Abstract
Description
本出願は、その全内容が参照により本明細書に組み入れられる2010年1月28日に出願された米国特許仮出願第61/299,162号の権利についての優先権を主張する。
モノクローナル抗体は、抗原を高度な特異性で結合できるその能力のため、研究、診断および治療用試薬として幅広く用いられている。抗原に対するその特異的結合に加えて、モノクローナル抗体は、そのFc領域を介して補体系およびエフェクター細胞を活性化し得る。抗体の生物学的特性を正確に解明するために、適切な対照が必須である。適切な対照なくしては、抗体の特異的結合活性と抗体の生化学的および生物学的な効果の間の因果関係を立証することは困難である。
本発明は、抗体の三次元構造を実質的に改変することなく抗原結合を減少させるかまたは除去する十分に特徴付けられた可変ドメインを有する、親和性が低下した新規の抗体を提供する。本発明をより簡単に理解できるようにするために、ある特定の用語および句を以下ならびに明細書全体を通して定義する。
本発明の親和性が低下した抗体は、抗原に対する結合を除去するかまたは減少させるために合理的に設計された抗体である。現時点で好ましい親和性が低下した抗体は、CDR領域を除いては全ての点で野生型(天然)抗体に類似し、かつ、野生型抗体に対して識別可能な三次元構造変化を殆ど示さない抗体である。例示的な抗体は、10倍低下した抗原に対する結合親和性を示すものである。好ましくは、親和性が低下した抗体は、100倍低下した抗原に対する結合親和性で抗原に結合する。より好ましくは、親和性が低下した抗体は、1000倍低下した抗原に対する結合親和性で抗原に結合する。最も好ましくは、親和性が低下した抗体は、抗原に対する実質的な結合を全く示さず、かつ、バックグラウンド結合レベルを超える抗原への特異的結合は検出できない。親和性は、表面プラズモン共鳴、または、放射性リガンド結合試験、または、フローサイトメトリーを含む一般的な実験方法により測定できる。本発明の親和性が低下した抗体は、抗原に対する特異的結合を保持するその野生型(天然)対応物のための対照試薬として、抗体の使用を含む多くのインビボおよびインビトロの適用において有用である。
本発明の一局面は、本発明のポリペプチドをコードする単離された核酸分子、ならびに、これらのポリペプチドをコードする核酸分子を同定するためのハイブリダイゼーションプローブとして充分な核酸断片、および、核酸分子の増幅または突然変異のためのPCRプライマーとして使用するための断片と関係する。本明細書中で使用される場合、「核酸分子」または「ポリヌクレオチド」という用語は、DNA分子(例えば、cDNAまたはゲノムDNA)およびRNA分子(例えば、mRNA)および、ヌクレオチドアナログを用いて作製されたDNAまたはRNAのアナログを含むよう意図されている。核酸分子は一本鎖または二本鎖であり得るが、好ましくは二本鎖DNAである。
本発明の一局面は単離されたポリペプチドに関係する。一態様において、本発明のポリペプチドは、細胞または組織源から、標準的なタンパク質精製技術を用いた適切な精製スキームにより単離できる。別の態様においては、本発明のポリペプチドは組換えDNA技術により産生される。あるいは、本発明のポリペプチドは標準的なペプチド合成技術を用いて化学的に合成できる。
本発明の別の局面は、本発明のポリペプチド(またはその一部)をコードする核酸分子を含有するベクター、好ましくは発現ベクターに関係する。本明細書中で使用される場合、「ベクター」という用語は、それに連結された別の核酸を運ぶことができる核酸分子を指す。ベクターの一つの型は、追加のDNAセグメントを連結することができる環状の二本鎖DNAループを指す「プラスミド」である。別の型のベクターは、ウイルスゲノム中に追加のDNAセグメントをすることが連結できるウイルスベクターである。あるベクターは、それが導入された宿主細胞中で自己複製できる(例えば、細菌の複製起点を有する細菌ベクター、および哺乳類のエピソームベクター)。その他のベクター(例えば、哺乳類の非エピソームベクター)は、宿主細胞への導入後、宿主細胞のゲノム中に組み込まれ、かつそれにより宿主ゲノムと共に複製される。さらに、あるベクターは作動可能に連結された遺伝子の発現を指示することができる。このようなベクターは本明細書中で「発現ベクター」と呼ばれる。一般に、組換えDNA技術において役立つ発現ベクターはしばしばプラスミドの形態をとる。プラスミドはベクターの最も一般的に使用される形態であるため、本明細書中において「プラスミド」および「ベクター」は互換的に使用できる。しかしながら、本発明は、同等の機能を提供するウイルスベクター(例えば、複製欠損レトロウイルス、アデノウイルスおよびアデノ関連ウイルス)等のこのような他の形態の発現ベクターも含むよう意図されている。
親和性が低下した抗体は、例えば、所定の治療レジメンの有効性を判定するために、臨床試験工程の一部として組織中のポリペプチドレベルを診断的または予後的にモニターするように設計されたアッセイ法において使用できる。抗体を検出可能な物質にカップリングすること(即ち、物理的に連結すること)により検出を容易にできる。検出可能な物質の例は、種々の酵素、補欠分子団、蛍光材料、発光材料、生物発光材料および放射活性材料を含む。好適な酵素の例は、西洋ワサビペルオキシダーゼ、アルカリホスファターゼ、P-ガラクトシダーゼまたはアセチルコリンエステラーゼを含み、好適な補欠分子団複合体の例は、ストレプトアビジン/ビオチンおよびアビジン/ビオチンを含み、好適な蛍光材料は、ウンベリフェロン、フルオレセイン、フルオレセインイソチオシアネート、ローダミン、ジクロロトリアジニルアミンフルオレセイン、ダンシルクロリドまたはフィコエリトリンを含み、発光材料の例はルミノールを含み、生物発光材料の例はルシフェラーゼ、ルシフェリンおよびエクオリンを含み、ならびに、好適な放射活性材料の例は125I、131I、35Sおよび3Hを含む。
抗体構造または発現に影響することなく抗原結合を減少させるかまたは除去するため、OKT3相補性決定領域のアミノ酸配列を改変した。本発明の対照抗体の例示的なCDR配列を表3に列挙する。親和性が低下した得られた抗体は、正常に発現し(図3)、かつ、ヒトジャーカット細胞(図4)またはヒト末梢血単核球(PBMC、図5)を結合する能力が最小限であるかもしくはそのような能力が無かった。
ヒト293T細胞は、10%ウシ胎児血清(FBS)(Invitrogen, Gibco, Carlsbad, CA)、2mM L-グルタミンおよび40ug/mlのゲンタマイシンを補充したダルベッコ変法イーグル培地(DMEM)中で維持された。抗ヒトCD3抗体産生ハイブリドーマ細胞株であるマウスOKT3は、10%FBS、1mMピルビン酸ナトリウム、2mM L-グルタミンおよび40ug/mlのゲンタマイシンを含有するIscoveの変法ダルベッコ培地(IMDM)中で生育させた。ヒト白血病T細胞ラインのジャーカットE6.1を10%FBS、2mM L-グルタミンおよび40ug/mlゲンタマイシンを補充したRPMI培地中で生育させた。全ての培地および補充物は、FBSを除いて、Lonza、Walkville、MDから購入した。
総RNAをOKT3ハイブリドーマ細胞株からInvitrogenのTRIzol試薬を用いて単離した。SuperScript(商標)III逆転写酵素(Invitrogen)およびオリゴ(dT)12-18プライマー(Invitrogen)を用いてcDNAを合成した。マウスOKT3重鎖(アクセッション番号A22261)および軽鎖(アクセッション番号A22259)ヌクレオチド配列のGenbank記録に基づいて、OKT3ハイブリドーマのdTプライミングされたcDNAを鋳型としてポリメラーゼ連鎖反応(PCR)により重鎖および軽鎖の全コード配列を増幅し、かつ、pMEと呼ばれるCMV最初期遺伝子プロモーターにより駆動される哺乳動物発現プラスミド中にクローニングした。プラスミドpME-wtOKT3 HC(図6)の全ヌクレオチド配列はSEQ ID NO:23に表され、かつ、プラスミドpME-wtOKT3LC(図7)の全ヌクレオチド配列はSEQ ID NO:24に表される。
CDR内の種々の抗原接触アミノ酸をアラニンで置換したOKT3重鎖および軽鎖バリアントを作製するために、標準的なPCR技術を用いて部位特異的突然変異誘発を行った。突然変異を持つPCR断片を、同じ発現プラスミドpME中にクローニングした。得られたプラスミドをシークエンシング反応により確認した。
標準的なリン酸カルシウム法を用いて、OKT3抗体およびバリアント抗体を一過性形質移入により産生した。簡単にいうと、形質移入の20〜24時間前に293T細胞を6ウェルプレート中に6×105細胞/3ml 293T培地/ウェルでまいた。形質移入の3時間前に培地を10%FBS、25mMヘペスバッファーおよび40ug/mlゲンタマイシンを補充したIMDMに替えた。形質移入工程は、最初に、OKT3重鎖および軽鎖プラスミドDNAを、各々8μg、31μlの2M CaCl2および滅菌水と最終容量250μlになるように混合することにより行った。その後DNA/カルシウム混合物をゆっくりと250μlの2×HBSバッファー(pH7.12において281mM NaCl、100mM HEPES、1.5mM Na2HPO4)に添加した。形質移入混合物を、室温で20分間インキュベートし、かつその後293T培養物にゆっくりと添加した。形質移入から12〜16時間後、培地を再び完全293T培地に替えた。形質移入から40〜48時間後、OKT3バリアント抗体を含有する培養上清を回収し、かつ続いての分析に使用した。
一過性形質移入により産生されたOKT3抗体およびバリアント抗体の量を、製造者によるプロトコールに従い、Guava RapidQuantマウスIgGキット(Guava Technologies, Hayward, CA)によって判定した。簡単にいうと、各形質移入由来の10μlの上清をIgG捕獲ビーズと40分間震盪しながらインキュベートした。続いて、FITC結合ヤギ抗マウスIgGを各試料に添加し、かつ60分間震盪しながらインキュベートした。試料の容量を200μlまで増やし、Guava EasyCyteシステム(Guava)に流した。各試料中の抗体濃度は、試料の平均蛍光チャンネル(MFI)を標準のそれと比較することにより算出した。
ヒト末梢単核細胞(hPBMC)をFicoll-paque勾配法を用いて単離した。簡単にいうと、ヒト全血を、1×リン酸緩衝塩類溶液(PBS)で1:1希釈し、かつ注意深くFicoll-paque(血液試料容量の0.4容量)(GE Healthcare, Uppsala, Sweden)の一番上に載せた。900gにおける30分間の遠心分離の後、血液とFicoll-paqueの境界面のhPBMCを注意深く抽出した。細胞懸濁液を1×PBSで1:3希釈し、かつ400gで10分間、8〜20℃において回転させた。細胞ペレットを、もう一回1×PBSにより400gで10分間、8〜20℃で洗浄し、かつ、フローサイトメトリー分析のためにFACSバッファー(2%FCS、0.1%NaN3を加えたPBS)で希釈した。
ヒトジャーカット細胞(96ウェルプレート中において1×105細胞/ウェル)またはhPBMC(96ウェルプレート中において3×105細胞/ウェル)を、100μlのOKT3およびバリアントの形質移入上清と共に4℃で60分間インキュベートし、かつ200μlのFACSバッファーで2回洗浄した。細胞を100μlのフィコエリトリン(PE)結合ヤギ抗マウス免疫グロブリンG(Invitrogen-Caltag, Carlsbad, CA)により4℃で60分間染色し、かつ200ulのFACSバッファーで2度洗浄した。その後、細胞を1%パラホルムアルデヒドで15分間、4℃において固定し、かつ、FACSバッファーで一度洗浄した。その後、Guava EaseCyte Plusシステム(Guava Technologies, Inc., Hayward, CA)における分析のために、細胞を200μlのFACSバッファーに再懸濁した。
総数1×105個のジャーカット細胞を連続的に希釈したOKT3抗体またはOKT3バリアント抗体、続いてPE結合ヤギ抗マウスIgGとインキュベートし、かつFACSにより分析した。1600ng/mlで、OKT3バリアントの結合は0.4ng/mlのOKT3抗体と匹敵する。従って、OKT3バリアントの結合親和性は親のOKT3抗体のそれよりもおよそ4000倍小さい(図8)。
バリアントIgG2a抗体の薬物動態分析。CDR突然変異重鎖(SEQ ID NO:26)およびCDR突然変異軽鎖(SEQ ID NO:47)の共発現によりIgG2a抗体を産生した。このバリアント抗体が親抗体OKT3の抗原であるCD3に最早結合しないことは明らかであるものの、この抗体がまだ別の抗原と思いがけなく相互作用するかどうかは不明である。これを試験する一つの方法は、内因性循環IgG2a抗体と類似のインビボ半減期を有するかどうか判定することである。バリアント抗体をB6マウスに投与し、かつ、21日目まで血清試料を集めた。組換えIgG2a抗体の濃度をアロタイプ特異的抗マウス抗体により測定した。抗体の半減期は、薬物動態プロファイルのβ相を当てはめることにより算出し、かつ、13.75日であると判定された(図9)。この半減期は、マウスIgG2a抗体について報告されている10〜15日の半減期と良く一致している(Talbotp, P.J.およびBuchmeie M.J. "Catabolism of homologous murine monoclonal hybridoma IgG antibodies in mice." Immunology 60: 485-489, 1987)。長い血清半減期は、この抗体が予期されない抗原と相互作用する可能性が低いこと、および、インビボシステムがこのバリアント抗体を同じアイソタイプの内因性の免疫グロブリンと区別できないことを示唆する。

本発明1016の、低下した結合能力を持つ抗体または抗体断片。
総RNAをOKT3ハイブリドーマ細胞株からInvitrogenのTRIzol試薬を用いて単離した。SuperScript(商標)III逆転写酵素(Invitrogen)およびオリゴ(dT)12-18プライマー(Invitrogen)(SEQ ID NO: 15)を用いてcDNAを合成した。マウスOKT3重鎖(アクセッション番号A22261)および軽鎖(アクセッション番号A22259)ヌクレオチド配列のGenbank記録に基づいて、OKT3ハイブリドーマのdTプライミングされたcDNAを鋳型としてポリメラーゼ連鎖反応(PCR)により重鎖および軽鎖の全コード配列を増幅し、かつ、pMEと呼ばれるCMV最初期遺伝子プロモーターにより駆動される哺乳動物発現プラスミド中にクローニングした。プラスミドpME-wtOKT3 HC(図6)の全ヌクレオチド配列はSEQ ID NO:23に表され、かつ、プラスミドpME-wtOKT3LC(図7)の全ヌクレオチド配列はSEQ ID NO:24に表される。
Claims (28)
- 改変された抗体または抗体断片が抗原に対する低下した結合能を有するように、低下した結合能力を持つ該抗体または該抗体断片を産生するための方法であって、以下の工程を含む方法:
(a)標的抗原の情報を取得する工程;
(b)該抗体内または該抗体断片内において、標的抗原と相互作用することができる結合アミノ酸を合理的に同定する工程;
(c)得られる抗体または抗体断片が該抗原との結合における低下した能力を有するように、該抗体内または該抗体断片内の結合アミノ酸配列を改変する工程;および
(d)改変された該抗体または該抗体断片の該標的抗原に対する結合親和性を試験する工程。 - 抗原との相互作用を減少させるかまたは除去するために、低下した特異的結合能力を持つ相補性決定領域(CDR)を産生するための方法であって、以下の工程を含む方法:
(a)重鎖可変領域または軽鎖可変領域であるCDRポリペプチド配列内から、該CDRポリペプチド配列中の少なくとも1つのアミノ酸を同定する工程であって、該アミノ酸が該抗原と相互作用することができる、工程;および
(b)該CDRポリペプチドのアミノ酸配列を改変する工程。 - 前記CDRポリペプチド配列中のアミノ酸を改変する前記工程が、少なくとも1つのアミノ酸を相互作用しないアミノ酸と置換することにより行われる、請求項2記載の方法。
- 1つのアミノ酸と前記抗原の間の相互作用が、水素結合、弱い共有結合性相互作用、極性力(polar force)、非極性力、塩橋およびファンデルワールス力からなる群より選択される分子間引力である、請求項2記載の方法。
- 前記CDRポリペプチド配列中の前記アミノ酸が、アスパラギン酸、グルタミン酸、リジン、アルギニン、セリン、スレオニン、チロシン、アスパラギン、ヒスチジンおよびグルタミンからなる群より選択される、請求項2記載の方法。
- 前記CDRポリペプチド配列中の前記アミノ酸が、チロシン、セリン、アスパラギン、アスパラギン酸、トリプトファン、アルギニン、フェニルアラニンおよびグリシンからなる群より選択される、請求項2記載の方法。
- 前記相互作用しないアミノ酸が、アラニン、バリン、ロイシン、イソロイシン、プロリンおよびメチオニンからなる群より選択される、請求項3記載の方法。
- 前記相互作用しないアミノ酸がアラニンである、請求項6記載の方法。
- 相互作用する前記アミノ酸を同定する前記工程が、前記CDRポリペプチド配列と前記抗原との複合体を表す結晶構造を分析することにより行われる、請求項2記載の方法。
- 前記CDRポリペプチド配列が抗体内または抗体断片内に位置する、請求項2記載の方法。
- 前記抗体または前記抗体断片が、
(a)免疫グロブリン分子全体;
(b)scFv;
(c)Fab断片;
(d)Fab'断片;
(e)F(ab')2;
(f)Fv;
(g)Fd断片;および
(h)ジスルフィド結合したFv
