Abstract

PROBLEM TO BE SOLVED: To provide an aromatic composition which is effective to a lifestyle related disease, especially to improvement of blood sugar level.SOLUTION: A Citrus Sudachi essential oil component is concentrated and separated from a Citrus Sudachi pericarp, and when living for several weeks under an atmosphere of an aromatic component originated in Citrus Sudachi, while using the Citrus Sudachi essential oil component, it is found that the blood sugar level is lowered significantly. Resultantly, the aromatic composition containing the Citrus Sudachi essential oil component as an active ingredient can be provided as aromatherapy for a lifestyle related disease, especially for improvement of blood sugar level.

近年、アロマテラピーとして、植物から抽出した精油をかいだり、塗ったりして、生理機能を整える療法がよく新聞紙上で紹介されている。また、精油には鎮静効果やストレス緩和効果などが知られている(特許文献1)。また、その精油の香りの効果は、芳香物質がきゅう覚を刺激し、交感神経系に対する活性化や副交感神経系に対する活性化によるものであるとされている(特許文献2、3)。
そして、これらの香り(香料)を利用して、肥満改善(痩身)に利用されたり(特許文献4)、脂肪蓄積抑制に使用されている(特許文献5)。更には、ホルモンバランスを改善し、エストロゲンの分泌促進の治療にも効果があると報告されている(特許文献6)。
これら以外にも皮膚免疫機能改善のためにも香料が使用されている(特許文献7)。このように植物から抽出した精油(香料)は、多くの疾患の症状改善や治療に利用されている。
しかしながら、このようなアロマテラピーにおいて、スダチ由来の精油が有効とする報告はなく、生活習慣病の症状改善や治療に有効であるとする報告も見出されていない。

本発明者らが、スダチの搾汁残渣あるいはスダチの果皮から得られた精油(芳香成分)に関する研究を進めていたところ、スダチの芳香成分が糖尿病モデルマウスの血糖値を改善あるいは降下させることを見出した。そこで、更に検討を進めたところ、スダチの芳香が消えると、改善されていた血糖値が上昇することを見出した。即ち、スダチ芳香成分には、血糖値改善(血糖値降下)作用を示す芳香成分が存在することが明らかとなった。そこで、他の柑橘類と同様に共通して含有される成分として、リナロール、p−シメンの血糖降下作用を個別に確認したが、具体的にこれらの成分に関して血糖降下作用を見出すことはできなかった。また、この血糖降下作用は、柚子の精油成分には見出すことができなかった。このことから、本発明者らは、スダチの精油成分に含まれる特有の成分(p−シメン、リナロール以外の精油成分)に、血糖降下作用があることを見出した。本発明は、これらの知見に基き完成したものである。   The present inventors have been conducting research on the essential oil (fragrance component) obtained from the juice residue of Sudachi or the skin of Sudachi, and it has been found that the fragrance component of Sudachi improves or lowers the blood glucose level of diabetes model mice. 1)エタノール抽出法:
スダチ果皮に対して、2倍量のアルコール水溶液(5%エタノール水溶液)を使用して、香気成分の抽出を行い、初期抽出液を得た。この抽出液を超音波霧化分留装置(松浦酒造製)による霧化分離を2回行うことにより、スダチ精油(回収油:100%)を分離した。分離条件は,溶液温度50℃,冷却部温度−20℃(1回目のみ−6℃)とした。1回目は、初期抽出液5000ml(回収油:0.18%)を霧化分離することで,分離液1000ml(回収油:0.74%)を得た。1回目の初期抽出液と分離液の品質比較を行い,分離液中の屈折示度および総酸が低値であることから,初期抽出液中の糖および有機酸は分離液中に移行しないことを確認した。続いて,一回目分離液の一部350mlを用いて、霧化分離することで,2回目の分離液70m(回収油:3.2%)を得た。この分離液を静置すると油水分離を生じ,上澄のスダチ精油分2mlを分離できた。上澄の精油分は水分,エタノールをほとんど含まず,無色透明でスダチの新鮮な香りを保持したスダチ精油であった。
上記スダチ精油分をガスクロマトグラフィー(ヒューレット・パッカード社、HP−5890A)で分析した。DB−WAXカラムを用い、オーブン温度を初期温度40℃3分、昇温速度5℃/分、到達温度210℃5分の条件で実施した。その結果を以下の表1(組成比)に示す。

(実施例2)GK−ラットを用いたスダチ芳香成分(スダチ精油)の血糖降下作用の評価
1)約10%のスダチ精油を含有した霧化分離液の効果:
上記霧化分離液に水を加えて超音波拡散させ、50倍希釈した希釈液(20000ppm)と500倍希釈液(2000ppm)を作製した。上記希釈液と水(プラセボ)を用いて、GK−ラット(30週令、雄性)の顔と手に、1日1回刷毛で塗布した。3週間の塗布試験後の結果を図1に示す。
図1に示されるように、50倍希釈した希釈液(20000ppm)の場合に、有意な血糖値降下作用が示されることが分かった。また、500倍希釈と比較し、その作用はスダチ精油の濃度依存性であった。
2)100%スダチ精油の効果:
スダチ精油分に水を加えて500倍に希釈した希釈液(20000ppm)を用いて、GK−ラット(30週令、雄性)の鼻に1日1回、3週間塗布を行った。また、対照群として上記希釈液の代わりに水を塗布した。対照群(1群5匹)とスダチ精油塗布群(1群5匹)を同じ室内の隣接箇所で飼育した。スダチ精油による芳香が飼育室内に拡散しており、そのため、スダチ精油の芳香による効果が、隣接して飼育した対照群でも評価できる。即ち、GK−ラットの鼻にスダチ精油を塗布した群における直接的なスダチの芳香効果と、対照群における室内芳香の間接的な効果が評価できることになる。なお、評価期間3週間の上記ラットの血糖値の変化を測定し、塗布終了の1週間後の血糖値を測定した。その結果を図2に示す。
図2に示されるように、500倍希釈液の場合に実施例1の場合と同様の血糖値降下の効果が得られることが明らかとなった。しかしも、スダチ塗布群だけでなく、対照群でも、図2に示されるように血糖値が有意に低下した。このことは、スダチの室内芳香によっても、血糖値降下が起こることを明らかにしたものである。更に、スダチ精油成分の塗布を中止したところ、血糖値が上昇に転じたことから、スダチ精油成分(芳香成分)による血糖値効果作用は明らかである。

Further, when the application of the Sudachi essential oil component was stopped, the blood sugar level started to rise, and thus the effect of the blood sugar level effect by the Sudachi essential oil component (fragrance component) is clear.

そこで、実施例1と同様の超音波霧化分留処理を行って得られた柚子の霧化分離液(約10%の柚子精油含量)と実施例1で得られたスダチ霧化分離液(約10%のスダチ精油含量)を使用した。各分離液を水で50倍に希釈した水溶液(20000ppm)を作製した。GK−ラット(30週令、雄性)を2群(1群5匹)に分けて、ラットの顔と手に上記水溶液を1日1回塗布した。3週間の塗布試験により、柚子とスダチの精油成分の効果の相違を検討した。その結果を図3に示す。
図3に示されるように、柚子精油成分では、血糖値は上昇傾向にあるが、スダチ精油分では僅かに血糖値が低下することが確認された。また、塗布開始から3週間後になるとスダチの方が柚子よりも有意に血糖値が低下する(p<0.05)ことが示された。
以上のことから、スダチには、柚子と異なり、血糖効果に有効な精油成分を含有することが明らかになった。

Therefore, the atomization separation liquid (about 10% eggplant essential oil content) of the eggplant obtained by carrying out the same ultrasonic atomization fractionation treatment as in Example 1 and the Sudachi atomization separation liquid obtained in Example 1 ( About 10% Sudachi essential oil content) was used. An aqueous solution (20,000 ppm) was prepared by diluting each separation solution 50 times with water. GK-rats (30 weeks old, male) were divided into 2 groups (5 animals per group), and the above aqueous solution was applied to the faces and hands of the rats once a day. The difference in the effects of the essential oil components of eggplant and sudachi was examined by a 3-week application test. The result is shown in FIG.
As shown in FIG. 3, the blood sugar level tends to increase with the coconut essential oil component, but it was confirmed that the blood sugar level slightly decreased with the Sudachi essential oil component. In addition, it was shown that blood glucose level was significantly lower in Sudachi than in eggplant after 3 weeks from the start of application (p <0.05).
From the above, it has been clarified that Sudachi contains an essential oil component effective for the blood sugar effect, unlike eggplant.

(実施例4)スダチ精油成分のp−シメン、リナロールの効果確認
d−リモネン以外の精油主要成分に関して、血糖値降下作用の有無を検証した。まず、柚子精油成分の第2主成分リナロール(34%)とスダチの成分p−シメン(4%)について、血糖値降下作用の有無を評価した。
GK−ラット(生後約1年、雄性)を1群5匹で3群(蒸留水塗布、リナロール塗布、p−シメン塗布)に分けた。実験開始時の血糖値を測定した後、ラットの鼻に0.1%のリナロール、p−シメン水溶液(蒸留水に市販のリナロール、p−シメンを添加し、超音波で分散させた)を1日1回塗布した。1週間毎に血糖値を測定し、3週間連日塗布を行った。
その結果、3週間後の3群のいずれにおいても血糖値に有意な差は見られなかった。即ち、リナロールとp−シメンにおいては、血糖値降下作用を示さないことが分かった。

As a result, there was no significant difference in blood glucose level in any of the three groups after 3 weeks. That is, it was found that linalool and p-cymene do not show a blood glucose lowering effect.
