大腸菌Top10F’(Invitrogen社)をプラスミドの増幅に使用する一方で、大腸菌BL21-Gold(DE3)(Invitrogen社)をタンパク質の発現に使用した。T7プロモーターをいずれも含有する、ベクターpET-23a(+)及びpET-15b(Novagen社)を、それぞれ、大腸菌細胞で、組換RoSBD及び組換AnSBDの発現のために使用した。pET23a(+)-RoSBDに関して、それぞれ、NdeI及びXhoI制限酵素認識部位を含む、フォワードプライマー 5’-CATATGGCAAGTATTCCTAGCAGT-3’ (SEQ ID NO. 1) 及びリバースプライマー 5’-CTCGAGTCATGTAGATACTTGGT-3’ (SEQ ID NO. 2)、並びに、pET15b-AnSBDに関して、それぞれ、NdeI及びXhoI制限酵素認識部位を含む、フォワードプライマー 5’-CATATGAGCAAGACCAGCACCAGT-3’ (SEQ ID NO. 3)及びリバースプライマー 5’-CTCGAGCTACCGCCAGGTGT-3’ (SEQ ID NO. 4)を、クローニング及び配列解析に使用した。
熱ショック方法を用いて大腸菌発現細胞をpET発現ベクターで形質転換した(Chou, W.I., Pai, T.W., Liu, S.H., Hsiung, B.K. & Chang, M.D. The family 21 carbohydrate-binding module of glucoamylase from Rhizopus oryzae consists of two sites playing distinct roles in ligand binding. Biochem J 396, 469-477 (2006))。100μlのコンピテント細胞及び100 ngのDNAまたは20μlのライゲーション混合物を混合し、次いで、氷上で30分間インキュベーションした。細胞を42℃にあるウォーターバス中で30から60秒間加熱し、次いで、氷上に2分間以上設置した。形質転換体の数を決定するために、形質転換体を、100μg/mLのアンピシリンを含んでいるLuria-Bertani(LB)寒天プレート上に播種し、37℃で一晩インキュベーションした。
組換タンパク質を含む大腸菌の細胞ペレットを100mlの結合バッファー(50 mM NaOAc、pH 5.5)中で再懸濁し、均質化により溶解させ(EmulsiFlex-C5 ホモジナイザー、AVESTIN)、細胞残屑を16,000 x gで30分間、4℃で遠心分離により除去した。得られた細胞溶解物を5 mLのアミロースレジンカラム(New England Biolabs)上にローディングした。次いで、カラムを50 mLの結合バッファー(50 mM NaOAc、pH 5.5)で洗浄し、吸着したタンパク質を溶出バッファー(10 mM グリシン-NaOH、pH 11)で溶出させた。精製SBDを限外ろ過装置(Centricon-10、Millipore)で遠心分離により濃縮し、バッファーを交換して50 mM NaOAc, pH5.5とした(Lin, S.C. et al. CBM21 starch-binding domain: a new purification tag for recombinant protein engineering. Protein Expr Purif 65, 261-266 (2009))。
組換タンパク質の分子量決定を液体クロマトグラフィー/質量分析計(LC/MS)(Waters)により実施した。完全なSBD (100 pmol)を50% (v/v)アセトニトリル中0.1 %のギ酸で酸性化し、通常のスキャン解像度の下、800-1800 m/zの範囲に対してデータを得た。複数の荷電イオンシリーズを有する元のエレクトロスプレーの質量スペクトルをデコンボリューションして質量スペクトルを得た。
精製したSBD (100 μlの100 nMの溶液)をSBDコーティングバッファー(50 mM Tris-HClバッファー、pH 8)中にまず希釈し、次いで、96ウェルELISAプレート(Greiner-Bio One)中でピペッティングした後、4℃で16時間インキュベーションした。同様に、100μlの100 nMの2G12 mAb (Polymum Scientific)を2G12コーティングバッファー(50 mM グリシン-HClバッファー、pH 3)中にまず希釈し、次いで、第2のELISAプレートでピペッティングして、4℃で16時間とする。以下の4つの試薬(100μl):(i) HIV-PC (HIV抗原及び抗体陽性血清、ID# 9144532;SeraCare Life Sicences)、(ii)1.0 μg/mLの抗RoSBD mAb (本筆者により製造され、要求があれば入手可能である)、(iii)HCV-PC (抗HCV Mixed Titer Performance Panel;膜10、ID# PHV205;SeraCare Life Sciences)及び(iv)HTLV-PC (抗HTLV I/II Mixed Titer Performance Panel;膜10、ID#PRP206;SeraCare Life Sciences)のそれぞれ一つを別々にSBDでコーティングした、または2G12でコーティングしたELISAプレートに添加し、37℃で1時間反応させた。HIV p24 ELISAキット(Perkin Elmer)及び抗HIV 1+2 ELISAキット(Abbott Diagnostics)を製造業者の指示に従ってそれぞれ用いて、HIV-PCがHIV抗原陽性であり、抗HIV抗体陽性であることを確認した。これに続いて、プレートをPBST (0.01 M リン酸緩衝食塩水+0.05% tween-20、pH 7.1)で三回洗浄した。
データの統計解析を双方向ANOVA、次いで不対の両側t検定(unpaired two-tailed t test)(GraphPad Prism Version 4; Graphpad Software)により実施した。P<0.05を有意とみなした。
直接サンドウィッチ酵素結合免疫吸着法(ELISA)を、HIV特異的なmAbまたはSBDでコーティングされたマイクロタイタープレート及び西洋ワサビペルオキシダーゼ(HRP)-共役組換gp120を用いて構築した;カットオフ指数(COI)>1を示すサンプルをポジティブとみなした。図1eは、HIV抗原及び抗体ポジティブコントロール(HIV-PC)中に存在するHIV抗原が、予期されたように、抗p24 mAb (N29)、抗gp41 mAb (AG10H9)、抗gp120 mAb (ED8.D4)、及び2G12により明確に検出されたことを示す。RoSBD及びAnSBDが同様なHIV検出能力を有することを強く示唆する、いずれかのSBDでコーティングされたプレートでシグナルが観察された。
RoSBD及びAnSBDとHIV-1糖タンパク質との特異性
以下のペプチド及びmAb:(i)ヒトIgG12次抗体([2C11];ヒトIgG1にサブクラス特異的であり、アロタイプ制限がない;Fc領域特異的)、(ii)HIV-1 p24ペプチド([N29]、HIV-1 p24のN末残基1-104に相当する合成ペプチド)、(iii)HIV-1 gp120ペプチド([ED8.D4];ヒトHIV-1 BH8分離gp160の残基427-448に相当する合成ペプチド)、(iv)HIV-1 gp41ペプチド([AG10H9],HIV-1 gp160の残基721-744に相当する合成ペプチド)、(v)2G12 mAb (HIV-1 gp120に対する組換ヒトmAb)、(vi)gp140 (HIVクレードA、株92/UG/037)を、競合的ELISA実験に使用した。Polymun Scientificから購入した2G12及びgp140を除いて、すべてをAbcamから入手した。
2つのSBDとHIV-1との間の分子間相互作用の特異性を、mAb、糖タンパク質及びグリカンを含む競合物質の存在下で評価した。図4a及び4bでは、ヒトIgG1(2C11)2次抗体の存在は阻害効果を何ら示さなかったので、これは、非特異的な競合が生じなかったことを示している。しかし、RoSBD及びAnSBDへのHIV-PCの結合に関するCOI値は、組換HIV-1 gp140、gp120として同じ高マンノース型グリカンを含むチャイニーズハムスター卵巣由来細胞(CHO)-発現エンベロープ糖タンパク質の存在下で、それぞれ、有意に、72%及び60%に減少した。これは、SBDとHIV糖タンパク質との間の分子間相互作用を示唆する。さらに、mAb 2G12、抗gp120 mAb、抗gp41 mAb及び抗p24 mAb競合物質の阻害率は、RoSBD-HIV PC結合に関してそれぞれ55%、42%、22%及び13%であった一方で、AnSBDのそれは、それぞれ33%、18%、6%及び10%であった。(gp120に対する)mAbと競合させた場合にSBD及びHIV-PC結合が明らかに減少することは、SBDがHIVを認識するという結論をさらにサポートするものである。データは、それぞれのSBDとHIV gp120との間の特別な相互関係を強く示唆しており、恐らくはgp120上のグリカン部分が関与する。
SBD及び2G12のHIV検出の比較
HIV ELISAにおいて使用するためのSBDの任意の実際的な用途を評価するために、HIV-1ポジティブであることが特徴付けされている、7つの一致したケース(incident)及び8つの一致した普及されているサンプルを含む、1セットの、HIV-1 発生/有病パフォーマンスパネル(PRB601; SeraCare Life Sicences)を用いて、抗p24、抗p41、抗gp120、及び2G12 mAb並びにRoSBD及びAnSBDのHIV検出能力を比較した。
図6は、全てのコーティング材料が反応性を示し、2つのSBDが明らかな検出シグナルを示したことを明らかにした。SBDまたはHIV特異的なmAbでコーティングされたアッセイのいずれかを用いて、15のサンプルに関して類似の検出プロファイルが観察されたにも関わらず、2G12及びRoSBDは比較的高い検出感受性を示した;明らかに、サンプル番号5、6、8、10、11及び13は、その他のものより多くのHIV-1抗原を含んだ。これらのデータは、本発明者らのSBDのHIV-1抗原認識能力を強く支持した。
RoSBDの特異的なマンナン結合活性
RoSBDのグリカンアレイスクリーニング
グリカンのマイクロアレイ解析を、糖鎖科学コンソーシアムのコアH機関(Consortium for Functional Glycomics)(CFG), Core H facility)で行った(Raman, R. et al. Advancing glycomics: implementation strategies at the consortium for functional glycomics. Glycobiology 16, 82R-90R (2006))。Mammalian Printed Assayは合計で377の異なる天然及び合成グリカンを含んだ。手短には、70μlのRoSBD (200μg/mL)をアレイのプリント表面(printed surface)に適用し、カバースリップをして、37℃で1時間、暗く湿気のあるチャンバー中でインキュベーションした。インキュベーションの後、カバースリップを取り除き、TSMバッファー(50 mM Tris-HCL, pH 7.5、 10 mM MgCl2、及び0.5 Mのスクロース)で4回すすいだ。70μlの抗RoSBD mAbをマイクロアレイのプリント表面に適用し、37℃で1時間、湿気のあるチャンバーでインキュベーションした。結合を検出するために、マイクロアレイを、PBSバッファー中5μg/mLの濃度にある、2次抗体である、Alexa Fluor 488共役ヤギ抗マウスIgG (Invitrogen)で、湿気のあるチャンバー中で37℃で1時間インキュベーションした後、洗浄工程を行った。結合の程度を、Perkin-Elmer Microarray XL4000スキャナーを用いて決定し、Imagene (V.6)イメージ解析ソフトウェアを用いて解析した。完全なグリカンアレイのデータセットは、cfg_rRequest_1340下にあるCFGデータアーカイブ中に見出してよい。
磁気減少(MR)測定(MagQu)のために、親水性の界面活性剤(例えばデキストラン)でコーティングされた63.2 nmの均質に分散された磁気ナノ粒子(MagQu)を含む溶液を使用した。複数外部交流磁界の下、磁気ナノ粒子は磁気相互作用を介して振動し、粒子表面と相互作用する分子の存在に感受性のある磁気特性としての、混合周波数磁化率(χac)を示す。χac値は、従って、磁気ナノ粒子とテストサンプル中の結合分子との間の会合に反応して減少し、MRシグナルを発する。
図8aは、RoSBDにより検出された2つの主要なシグナルをはっきりと示した:グリカン54 Fucα(1,2)Galβ(1,3)GalNAcβ(1,3)Galα、及びグリカン187 Manα(1,2)Manα(1,2)Manα(1,3)Manα(Man4)。前者はヒト血液型O抗原のグリコシル化(Korchagina, E.Y. et al. Design of the blood group AB glycotope. Glycoconj J 22, 127-133 (2005))と関係し、後者はHIV gp120におけるM9の合成多糖成分(Wang, J., Li, H., Zou, G. & Wang, L.X. Novel template-assembled oligosaccharide clusters as epitope mimics for HIV-neutralizing antibody 2G12. Design, synthesis, and antibody binding study. Org Biomol Chem 5, 1529-1540 (2007))並びにSBAである。宿主上のCD4レセプターを認識するMan4がHIV-1進入の重要な成分として作用するので、これらのデータは、RoSBDとHIV gp120との間の相互作用に関する追加の、そして独立した一連の証拠を提供した。興味深いことに、グリカンアレイ解析及び他の方法を用いて、2G12がHIV-1 gp120のこのMan4構造に結合することも示されている(Ji, X., Gewurz, H. & Spear, G.T. Mannose binding lectin (MBL) and HIV. Mol Immunol 42, 145-152 (2005)及びLiu, W.T. et al. Identification and characterization of a novel fibril forming peptide in fungal starch binding domain. Biochem Biophys Res Commun 377, 966-970 (2008))。
SBDとMan4結合タンパク質との間の構造的類似性及びRoSBD上のMan4結合部位のマッピング
推定されるMan4結合部位の分子モデリング
RoSBDの推定されるMan4結合部位のモデリングを、Daliサーバーを用いて、RoSBD複合体(PDB ID: 2V8M)及び2G12-Man4複合体(PDB ID: 1ZLS)の3次元構造を重ね合わせることにより取り組んだ。5Åの半径内でのRoSBD上の推定される結合残基を発見するために単糖Man4を用いた。さらに、2G12-Man4複合体と適合する(coordinate from)Man4を用いて、結合部位I及びIIと最も保存されたヘキソース環が相互作用できるように、RoSBD-G7構造のG7結合部位中にMan4をフィットさせた。予測されるRoSBD-Man4複合体におけるMan4の5Åの半径内の残基を標識した。
RoSBD及びその変異体を、テンプレートとしてpET23a-ROSBDを用いて、PCRに基づくQuikChange(登録商標)部位特異的突然変異法(Stratagene)を使用して、生成した。SBD誘導体を増幅させるのに使用するプライマーペアーを設計して表1にリスト化した。10 ngのテンプレート、0.625μlの各プライマー(10μM)、2.5μLの反応バッファー(10x)、3.125μL dNTP (2.5 mM)、0.25 μL Pfu Turbo DNAポリメラーゼ(20 U/μL)(Stratagene)を含むPCR混合物をddH2Oを用いて最終体積25μLにした。この研究で使用した熱サイクル条件は、95℃で5分間を1サイクル及び95℃で1分間を20サイクル(変性)、45℃から65℃で30秒間(アニーリング)及び68℃で8分間(伸長)、及び最後に68℃で10分間を1サイクルであった。PCR反応をサーマルサイクラー(9700, Applied Biosystems)で実施した。各変異体のプラスミドの配列をDNA配列決定解析により確認した。
現在、10のMan4結合タンパク質がCFG Core Hデータベースで入手可能であり、そのうちの2つは、主にβが豊富な構造を有するカルシウム依存性レクチン、Chromobacterium violaceum由来のCvレクチン(CV-IIL)及びBurkholderia cenocepacia由来のBclAレクチンであり、AnSBDとして、同一の相同のCATHスーパーファミリー(2.60.120.400)に属している。これらのレクチンは、メチルα-D-フコシド(Emau, P. et al. Griffithsin, a potent HIV entry inhibitor, is an excellent candidate for anti-HIV microbicide. J Med Primatol 36, 244-253 (2007))及びメチルα-D-マンノシドに対して最も高い親和性を有する病原菌において発見された毒性因子である。これらの可能性のある役割は、宿主認識、接着及びバイオフィルム形成に関与している可能性がある(Fromme, R. et al. A monovalent mutant of cyanovirin-N provides insight into the role of multiple interactions with gp120 for antiviral activity. Biochemistry 46, 9199-9207 (2007))。これらの高い構造類似性は、2つの真菌性SBDとマンナン結合タンパク質との間の機能的相関関係に対する強力な証拠を提供する。
RoSBDによるHIV-1感染の阻害及びそれらの細胞毒性
細胞、ウイルス株及び抗ウイルスアッセイ
以下の細胞及びウイルス株をAIDS Research and Reference Reagent Program、Division of AIDS、National Institute of Allergy and Infectious Diseaseから入手した:H9細胞及びHIV-1RTMF (AZT耐性ウイルス)のウイルス株。H9細胞を10%のウシ胎児血清、100ユニット/mlのペニシリン及び100 mg/mLのストレプトマイシンが配合されたRPMI 1640培地で育成しかつ維持した。H9細胞を37℃で2時間、HIV-1ウイルスとともにインキュベーションした。H9細胞を1 mLあたり1 x 105細胞で培地中で懸濁し、0.1の感染の多重度でHIVに感染させた。感染後、細胞をPBSで2回洗浄し、次いで培地で洗浄した。細胞懸濁液(100μL)を96ウェルプレートの各ウェルに添加し、次いで、増加させた濃度のSBD (0, 1.25, 2.5, 5, 10, 20, 40 & 80 μM)を感染細胞に添加した。4日のインキュベーション後、細胞を適切な濃度のSBDを含む新鮮な培地で継代して、さらに4日インキュベーションした。H9細胞におけるHIV-1RTMFウイルスのウイルス複製に対するSBDの活性を試験した。抗ウイルス活性をHIV-1 p24抗原ELISAを使用して決定した(di Marzo Veronese, F. et al. Monoclonal antibodies specific for p24, the major core protein of human T-cell leukemia virus type III. Proc Natl Acad Sci U S A 82, 5199-5202 (1985))。
H9細胞に対するSBDの細胞毒性をMTTアッセイを用いて解析した。手短には、指数関数的に増殖しているH9細胞を3x104細胞/ウェルの密度で96ウェルプレート上に播種し、薬剤への暴露に先立ち、37℃で24時間インキュベーションした。処置の日に、一連のSBD濃度(0, 1.25, 2.5, 5, 10及び20 μM)を用いて、H9細胞におけるそれらの細胞毒性を規定通りに試験した(Abd-Elazem, I.S., Chen, H.S., Bates, R.B. & Huang, R.C. Isolation of two highly potent and non-toxic inhibitors of human immunodeficiency virus type 1 (HIV-1) integrase from Salvia miltiorrhiza. Antiviral Res 55, 91-106 (2002))。インキュベーションの8日後に、50μlのMTTを添加し(最終濃度1 mg/mL)、プレートを37℃で4時間インキュベーションして、代謝的に活性な細胞と反応することによりMTTからホルマザンの結晶が形成されるようにした。ホルマザンの結晶をDMSOで溶解させた。各ウェルの吸光度を光学密度540 nmにおいてマイクロタイターリーダーで測定した。反応は特異的であった;死んだ細胞では有意な量のホルマザンは検出することができなかった。
異なるRoSBD濃度の不在及び存在下におけるウイルス複製を、HIV-1 p24抗原ELISAを用いて、8日後に測定した。H9細胞におけるHIV-1感染に対するRoSBDの抗ウイルス活性をmaterials and methodsに記載されるようにして決定した。RoSBDは、用量反応曲線(図11
から計算されるように、4.5 μMのIC50値でAZT耐性ウイルスを阻害した。1.25から80 μMの範囲にある一連のRoSBD濃度を、H9細胞におけるRoSBDの細胞毒性を試験するために使用した。培地におけるH9細胞の生存を決定するためにMTTアッセイを用い、SBDがより高濃度(40及び80 μM)であってこの場合にはH9細胞が死滅した場合を除けば、全培養期間の間、細胞が生存したことが証明された。20 μMの濃度に関しては、細胞の生存は約79%であったが、10 μmでは88%であった。

- HIV糖タンパク質上のエピトープに特異的に結合する炭水化物結合モジュール(CBM)の抗体模倣体を有効量で対象に投与することを含む、前記対象の予防的または治療的なHIV感染の阻害方法。
- CBMがHIV糖タンパク質のエピトープに存在するグリカン構造に特異的に結合する、請求項1に記載の方法。
- HIV糖タンパク質がHIV1-gp120である、請求項1に記載の方法。
- グリカン構造がN結合型高マンノースグリカンMan9GlcNAc2である、請求項2に記載の方法。
- CBMがデンプン結合ドメイン(SBD)である、請求項1に記載の方法。
- SBDがCBMファミリー20、21、25、26、34、41、45、48及び53からなる群から選択されるメンバーである、請求項5に記載の方法。
- SBDがCBMファミリー20である、請求項6に記載の方法。
