典型的な粒子は、タングステン、白金および好ましくは金よりなるものを含む。爆撃のために、懸濁液中の細胞は、フィルターまたは固形培養培地で濃縮される。別法として、未熟胚または他の標的細胞は固形培養培地に配置し得る。爆撃すべき細胞は、マクロプロジェクタイル停止プレート(macroprojectile stopping plate)下の適当な距離に位置する。

Claims (12)
- 高められたブリックスを付与するリコペルシコン・ヒルスタム(Lycopersicon hirsutum)のhir4対立遺伝子を含むトマト植物であって、ここに該植物は貧弱な植物習性を付与するリコペルシコン・ヒルスタムのhir4対立遺伝子に遺伝学的に連鎖する対立遺伝子を欠く該トマト植物。
- 請求項1記載のトマト植物の植物部分。
- 該部分が細胞、種子、根、茎、葉、果実、花または花粉である請求項3記載の植物部分。
- 高められたブリックスを含むトマト植物を得るための方法であって、
(a)高められたブリックスを付与し、かつ、貧弱な植物習性を付与するリコペルシコン・ヒルスタムの対立遺伝子に植物において遺伝学的に連鎖するリコペルシコン・ヒルスタムからのhir4対立遺伝子についてヘテロ接合であるトマト植物を得;
(b)植物の子孫を得;ついで
(c)子孫がhir4を含むが、貧弱な植物習性を付与する対立遺伝子は含まないように遺伝子組換えが生じた少なくとも第1の子孫植物を選択する
ことを含む該方法。 - 子孫植物を選択することが、(1)リコペルシコン・ヒルスタムにおけるhir4対立遺伝子に遺伝学的に連鎖する遺伝子マーカーを含み、および/または、リコペルシコン・エスキュレンタム(Lycopersicon esculentum)からの対応する遺伝子座に存在する遺伝子マーカーを欠き、および(2)リコペルシコン・ヒルスタムにおいて貧弱な植物習性を付与する対立遺伝子に遺伝学的に連鎖した遺伝子マーカーを欠き、および/または、リコペルシコン・エスキュレンタムからの対応する遺伝子座に存在する遺伝子マーカーを含む、子孫植物を同定することを含む請求項4記載の方法。
- 子孫植物を選択することが、TG155対立遺伝子、ANTL対立遺伝子、C2_At3gl6150対立遺伝子、cTOE-6-D11対立遺伝子、CT50対立遺伝子、C2_At4g33350対立遺伝子、CT73対立遺伝子、TG443対立遺伝子、TG500対立遺伝子、NL0233688対立遺伝子、NL0233750対立遺伝子、NL0233794対立遺伝子、NL0233797対立遺伝子、NL0233922対立遺伝子、NL0234257対立遺伝子、NL0234328対立遺伝子、NL0234071対立遺伝子、NL0234055対立遺伝子、NL0233999対立遺伝子およびNL0215788対立遺伝子よりなる群から選択される少なくとも1の対立遺伝子を検出することを含む請求項4記載の方法。
- (a)対立遺伝子を、オリゴヌクレオチド・プライマー対(またはその複数)を用いるPCRベースの方法によって検出し;かつ
(b)TG155対立遺伝子は配列番号:1および配列番号:2を含むプライマー対を用いて検出し;ANTL対立遺伝子は配列番号:9および配列番号:10を含むプライマー対を用いて検出し;C2_At3gl6150対立遺伝子は配列番号:11および配列番号:12を含むプライマー対を用いて検出し;cTOE-6-D11対立遺伝子は配列番号:13および配列番号:14を含むプライマー対を用いて検出し;CT50対立遺伝子は配列番号:15および配列番号:16を含むプライマー対を用いて検出し;C2_At4g33350対立遺伝子は配列番号:17および配列番号:18を含むプライマー対を用いて検出し;CT73対立遺伝子は配列番号:19および配列番号:20を含むプライマー対を用いて検出し;TG443対立遺伝子は配列番号:21および配列番号:22を含むプライマー対を用いて検出し;TG500対立遺伝子は配列番号:5および配列番号:6を含むプライマー対を用いて検出し;NL0233688対立遺伝子は配列番号:23および配列番号:24を含むプライマー対を用いて検出し;NL0233750対立遺伝子は配列番号:27および配列番号:28を含むプライマー対を用いて検出し;NL0233794対立遺伝子は配列番号:31および配列番号:32を含むプライマー対を用いて検出し;NL0233797対立遺伝子は配列番号:35および配列番号:36を含むプライマー対を用いて検出し;NL0233922対立遺伝子は配列番号:39および配列番号:40を含むプライマー対を用いて検出し;NL0234257対立遺伝子は配列番号:43および配列番号:44を含むプライマー対を用いて検出し;NL0234328対立遺伝子は配列番号:47および配列番号:48を含むプライマー対を用いて検出し;またはNL0234071対立遺伝子は配列番号:51および配列番号:52を含むプライマー対を用いて検出する請求項6記載の方法。 - 対立遺伝子を検出することが、配列番号:55または配列番号:56のヌクレオチド46;配列番号:57または配列番号:58のヌクレオチド61;配列番号:59または配列番号:60のヌクレオチド61;配列番号:61または配列番号:62のヌクレオチド61;または配列番号:63または配列番号:64のヌクレオチド61に対応する遺伝子移入エル・ヒルスタム配列中のポジションの単一ヌクレオチド多型を検出することを含む請求項6記載の方法。
- 請求項5記載の方法によって生産した植物。
- 植物が、高められたブリックスを付与し、かつ、貧弱な植物習性を付与するそれに遺伝学的に連鎖したリコペルシコン・ヒルスタム対立遺伝子を欠く、リコペルシコン・ヒルスタムからのhir4対立遺伝子を含む請求項8記載の植物。
- 細胞、種子、根、茎、葉、果実、花および花粉よりなる群から選択される請求項9記載の植物の部分。
- (a)高められたブリックスを付与するリコペルシコン・ヒルスタムのhir4対立遺伝子を含む染色体セグメントを植物に遺伝子移入することを含み、ここに該セグメントが貧弱な植物習性を付与するリコペルシコン・ヒルスタムのhir4対立遺伝子に遺伝学的に連鎖する第2の対立遺伝子を欠く、高められたブリックスを含むトマト植物を生産する方法。
