク質のバリアント形態など)の使用を提供する。本発明は、BTNL9タンパク質(そのバリアントが含まれる)、かかるタンパク質の使用、および上記の全タンパク質をコードする核酸を提供する。BTNL9タンパク質は、T細胞の活性化、増殖、およびサイトカイン産生を低下させることによってT細胞機能を変化させることができる。かかる効果により、T細胞媒介性の自己免疫疾患または炎症性疾患(とりわけ炎症性腸疾患および線維症など)を有効に処置することができる。BTNL9のインヒビターは、BTNL9がT細胞の活性化、増殖、およびサイトカイン分泌を低下させるのを防止し、したがって、全体としてT細胞活性化を増大させるように機能することができる。かかる効果は、疾患(癌など)の処置またはワクチンの効力の増強に有用であり得る。BTNL9のアゴニストは、例えば、BTNL9タンパク質を発現するB細胞の活性化状態を変化させることによって免疫細胞機能を変化させることができる。
「抗体」は、本明細書中で意味する場合、免疫グロブリンの重鎖可変領域および/または免疫グロブリンの軽鎖可変領域を含む。抗体は、軽鎖可変領域(VL)、軽鎖定常領域(CL)、重鎖可変領域(VH)、第1の重鎖定常領域(CH1)、ヒンジ領域、第2の重鎖定常領域(CH2)、および第3の重鎖定常領域(CH3)を含む全長四量体抗体(IgG、IgA、IgD、IgM、またはIgE抗体など)であり得る。あるいは、抗体は、フラグメント(Fabフラグメントなど)または必要に応じて組換えフラグメント(scFvフラグメントなど)であり得る。単一の可変領域(VH領域またはVL領域のいずれか)を含む単一ドメイン抗体も本明細書中で意味する抗体である。単一ドメイン抗体は、米国特許出願公開第2006/0062784号(単一ドメイン抗体を記載している部分が、本明細書中で参考として援用される)に記載されている。さらに、種々の1価の形態(一本鎖抗体、例えばscFv、Fab、scFv−Fc、ドメイン抗体、および、例えば、国際特許出願公開第WO2009/089004号および米国特許第5,837,821号(その記載部分が本明細書中で参考として援用される)に記載の種々の形式が含まれる)および多価分子(F(ab)2および、例えば、国際特許出願公開第WO2009/089004号および米国特許第5,837821号(その記載部分が本明細書中で参考として援用される)に記載の多価分子など)は、「抗体」の意味の範囲に包含される。
BTNL9は、そのドメイン構造に基づいて、ブチロフィリン様(BTNL)ファミリー内のタンパク質に属している。例えば、Arnett et al.(2008),Current Immunology Reviews 4:43−52およびArnett et al.(2009),Cytokine 46:370−75を参照のこと。BTNLファミリー中のヒトタンパク質にはBTNL2、BTNL3、BTNL8、BTNL9、ERMAP、およびMOGが含まれ、これらのタンパク質のドメイン構造を図1に図式的に示す。図1から明らかなように、BTNL2は、その細胞外領域中に4つの免疫グロブリン様(Ig様)ドメイン(2つのIgV様ドメインおよび2つのIgC様ドメイン)を有する唯一のファミリーのメンバーである。MOGおよびERMAPはそれぞれたった1つのIg様ドメインを有する。BTNL3、BTNL8、およびBTNL9はまた、明確にIg様ドメインである一方の細胞外ドメインおよびIg様ドメインのいくつかの特徴を欠くがIg様ドメインであるためのほぼ正確なサイズである別のドメインを有する。全BTNLファミリーメンバーは膜貫通ドメインを有する。BTNL2およびMOGは短い細胞内領域を有するのに対して、BTNL3、BTNL8、BTNL9、およびERMAPは、B30.2ドメインを含むより長い細胞内領域を有する。いくつかのタンパク質のB30.2ドメインの変異は一定の疾患と関連しているにも関わらず、細胞内B30.2の機能は未知である。Henry et al.(1998),Mol.Biol.Evol.15:1696−1705(その関連する開示が本明細書中で参考として援用される)を参照のこと。さらに、いくつかのB30.2ドメインの結合パートナーが同定されている。例えば、Jeong et al.(2009),J.Biol.Chem.284:22444−22456を参照のこと。
本発明は、BTNL9の分泌性、可溶性バージョン、ならびに細胞表面に発現することができる膜貫通ドメインを含むバージョンを包含する。かかるタンパク質を単離することができる(すなわち、BTNL9タンパク質が調製物中に存在するタンパク質の少なくとも80%または少なくとも90%を構成する精製タンパク質調製物の一部であり得る)。本発明は、さらに、下記のBTNL9核酸によってコードされるBTNL9タンパク質を含む。BTNL9タンパク質は、本明細書中で意味する場合、配列番号2のアミノ酸配列、ならびにそのフラグメント、誘導体、およびバリアントを含むタンパク質(上記および下記で論じる融合タンパク質および多量体が含まれる)を包含する。配列番号2のアミノ酸配列は、1位から開始され、約29位〜約38位、必要に応じて34位で終結するシグナル配列を含む。したがって、成熟BTNL9のアミノ酸配列は、配列番号2の約30〜約39位の位置から開始される。必要に応じて、BTNL9の成熟アミノ酸配列は、配列番号2の35位から開始される。
本発明は、本明細書中に記載のBTNL9タンパク質(配列番号2のアミノ酸配列ならびにそのフラグメントおよび/またはバリアントを含むタンパク質が含まれる)をコードする単離された核酸(例えば、DNAおよびRNAが含まれる)を包含する。これらの核酸は、とりわけ、組換えタンパク質の産生および組織サンプル中のBTNL9核酸の存在の検出(例えば、診断での使用のため)に有用である。かかる核酸は、ゲノムDNAまたはcDNAであり得る。核酸は、BTNL9タンパク質をコードする中断されていない読み取り枠を含むことができる。本発明の核酸分子には、一本鎖形態および二本鎖形態の両方のDNAおよびRNAならびに対応する相補配列が含まれる。「単離された核酸」は、天然に存在する供給源から単離された核酸の場合、核酸が単離された生物のゲノム中に存在する隣接遺伝子配列から分離された核酸である。化学合成された核酸(オリゴヌクレオチドなど)またはテンプレートから酵素的に合成された核酸(ポリメラーゼ連鎖反応(PCR)産物またはcDNAなど)の場合、かかるプロセスに起因する核酸が単離された核酸であると理解される。単離された核酸分子は、個別のフラグメントの形態またはより大きな核酸構築物の成分としての核酸分子をいう。
BTNL9タンパク質または抗BTNL9抗体(または抗イディオタイプ抗体)を下記のように作製することができる。BTNL9タンパク質または抗BTNL9抗体をコードする核酸を、本明細書中に記載のように、ベクターに導入することができ、このベクターを宿主細胞に導入することができる。BTNL9タンパク質または抗BTNL9抗体をコードする核酸を含むベクターおよび宿主細胞は、本発明に包含される。BTNL9タンパク質または抗BTNL9抗体をコードする核酸を含む宿主細胞を、BTNL9タンパク質または抗BTNL9抗体を発現することができるような条件下で培養することができる。次いで、発現したBTNL9タンパク質または抗BTNL9抗体を、上記細胞を培養した培地または上記細胞から得て、当該分野で公知の多数の適切な任意の手段によって精製することができる。さらに、BTNL9タンパク質産生のための遺伝子工学的方法には、公知の方法に従った無細胞発現系での、細胞宿主での、組織での、および動物モデルでのポリヌクレオチド分子の発現が含まれる。
本明細書中に記載のBTNL9タンパク質に特異的に結合する抗体、抗BTNL9抗体に結合する抗イディオタイプ抗体、およびこれらの抗体の使用は、本発明によって包含される。抗BTNL9抗体は、配列番号2のアミノ酸配列またはそのフラグメント(配列番号2のアミノ酸35〜257など)からなるポリペプチドに結合することができる。本明細書中で使用する場合、第1の抗体によるBTNL9タンパク質上のエピトープの特異的結合とは、第1の抗体と競合する別の抗体によって、第1の抗体がBTNL9タンパク質から移動させられることができるが、第1の抗体と結合を競合しない他の抗BTNL9抗体によって置き換えることができないことを意味する。多数の競合的結合アッセイが当該分野で公知である。
アンタゴニスト抗体またはアゴニスト抗体に加えて、BTNL9タンパク質に特異的に結合することができる他の抗体関連分子(アフィボディ(Roennmark et al.(2002),J.Immunol.Methods 261(1−2):199−211(アフィボディを記載している部分が、本明細書中で参考として援用される))およびBTNL9に特異的に結合してBTNL9タンパク質の生物学的活性を阻害する国際特許出願公開第WO00/24782号(これらのペプチドを記載している部分が、本明細書中で参考として援用される)に記載の生物学的に活性なペプチドなど)が本発明に包含される。さらに、BTNL9アンタゴニストには、BTNL9タンパク質および/またはmRNAの発現を調整するのに有用な上記核酸(例えば、干渉RNA(またはこれをコードするDNA)またはアンチセンスRNAもしくはDNAなど)が含まれる。
BTNL9.Fc融合タンパク質が活性化T細胞の増殖を阻害することができることを本明細書中で証明する。BTNL9はまた、活性化T細胞によるサイトカイン(IL−2、TNFα、IFNγ、およびIL−17など)の産生を阻害する。BTNL9がB細胞に結合することができ、わずかな程度T細胞に結合することができることを蛍光標示式細胞分取(FACS)によってさらに証明する。これらの知見は、BTNL9またはBTNL9経路をアゴナイズする能力を有する分子がT細胞によって媒介される自己免疫疾患または炎症性疾患を処置するための治療薬として有用であり得ることを示す。かかる疾患には、例えば、全身性エリテマトーデス、クローン病、潰瘍性大腸炎、喘息、多発性硬化症、関節リウマチ、乾癬、サルコイドーシス、および線維性疾患(アテローム性動脈硬化症、慢性閉塞性肺疾患(COPD)、肝硬変、強皮症、腎臓移植線維症、および肺線維症が含まれる)が含まれる。
実施例1:ヒト免疫細胞および成体ヒト組織におけるヒトBTNL9タンパク質をコードするmRNAの発現
初代ヒト免疫細胞および種々の組織でのヒトBTNL9をコードするmRNAの発現に関する情報を収集するために、以下の実験を行った。
以下は、ヒトBTNL9の細胞外領域およびヒトIgG1抗体のFc部分を含む融合タンパク質をどのようにして作製したかを記載している。リンカー+ヒトIgG1 Fcフラグメントに融合したヒトBTNL9の細胞外ドメインをコードするcDNAを含む適切なベクターを構築した。配列番号18はこのcDNAの配列を提供し、配列番号19はこのcDNAによってコードされるBTNL9.Fcタンパク質のアミノ酸配列を提供する。Cos PKB細胞を、LIPOFECTAMINE(商標)2000(Invitrogen)を使用してBTNL9.Fc哺乳動物発現構築物でトランスフェクトし、0.5%低Ig血清を含む完全ダルベッコ改変イーグル培地(DMEM)で培養した。これらの方法は、Ettehadieh et al.,OVEREXPRESSION OF PROTEIN KINASE BA ENHANCES RECOMBINANT PROTEIN EXPRESSION IN TRANSIENT SYSTEMS in Animal Cell Technology:From Target to Market:Proceedings of the 17th ESACT Meeting,Tyloesand,Sweden,June 10−14,2001,Vol.1,Lindner−Olsson et al.,eds.,pp.31−35,Springer,2001に詳述されている。組換えタンパク質の作製方法を記載しているこの参考文献の部分が、本明細書中で参考として援用される。トランスフェクション7日後、上清を採取し、BTNL9.Fcタンパク質をプロテインAカラムクロマトグラフィ(MABSELECT(商標)SuRe column,GE Healthcare)によって精製した。

標準的な抗CD3増殖アッセイでは、上記のように、ヒトCD4+T細胞を単離し、BTNL9.Fc(T細胞の増殖を阻害することが以前に示されている濃度で)または種々の他のFc含有タンパク質を使用するか使用しないで、抗CD3抗体で刺激した。刺激72時間後、100μlの上清を各条件から採取した。次いで、上清を、Meso Scale Discovery of Gaithersburg,Marylandから販売されている複数のサイトカイン(IL2、IL4、IL5、IL10、IL13、IL17、GM−CSF、TNFα、IFNγ、およびIL1β)を同時に検出するためのカスタマイズされた市販のキットを使用してサイトカインレベルについてアッセイした。かかるキットアッセイは、原理上はELISAアッセイに類似しているが、複合検出テクノロジーを使用している。
特異的細胞型BTNL9が何に結合するかを決定するために以下の実験を行った。マウス脾細胞の単一細胞懸濁物を生成し、次いで、2μg/mlの抗CD3抗体(2C11−マウス;OKT3−ヒト)、コンカナバリン(conconavalin)A(Con A)、または細菌リポ多糖(LPS)で48時間活性化した。コントロールとして非刺激細胞も含めた。非刺激細胞および刺激細胞を、huBTNL9.FcまたはコントロールFc含有タンパク質を使用して氷上で60分間染色した。洗浄後、結合したFcタンパク質を、FACSを使用してフィコエリトリン(PE)結合体化F(ab’)2ヤギ抗ヒトFc(Jackson ImmunoResearch)で検出した。さらに、これらの染色された細胞をアロフィコシアニン(APC)結合体化CD3またはCD19(BD Biosciences)で同時染色して、混合細胞集団中のT細胞およびB細胞をそれぞれ特異的に同定した。サンプルを固定し、FACSCALIBUR(商標)フローサイトメーター(BD Immunocytometry Systems,San Jose,CA,USA)を使用して分析した。
BTNL9.Fcによる活性化ヒトT細胞の増殖の阻害が細胞死に起因するかどうかを決定するために以下の実験を行った。BTNL9.Fcまたはコントロールタンパク質の存在下または非存在下でのヒトCD4+T細胞の抗CD3抗体による刺激後の乳酸デヒドロゲナーゼ(LDH)放出を検出するためのアッセイを使用して、細胞傷害性を検出した。LDHは、原形質膜の損傷および細胞死の際に上清に放出される安定な細胞質酵素である。LDHを、製造者の指示(LDH Cytotoxicity Detection Kit,Clontech Laboratories,Inc,Mountain View,CA,USA)に従った72時間の刺激後の比色反応によって検出した。Triton−Xで溶解した細胞を、細胞死による最大LDH放出のポジティブコントロールとして使用した。同一の実験用アッセイウェルを使用して、細胞増殖阻害およびLDH放出を検出した。
BTNL9が正常な結腸組織と比較して炎症性腸疾患(クローン病または潰瘍性大腸炎)を罹患しているドナー由来の結腸組織中で過剰発現または過小発現するかどうかを決定するために以下の実験を行った。炎症性腸疾患を罹患していないドナーおよび潰瘍性大腸炎またはクローン病のいずれかを罹患しているドナー由来の結腸組織におけるヒトBTNL9発現を、ABI PRISM(登録商標)7900HT配列検出システム(Applied Biosystems Inc,Foster City,CA,USA)を使用した定量的リアルタイムRT−PCRによって測定した。BTNL9mRNA発現の検出量を、ハウスキーピング遺伝子(β−アクチン)の発現に対して正規化した。cDNAを生成するために、罹患組織または正常組織由来のDNアーゼ処理した(DNA−free,Ambion)総RNA20ngを、TAQMAN(登録商標)逆転写キット(Applied Biosystems Inc.)を使用して逆転写した。このcDNAを、TAQMAN(登録商標)ユニバーサルバッファー(Applied Biosystems Inc.)およびhuBTNL9プローブセット(Applied Biosystemsから購入;プローブセットHs_00537320_m1)を使用した定量的リアルタイムRT−PCRにおけるテンプレートとして使用した。PCR条件は、50℃で2分間、次いで95℃で10分間、次いで以下の温度レジメで40サイクル:95℃で15秒間、その後に60℃で1分間。各PCR反応を、研究に含まれる各生物サンプルについて三連で行った。
本実験の目的は、BTNL9.Fcの凝集の状態がT細胞の増殖を阻害するその能力で役割を果たすかどうかを決定することであった。様々な凝集の状態にあるBTNL9.Fcの精製画分を以下のように得た。BTNL9.Fcを発現する哺乳動物細胞の培養上清から得たBTNL9.Fcを、プロテインAクロマトグラフィによって精製した。より具体的には、BTNL9.Fcを、25mM Tris、150mM NaCl(pH7.4)中のプロテインAカラムにロードした。カラムを25mM Tris、0.5M L−アルギニン(pH7.5)で洗浄後、25mM Tris、150mM NaCl(pH7.4)で洗浄した。BTNL9.Fcタンパク質を50mMクエン酸ナトリウム、1M L−アルギニン(pH3.5)で溶出し、1M Tris(pH8.0)を使用して中性pHまで滴定した。BTNL9.Fcを、154mM NaCl、3.89mM KH2PO4、12mM Na2HPO4(pH7.2)(PBS)にて実施したサイズ排除クロマトグラフィ(SEC)によってさらに精製した。各々の個別画分を分析SECによって分析し、次いで、保存のために3つの画分にプールした。プールした画分を濃縮し、50μM EDTAを含むPBSで処方し、分析SECによって分析し、その結果を図9に示す。
BTNL9が脾臓中の血管組織で発現されるかどうかを決定するために以下の実験を行った。凍結ヒト脾臓組織を、75%アセトン/25%エタノールを使用して固定し、ヒトBTNL9およびヒトCD31に特異的な抗体と組み合わせて染色した。CD31は、血管内皮上に優先的に発現する。インキュベーションおよび洗浄後、二次抗体を、免疫蛍光による検出のために組織に添加した。最終洗浄後、切片をDAPIで染色し、画像処理した。BTNL9およびCD31染色の共局在は、脾臓における毛細血管内皮でのBTNL9発現を実証する。脾臓はまた、より弱く染色されるBTNL9+/CD31−細胞を含む。したがって、これらの結果は、BTNL9が脾臓における血管組織および非血管組織の両方に発現することを示す。

- BTNL9タンパク質および別のポリペプチドを含む融合タンパク質をコードする核酸であって、該核酸が、
(a)ポリペプチドをコードするポリヌクレオチドであって、該ポリヌクレオチドが、
(i)配列番号1のヌクレオチド334〜1002のヌクレオチド配列からなるか、または
(ii)ストリンジェントな条件下で(i)のポリヌクレオチドとハイブリッド形成する、ポリヌクレオチド、および
(b)配列番号1の配列からなるポリヌクレオチドとハイブリッド形成せず、(a)のポリヌクレオチドによってコードされるポリペプチドをインフレームで有するポリペプチドをコードするポリヌクレオチド

- BTNL9タンパク質の作製方法であって、
請求項48に記載の宿主細胞を前記核酸の発現に適切な条件下の培地中にて培養する工程、および
該細胞または培養培地から、発現したタンパク質を回収する工程
を含む、方法。 - 自己免疫疾患または炎症性疾患を有する患者の処置方法であって、
(a)配列番号2のアミノ酸35〜257のアミノ酸配列、
(b)配列番号2のアミノ酸35〜257と少なくとも90%同一のアミノ酸配列であって、該アミノ酸配列の、配列番号2のアミノ酸35〜257とのアラインメントウィンドウが少なくとも80アミノ酸長である、アミノ酸配列、または
(c)配列番号2のアミノ酸35〜257の配列と比較して単一のアミノ酸の20個以下の挿入、欠失、または置換を有するアミノ酸配列
を含む治療有効用量のBTNL9タンパク質を該患者に投与する工程を含み、
該BTNL9タンパク質が抗CD3抗体によって刺激されたT細胞の増殖を阻害することができる、方法。 - 前記自己免疫疾患または炎症性疾患が、全身性エリテマトーデス、関節リウマチ、炎症性腸疾患、クローン病、潰瘍性大腸炎、乾癬、サルコイドーシス、喘息、または線維性疾患からなる群から選択される、請求項50に記載の方法。
- 前記自己免疫疾患または炎症性疾患がクローン病である、請求項51に記載の方法。
- 前記自己免疫疾患または炎症性疾患が潰瘍性大腸炎である、請求項51に記載の方法。
- 前記自己免疫疾患または炎症性疾患が線維性疾患である、請求項51に記載の方法。
- T細胞の増殖を阻害するための方法であって、
(a)配列番号2のアミノ酸35〜257のアミノ酸配列、
(b)配列番号2のアミノ酸35〜257と少なくとも90%同一のアミノ酸配列であって、該アミノ酸配列の、配列番号2のアミノ酸35〜257とのアラインメントウィンドウが少なくとも80アミノ酸長である、アミノ酸配列、または
(c)配列番号2のアミノ酸35〜257の配列と比較して単一のアミノ酸の20個以下の挿入、欠失、または置換を有するアミノ酸配列
を含むBTNL9タンパク質を該T細胞に添加する工程を含み、
該BTNL9タンパク質が抗CD3抗体によって刺激されたT細胞の増殖を阻害することができる、方法。 - 前記阻害がin vitroで起こる、請求項55に記載の方法。
- 前記阻害がin vivoで起こる、請求項55に記載の方法。
- 自己免疫疾患または炎症性疾患を有する患者の処置方法であって、治療有効用量の抗BTNL9抗体を該患者に投与する工程を含み、
該抗BTNL9抗体が配列番号2のアミノ酸35〜257の配列を含むBTNL9タンパク質によるT細胞の増殖阻害を増大させ、
該抗BTNL9抗体が配列番号2のアミノ酸35〜257のアミノ酸配列からなるタンパク質に結合する、方法。 - 治療有効量の配列番号2のアミノ酸35〜257からなるBTNL9タンパク質に結合する抗体を癌患者に投与する工程を含む、癌患者の処置方法。
- 前記癌が、急性または慢性白血病、リンパ腫、非ホジキンリンパ腫、ホジキン病、リンパ性白血病、リンパ性リンパ腫または皮膚リンパ腫、癌腫、肉腫、胸腺腫、縦隔の新生物、乳癌、前立腺癌、頭頸部癌、肺癌、非小細胞肺癌、小細胞肺癌、各種皮膚癌、膀胱癌、悪性神経膠腫、食道癌、胃癌、膵臓癌、肝胆管新生物、小腸、結腸、または直腸の癌、腎臓または尿管の癌、睾丸癌、尿道または陰茎の癌、婦人科系腫瘍、卵巣癌、骨の肉腫、内分泌系の癌、皮膚黒色腫、眼内黒色腫、中枢神経系の新生物、および形質細胞新生物からなる群から選択される、請求項59に記載の方法。
- 前記抗体がアンタゴニスト抗体である、請求項59または60に記載の方法。
- 癌に対して患者にワクチン接種するための方法であって、癌細胞に高度に発現する抗原および配列番号2のアミノ酸35〜257からなるタンパク質に結合するアンタゴニスト抗体を該患者に投与する工程を含む、方法。
- 自己免疫状態または炎症状態を有する患者を処置するための方法であって、以下の工程:
(a)該患者からT細胞を取り出す工程、
(b)該T細胞を、抗CD3抗体を含むタンパク質とBTNL9タンパク質との組み合わせで刺激する工程であって、該BTNL9タンパク質が、
(i)配列番号2のアミノ酸35〜257のアミノ酸配列、
(ii)配列番号2のアミノ酸35〜257と少なくとも90%同一のアミノ酸配列であって、該アミノ酸配列の、配列番号2のアミノ酸35〜257とのアラインメントウィンドウが少なくとも80アミノ酸長である、アミノ酸配列、または
(iii)配列番号2のアミノ酸35〜257の配列と比較して単一のアミノ酸の20個以下の挿入、欠失、または置換を有するアミノ酸配列
を含む、刺激する工程、
(c)刺激されたT細胞を採取する工程、および
(d)採取したT細胞を該患者に戻す工程
を含み、
該BTNL9タンパク質が抗CD3抗体によって刺激されたT細胞の増殖を阻害することができる、方法。 - 前記BTNL9タンパク質が、(b)(iii)のBTNL9タンパク質であり、配列番号2のアミノ酸35〜257の配列と比較して単一のアミノ酸の10個以下の挿入、欠失、または置換を有する、請求項63に記載の方法。
- 前記BTNL9タンパク質が、配列番号2のアミノ酸35〜257の配列と比較して単一のアミノ酸の5個以下の挿入、欠失、または置換を有する、請求項64に記載の方法。
- 前記BTNL9タンパク質が(b)(i)のBTNL9タンパク質である、請求項63に記載の方法。
- 前記自己免疫状態または炎症状態が、全身性エリテマトーデス、関節リウマチ、炎症性腸疾患、クローン病、潰瘍性大腸炎、乾癬、サルコイドーシス、喘息、または線維性疾患からなる群から選択される、請求項63から66のいずれか1項に記載の方法。
