Abstract
Description
アンチセンスヌクレオチドは、ワトソン・クリック型塩基対で定義されることによって、DNA、未成熟−mRNAまたは成熟したmRNAの相補的塩基配列に結合(混成化)して、DNAからタンパク質として遺伝子情報の流れを妨げるものである。標的配列に特定性があるアンチセンスヌクレオチドの性質は、それらを例外的に多機能にする。アンチセンスのヌクレオチドは、モノマー単位の長い鎖であるため、これらの標的RNA配列に対して簡単に合成することができる。最近の多くの研究は、標的タンパク質の研究のための生化学的手段としてアンチセンスヌクレオチドの有用性を証明した(Rothenbergら.,J.Natl.Cancer Inst,1999年,第81巻,p.1539−1544)。オリゴヌクレオチド化学および向上した細胞株吸着、標的結合親和度およびヌクレアーゼ耐性を示すヌクレオチド合成分野で、最近多くの進歩があったため、アンチセンスヌクレオチドの使用は、新しい形の抑制剤として考慮することができる。
前記のペプチドミネティックスは、TIP41活性を率いるTIP41タンパク質の結合ドメインを抑制するペプチドまたはペプチドである。非加水分解性ペプチド類似体の重要な残基としては、βターンペプチドコア(Nagaiら,Tetrahedron Lett,1985年,第26巻,p.647)、ケトメチレンシュードペプチド類(Ewensonら,J Med chem,1986年,第29巻,p.295;およびEwensonら,in Peptides:Structure and Function(Proceedings of the 9th AmeriCan Peptide Symposium)Pierce chemiCal co.Rockland,IL,1985年)、アゼピン(Huffmanら,in Peptides:chemistry and Biology,G.R.Marshall ed.,EScOM Publisher:Leiden,Netherlands,1988年)、ベンゾジアゼピン(Freidingerら,in Peptides;chemistry and Biology,G.R.Marshall ed.,EScOM Publisher:Leiden,Netherlands,1988年)、βアミノアルコール(Gordonら,Biochem Biophys Res commun,1985年,第126巻,p.419)、および置換されたγ−ラクタム環(Garveyら,in Peptides:chemistry and Biology,G.R.,Marshell ed.,EScOM Publisher:Leiden,Netherlands,1988年)を用いて作成することができる。
センスRNAとアンチセンスRNAが二重鎖RNA分子を形成し、ここで、センスRNAがTIP41 mRNA中の一部の連続ヌクレオチドの標的配列と同一な核酸配列を含むsiRNA分子であることが好ましい。前記のsiRNA分子は、TIP41遺伝子の塩基配列内で選択される10〜30個の塩基からなるセンス配列および、前記のセンス配列に相補的に結合するアンチセンス配列からなることが好ましいが、これに限られるわけではなく、TIP41遺伝子の塩基配列を対象に相補的に結合できるセンス配列を有する二重鎖RNA分子ならすべて使用可能である。前記のセンス配列は、センス配列と相補的な配列を有していることが最も好ましい。
TIP41抗体は、TIP41注入によって製造されたもの、または市販され購入したものなどすべて使用可能である。また、前記の抗体は、多クローン抗体、モノクローナル抗体およびエピトープと結合することのできる断片等を含む。
アプタマーは、それ自体で安定した三次構造を有しながら標的分子に高い親和性と特異性で結合することができる特性を有する単一鎖核酸(DNA、RNA、または変形核酸)である。アプタマーは、SELEX(Systematic Evolution of Ligands by EXponential enrichment)と呼ばれるアプタマー発掘技術が最初に開発された後(Ellington,AD and Szostak,JW,Nature,1990年,第346巻,p.818−822)、低分子有機物、ペプチド、膜タンパク質まで、多様な標的分子に結合することができる多くのアプタマーが、継続して発掘された。アプタマーは、固有の高い親和性(普通pM水準)と特異性で標的分子に結合することができるという特性によって、単一抗体と比較され、特に「化学抗体」と呼ばれる程、代替抗体としての高い可能性がある。
(1)実験群として、癌細胞株に被検物質を処理する工程、
(2)工程(1)の処理された細胞でTIP41とPP2Acタンパク質間の結合水準またはTIP41とMKK7タンパク質間の結合水準を確認する工程、および
(3)工程(2)のTIP41とPP2Acタンパク質間の結合水準またはTIP41とMKK7タンパク質間の結合水準を対照群と比較して減少させる被険物質を選別する工程。
(1)実験群として、癌細胞株に被検物質を処理する工程、
(2)工程(1)で処理された細胞株でMKK7タンパク質の活性程度を測定する工程、および
(3)工程(2)のMKK7タンパク質の活性を対照群と比較して増加した被険物質を選別する工程を含む、癌の予防または治療剤のスクリーニング方法。
(1)実験群として、癌細胞株に被検物質を処理する工程、
(2)工程(1)で処理された細胞株でPP2Acタンパク質の発現程度を測定する工程、および
(3)工程(2)のPP2Acタンパク質の発現量を対照群と比較して減少させた被検物質を選別する工程。
<1−1>免疫染色方法を用いたTIP41発現の確認
患者の肝癌組織(忠南大学校医科大学キム・ジンマン教授チーム)を10%中性バッファーホルマリン溶液で固定して、パラフィンを添加した後、5μm厚に切断した。切断されたセクション(section)を10mMのクエン酸バッファー(pH6.0)で、4分間処理した後、3%過酸化水素(H2O2)が含まれている0.1M Tris緩衝食塩水(TBS、pH7.4)に30分間置いた。前記のセクションをProtein Block Solution(DAKO)で、常温で20分間処理した後、アンチTIP41抗体と30分間反応させた。0.1M TBST(0.01% ツイーン20を含む0.1M TBS)で洗浄し、セクションをエヌビジョンアンチウサギポリマー(nVision anti−rabbit polymer)(DAKO)と30分間反応させた。ペルオキシダーゼが結合した抗体を3,3'−ジアミノベンジジン色原体基質溶液(3,3 '−diaminobenzidine(DAB)chromogen substrate solution)(DAKO)と反応させて、可視化した。顕微鏡下で観察して、適切に染色された時、0.1M TBSで洗浄して反応を終了させた後、オリンパスBX51顕微鏡(オリンパス、日本)を用いて観察し、オリンパスDP70カメラ(オリンパス、日本)でイメージを得た。
B型肝炎ウイルス(HBV)によって発病した肝癌患者7人とそうでない肝癌患者7人の肝癌組織および隣接正常組織(忠南大学校医科大学)から抽出されたタンパク質を用いて、TIP41タンパク質発現量をウエスタンブロット方法を用いて確認した。
韓国の細胞株銀行から分譲されたヒトのHuh7肝癌細胞株は、10%の牛胎児血清を含んだダルベッコの改変イーグル培地(Dulbecco's modified eagle's medium)(DMEM)で37℃、5%CO2で培養した。SiRNAをトランスフェクションさせるためにHuh7肝癌細胞株1×106細胞/100mm皿に植えて24時間培養し、その後、トランスフェクションを行なった。
TIP41のTRAILセンシタイザーとしての機能を確認するためにsiRNA(Dharmacon RNAi Technologies,米国)を選別して効果的にTIP41タンパク質の発現を抑制する配列を構築した。
<2−1>核染色法を用いたTIP41タンパク質発現抑制で誘導されるアポトーシス分析
6ウェルプレートで前記実験例<1−1>の方法で培養されたTIP41発現抑制された細胞株を2×105細胞/ウェルずつ分注した。24時間後、TRAILを100ng/mlで各々0、2、4、6、8時間処理した後、Hoechst33342を5μg/mlで30分間室温で染色した。Hoechst33342で核を染色した後、蛍光顕微鏡で死滅した細胞数をカウントして、アポトーシスを測定した。
前記の実験例<1−1>の方法で培養したHuh7肝癌細胞株にTIP41 siRNAをトランスフェクションさせて、細胞内TIP41発現量を減少させた後、TRAILを100ng/mlで処理した後、0、2、4、6、8時間帯にアポトーシスをアネキシンV−FITc/PI染色法で死滅した細胞株と正常細胞株を区別して染色した後、FACS(蛍光活性化セルソーター)を用いてアポトーシスを測定した。
<3−1>カスパーゼの活性化
TIP41 siRNAとTRAILを処理した後に誘導されるアポトーシスの経路を確認するために、TIP41 siRNAをトランスフェクションさせた72時間後、TRAILを100ng/mlで0、0.5、1、2、4、6時間処理した後、カスパーゼ−3、−9、−9およびポリ(ADPリボース)重合酵素(PARP)の発現をウエスタンブロットを用いて測定した。
アポトーシスの経路を確認するために、ミトコンドリア(mitochondria)のシトクロムC(Cytochrome C)が細胞質に放出されるかどうかを確認するために、細胞分画(subcellular fractionation)およびウエスタンブロットを用いた。前記の実験例<1−1>の方法で培養したHuh7肝癌細胞株にTIP41 siRNAをトランスフェクションさせて、細胞内TIP41発現量を減少させた後、60mm皿にTIP41の発現が抑制された細胞株を2×106細胞/皿に分注した。24時間後、TRAILを100ng/mlで、各々0、1、2、4時間処理した後、細胞分画を行なった。細胞分画は、Subcellular Proteome Extraction kit(Calbiochem社)を用いて行ない、キット(kit)は、4種類のバッファーで構成され、各バッファーの使用に応じて細胞質、ミトコンドリア、核および細胞骨格タンパク質の順で細胞分画が分離された。溶出バッファー1(extraction buffer 1)500μlを各々の細胞株に添加して、4℃で10分間撹拌した後、遠心分離して細胞質分画を得、溶出バッファー2 500μlを4℃で30分間撹拌した後、遠心分離してミトコンドリアの分画を得た。分画された細胞質とミトコンドリアにウエスタンブロットを用いた。
<4−1>TIP41タンパク質発現抑制およびTRAIL処理で誘導されるJNK伝達経路のタンパク質活性
自己アポトーシスの場合、JNKリン酸化による経路が多くの論文で報告されており、p38の活性化もまたアポトーシスを誘導する可能性があるという報告があった。
JNK伝達経路とTIP41抑制とTRAIL処理で誘導されるアポトーシス間の関連性を確認するために、実験例<4−1>と同一な方法で細胞株を培養し、TIP41 siRNAを処理した後、JNK阻害剤であるSP600125(10μg/ml)を1時間処理してJNK伝達経路を遮断した。TRAIL刺激で誘導されるアポトーシスをアネキシンV−FITc/PI染色法を用いたFACS分析法を用いて測定した。
<5−1>肝癌細胞でp53非依存的アポトーシス経路の確認
現在使用されている抗癌剤の最も大きな問題点は、正常細胞株と癌細胞株を区別できず、正常細胞株を殺すという副作用を有しており、このような正常細胞の死滅機序は、p53経路の活性化を通して起きることが報告された。P53依存的アポトーシスを誘導する抗癌剤は、癌細胞株のみならず、正常細胞株にも影響を与えるので、TIP41 siRNAを処理した後、TRAILによって誘導されるアポトーシスがp53を通じて行なわれるのかどうかを確認した。
HCT116細胞株は、正常的にp53タンパク質を作る大腸癌細胞株であり、TRAIL耐性細胞株にTRAIL単一処理によってアポトーシスが頻繁に発生しないことが知られている。HCT116 p53欠如細胞は、Dr.B.Vogelsteinによって1998年に確立された細胞株で、p53遺伝子が欠損している(Vogelstein,B.,Science,1998年)。前記の細胞株の分譲を受けて実験に用い、細胞株は10%牛胎児血清、100mg/ml、ストレプトマイシン、そして100 IU/mlアンピシリンを含んだDMEM(Hyclone,米国)培地に、安定な抗生剤マーカーG418を100μg/ml添加して、3日周期で継代培養した。
P53非依存的である可能性を確認するために、前記の実験例<5−2>制作したHCT116 p53欠損細胞株にTIP41 siRNAをHuh7肝癌細胞株の場合と同じ実験方法でトランスフェクションした後、同一な条件でTRAILを処理して、アポトーシスを誘導した後、アネキシンV−FITc/PI染色法を用いたFACS分析法を行なった。
<6−1>細胞毎週TRAIL受容体の発現の確認
TRAIL抵抗性を克服させて、癌細胞株特異的な治療剤としての効能を高めるために、癌細胞株のアポトーシスを増加させる方法として、TRAIL受容体の発現を増加させる研究結果が報告された。このような研究と関連してTIP41遺伝子の発現抑制を通じたアポトーシスとTRAIL受容体との相関関係を確認するために、正常細胞株と肝癌および肺癌細胞株でTRAIL受容体の発現様相を確認した。TRAIL受容体は4種類に区分され、TRAIL−R1(DR4)、TRAIL−R2(DR5)、TRAIL−R3(DcR1)およびTRAIL−R4(DcR2)で、R1とR2は、細胞死受容体に作用してTRAILが結合すると、細胞株を自己アポトーシスが起きるように誘導するが、R3とR4はデコイ受容体で、TRAILがR1とR2に結合することを阻害してTRAILによるアポトーシスを阻害すると報告されている。
TIP41タンパク質のノックダウン(knockdown)で誘導されたアポトーシスとTRAIL受容体との相関関係を確認するために、TRAILおよびTIP41 siRNAで処理されたHuh7肝癌細胞株でのTRAIL受容体の発現水準を測定した。TIP41 siRNAをHuh7肝癌細胞株にトランスフェクション72時間後、100ng/mlのTRAILを4時間および6時間処理した。TRAILを処理した後、定量的リアルタイムPCRを行なうためにHuh7肝癌細胞株をトリゾールを用いてRNAを抽出した後、抽出したRNA1μgの逆転写酵素superscriptII(invitrogen)を用いてcDNAを合成し、それを鋳型に、TIP41、TRAIL−R1(Death receptor(DR)4)、TRAIL−R2(DR5)、TRAIL−R3(DcR1)およびTRAIL−R4(DcR2)遺伝子のプライマーを用いて、定量的リアルタイムPCRを行なった。PCRの結果は、2−DDct比較法を用いて、各遺伝子の相対的発現量を測定した。
肝癌組織とそれに隣接する組織で、TRAIL受容体の発現様相をリアルタイム逆転写重合反応(Reverse Transcriptase−PCR)法で測定した。実験に使用した組織は、エドモンソン(Edmondson)及びステイナー(Steiner)分類法によってI(n=5)、II(n=5)、III(n=5)およびIV(n=4)に分類した、各肝癌段階にある患者の肝癌組織(hepatocellular Carcinoma、HCC)と周辺の正常組織を含む19の組織を、カトリック医大(Catholic University of Medicine in Seoul,韓国)で外科手術前の患者の同意の下で採取した。
<7−1>FACS分析によるTIP41発現の抑制とTRAIL処理による正常細胞株でのアポトーシス分析
TIP41は、癌細胞株に特異的に発現するが、正常細胞株でも発現するため、TIP41の発現を抑制した時、癌細胞株のように正常細胞株のアポトーシスが誘導される可能性があるため、TIP41タンパク質の発現抑制とTRAIL処理で誘導されるアポトーシスが癌細胞株特異的な反活性であることを確認するために、正常細胞株であるHAECにTIP41タンパク質発現抑制とTRAIL処理によってアポトーシスを誘導した。
TIP41 siRNAを前記と同一な実験方法で正常細胞株にトランスフェクションさせた後、カスパーゼ−8、−3、JNK、PARPタンパク質の活性化を確認した。正常細胞株に100ng/TRAILを0、2、4時間処理し、その後、アポトーシス促進性タンパク質の発現および活性を前記で記載したウエスタンブロット方法を用いて確認した。
<8−1>肺癌細胞株でTIP41タンパク質発現抑制を通じたTIP41媒介アポトーシス
<8−1−1>ウエスタンブロット方法によるTIP41タンパク質発現抑制で誘導される肺癌細胞株のアポトーシスの確認
前記の<実験例>で用いた肝癌細胞株以外のTRAIL耐性癌細胞株でTIP41タンパク質発現抑制によるTRAILの耐性克服するセンシタイザーとしての機能を検証するために、肺癌細胞株のA549細胞株で肝癌細胞の場合と同じ方法でTIP41 siRNAをトランスフェクションさせた後、72時間経過後に100ng/mlのTRAILを0、1、2、3、4時間処理した後、アポトーシスに関連したタンパク質の活性化をウエスタンブロット方法を用いて確認した。
肺癌細胞株のA549細胞株にTIP41 siRNAをトランスフェクションさせて、細胞内TIP41発現量を減少させた後、TRAILを100ng/mlで処理した後、0、1、2、4、6時間帯にアポトーシスを核染色法を用いて測定した。
細胞内TIP41発現量を減少させるために、A549細胞株にTIP41 siRNAをトランスフェクションして、TRAILを100ng/mlで処理した後、0、1、2、4、6時間帯にアポトーシスをアネキシンV−FITc/PI染色法で死滅した細胞株と正常細胞株を区別して染色した後、FACSを用いてアポトーシスを測定した。
HCT116大腸癌細胞での場合と同一な方法で、TIP41 siRNAをトランスフェクションさせた後、72時間経過後、TRAILを0、50、100ng/mlで処理し、ビヒクル(vehicle)には、0.3%DMSOを処理した後、アポトーシスを誘導する実験を行った。死滅した細胞株を前記記載の方法と同一にアネキシンV−FITc/PI染色した後、アポトーシスを測定するためにFACS分析法を用いた。
前記の<実験例>で用いたHuh肝癌細胞株以外のTRAIL耐性癌細胞株でTIP41タンパク質発現抑制によるTRAILの耐性を克服するセンシタイザーとしての機能を検証するために、HepG2(肝臓肝細胞の細胞(liver hepatocellular cell))細胞とSK−Hep1細胞にTIP41 siRNAをトランスフェクションさせた後、72時間経過後、100ng/mlのTRAILを0、1、2、4、6時間処理した後、アポトーシスに関連したタンパク質の活性化をウエスタンブロット方法を用いて測定した。
<9−1>Huh7肝癌細胞株をヌードマウスに移植
ヌードマウスの右側の背部分に肝癌細胞株のHuh7肝癌細胞株を2×106程度移植した後、腫瘍を一定のサイズ(50〜100mm3)に成長させた。実験は、個体群ごとに7匹ずつ(n=7)、総28匹のヌードマウスをJapan SLc Inc.から購入して行なった。
実験例<9−1>に記載した方法でHuh7肝癌細胞株をヌードマウスの背に移植して、腫瘍を形成した後、12日間、0、4、6、8日に対照 siRNA(control siRNA)とTIP41 siRNAをヌードマウスの背に形成した腫瘍に注入し、2、5、7、9、10、11日にTRAILを注入した後、12日にマウスを犠牲にした。50nMの濃度でリポフェクタミンRNAiMax試薬を用いて混合物を作成し、形成された腫瘍に注入し、2、5、7、9、10、11日にTRAILを2.5μg/kgずつ注入した後、4つのグループ(control siRNA+vehicle、control siRNA+TRAIL、TIP41 siRNA+vehicle、TIP41 siRNA+TRAIL)に分け、TIP41 siRNAとTRAIL処理による腫瘍のサイズを確認した。
SiRNAを処理していないヌードマウスと、control siRNA、TIP41 siRNAを処理したヌードマウスにTRAILを処理した群と、処理しない群に分けて、6群で生成された腫瘍を採取してホルマリンに固定した後、パラフィンに包埋した。5μmの厚さに切片をつくり、キシレンを用いてパラフィンを除去した後、エタノール濃度による再水和(rehydration)させた。4%パラホルムアルデヒド溶液を処理して、組織切片を固定し、プロテイナーゼK溶液を処理して、組織を透過可能にした後、4%パラホルムアルデヒド溶液に再度固定した後、TUNEL staining kit(promega,米国)を用いて、実験を行なった。平衡バッファーを用いて組織切片を平衡化させ、その間にrTdT反応溶液を準備して平衡化した部位にそれを処理した。最後に、ストレプトアビジンHRP溶液を処理してDAB混合物を用いて発色反応させた後、光学顕微鏡で観察した。
4つのグループ(control siRNA+vehicle、control siRNA+TRAIL、TIP41 siRNA+vehicle、TIP41 siRNA+TRAIL)から採取した腫瘍を溶解させてタンパク質を抽出した後、アポトーシスの標識因子であるカスパーゼ−8抗体を用いて、ウエスタンブロット方法でアポトーシスを確認した。
<10−1>発現ベクターの構成
TIP41、PP2Ac、MKK7、α(alpha)4およびPR65遺伝子を発現するベクターを製造するために、前記の肝癌細胞株からRNAを分離して、発現ベクターにクローニングした。逆向RNA(reserved RNA)を鋳型に用いて、個々の遺伝子に該当するプライマーを用いてPCRを行なった。TIP41正方向プライマー(配列番号 15:5'−cg ggt acc aa atg atg atc cac ggc ttc'−3')とTIP41逆方向プライマー(配列番号16:5'−ccc gga tcc tta ttc cac ttg tgt act−3')、PP2Ac正方向プライマー(配列番号17:5'−cg gga tcc atg gac gag aag gtg ttc−3')、PP2Ac逆方向プライマー(配列番号18:5'−a tag ttt agc ggc cgc tta cag gaa gta gtc tgg−3')、MKK7正方向プライマー(配列番号19:5'−ccg ctc gag atg gcg gcg tcc tcc ctg−3')、MKK7逆方向プライマー(配列番号20:5'−gg ggt acc cct gaa gaa ggg cag gtg−3')、α4正方向プライマー(配列番号21:5'−cg gga tcc atg gct gct gag gac gag−3')、α4逆方向プライマー(配列番号22:5'−a tag ttt agc ggc cgc tca gcc cat gtt ctg tcg−3')、PR65正方向プライマー(配列番号23:5'−cg gga tcc atg gcg gcg gcc gac ggc−3')、およびPR65逆方向プライマー(配列番号24:5'−a tag ttt agc ggc cgc tca ggc gag aga cag aac−3')を用いてPCRを行なった。前記のPCRの条件は、95℃で3分間で変性させた後、95℃で1分、58℃で1分、72℃で1分30秒の条件で30サイクル反復した後、72℃で10分間伸長した後、4℃に冷やした。
韓国細胞株銀行から分譲されたHEK293T細胞株に10%の牛胎児血清を含んだダルベッコの改変イーグル培地(DMEM)で37℃、5%CO2で培養した。SiRNAをトランスフェクションするためにHEK293T細胞株を1×106細胞/100mm皿に植えて24時間培養し、その後、トランスフェクションを行なった。
本発明者らは、前記の実験を通じて、TIP41発現抑制およびTRAIL処理によるアポトーシスの原因が、JNKの継続的な活性化の結果であることを解明したが、それは、TIP41の発現抑制がJNK活性化作用機序を誘導するということを意味する。TIP41と結合することが知られているタンパク質によってJNK活性が調整されるかどうかを確認するために、最近の報告で、TIP41と結合することが知られたタンパク質ホスファターゼ2Ac(Protein phosphatase 2Ac、PP2Ac)を発現させる発現ベクターを、前記の実験例<10−1>で製造した。
TIP41とJNKキナーゼとして知られているPP2Acとの結合を確認するために、実験例<10−1>で製造された発現ベクターをHEK293T細胞株に前記と同一な方法でトランスフェクションさせた後、GSTプルダウンしてウエスタンブロットを行なった。
<11−1>MKK7過発現後、TIP41とMKK7間の相互作用確認
TIP41とJNKキナーゼとして知られているMKK7との間の結合を確認するために、前記の<実験例10>で製造したMKK7発現ベクターをHEK293T細胞にトランスフェクションさせて過発現させた後、<実験例10>と同一な方法でGSTプルダウン分析を行なった。
TIP41とMKK7間の相互作用を確認するために、各々の抗体を用いて、免疫沈降法を行なった後、ウエスタンブロットを行なってTIP41とMKK7間の結合を確認した。免疫沈降法は、沈降させたタンパク質の抗体を細胞溶解物1mgに2μgずつ入れて、4℃で2時間反応させた後、プロテインGセファロースビーズ30μlずつ入れて、4℃で12時間反応させる。その後、PBST[0.1%のツイーン20を含むPBS]バッファーを1mlずつ入れて洗浄し、それを4回反復した。その後、洗浄されたビーズに1×サンプルバッファー(ウエスタンブロット用)100μlずつ添加した後、95℃ヒーティングブロックで5分間沸かした後、氷で2分間冷却した。その後、MKK7、TIP41の抗体を用いて、ウエスタンブロットを行なった。
TIP41とPP2AcおよびMKK7間の相互作用をより正確に確認するために、細胞内でTIP41とMKK7の相互作用ではない、E.coliで組換えタンパク質を合成し、合成されたタンパク質を1:1の反応率に反応させるGSTプルダウン分析を行なった。組換えタンパク質は、前記の実験例<10−1>で製造されたE.coli用発現ベクターをBL21 E.coli菌株に形質転換させて製造した。誘導は、形質転換したE.coliをLB培地に接種して、O.D値が0.4〜0.6の間になるように培養した後、イソプロピルβ−D−1−チオガラクトピラノシド(IPTG)を最終濃度が1mMになるように添加した後、2時間30℃で培養した。その後、超音波破砕を用いて溶解物を得てGSTプルダウンに用いた。
MKK7と結合するTIP41の結合部位を確認するために、TIP41の断片発現ベクターを前記<実験例10>に記載されている方法でクローニングした。
TIP41と結合するMKK7の結合部位を確認するために、MKK7断片発現ベクターを前記<実験例10>に記載されている方法でクローニングした。MKK7−D1正方向プライマー(配列番号32:5'−cg gga tcc cgc agc atg gag agc att−3')とMKK7−D1逆方向プライマー(配列番号20)、MKK7−D5正方向プライマー(配列番号33:5'−cg gga tcc gcc ggc tgt gcc gcc tac−3')、MKK7−D5逆方向プライマー(配列番号20)、MKK7−D6正方向プライマー(配列番号19)、MKK7−D6逆方向プライマー(配列番号34:5'−at agt tta gcg gc cg cta aat gcg ctc ggg gat ggg−3')を用いてPCRを行なった。図13のAに示すように、全長MKK7をD1、D5およびD6の3つの断片に分けて、それぞれに該当する前記のプライマーを用いて増幅させた後、pGST発現ベクターに挿入した。
<15−1>MKK7発現抑制によるアポトーシスの減少
アポトーシス耐性がMKK7とTIP41間の相互作用によって媒介されることを確認するために、MKK7に対するsiRNAによるアポトーシス耐性の減少を調べた。
実際にTIP41発現抑制が直接的にMKK7の活性化に関与して、それによってMKK7下部信号伝達に与える影響を確認するために、試験管内(in vitro)共免疫キナーゼ(immuno−co−kinase)分析を行なった。共免疫キナーゼの分析は、免疫沈降法の一種で、MKK7を沈降させた後、それを分離して試験管内(in vitro)状態で人為的にMKK7の基質およびMKK7の下部信号伝達に関与する基質を同時に定量的に添加して、リン酸化される程度を確認する実験方法である。前記のような方法でMKK7抗体を用いて免疫沈降をさせ、MKK7タンパク質に結合しているプロテインGビーズを得た後、キナーゼバッファーにMKK7の基質である組換えGST−JNK1とJNK1の基質であるGST−c−Junおよび標識因子して同位元素(−32P)を入れて、37℃で30分間反応させた。その後、1×タンパク質ローディングダイ(protein loading dye)を添加した後、95℃で5分間沸かした。10%のSDS−PAGEゲルに電気泳動した後、ゲルドライヤー機でゲルを乾燥させた後、BASリーダー(放射線測定機器、富士通、日本)で感光した。
<1−1>散剤の製造
TIP41タンパク質発現または活性阻害剤 2g
乳糖 1g
前記の成分を混合し、気密包に充填して散剤を製造した。
TIP41タンパク質発現または活性阻害剤 100mg
トウモロコシ澱粉 100mg
乳糖 100mg
ステアリン酸マグネシウム 2mg
前記の成分を混合した後、通常の錠剤の製造方法によって、打錠して錠剤を製造した。
TIP41タンパク質発現または活性阻害剤 100mg
トウモロコシ澱粉 100mg
乳糖 100mg
ステアリン酸マグネシウム 2mg
前記の成分を混合した後、通常のカプセル剤の製造方法によって、ゼラチンカプセルに充填して、カプセル剤を製造した。
TIP41タンパク質発現または活性阻害剤 1g
乳糖 1.5g
グリセリン 1g
キシリトール 0.5g
前記の成分を混合した後、通常の方法によって1丸薬当り4gになるように製造した。
TIP41タンパク質発現または活性阻害剤 150mg
大豆エキス 50mg
ブドウ糖 200mg
澱粉 600mg
前記の原料を混合し、30%のエタノール100mgを追加して、60℃で乾燥して顆粒形成した後、包に充填した。
Claims (31)
- TIP41タンパク質の発現または活性抑制剤を含有する、TRAIL感受性増進用組成物。
- 前記のTIP41タンパク質が、配列番号1で表されるアミノ酸配列を有していることを特徴とする、請求項1に記載の組成物。
- 前記のTIP41タンパク質の発現抑制剤が、TIP41遺伝子のmRNAに相補的に結合するアンチセンスヌクレオチド、低分子干渉RNA(short interfering RNA)および低分子ヘアピン型RNA(short hairpin RNA)からなる群から選択されたいずれか1つであることを特徴とする、請求項1に記載の組成物。
- 前記のTIP41タンパク質の活性剤が、TIP41タンパク質に特異的に結合する化合物、ペプチド、ペプチドミメティックス、アプタマー、抗体、および天然物からなる群から選択されたいずれか1つであることを特徴とする、請求項1に記載の組成物。
- TRAILを用いた癌、炎症性疾患または自己免疫疾患の治療に使用されることを特徴とする、請求項1に記載の組成物。
- 前記の癌が、肝癌、大腸癌、子宮頸癌、腎臓癌、胃癌、前立腺癌、乳癌、脳腫瘍、肺癌、子宮癌、結腸癌、膀胱癌、血液の癌および膵臓癌からなる群から選択されるいずれか一つであることを特徴とする、請求項5に記載の組成物。
- TIP41タンパク質の発現または活性抑制剤を含有する抗癌補助剤。
- 前記のTIP41タンパク質が、配列番号1で表されるアミノ酸配列を有していることを特徴とする、請求項7に記載の抗癌補助剤。
- 前記のTIP41タンパク質の発現抑制剤が、TIP41遺伝子のmRNAに相補的に結合するアンチセンスヌクレオチド、低分子干渉RNA(short interfering RNA)および低分子ヘアピン型RNA(short hairpin RNA)からなる群から選択されたいずれか1つであることを特徴とする、請求項7に記載の抗癌補助剤。
- 前記のTIP41タンパク質の活性剤が、TIP41タンパク質に特異的に結合する化合物、ペプチド、ペプチドミメティックス、アプタマー、抗体および天然物からなる群から選択されたいずれか1つであることを特徴とする、請求項7に記載の抗癌補助剤。
- 前記の癌が、肝癌、大腸癌、子宮頸癌、腎臓癌、胃癌、前立腺癌、乳癌、脳腫瘍、肺癌、子宮癌、結腸癌、膀胱癌、血液の癌および膵臓癌からなる群から選択されるいずれか一つであることを特徴とする、請求項7に記載の抗癌補助剤。
- 前記のTIP41タンパク質の発現または活性阻害剤が、TRAIL感受性を増進させることを特徴とする、請求項7に記載の抗癌補助剤。
- 請求項7〜請求項12のいずれか1項に記載の抗癌補助剤を含む、癌の治療または予防用組成物。
- (1)実験群として、癌細胞株に被検物質を処理する工程、
(2)工程(1)の処理された細胞株でTIP41とPP2AcまたはMKK7タンパク質の結合水準を確認する工程、および
(3)工程(2)のTIP41とPP2AcまたはMKK7タンパク質の結合水準を対照群と比較して減少させる被検物質を選別する工程
を含む、癌の予防または治療剤のスクリーニング方法。 - 癌細胞株が、TRAIL耐性の癌細胞株であることを特徴とする、請求項14に記載の癌の予防または治療剤のスクリーニング方法。
- 工程(2)のタンパク質の結合水準が、免疫沈降法(immunoprecipitation)、酵素免疫分析法(ELISA)、ウエスタンブロット法(Western Blotting)、グルタチオン−S−トランスフェラーゼ(Glutathione−S−Transferase)(GST)プルダウン分析、タンパク質チップ(Protein Chip)、蛍光共鳴エネルギー移動(Fluorescence Resonance Energy Transfer、FRET)、蛍光蛋白質再構成法(Bimolecular fluorescence complementation、BiFC)および酵母ツーハイブリッド(Yeast two−hybrid、Y2H)からなる群から選択されたいずれか1つで測定することを特徴とする、請求項14に記載の癌の予防または治療剤のスクリーニング方法。
- (1)実験群として、癌細胞株に被検物質を処理する工程、
(2)工程(1)の処理された細胞株でMKK7タンパク質の活性程度を測定する工程、および
(3)工程(2)のMKK7タンパク質活性が対照群と比較して増加した被検物質を選別する工程
を含む、癌の予防または治療剤のスクリーニング方法。 - 癌細胞株が、TRAIL耐性癌細胞株であることを特徴とする、請求項17に記載の癌の予防または治療剤のスクリーニング方法。
- MKK7タンパク質の活性程度が、免疫蛍光法、酵素免疫分析法(ELISA)、質量分析およびタンパク質チップからなる群から選択されたいずれか1つで測定することを特徴とする、請求項17に記載の癌の予防または治療剤のスクリーニング方法。
- (1)実験群として、癌細胞株に被検物質を処理する工程、
(2)工程(1)で処理された細胞株でPP2Acタンパク質の発現程度を測定する工程、および
(3)工程(2)のPP2Acタンパク質の発現量を対照群と比較して減少させた被検物質を選別する工程
を含む、癌の予防または治療剤のスクリーニング方法。 - 癌細胞株が、TRAIL耐性癌細胞株であることを特徴とする、請求項20に記載の癌の予防または治療剤のスクリーニング方法。
- 工程(2)のPP2Acタンパク質の発現程度が、逆転写重合酵素の連鎖反応(Reverse Transcription−Polymerase chain Reaction、RT−PCR)、酵素免疫分析法(ELISA)の免疫組織化学、ウエスタンブロット法(Western Blotting)および流細胞分析法(FACS)からなる群から選択されたいずれか1つで測定することを特徴とする、請求項20に記載の癌の予防または治療剤のスクリーニング方法。
- 薬学的に有効な量のTIP41の発現または活性抑制剤を、TRAIL媒介アポトーシスと関連した疾病にかかった個体に投与する工程を含む、TRAILに対する感受性を増進させる方法。
- 前記のTRAIL媒介アポトーシスと関連した疾病が、癌、炎症性疾患または自己免疫疾患であることを特徴とする、請求項23に記載の方法。
- 前記の癌が、肝癌、大腸癌、子宮頸癌、腎臓癌、胃癌、前立腺癌、乳癌、脳腫瘍、肺癌、子宮癌、結腸癌、膀胱癌、血液の癌および膵臓癌からなる群から選択されるいずれか一つであることを特徴とする、請求項24に記載の方法。
- 前記の炎症性疾患が、皮膚炎、アレルギー、アトピー、結膜炎、歯周炎、鼻炎、中耳炎、咽頭炎、扁桃腺炎、肺炎、胃潰瘍、胃炎、クローン病、大腸炎、痔、痛風、強直性脊椎炎、リウマチ熱、ループス、線維筋痛症(fibromyalgia)、乾癬性関節炎、骨関節炎、関節リウマチ、肩関節周囲炎、腱炎、腱鞘炎、腱周囲炎、筋肉炎、肝炎、膀胱炎、腎臓炎、シェーグレン症候群(sjogren's syndrome)、多発性硬化症、急性および慢性の炎症性疾患からなる群から選択されるいずれか一つであることを特徴とする、請求項24に記載の方法。
- 前記の自己免疫疾患が、関節リウマチ、多発性硬化症、重度の筋無力症、クレブス病、橋本氏甲状腺炎、アジソン病、白斑症、硬皮症、グッドパスチャー症候群、ベーチェット病、クローン病、強直性脊椎炎、ブドウ膜炎、血小板減少性紫斑病、尋常性天疱瘡、幼児糖尿病、自己免疫溶血性貧血、クリオグロブリン血症、副腎白質異栄養症および全身性紅斑性狼瘡からなる群から選択されることを特徴とする、請求項24に記載の方法。
- 薬学的に有効な量のTIP41の発現または活性抑制剤を個体に投与する工程を含む、癌の予防方法。
- 薬学的に有効な量のTIP41の発現または活性抑制剤を抗癌補助剤として癌にかかった個体に投与する工程を含む、癌の治療方法。
- TRAIL媒介アポトーシスと関連した疾病の治療時にTRAIL感受性増進のために使用するための、TIP41の発現または活性抑制剤。
- 抗癌補助剤として使用するためのTIP41の発現または活性抑制剤。
