Abstract
【解決手段】 患者のインシュリン生成を促進すべく1型糖尿病患者の眼に効果が得られる量のインシュリン生成細胞を移植する。

Description
(政府支持の宣言)
ここで開示される発明は一部国立衛生研究所認可番号DK−58508及びDK−075487号の記載に基づくものであるため、米国政府は本発明に所定の権限を有する。
しかしながら、体内の実験により得られるものはより生理学的な設定における全器官、即ち生命組織の細胞の能力を説明するために十分なものではない。体内システムから生理学的状況を観察するために、体内条件下において研究をする必要がある。近年撮像技術を使用して原位置の細胞機能を調査するためのアプローチが増加している。
(例1)
インシュリン解放の減少による体内のグルコース恒常性の異常は糖尿病の顕著な特徴である(1)。生理的条件下において、膵臓β細胞からのインシュリン解放は、細胞の代謝活性、自己分泌又はパラクリンシグナル伝達、並びにホルモン及び神経伝達物質からの連続した入力の複雑な協調作用によって調整される。β細胞はその他の内分泌細胞タイプの細胞と共に膵臓内分泌部、即ち血管が密に新生されたランゲルハンス島内に位置され(3)、多量に神経を支配される(4)。従って、健康状態及び罹患した状態におけるインシュリン解放を制御するβ細胞シグナル伝達及び機構の複雑さを十分に理解するために、体内の島の血管新生及び神経支配の研究が必要である。ここで、ハツカネズミの目の前房内に移植される膵島の体内蛍光撮像のための新規な非侵襲的技術基盤を開示する。眼の前房、即ち虹彩に移植される島は血管が新生され、細胞の構造体を保持し、刺激に反応するようになる。非侵襲的なレーザ走査顕微鏡法(LSM)により、島の血管新生の他、β細胞の機能及び細胞死の体内撮像が可能となる。従って、通常及び糖尿病の両条件下において長期にわたって実施される、β細胞のシグナル伝達の繰り返しの非侵襲的な体内調査の基礎が得られる。(方法及び材料)
ハツカネズミモデル。C57BL6、及びTie2−GFPのハツカネズミ(STOCK Tg(TIE2GFP)287Sato/J)がジャクソンラボラトリーズ(メイン州Bar Harborに所在)から購入した。RIP−GFPハツカネズミはKarolinska Institutetにおけるコア施設にて生産され、通常の耐糖能及びGFPのβ細胞限定発現(方法を参照のこと)を特徴とする。全ての実験はKarolinska Institutet及びマイアミ大学におけるローカル動物倫理委員会によって承認された。
(膵島の眼の前房への移植。)
膵島は上述したように分離され、培養される(25)。30乃至300の島が培地から無菌PBSに移され、1ミリリットルのHamilton注射器(ネバダ州Renoに所在するHamilton社)に連結された27Gの眼のカニューレ内に0.4mmのポリエチレンチューブ(英国ケント州に所在するPortex Limited)を通じて吸引される。ハツカネズミはイソフルレン(スウェーデンSolnaに所在するScandinavia AB社のイソフルレン)を使用して麻酔され、Temgesic(登録商標)(ニュージャージー州Schering−Plough社)が術後の痛みを低減すべく皮下注射される。立体顕微鏡下において、27G針を使用して眼の底部における強膜近傍の角膜を穿刺する。虹彩を損傷し、出血させないように細心の注意を要する。次に眼のカニューレが徐々に挿入され、島は前房内に徐々に注入され、虹彩に位置される。注入後、カニューレが注意深く取り払われ、動物は覚醒するに先立って横たえられる。ハツカネズミは迅速に意識を回復するが、移植された眼からは圧迫感や刺激の兆候は見られない。(眼の前房へ移植される島の生体内撮像。)
上述した移植されたハツカネズミは40%の酸素と2%までのイソフルレン(イソフルレン)との混合体により麻酔され、加熱パッドに載置される。ハツカネズミの頭部は定位の頭部保持部(日本国東京Narishige社のSG−4N)を使用して保持され移植された島を含む眼が上方に配向されるように位置される。眼瞼は注意深く引き戻され、眼はUST−2中実自在継ぎ手(Narishige社)に取り付けられた一対のピンセットを使用して強膜角膜連結部に徐々に保持される。ピンセットの先端は2つの先端の間にループを形成する一片のポリエチレンチューブにより覆われる。上記構造体により、損傷を付与したり眼の血液循環を中断させることなく可撓性を備えつつ頭部及び眼を安定して固定することができる。TCS−SP2−AOBSの2光子励起25の共焦点スキャナ及びレーザを備えたLeica社DMLFSA正立顕微鏡が、濾過された生理的食塩水を浸漬液として使用する長距離浸水レンズ(Leica社、HXC APO、10x0.3W、20x0.5W、40x0.8W)と共に撮像に使用される。
(RIP−GFPハツカネズミの生産)
RIP−GFPハツカネズミはRIP1.EGFP 発現カセット(rat insulin-1 promoter -410/+1bp-EGFPSV40polyA)をB6CBAF1/Crl ドナーからの単細胞段階の胚に注入することにより生産される。得られたFO生産はPCR分析によるRIP1.EGFP ゲノムの組み込みのために切れ目を入れられる。RIP1.EGFP 導入遺伝子は7匹の初代遺伝子導入動物(17.5%)に見られ、これらはF1動物を生産すべくC57 BI/6NCrl の同系交配のハツカネズミと
交配される。初代の系列は1)免疫染色によって決定されるβ細胞中のGFPの表示、及び2)動物及び細胞の生理学に対して表示される。RIP1.EGFP の初代系列ナンバー29は調整された動物、及びGFPのβ細胞抑制発現と比較して通常の耐糖能を有し、ホモ接合体交配のために選択される。
(免疫組織化学。)
ハツカネズミは、眼球内の島移植(n=12)に続いて3日後、7日後、14日後、及び28日後に頸椎脱臼に続く濃縮二酸化炭素暴露により死亡する。移植物を有する眼が取り払われ、4%のパラホルムアルデヒドに1時間後置される。スクロース置き換え(PBS中10%、20%、及び30%)による冷却保護の後、眼の縦の区分が低温保持装置にて切断される(14マイクロメートル)。区分はPBSにて洗浄され(3X10分)、5%のウシ血清アルブミン及び0.1%のトリトンを含むPBSにて保温される(1時間)。その後、区分は抗インスリン(ニューヨーク、Accurate Chemical & Scientific Corp.、1:500)及び抗グルカゴン(ミズーリ州セントルイスに所在するSigma社、1:5000)を含むPBSにて一晩中保温される。免疫染色はAlexa488やAlexa568(Molecular Probes社、1:500)共役二次抗体を使用して視認される。
America Inc.のOlympus Fluoview)を使用して試験される。。全ての免疫染色画像はデジタル的に得られ、再コンパイルされる(フォトショップ5.0、カリフォルニア州サンジョゼに所在するアドビ社)。区分は10倍及び40倍に拡大され視認される。Zeiss Axiovisionソフトウェアを使用してデジタル化された蛍光顕微鏡による画像を分析する。計測されたパラメータ(即ち、インシュリン免疫反応細胞と、グルカゴン免疫反応細胞との比率)が眼毎の少なくとも2つの別体の島からの少なくとも3つの隣接する区分に由来する平均値として計算される。3つの眼からの計算結果の平均値が求められる。明瞭に標識された核(DAPI染色)を有する細胞のみが分析に含まれる。データは平均値の標準誤差SEMのプラス−マイナスとして示される。
(島移植物の血管新生の定量化。)
血管密度は移植物領域当たりの血管密度の数として決定される。血管密度は1本の血管や血管の枝として規定される。β細胞GFP蛍光透視像が移植物領域を形成することに使用される。2つの光学的区分が各島移植物から定量化される。光学的区分はzスタックの一連の画像から選択される。第1の区分は信号の損失なく移植物のもっとも深いレベルにて選択される。第2の区分は移植物の表面及び最も深い区分の間における中間部にて選択される。定量化のための画像は10倍又は20倍のレンズ、及び2.0倍以上のズームの倍率を使用して撮像される。データは平均値の標準誤差SEMのプラス−マイナスとして示される。血管新生の定量化はLeica共焦点ソフトウェア(バージョン2.61)を使用してなされる。([Ca2+]の変化の体内記録。)
体内β細胞の機能を評価するために、眼の前房に移植される島はCa2+標識であるFluo−4及びFura−Redの混合物を装填される。上記2つの染料を同時に作用させることにより、視認可能な波長(29)における励起スペクトルによる[Ca2+]iのラシオメトリックな計測が可能となる。装填すべく、500マイクロメートルの各AMエステルのFluo−4及びFura−Redを含む、(単位:ミリモル)140NaCl、5KCl、2NaHCO3、1NaH2PO4、1.2MgCl2、2.5CaCl2、10HEPES、3グルコース(pH7.4のKOH)からなる細胞外溶液が前房を潅流する。潅流のために、ハツカネズミはヒプノルム(英国Leedsに所在するVetaPharma社のフェンタニル0.315ミリグラム/ミリリットル、及びフルアニソン10ミリグラム/ミリリットル)を1、ドルミカム(スイス国Baselに所在するRoche社、5mg/ml)を1、滅菌水を2とした10ミリリットル/キログラムの混合物により麻酔される。麻酔期間を延長すべく30乃至40分毎に混合物(4ミリリットル/キログラム)の注射が行われる。
膵島は体内監視においてアクセスが困難である。その理由として、これらは深部に位置され、膵臓外分泌組織に分散され、膵臓容積の1,2%を構成することが挙げられる。従って、今日のβ細胞の機能の研究の殆どは体内の分離した島又は細胞に対して行われる。分離した島(6)、特に膵臓の薄切り(7)により、多数の細胞環境におけるβ細胞の機能の研究が可能となる。しかしながら、上記準備は形成された端点までの制約を受け、部分的に血管及び神経の連結部からの入力を欠く。通常の健康状態下におけるインシュリン解放の調整に関する複雑なシグナル伝達網、及びなぜこれらが2型糖尿病において適切に機能しないかを理解するためにβ細胞の機能の体内監視が迅速に要求されている。これは1型糖尿病の治療として挙げられる臨床における島の移植の状況にも当てはまる(8)。今日まで実験及び臨床の島移植後におけるβ細胞のシグナル伝達の監視は可能ではなかったため、移植物機能の特性及び新規な関与の評価の両者が深刻に妨害されてきた(9)。LSMは分離した島及び準備された細胞を使用したβ細胞の多数のシグナル伝達経路の撮像に好適に使用される。しかしながら、生体内においてLSMをβ細胞の生理学の研究に使用することは記録にない。
高拡大図により、殆どのアネキシンV−APC標識はGFP蛍光を含まない移植領域にて視認されることがわかる。図4I及び4Lに示すように、いくつかのアネキシンV−APC蛍光はGFP蛍光β細胞の表面にて視認され、このことは自然死の細胞を標識することを示す。即ち、眼の前房に移植される島の体内状況下においてβ細胞死は非侵襲的に、且つ長期にわたって撮像可能である。
(まとめ)
体内において生理学及び病態生理学の両者における島の細胞の非侵襲的な研究のための新規な基盤が開示された。島細胞の調査のために眼の前房を体内モデルとして使用することにより、形態、血管新生、神経支配、細胞死、及び細胞シグナル伝達の連続した監視が可能となる。体内における島の細胞のシグナル伝達を研究するためのこの基盤により、ホルモン系及びニューロン系の他、内分泌細胞や血管細胞の自己分泌シグナル又はパラクリンシグナルからの調整入力の効果を明瞭にすることが補助される。更に、上記は健康状態又は糖尿病状態下におけるβ細胞の機能及び生存の非侵襲的な体内研究のための新規なアプローチとして機能する。上記基盤は膵臓β細胞の信号伝達の研究に限定されるものではなく、体内の多数の様々な細胞タイプ及び器官組織の調査に拡張可能であることに注目すべきである。従って、眼の前房は体内条件下における細胞レベルにて複雑なシグナル伝達ネットワークの統合を初めて明瞭に示す多目的自然体窓として使用される。

この例において、単細胞解像度における体内の非侵襲的な長期にわたる細胞生物学研究のための段階的な手順が開示される。ここで、自然体窓として機能する角膜が効果的である。この目的において、部位の組織は眼の前房内に移植され、細胞の生物学的パラメータは角膜を通じてLSMにより測定される。眼の前房は様々な組織の研究のために移植部位として頻繁に使用されてきた3−7。本来眼の前房は免疫特権を付与された部位としての特徴により、移植部位として選択されたが8、殆どの研究において前房は容易にアクセス可能であり、角膜により移植される組織の巨視的観察が可能となるため、同系の移植設定に使用された。付加的に、前房の基部を形成する虹彩は体内において最も高密度な血管及び自律神経を有する器官の1つである。従って、移植物の迅速な神経支配9及び血管新生10が可能である。今日まで前房を移植部位として使用する研究は主に移植物生理学を研究するために巨視的観察5を使用してきた。上記は低解像度にて観察可能なパラメータに対する長期的な研究を制約してきた。移植物を取り払った後に、体内電気生理学7の他、組織学3、及び様々なその他の体内技術により、形態学及び細胞の機能の評価が可能となるが、これは研究の終点を設定し、長期にわたる監視を妨害する。

LSMのために、アルゴンを備えたLeica社TCS−SP2−AOBS共焦点レーザ走査、及びLeica社DMLFSA顕微鏡に連結されたHeNeレーザが使用される。2光子励起はTkSapphireレーザ(米国Spectra−Physics社Tsunami)を使用して82MHzまで、且つ100fsまでの励起が実施される。
(頭部保持アダプタ)
外科手術及び撮像をすべくハツカネズミの頭部を固定するために、日本国Narishige社製造による頭部保持アダプタSG−4N−Sが使用される。使用される麻酔薬の種類に応じて、頭部保持部はガスマスク(GM−4−S)又はノーズピースを備える。頭部保持部は顕微鏡の特注ステージに適合する金属板に固定される。金属板は加熱パッドにより覆われる。体温は加熱パッドの温度を調整する直腸プローブを通じて調整される。
(眼球固定器)
眼瞼を収縮させ、眼を付加的に固定すべく特注の支持装置が使用される。ナンバー5Dumont鉗子を小型の金属バーに固定子、UST−2中実自在継ぎ手に金属バーを挟持する。目の高さと同じ位置に合わせた頭部保持部と同じ位置にて自在継ぎ手を金属板に固定するため、鉗子の先端部を眼に至らしめることができる。ポリウレタンチューブの一部により鉗子の先端部を覆い、先端部間にループを形成する。鉗子の前方部分にネジを固定するため、鉗子の先端部間の距離を調整することができる。
(前房潅流)
流出部:内径0.9mm且つ外径1.2mmからなる35cmまでのポリエチレンチューブの一端を毛細血管保持部に連結し、ピストンを備えない10ミリリットルの注射器(即ち、開放されたタンク)にチューブの他端を連結する。
流入部:内径0.76mm且つ壁厚0.86mmからなる35cmまでのタイゴン(登録商標)チューブの一端を毛細血管保持部に連結し、1ミリリットルの注射器にチューブの他端を連結する。
(撮像工程)
共焦点LSM及びTPLSMによりキャプチャされた画像のノイズを取り払うべくウェーブレットフィルタ15が使用される。分析及び画像表示のために例えばVolocity及びLeica共焦点ソフトウェア等の処理ソフトウェアを使用する。
(非制限的例における処理)
(膵島の眼の前房への移植)
1.上述したようにハツカネズミの膵島を分離する16,17。
2.内径0.4mmを有する約10cmのポリエチレンチューブを備えた27G針を0.5ミリリットルのHamilton注射器に連結し、チューブの他端に鈍い27Gカニューレを挿入する。
3.30乃至40の小島を培地から殺菌PBSを備えた皿に移し、小島を皿の中央部にできるだけ密集させて位置させる。
4.小島を鈍い27Gカニューレ及び連結されたポリエチレンチューブ内に吸引する。小島は前房内に容易に注入できるように、好適に最小限の容積(例えば、20マイクロリットル以下)にて吸引される。
5.選択肢(A)又は(B)に従って眼の前房に膵島を移植する。
(A)頭部保持部を使用した膵島の移植
i.1つの脱脂綿を50ミリリットルの試薬チューブ内に位置させ、約1ミリリットルのイソフルレンを脱脂綿に滴下する。試薬チューブ内にて数秒間ハツカネズミを保持することにより、ハツカネズミを気絶させる。
ii.立体顕微鏡下の頭部保持部にハツカネズミを位置させ、移植のために選択される眼のある頭部を上方に配向させて固定する。
iii.40%の酸素及び60%の窒素中にて2乃至2.5%のイソフルレンを使用してハツカネズミを麻酔する。
麻酔の領域が吸引され、操作者を保護する。
iv.ブプレノルフィン(0.05ミリグラム/キログラム)を皮下注射し、手術後の痛みを低減する。
v.眼瞼を注意深く引き戻し、眼球固定器のポリエチレンチューブのループを強膜角膜連結部下にそっと載置する。
vi.容易に取り扱えるように、27G針を1ミリリットル注射器に連結する。虹彩への損傷及び出血を避けるように留意して、27G針を使用して強膜近傍の角膜を穿刺する。vii.針により形成された孔を通じて眼の前房内に鈍いカニューレをそっと挿入する。小島を前房内に徐々に注入する。
viii.覚醒するに先立って付加的な10乃至15分間ハツカネズミを頭部保持部に残しておく。
ix.乾燥を防止すべく眼にViscotearsを滴下する。
体内撮像まで小休止する。時間は研究パラメータにより決定される。
(B)頭部保持部を使用しない膵島の移植
i.5ミリリートルノプラスチック製注射器からピストンを取り払い、底部から1cmまでの注射器の部分を切断することにより小型のガスマスクを形成する。麻酔ポンプのチューブを針接続部に連結させる。
ii.脱脂綿及び約1ミリリットルのイソフルレンを備えた50ミリリットルの試薬チューブにハツカネズミを数秒間保持することにより、ハツカネズミを気絶させる。
iii.移植するために選択された眼を有するハツカネズミを立体顕微鏡下の加熱パッドに上方に配向されるように載置する。ハツカネズミの鼻を準備したガスマスク内に位置させる。
iv.40%の酸素及び60%の窒素中にて2乃至2.5%のイソフルレンを使用してハツカネズミを麻酔する。
v.ブプレノルフィン(0.05ミリグラム/キログラム)を皮下注射し、手術後の痛みを低減する。
vi.眼の周囲の皮膚を収縮させ、眼の強膜角膜連結部を視認できるように露出させ、ハツカネズミの呼吸や血液循環を妨害することなく頭部の位置をそっと固定する。
vii.上記(A)をviからixまで続ける。
(眼の前房に移植される小島の撮像)
6.工程1乃至5により小島を移植する。研究の目的に応じてドナーとなるハツカネズミ及び受容するハツカネズミを選択する。
7.イソフルレンに短期間暴露することにより受容するハツカネズミを気絶させる。
8.ハツカネズミを頭部保持部に位置させ、移植される小島を含む眼を有する頭部を上方に配向されるように固定する。
9.40%の酸素及び60%の窒素中にて2乃至2.5%のイソフルレンを使用してハツカネズミを麻酔する。
10.眼瞼を注意深く引き戻し、眼球固定器のポリエチレンチューブのループを強膜角膜連結部下にそっと載置する。
11.共焦点及び2光子LSMを具備した正立顕微鏡下にハツカネズミと共に頭部保持部を載置する。
12.概観を得るべく低倍率の対物レンズ(2.5及び5倍)を使用する。高解像度を得るべくLSMは濾過した生理食塩水やViscotearsをレンズ及び角膜の間の浸液として使用する長い作業距離を有する浸水対物レンズ(10、20、及び40倍)を使用する。
13.部位の生物学的パラメータを撮像する。
(A)移植物形態の撮像
i.細胞特有の形態を視認すべく、ベータ細胞に蛍光タンパク質を発現する遺伝子組み換えのハツカネズミの島(例、RIP−GFP)が移植に使用される。GFP蛍光を488ナノメートルのレーザにて励起し、495乃至530ナノメートルの範囲の発光を検知する。島形態も反射像の検知により撮像可能である。レーザ(例、633ナノメートル)を選択し、AOBS制御を設定し、反射像検知を最適化する。プラスマイナス4ナノメートルのレーザ波長の範囲に発光を集束する。
(B)虹彩及び移植される島の血管新生の撮像
内皮細胞又は血管ルーメンをのいずれかを撮像することにより血管新生を視認化する。i.内皮細胞を撮像すべく、Tie2−GPFハツカネズミを移植用に使用する。
ii.GFP蛍光を488ナノメートルのレーザにて励起し、495乃至530ナノメートルの範囲の発光を検知する。
iii.血管ルーメンの撮像のために、蛍光標識された10ミリグラム/ミリリットルのデキストラン(70キロダルトン)(約70キログラム/モル)を0.1ミリリットル尾静脈注射する。
iv.蛍光標識されたデキストランの注射後、選択されたデキストランに好適な設定を使用して移植される島を撮像する。
(C)ベータ細胞死の撮像
i.アロキサン(75ミリグラム/キログラム(体重))の経皮的注入により移植された島においてベータ細胞死を誘発する。
ii.ハツカネズミが高血糖状態になったことを確認すべくアロキサンを投与して24時間後に血液グルコースレベルを測定する。
iii.アネキシンV−APCを0.1ミリリットル尾静脈注射する。
iv.APC蛍光に好適な設定を使用してアネキシンV−APC投与後4乃至6時間移植された島におけるベータ細胞死を撮像する。APCを633ナノメートルにて励起し、発光を645乃至680ナノメートルの範囲にて集束する。
(D)眼の前房の潅流によりCa2+標識を移植物に装填した後の細胞質の遊離Ca2+濃度の撮像
前房潅流は参照文献18から変形される。
i.ピペットを30乃至40マイクロメートルの先端径にて破断する。先端部を35°斜角をなして最終的な径を70乃至90マイクロメートルとする。流出側のピペットは流入側のピペット(70マイクロメートルまで)より僅かに大きい(90マイクロメートルまで)。
ii.注射器、チューブ、及び毛細血管保持具を細胞外溶液にて満たし、ピペットを毛細血管保持具の毛細血管グリップの頭部に載置する。
iii.ヒプノルム/滅菌水/ドルミカムの混合物(1:2:1)を100マイクロリットル/10グラム(体重)にて腹腔内注射によりハツカネズミを麻酔する。麻酔は1、2分に行う。30分後及び60分後にヒプノルム/滅菌水の混合物(1:3)を50マイクロリットル/10グラム(体重)にて注射し麻酔時間を延長する。必要に応じて90分後に最初のヒプノルム/滅菌水/ドルミカムの混合物(1:2:1)を50マイクロリットル/10グラム(体重)にて注射することにより麻酔時間を更に延長する。
v.眼瞼を注意深く引き戻し、眼球固定器のポリエチレンチューブのループを強膜角膜連結部下にそっと載置し、保持部及びハツカネズミを正立顕微鏡下に載置する。
vii.2.5倍の対物レンズを使用し、眼全体を監視すべく流出ピペットを極微操作装置と共に前房内に挿入する。
viii.Fluo−4及びFura−Redの混合物(1:1、各500マイクロメートル)を130マイクロリットルまで流入毛細血管内に吸引し毛細血管保持具を極微操作装置に固定する。染料を含まない洗浄のために十分な細胞外溶液が注射器内に含まれることを確認する。
ix.流入ピペットを流出ピペットに対向する前房内に挿入する。
工程vii.と同様に角膜を穿刺する。
x.最初に房水を潅流液に迅速に(10マイクロリットルまでを30秒以内)交換する。潅流の機能性を監視する。
xi.毎分3マイクロリットルの速度にて40分間、眼の前房を連続して潅流する。
xii.装填後、前房を高い速度にて(毎分10マイクロリットルまで)潅流することにより前房内の染料を洗い流す。
xiii.染料の洗浄後、潅流を停止する。ピペットは取り払わない。
xiv.細胞質の遊離Ca2+の濃度の変化を撮像すべく、より拡大率の高い浸水対物レンズ(10、20、或いは40倍)に交換し、Viscotearsを浸液として使用する。
xv.Fluo−4及びFura−Redを同時に撮像することにより、細胞質の遊離Ca2+の濃度の変化のラシオメトリック計測が可能となる。Fluo−4及びFura−Redは488ナノメートルにて励起され、発光はFluo−4が495乃至535ナノメートル、Fura−Redが600乃至700ナノメートルに集束する。
xvi.部位の細胞内のFluo−4及びFura−Red蛍光を一連の時点においてデータの取得を開始する。
xvii.刺激を受けない蛍光レベルを基準線とし、グリベンクラミド(1ミリグラム/キログラム)を尾静脈注射することによる刺激により全身にわたってインシュリンを解放する。島移植物内のベータ細胞における細胞質の遊離Ca2+の濃度の変化は注入後数秒間のうちに監視する必要がある。
xviii.撮像後にピペットを眼から注意深く取り払う。
xix.術後の痛みを低減すべくハツカネズミにブプレノルフィン(0.05ミリリットル/キログラム)を皮下注射する。
xx.ハツカネズミを覚醒するまで温暖な環境(30℃まで)に載置する。ヒプノルム又はドルミカム麻酔の後、数時間要する。
(時間的調節)
島の分離:4時間まで
前房への島の移植(工程2乃至5):25分まで(ハツカネズミの場合)
移植物形態の撮像(工程13A):1時間まで(ハツカネズミの場合)移植物の血管新生の撮像(工程13B、i及びii、又はiii及びiv):1時間まで(ハツカネズミの場合)
ベータ細胞死の撮像(工程13C、i及びii):24時間まで、(工程13Ciii及びiv):5時間まで
眼の前房を潅流することにより移植物にCa2+標識を装填した後の細胞質の遊離Ca2+濃度の撮像(工程13Di):1.5時間まで(4乃至6のピペットの場合)、(工程13Dii乃至xx):2時間まで(ハツカネズミの場合)
(予期される結果)
ここで開示される基盤により、部位の組織にアクセスすべく侵襲的な外科手術を行うことなく体内における複数の生物学的パラメータの長期にわたる評価が可能となる。眼の前房への膵臓ランゲルハンス島の移植後に、上記手順により反射像や蛍光によって容易に島移植物の形態が検知可能である。撮像反射像による膵島移植物の形態学的特徴付け及びGFPが実施され、ここでインシュリンプロモータ(緑色のベータ細胞)下においてGFPを発現するハツカネズミの島はTie2プロモータ(緑色の内皮細胞)下においてGFPを発現するハツカネズミに移植される。GFPは35%のレーザ出力にて488ナノメートルにて励起され、発光は495乃至530ナノメートルの範囲にて測定される。反射は35%のレーザ出力にて633ナノメートルにて励起されることにより反射像をなし、発光は632乃至639ナノメートルの範囲にて測定される。

- 患者のインシュリン生成を促進すべく1型糖尿病患者の眼に効果が得られる量のインシュリン生成細胞を移植する工程を含むことを特徴とする1型糖尿病の患者を処置する方法。
