(5)ここで定義されたデバイスの用途は、放射線量を測定するための用途である。「放射線測定」は、間接的または直接的にイオン化放射線に晒された結果生じる物質および生体組織内に吸収された線量を計算することである。ここで記述されたデバイス(好ましくは、HEMTベースのデバイス、より好ましくはAlGaN/GaNヘテロ構造を構成するデバイス)は、例えば、好ましくはリアルタイムで、人間の患者上(内)での皮膚線量測定用に用いられてよい。

後述の文章は、本発明のヘテロ構造の好ましいレイアウトを説明するものであるが、本発明はそれらの設定に限定されるものではないことが理解されなければならない。GaNチップは電気コンタクトを有しており、これらは細胞電位の測定のために用いられている。サファイア基板がAlN核形成層で被覆されている。動作領域は2500nmの膜厚のGaN層と28%のアルミニウム量を含む25nmの膜厚のAlGaNバリア層とから構成されている。これら二つの物質間における二次元電子ガスキャリアの密度は、6.1×1012cm−2(Steinhoff G.et al.,2003b)である。上記デバイスは、3nmの膜厚を有するGaNキャップ層によって被覆されており、この層が、細胞が培養される表面である。
後述する文章は、本発明のヘテロ構造の好ましい設定を説明するものであるが、本発明はそれらの設定に限定されるものではないことが理解されなければならない。チップは、配線および不動態化の前に、超音波槽中のアセトンおよびイソプロパノールを用いて洗浄された。それぞれの単一の測定の前に、ちょうどGaNサンプルのように、70%のエタノールを用いて洗浄された。研磨された裏面を有する利用可能な1つのGaNサンプルが存在した。全てのその他の使用されたGaNサンプルとチップとは、研磨された裏面を持っていない。これらのサンプルを用いた光学顕微鏡検査はできなかった。
後述する文章は、本発明のヘテロ構造の好ましい環境を説明するものであるが、本発明はそれらの環境に限定されるものではないことが理解されなければならない。GaNチップはサンプルホルダーの上に搭載されて、金配線に接続されており、当該金配線は0.1mmの直径を有している。データの取得は、Keithley Source and Multimeters(Keithley Instruments Inc.,Cleveland, Ohio 44139)を用いて実現される。それぞれのGaNチップはKeithely 2400 Source−meterに接続されており、当該Keithely 2400 Source−meterは電圧の印加と電流の測定とを同時に実現することを可能にする。SourceMeterは、正確で、低電圧で、高安定のDCパワー供給を、低ノイズで、高反復可能性で、高インピーダンスのマルチミーターに組み合わせる。それは、5−1/2−デジタル解像度を有する0.012%の基礎精度を持つ。5−1/2ディジットにて、SourceMeterは、IEEE488バス(Model2400Seies SourceMeter User‘s Manual, Revision G, 1988, Keithley Instruments,inc.)上で一秒間に520リードを行う。電流を同時に測定しながら、120mVの定電圧がGaNトランジスタのソースドレインコンタクトに与えられる。所定の精度は、デバイスの基礎設定に対して有効である。リードのための異なるインテグレーション時間に応じて、その精度は所定の値から異なり得る。Keithleyデバイスはこれらのデータを、Windows XPの基礎PC中にインストールされたGBIP Interface Board,type KPCI−488LP IEEE−488.2(Keisthley Instruments Inc., Vleveland, Ohio 44139)に送る。測定デバイスの制御および読み出しに加えてサンプルデータの蓄積は、LabView 8.6.1 Express(National Instruments Dtl.,Munich, Germany)を用いてなされる。全ての放射線照射実験のために、医療X線デバイス、Stabilipan TR300fのタイプ (Seimens AG, Munich, Germany)が用いられた。
細胞電位測定を開始する前に、異なる物理的環境の影響へのトランジスタデバイスの依存性を評価することが考えられる。GaNチップは、光、温度、およびpH値の変化に対して非常に敏感であって、且つ、X線放射に対する応答を示す。測定されたデータの適切な解釈を行うためには、トランジスタの幅広い特徴付けを行うことが、物理的および電気的特性を知るために望ましい。好ましくは、それぞれの単一のチップ(本発明のヘテロ構造またはデバイス)の個別の特徴づけが実行される。
細胞がアポトーシスを経験するときのpH値の変化を記述する幾つかの刊行物が存在している(Lagadic−Gossmann D.et al.、 2004; Tannock I.F., 1989)。細胞放射線照射実験中における予期されたシグナルの変化の感触を得るために、GaNトランジスタ電流がpH値に依存して測定される。この目的のために、異なるpH値を持つ溶液が、HCLとNaOHとを混合することによって生成された。その溶液のpH値は、pHメータ、CyberScan500のタイプを用いて決定された(Eutech Instruments Europe, Nijkerk,Netherlands)。
幅広い細胞成長実験は、GaN表面上の細胞が生存していること、および基板物質から細胞の影響が何も存在しないことを保証するために、実行された。その形態試験および成長研究は、光学顕微鏡またはレーザー走査顕微鏡および原子間力顕微鏡によって実行された。細胞が生存しているか死滅しているかを決定する最も早い方法の一つは、その外部の形状を調べることである。GaNチップ上の細胞成長は、通常の顕微鏡スライド上での細胞成長と比較された。GaN表面上の細胞成長とガラス基板上の細胞成長との間における視認できる相違は存在していない。

細胞通信経路は、一次メッセンジャーと二次メッセンジャーの細胞膜を横断する移動をもたらす。これらの細胞通信経路の解読は、高度な細胞ホメオスタシスを理解する鍵である。ここで、我々は、長期間の測定における放射線照射実験の間、帯電された外部細胞メッセンジャーによって引き起こされる細胞電位応答を記録することができる、半導体ナノ構造に基づいた測定アプローチを提示する。AlGaN/GaNヘテロ構造は、電子トランジスタおよびイオン感応型トランジスタのゲート層を提供する。電気特性とそれゆえシステムの導電性とは、細胞膜を横断するイオン束によって変化するチップ表面電位に大きく依存している。このことにより、細胞応答とそれらの時間順序に対する特別な調査が可能となる。本デバイスは放射線に敏感であるが、ヘテロ構造は、X線に晒されている間、安定的で反復可能性のある挙動をもって動作可能である。我々は、放射線照射された細胞によって引き起こされた、60秒以内で0.13μAまでのトランジスタのシグナルの変化を測定した。放射線照射の後のみならず、放射線照射の間についても、細胞電位を測定できることは、試験の制約をかなり広げることである。

我々は、電荷およびpH感応型溶液ゲートAlGaN/GaN高電子移動度トランジスタのリアルタイムX線照射応答を提示する。本デバイスは、μGyの範囲に入る線量に対するリニアな統合応答を含む、X線照射下および続いて起きるX線照射に対して安定的で且つ再現可能な挙動を示す。溶液中のデバイスの適定測定によると、リニアなpH反応と感度が、X線照射下で保持されるばかりではなく、放射線応答が測定され得ることを明らかにしている。デバイスが生体適合性であって、当該デバイスは液体中で且つ激しい放射線下で同時に動作され得るので、当該デバイスは、医療放射線測定およびバイオセンシングのアプリケーションの双方に対して大変適している。生体システムを原位置でモニターできることは、近代の医療社会にとって極めて重要である。今回の研究の一側面は、放射線暴露の前に、間に、および後に、細胞に対する放射線のリアルタイムの影響を研究する方法を開発したことである。近年、広いバンドギャップの半導体はこの研究分野の入口に立ち、GaNは特に有望な特徴を示してきた。たとえば、AlGaN/GaN高電子移動度トランジスタ(HEMTs)は生体適合的であることが報告されている1,2。GaNキャップ層を付加的に用いることで、このデバイスは、水溶液中で動作され得、かなりの程度に化学的に不活性であり、そして、高リニアなpH感度を示す3。さらに、これらのデバイスは、気体、極性流体、特定イオン、および細胞応答の検知のために用いられてきた1−4。ここで、我々は、同時液相pH測定と、AlGaN/GaNのHEMTに基づくイオン感応型電界効果トランジスタ(ISFETs)を用いた放射線検知とを実証することにより、アプリケーションの範囲を広げる。本デバイスは、X線に対して安定的で、反復可能で、そして敏感な応答を示し、放射線照射中のpH感度を保持する。これらのサイズの小ささと湿潤な環境における動作性のために、そのようなデバイスは、生体内測定にとって大変適しており、これらは激しい環境における生化学応答および放射線応答を同時に測定するための手段を提供する。この研究のために用いられるHEMTデバイスは、2.6nmの膜厚のGaNキャップ層を用いたTopGaN(Warsaw,Poland)による工業用MOCVDベースの成長プロセスを用いて製造された。HEMTデバイスの製造方法および物理的特性は、従前の刊行物に記述されている5,6。測定環境は、標準的なガラス電極(pH/Ion−Meter Metrohm 781)、Ptカウンター電極、Ag/AgCl参照電極、および作動電極としてのISFETを備えた単一電気化学的細胞から構成された。トランジスタの活性領域(0.88mm2)は、動作中、4mmのAlフィルターを用いた医療用X線システム(Stabilipan TR300f,Siemens AG)によって放射線照射がなされ、空気中の線量の参照測定は面積線量計(Diamentor M4, PTW)を用いて記録された。低線量実験用には、キャビネットX線システム(MX−20,Faxitron X−ray LLC)が利用された。溶液測定は、10mMのHEPESバッファおよび0.1MのNaClまたはKClの溶液中で実行された。この溶液は、バッファで希釈されたNaOHまたはKOHおよびHCLを用いて適定されたものであった。すべての実験は、トランジスタの光感度のために、完全に暗い環境で実行された。X線照射下およびそれに続くX線照射におけるデバイス性能の安定性および再現可能性を確認するために、多数のシリーズの線量率依存性輸送測定が実行された。図5は、120mVの固定電位と線量率に対してプロットされたフローティングゲート電位での、ソースドレイン電流ISDの変化に伴う、150kVのパルスX線照射シリーズの結果を示している。図5の差込図に示されるように、10分間のパルスで15分の間隔を間に設けて放射線照射が実行された。光励起キャリアの生成により、X線の線量率に応じたソースドレイン電流の単調増加が観察された。放射線シリーズの繰り返しから得られた結果を比較することで、トータルの線量が60Gyを超えた後には、何らの顕著な永久性能の変化を伴うことのない素晴らしい再現性が得られた。これらの小さい偏差は、本デバイスが放射線測定のアプリケーションにとって大変適していること、且つ、本デバイスが溶液動作下で絶対受容線量率に対する内部較正を提供することを実証している。我々は、溶液条件中でのソースドレイン電流反応の測定が、本デバイスのいかなる特別の表面処理を行うことなく、同様の再現性を示したことを指摘する。

Claims (13)
- 外部刺激に応答して生体細胞によって生成される、電気信号のような細胞シグナルをモニターするデバイスであって、
(a) 高電子移動度トランジスタ(HMET)を含む、少なくとも一つの分離領域と、
(b) 前記HMETに付着した非興奮性細胞とを備えている、デバイス。 - 請求項1に記載のデバイスの用途であって、
任意のリアルタイムで、外部刺激に応答して生体細胞によって生成される、電気信号のような細胞シグナルをモニターするために用いられる、用途。 - 請求項1に記載のデバイスの用途であって、
(a)放射線量率の測定および/または(b)放射線量の測定のために用いられる、用途。 - 請求項1に記載のデバイスの用途であって、
任意のリアルタイムで、表面電位(pH)の変化を測定すると同時に(a)放射線量の測定および/または(b)放射線量率の測定を行うために用いられる、用途。 - 請求項3または4に記載のデバイスの用途であって、
高電子移動度トランジスタ(HMET)を含む少なくとも一つの分離領域は、前記HMETに細胞が付着されていない、用途。 - 請求項3または4に記載のデバイスの用途であって、
外部刺激に応答して生体細胞によって生成される細胞シグナルをモニターすることを含んでいる、
用途。 - 請求項1に記載のデバイスの用途であって、
前記細胞に所望の効果を与える試験化合物をスクリーニングするために用いられる、用途。 - 請求項2〜7のうちのいずれか1項に記載の用途であって、
低量の放射線および/または高量の放射線の測定のために用いられる、用途。 - 請求項2〜8のうちのいずれか1項に記載の用途であって、
マイクログレイ(μGy)の領域で、放射線を測定する、用途。 - 請求1〜9のうちのいずれか1項において、
前記HEMTは、AlGaN/GaN−ヘテロ構造である、デバイスまたは用途。 - 外部刺激に応答して生体細胞によって生成される、電気信号のような細胞シグナルをモニターする方法であって、
(a) 上記で定義されたデバイスを用いる工程と、
(b) 外部刺激に応答して生体細胞によって生成される、電気信号のような細胞シグナルをモニターする工程とを備える、方法。 - 外部刺激に応答して生体細胞によって生成される、電気信号のような細胞シグナルをモニターする方法であって、
(a) HEMTを含む、少なくとも一つの分離領域を設ける工程と、
(b) 前記分離領域上に非興奮性細胞を成長/付着させる工程と、
(c) 外部刺激に応答して生体細胞によって生成される、電気信号のような細胞シグナルをモニターする工程とを備える、方法。 - HEMTを含む少なくとも一つの分離領域の用途であって、
任意のリアルタイムで、外部刺激に応答して生体細胞によって生成される、電気信号のような細胞シグナルをモニターするデバイスを製造するために用いられる、用途。
