Abstract
Description
本発明は、誤って折り畳まれたプリオンタンパク質に特異的な抗体およびエピトープに関する。より具体的には、本発明は、誤って折り畳まれたプリオンタンパク質のYMLエピトープに特異的な抗体およびエピトープを提供する。
プリオン疾患(例えば、クロイツフェルトヤコブ病、牛海綿状脳症、ヒツジスクレイピーならびにシカおよびエルク(elk)の慢性消耗病)は、正常な細胞プリオンタンパク質(PrPC)から異常なプロテアーゼ耐性アイソフォーム(PrPSc)への鋳型誘導変換(template−directed conversion)を一般に特徴とする。一部のプリオン疾患は遺伝することがあり、PNRP遺伝子の突然変異を含むことがあるが、他は散発性または感染性である。遺伝性の形の様々な突然変異が同定されており、突然変異は、PrPCを、異常で疾患関連のPrPScの形への変化をより受けやすくすることがある。
本発明は、一部、誤って折り畳まれたプリオンタンパク質に特異的な抗体およびエピトープ、例えば、誤って折り畳まれたプリオンタンパク質のYMLエピトープに特異的な抗体およびエピトープを提供する。
以下の記載では、いくつかの用語が広く用いられ、以下の定義は、本発明の様々な態様の理解を容易にするために提供される。用語の例を含む、本明細書での例の使用は、例示目的だけのためであり、本明細書の本発明の実施形態の範囲および意味を限定するものでない。数値範囲は、その範囲を定義する数を含む。本明細書では、単語「含んでいる」は限定なしの用語として用いられ、句「含むが、これに限定されない」に実質的に同等であり、単語「含む」は対応する意味を有する。
別の実施形態では、本発明は、プリオンの誤った折畳みに関連する疾患または障害の処置のための化合物を同定する方法を提供する。PrPScアイソフォームまたは誤って折り畳まれたPrPの他の形によって提示されるYMLエピトープに特異的な抗体は、試験化合物の存在下および非存在下で、標的抗原、例えば本明細書に記載のPrPScを含む試料と組み合わされる。結合した抗体およびPrPScの複合体は収集され、複合体中のPrPScの相対量について本明細書で記載のように分析される。試験化合物の非存在下での結合レベルよりも低い、試験化合物の存在下でのPrPSc特異的抗体の結合レベルは、試験化合物が、プリオンの誤った折畳みに関連する疾患または障害の処置または改善のための可能性のある治療化合物であるかもしれないことを示す。一部の実施形態では、抗体は、配列番号7を含むエピトープに結合する。
包装材料、および哺乳動物のPrPScなどの誤って折り畳まれたPrPに特異的な抗体または抗血清を含む組成物を含む製造物も提供される。組成物は、生理的または薬学的に許容される賦形剤を含み、包装材料は、組成物の有効成分(例えば抗血清または抗体)を示す表示を含むことができる。表示は、例えば本明細書に示すキットで用いられる診断試薬としての、組成物の使用目的をさらに含むことができる。
本明細書で提供される1つまたはそれより多くのペプチドを含む組成物を含むキットは、YML特異的抗体または抗血清の同定のための抗体の産生またはスクリーニングのための、化合物または組成物の使用説明書と共に提供される。キットはYML特異的抗体または抗血清の産生および/または同定のために役立つことができ、説明書は、例えば、投与濃度、投与間隔、好ましい投与方法、免疫学的スクリーニングまたは試験の方法などを含むことができる。
方法
一般参考文献:263Kハムスター順応プリオンは、Kimberlinら、1978年によって記載されている。RMLマウス順応プリオンは、Chandler, R. L.(1961年)(Lancet1巻、1378〜1379頁)によって記載されている。これらは、当技術分野で公知の方法、例えばBueler Hら、1993年(Cell73巻:1339〜1347頁)、Oldstoneら、2002年、またはMeade−Whiteら、2009年のものを用いて、マウスまたはハムスターを感染させるために用いることができる。Boltonら、1987年は、スクレイピー因子の単離および精製のために用いることができる方法を記載している。Carlsonら、1986年(Cell、46巻:503〜511頁)は、マウスおよびハムスターでのスクレイピーの臨床診断のために用いることができる方法を記載している。PrPを過剰発現するか、部分的に発現するか、または発現しない様々な遺伝子導入マウスが、Fischerら、1996年(EMBO J15巻:1255〜1264頁)およびWeissmannら、2003年(British Medical Bulletin66巻:43〜60頁)によって記載されている。
脳組織(正常およびスクレイピー感染マウスまたはハムスター)を、Fischerら2000年(Nature408巻:479〜483頁)によって改変された通りに処理し、分析した。簡潔には、PBS、0.5%デオキシコール酸(Sigma)、0.5%NP−40中で10%ホモジネートを作製した。ホモジネート中の総タンパク濃度をBCAアッセイ(Pierce)で決定し、ホモジナイゼーション緩衝液で5mg/mlに調節した。PK耐性物質の検出のために、ホモジネート1.5μlを0.15μgのPK(Sigma)と一緒に、またはそれなしで、37℃で60分間インキュベートした。20mMのPMSF(Sigma)の添加によって消化を停止した。
Paramithiotis、2003年によって改変された通りに、抗体を磁気ビーズにコンジュゲートさせ、免疫沈降実験に用いた。簡潔には、7×108個の磁気ビーズ(1mlのPBS中)(Dynal;Lake Success、New York)を、製造業者の説明書に従って1A1、8B4、4E4またはIgMアイソタイプ対照に結合した。製造業者の推奨に従ってコンジュゲートしたビーズを洗浄およびブロックし、1mlのPBSに次に再懸濁させた。
記載されている通りに(Paramithiotisら、2003年)、イムノブロッティングを実施した。タンパク質を、PVDF膜(Invitrogen)に移した。5%(w/v)乾燥脱脂乳で膜をブロックした。TBST(25mMトリス−HCl、0.2M NaCl、0.5%Tween20)中ですべてのインキュベーションを実行した。化学発光、増強ECL(Amersham)またはスーパーWest Dura(Pierce;Rockford、Illinois)によってペルオキシダーゼ活性を検出した。
抗体作出
ヒトプリオンタンパク質のアミノ酸126〜132に対応するKLH結合ペプチドGGYMLGS(配列番号8)でBalb/cマウスを免疫することによって、抗体を作出した。このペプチドは最初のβ鎖に位置しており、誤って折り畳まれたPrPScでは折り畳まれておらずにアクセスしやすいが、天然のPrPCではそうでないと予想されている。モノクローナル抗体は、標準のハイブリドーマ法によって作出した。BSAに結合した免疫原ペプチドへの結合(ELISAによる)に基づいて抗体を選択した。
PrPScの免疫沈降
免疫沈降させた試料は、6D11−ビオチンを一次抗体(1:5000)として、およびStrep−HRPを二次抗体(1:5000)として、ウェスタンブロット法(図1)によって分析した。8B4ビーズは陽性対照として働き、PrPノックアウトマウス(K/O)を除くすべての脳ホモジネート試料からPrPを免疫沈降させることができた。ビーズのみ、IgMアイソタイプビーズおよび4E4ビーズは陰性対照として働き、4E4によって免疫沈降したRMLおよび263Kのレーンの2つの非常にかすかなバンドを除き、PrPは予想通り免疫沈降しなかった。1A1、PrPのβ−1鎖に対して生成されたIgM抗体は、スクレイピータンパク質をRML(マウススクレイピー株)および263K(ハムスタースクレイピー株)の両方から免疫沈降させることができた。おそらく過剰発現PrPマウス脳における誤って折り畳まれたPrPの小さな発現のために、Tg20レーンにかすかなバンドがある。我々のデータは、マウスおよびハムスターの両方の脳で、1A1がスクレイピーPrPだけを認識することができ、野生型PrPはできなかったことを示した。
YML腫瘍標的
1A1抗体を、正常細胞および腫瘍細胞の両方に結合するその能力について試験した。対照として、各細胞型でのPrPの発現レベルを特定するために、抗PrPC抗体6D11を用いた。細胞に結合する抗体の能力は、正常細胞型としてHUVEC(ヒト臍静脈内皮細胞)を用いて、フローサイトメトリーによって観察した。8つの型の腫瘍細胞への結合についてのデータを示す。試験した癌細胞の5つは、マウス(B16−黒色腫、NSC34−モーターニューロン/神経芽細胞腫ハイブリッド)およびヒト(HL60−前骨髄球白血病、MO3.13−オリゴデンドロサイト/筋肉ハイブリッド、SiHa−子宮頸癌腫)に由来する不死化細胞系である。試験した残りの癌細胞は、British Columbia Cancer AgencyのLiving Tumor Laboratory(LTL)によって増殖された一次腫瘍細胞である。一次ヒト腫瘍は、免疫不全マウスの腎被膜下で増殖される。これは、当初の腫瘍構造および表現型が、当初収集された腫瘍との一貫性を維持することを可能にする。1A1抗体への結合について試験された3つの腫瘍は、LTL−013(びまん性大細胞型B細胞リンパ腫)、LTL−257(結腸直腸肉腫)およびLTL−323(黒色腫)である。
腫瘍モデルでの効力
雌C57B1/6マウスにおけるマウスの黒色腫(B16)の成長を変更するその能力について、1A1抗体を試験した。試験の0日目に、3×105個の腫瘍細胞をマウス12匹のわき腹に皮下移植した。マウスを2つの処置群に無作為に割り付けた。第1群は、PBSで処置した。第2群は、10mg/kgの1A1抗体で処置した。マウスは、−1、2および5日目に処置した。
