Huh7細胞を通常の増殖条件下で増殖させ、0.5%のDMSOを含む溶媒中で指示された量の薬剤で処理した。細胞を5時間薬剤の存在下で増殖させ、次いで、例えば、ATCC#VR‐129Bから入手した、マウスの脳心筋炎ウイルス(EMCV)(250pfu)に感染させた。感染細胞をさらに18時間増殖させ、次いで、MTSアッセイを用いて、細胞生存率を測定した。負の対照の細胞を、0.5%DMSOのみを含む緩衝液で単独で処理した。インターフェロン処理を、正の対照として、ウイルス阻害に用い、最終濃度10 IU/mLの、例えば、インターフェロン‐α:シェリルプラウ(Schering‐Plough)社製のイントロンAを、薬剤処理と同様に加えた。Promega#G3580からのセルタイター96(CellTiter96)(登録商標)のアクエアス・ワン・ソリュージョン・細胞増殖アッセイ(AQueous One Solution Cell Proliferation Assay)(MTS)等のMTSアッセイを用いて、細胞生存率を測定した。
本実施例は、KIN400化合物の抗ウイルス作用の有効性の増加における最適化について、説明する。最適化において、2段階QSARアプローチを用いる;誘導体開発前の構造クラスを確定する、類縁体小集合から開始する。初期に同定した活性誘導体を用いて、第2段階における関心を惹くさらなる最適化の構造クラスの部分集合を確定する。
細胞培養感染モデルにおける抗ウイルス作用。本明細書に開示するKIN400化合物は、HCV遺伝子型2aおよびインフルエンザウイルスWSN株に対して有効な活性を有する。さらに最適化した分子の広範囲の抗ウイルス活性を特性評価するために、細胞培養感染モデルを使用して、種々のHCV遺伝子型およびインフルエンザウイルス菌株を分析する。さらに、新たに公衆衛生に関係する、西ナイルウイルス(WNV)に対する最適化した化合物の活性を試験する。試験には、感染2〜12時間前に、化合物で細胞を処理すること、または感染8時間後に、細胞を処理することが含まれる(表3)。ウイルス生産および細胞のISG発現を経時的に評価して、主要構造クラスの代表的な化合物の抗ウイルス効果を分析する。IFNβ処理を正の対照として用いる。
臨床前薬物動態および耐性のプロファイリング。インフルエンザウイルスおよびWNV感染動物モデルにおけるKIN400化合物の抗ウイルス活性のさらなる特性評価を行うために、KIN400化合物のインビボ薬物動態(PK)プロファイル、および耐性/毒性を評価する。マウスは、WNVおよびインフルエンザの最も一般に使用されるげっ歯動物モデルであるため、マウスをこれらの試験のために選択する。
・第1段階:pH(pH3〜9)、緩衝液、および浸透圧の調整
・第2段階:エタノール(<10%)、プロピレングリコール(<40%)、ポリエチレングリコール(PEG)300または400(<60%)の共溶媒を加えることによる、溶解度の向上
・第3段階:必要に応じて、N‐N‐ジメチルアセトアミド(DMA、<30%)、N‐メチル‐2‐ピロリドン(NMP、<20%)、および/またはジメチルスルホキシド(DMSO、<20%)の共溶媒、またはシクロデキストリン(<40%)を加えることによる、さらなる溶解度の改善。
2.08g(10mmol)のカルコンII、および1g(10mmol)のシアノチオアセトアミドIIIを、メタノール中8.5%ナトリウムメトキシド溶液(15ml)中で溶解し、その溶液を1.5時間、ウォーターバスで加熱する。酢酸で沈殿物を再結晶化し、2.1g(73%)のIV mp 228〜230℃を得る。
10%水酸化カリウム溶液の10mlの試料を、30mlのDMF中ピリジン‐2‐チオールIVの10mmol懸濁液に加え、その後、5mlのDMF中10mmolのN‐クロロ‐アセチルモルホリンV溶液を滴加する。30分後、反応混合物を水で希釈し、沈殿物をろ過によって除去する。試験中の反応混合物の温度は、15〜20℃以内に維持する必要がある。ベンゼンによる再結晶化によって、化合物Iを得る(収率75%)。

- 次に示す構造を有する化合物を含む、医薬組成物:

XおよびYはそれぞれ、独立して、S、O、NH、NR、低級アルキル、置換された低級アルキル、ヘテロアルキル、または置換されたヘテロアルキルであり;
Zは、OH、OR8、NR8R9、CO2H、CO2R8、CONH2、CONR8R9、CONR8OH、CONR8OR9、NR8(C=O)NR9R10、NR8(C=O)R9、NOH(C=O)NR8R9、NR8(C=O)NOR9R10、N‐H テトラゾール、N‐アルキルテトラゾール、1‐アミジン、2‐アミジン、グアニジン、スルホニルアミジン、スルファモイルアミジン、スルホニルグアニジン、スルファモイルグアニジン、N‐シアノアミジン、N‐シアノグアニジン、N‐ニトロアミジン、N‐ニトログアニジン、アミノスクアリン酸、アミノスクアリン酸アミド、CS(OR8)、SO2R8、COR8、CONR8R9、(SO)nNR8R9、NR8(SO)nNR9R10、またはNR8SO2R9から選択され;
R8、R9およびR10はそれぞれ、独立して、H、低級アルキル、アリール、アルケニル、アルキニル、アルキルアリール、アリールアルキル、アルコキシ、アリールオキシ、アリールアルコキシ、ヘテロアルキル、ヘテロアリール、ヘテロアリールアルキル、環状へテロアルキル、環状へテロアルキルアルキルから選択されるか、または、対で一緒になる場合、モルホリン、ピペリジン、ピロリジン、ピラゾール、イミダゾール、もしくはテトラゾールを含むが、これらに限定されない、環状基を形成してもよく;
W1、W2、W3はそれぞれ、独立して、CまたはNであり;かつ、
n=0、1、2、3、4または5である)。 - 請求項1記載の化合物もしくはその医薬として許容し得る塩、これらの互変異性体、異性体および/またはプロドラッグを含む、請求項1記載の医薬組成物。
- 脊椎動物に、請求項2記載の医薬組成物を投与することを含む、脊椎動物におけるウイルス感染を治療するかまたは予防する方法。
- 前記ウイルス感染が、次に示すファミリーの1以上のウイルスによって発生する、請求項9記載の方法:アレナウイルス科(Arenaviridae)、アストロウイルス科(Astroviridae)、ビルナウイルス科(Birnaviridae)、ブロモウイルス科(Bromoviridae)、ブニヤウイルス科(Bunyaviridae)、カリシウイルス科(Caliciviridae)、クロステロウイルス科(Closteroviridae)、コモウイルス科(Comoviridae)、シストウイルス科(Cystoviridae)、フラビウイルス科(Flaviviridae)、フレキシウイルス科(Flexiviridae)、ヘペウイルス(Hepevirus)、レヴィウイルス科(Leviviridae)、ルテオウイルス科(Luteoviridae)、モノネガウイルス目(Mononegavirales)、モザイクウイルス(Mosaic Viruses)、ニドウイルス(Nidovirales)、ノダウイルス科(Nodaviridae)、オルソミキソウイルス科(Orthomyxoviridae)、ピコビルナウイルス(Picobirnavirus)、ピコルナウイルス科(Picornaviridae)、ポチウイルス科(Potyviridae)、レオウイルス科(Reoviridae)、レトロウイルス科(Retroviridae)、セキウイルス科(Sequiviridae)、テヌイウイルス(Tenuivirus)、トガウイルス科(Togaviridae)、トンブスウイルス科(Tombusviridae)、トチウイルス科(Totiviridae)、ティモウイルス科(Tymoviridae)、ヘパドナウイルス科(Hepadnaviridae)、ヘルペスウイルス科(Herpesviridae)、パラミキソウイルス科(Paramyxoviridae)、またはパピローマウイルス科(Papillomaviridae)。
- 前記ウイルス感染が、インフルエンザウイルス、C型肝炎ウイルス、西ナイルウイルス、SARSコロナウイルス、ポリオウイルス、麻疹ウイルス、デング熱ウイルス、黄熱ウイルス、ダニ媒介性脳炎ウイルス、日本脳炎ウイルス、セントルイス脳炎ウイルス、マーレーバレーウイルス(Murray Valley virus)、ポワサンウイルス(Powassan virus)、ロシオウイルス(Rocio virus)、跳躍病ウイルス、バンジウイルス(Banzi virus)、イルヘウスウイルス(Ilheus virus)、ココベラウイルス(Kokobera virus)、クンジンウイルス(Kunjin virus)、アルフィウイルス(Alfuy virus)、ウシ下痢症ウイルス、キァサヌール森林病ウイルス(Kyasanur forest disease virus)、またはヒト免疫不全ウイルス(HIV)である、請求項9記載の方法。
- 前記方法が、インフルエンザウイルス、C型肝炎ウイルス、西ナイルウイルス、SARSコロナウイルス、ポリオウイルス、麻疹ウイルス、デング熱ウイルス、黄熱ウイルス、ダニ媒介性脳炎ウイルス、日本脳炎ウイルス、セントルイス脳炎ウイルス、マーレーバレーウイルス(Murray Valley virus)、ポワサンウイルス(Powassan virus)、ロシオウイルス(Rocio virus)、跳躍病ウイルス、バンジウイルス(Banzi virus)、イルヘウスウイルス(Ilheus virus)、ココベラウイルス(Kokobera virus)、クンジンウイルス(Kunjin virus)、アルフィウイルス(Alfuy virus)、ウシ下痢症ウイルス、キァサヌール森林病ウイルス(Kyasanur forest disease virus)、またはヒト免疫不全ウイルス(HIV)に対するワクチンをさらに接種することにより、脊椎動物にワクチン接種することを含む、請求項9記載の方法。
- 真核細胞に、請求項2記載の化合物を投与することを含む、真核細胞における自然免疫応答を調節する方法。
